Abstract

Objectives: To analyze the differences in the expression of Brain Derived Neurothropic Factor (BDNF) in Rattus norvegicus cerebrum and cerebellum of newborn between those exposed to Mozart's music composition in default sequence, reverse sequence, and without exposure in the womb.Materials and Methods: Analytical laboratory experimental study with randomized post test only control group design using animal models Rattus norvegicus. The animal models were divided into three groups: control group without any exposure, the treatment groups with exposure to Mozart's music in default sequence and another group in reverse sequence since day 10 of gestation. We used a comparison test in the analysis of BDNF expression.Results: We found significant difference in BDNF expression with p value 0.004 (mean 8.98±1.31 default sequence group, 5.58±3.08 reverse sequence group, 6.80±1.95 control) in the cerebrum. We found significant difference of BDNF expression with p value 0.003 (mean 9.48±1.41 default sequence group, 6.02±3.25 reverse sequence group, 7.14±2.54 control) in the cerebellum. In cerebrum dan cerebellum we found significant difference between standard Mozart’s music and control (cere-brum p=0.018, cerebellum p=0.001), and we found significant difference between standard Mozart’s music and reverse Mozart’s music (cerebrum p=0.001, cerebellum p=0.008) and no significant difference in reverse Mozart and control (cerebrum p=0.264, cerebellum p=0.490)Conclusion: Sequence in Mozart’s music is very important in increase expression of BDNF.

Abstract

Objectives: To compare the TLR2 expression in the placenta between treated by sambiloto EA-96 fraction tablet (AS201-01) and dihydroartemisinin-piperaquine phosphate (DHP)Materials and Methods: Experimental study using 24 pregnant mice. All sample divided into 4 groups with randomization are uninfected group, Plasmodium infected group and given placebo, sambiloto (AS201-01) and DHP. Then performed surgery and placental sampling were staining with adopting in tunnel assay method to measure the TLR2 expression of placental.Results: The expression of TLR2 in uninfected group has the lowest rate compared to other groups. The infected and placebo treated group has the highest TLR2 expression campared with sambiloto and DHP. The sambiloto group has not differ signi-ficantly with the group uninfected and lower than DHP.Conclusion: Tablet of sambiloto EA-96 fraction (AS201-01) decreased TLR2 expession better than with DHP tablet.

Abstract

Objectives: To determine the effect of Sambiloto tablet (AS201-01) in reducing the placental Chondroitin Sulfate A (CSA) Expression of pregnant mice infected Plasmodiumberghei.Materials and Methods: Experimental study using 24 pregnant mice were divided into 4 groups with randomization. Uninfected group, the placebo group, the Sambiloto tablet (AS201-01) group and the DHP tablet (as a standart drug) group. The last three groups, were infected with P. bergheion day 9th of pregnancy, and the treatment was started at day 11th of pregnancy, and samples were terminated at day 15th of pregnancy by surgery. Placental sampling were stained with Tunnel assay to measure placental CSA antibodies.Results: The placental Chondroitin Sulfate A (CSA) expression. Uninfected group compared to Sambiloto tablet (AS201-01) groups was not significantly different (p>0.05), uninfected group compared with the other treatment groups differ meaningfully (p<0.05). Placebo group compared with all groups significantly different (p<0.05). Sambiloto tablet (AS201-01) group compared to uninfected group (p>0.05) was not significantly different, with another group was significantly different (p<0.05). DHP tablet group compared to all the groups was significantly different (p<0.05).Conclusion: Placental Chondroitin Sulfate A (CSA)expression of mice infected by Plasmodium berghei treated with Sambiloto tablet (AS201-01) lower than DHP tablet.

Abstract

Objective: To compare the administration of sambiloto tablets (AS201-01) and dihydroartemisinin-piperaquine phosphate tablets in pregnant mice infected by P. berghei on the expression of MIF in the placenta.Materials and Methods: Experimental laboratory research, 24 pregnant mice were divided into 4 groups with randomization, ie. non-infected, placebo, sambiloto (AS201-01) and DHP groups. On day 9 P. berghei was infected, on day 11 the treatment was given, day 15 the surgery was performed, placental samples were taken, immunohistochemical staining was given, and MIF expression was assessed.Results: The expression of MIF in the group not infected with P. berghei had the lowest mean, while the highest mean was found in the placebo group. Uninfected groups were not significantly different from sambiloto (AS201-01) tablet group. Sambiloto tablet group (AS201-01) had lower MIF expression than DHP group, but it was not significantly different. Sambiloto tablet group (AS201-01) showed lower MIF expression than placebo. MIF expression in DHP group was lower than that in placebo group. From the lowest, the MIF expressions were as follows: group not infected with P. berghei, group receiving sambiloto (AS201-01) tablet, DHP group and placebo group.Conclusion: MIF expression in the placenta of pregnant mice infected with P. berghei and receiving sambiloto (AS20-01) tablets was not different from those receiving DHP tablets.

