Abstract

Diminazene aceturate has been reported to inhibit the reverse transcriptase activity by intercalating action mechanism of a number of protozoon eucaryot parasitic like Trypanosoma species. The phamacokinetics of diminazene in the blood plasma of five infected goats treated with single intramuscular doses of 7 mg diminazen base kg-1 body weight was investigated. The concentrations of the drug were determined by reverse phase high performance liquid chromatography. Results show that the mean (± SD) Absorption, Distribution, Metabolism and Excretion (ADME) of the drug plasma followed a tri-exponential process with Ka (minutes-1) were obtained at 5.10-2 ± 26.10-3, α (