Abstract

Objectives: To determine the effect of bone marrow mesenchy-mal stem cells (BMSCs) on BMP-15 expression and Graafian follicle count in endometriosis mice.Material and Methods: This study was a laboratory randomized clinical trial on Mus musculus. The object of the study was 42 mice which were divided into 3 groups, the control, endome-triosis, and endometriosis + BMSCs groups. Comparison of BMP-15 expression and Graafian follicle count between groups was evaluated.Results: Immunohistochemical analysis showed that BMP-15 expression in control, endometriosis, and endometriosis + BMSCs groups had p=0.551, p=0.446 and p=0.917 with ANOVA test p=0.273, indicating no statistically significant differences between groups . Graafian follicular count in the three groups had p=0.31, p=0.001, and p=0.006, with the Kruskal-Wallis test p=0.001. Graafian follicles in the endometriosis + BMSCs group were higher than those in control and endometriosis groups.Conclusion:In the endometriosis mouse model with bone mar-row stem cell transplantation the BMP-15 expression in each group did not show a difference, but a significant difference was found in the number of Graafian follicles.

Abstract

Objectives: To analyze the influence of 50 percent food  restriction during pregnancy to the dendritic density of cerebellum and cerebellum of newborn Rattus norvegicus.Materials and Methods: Laboratory experimental study with single blind randomized post-test only control group design using animal model; pregnant Rattus norvegicus as treatment models. Subjects were divided into two groups: control group and treatment group which was exposed to 50% food restriction (FR 50%). At day 21, both group sacrificed and the cerebrum and cerebelum of the offsprings were prepared and stained with silver impregnation. We used parametric independent t-test in analyzing dendritic density.Results: In the cerebrum there was a significant difference in dendritic density between control (4.98+2.17) and treatment (2.69+0.76) groups with p=0.001 (p<0.05). In the cerebellum there was ALSO a significant difference in dendritic density between control (7.37+2.23) and treatment groups (3.01+0.64) with p=0.000 (p<0.05).Conclusions: The dendritic density of cerebrum and cerebellum of newborn Rattus norvegicusexposed to 50 percent of food restriction during pregnancy  were lower than control.

Abstract

Objectives: This study aim to determine sequence of Mozart songs in the analyzing differences in dendritic density of cerebellum and cerebellum of new born Rattus novergicusMaterials and Methods: Experimental study randomized post test only control group design using Rattus norvegicus. Animal subjects were divided into three groups which were control group, and the treatment group that were given exposure to default sequence and reversed sequence of Mozart's music from gestation day 10. We used a comparison test in the analysis expression of BDNF.Results: In the cerebrum there was a significant difference in the Mozart group in reverse rather than with Mozart standard sequence and in standard Mozart group with no exposure group, with p=0.003 and p=0.000. In the cerebellum there was a significant difference in the group in reverse rather than with standard Mozart and in the standard Mozart group with the control group, with p=0.000 and p=0.000. However, there was no significant difference between control group and Mozart group upside down in cerebrum and cerebellum with p=0.109 and p=0.077Conclusion: The density of dendrites in the cerebrum and cerebellum of Rattus norvegicus newborn exposed to Mozart's music during pregnancy with standard Mozart sequence was higher than that in those receiving reverse order and without exposure. There were no significant differences between the density of the cerebrum and cerebellum dendrites between groups exposed to reversed sequence of Mozart composition and those without exposure.

Abstract

Objectives: To analyze the difference neuronal and glial (astrocytes, oligodendrocyte, microglia) cell count in cerebrum and cerebellum of Rattus norvegicus newborns with 50% food restriction and control group.Materials and Methods: This was an analytical experimental study with single blind randomized post test only control group design using animals subjects Rattus norvegicus. This study was conducted at Animal laboratory, Veterinary Faculty, Universitas Airlangga. Animal subjects were divided into FR50% group and control. Neuron and glial (astrocytes, oligodendrocytes, microglia) counts were analyzed using comparison test, with CI 95%.Results: There was a significant difference in cerebrum and cerebellum neuron cell count between intervention and control group (9.88+3.59 vs 16.88+2.553; p=0.000 and 7.5+1.789 vs 11.44+4.56; p=0.02). There was no difference in cerebrum and cerebellum glial cell count. There was a significant difference in cerebellum astrocyte between intervention and control group (80.94+24.255 vs 59.69+18.77; p=0.02) but no difference in cerebrum. There was a significant difference in cerebrum and cerebellum oligodendrocyte between intervention and control group (14.06+12.195 vs 25.13+8.609; p<0.000 and 13.63+6.712 vs 24.00+8.862; p=0.001), and there were significant difference in cerebrum and cerebellum microglia cell between intervention and control group (5.25+3.435 vs 4.94+3.838; p=0.620 and 8.81+4.119 vs 5.25+1.483; p=0.004).Conclusion: Food Restriction 50% (FR50%) in Rattus norvegicus decreased cerebrum and cerebellum neuron cell and oligodendrocyte count and increased cerebrum and cerebellum microglial count.

Abstract

Objective: To create an animal model of vesicovaginal fistula as a modality to develop therapy.Material and Methods: This study was an preliminary experimental study to create new fistula vesicovaginal formation in a rabbit model. Six female New Zealand white rabbits of Stem Cell Research and Development Center of Universitas Airlangga were used in this study and were divided into 3 groups in which there were 2 rabbits in each group. Vesicovaginal fistula formation was performed through laparotomy surgery. After the abdomen is opened, the Nasogastric tube is inserted vaginally into the anterior vaginal wall. The incision is made on the anterior vaginal wall and the posterior wall of the bladder. The Nasogastric tube that comes out of the vagina is inserted into the bladder incision to maintain the formation of vesicovaginal fistula. Both mucosal walls are sutured and tested to determine if there is any leak. Evaluation the model of vesicovaginal fistula was carried out 3 weeks postoperatively according the group.Result: Four out of six rabbits have formed vesicovaginal fistulas, which are from groups two and three. There are no complications during the treatment period until terminationConclusion: Rabbits have the potential to become experimental animals for the vesicovaginal fistula model. The best time to release a nasogastric tube to maintain a fistula is 2 weeks.

Abstract

HIGHLIGHTS 1. Endometriosis treatment remains controversial, whether it should be managed with hormonal modulation and/or surgical removal of the lesion.2. NF-kB becomes potential therapeutic target as it has constitutive activation in peritoneal endometriosis.3. It has been recognized that NF-kB expression decreases when red fruit extract is given.4. Effect of red fruit extract, compared to leuprolide, a widely used treatment for endometriosis, on NF-kB expression was examined.5. NF-kB was significantly lower in endometriotic group receiving red fruit extract than in group receiving leuprolide.   ABSTRACT Objectives: This study aimed to prove the effect of different administration of red fruit (Pandanus conoideus Lam) extract and leuprolide on the expression of NF-kB and endometrial implant width in mouse models of endometriosis. Materials and Methods: This study used completely randomized design with pre-post separated sample. The sample size was 36 female mice (Mus muculus), which were divided into 3 groups randomly. They were given with cyclosporine A, estrogen and endometrial tissue to form endometriosis model. Results: The mean expression of NF-kBin in treatment group receiving leuprolide (4.13 + 0.70) and red fruit (2.70 + 1.08) were significantly lower than that in control group (6.94 + 1.007). Whereas, in treatment group receiving red fruit (2.70 + 1.08), it was significantly lower than in those receiving leuprolide (4.13 + 0.70). Mean endometrial implant width in treatment group receiving leuprolide (13.62 + 3.21) and red fruit (8.93 + 2.74) were significantly lower than that in control group (28.89 + 8.28). Whereas, in treatment group receiving leuprolide (13.62 + 3.21), it was not significantly higher than in those receiving red fruit (8.93 + 2.74). Conclusion: The expression of NF-kB was significantly lower after the administration of red fruit than leuprolide. The width of endometrial implant was not significantly lower after the administration of red fruit than after the administration of leuprolide.
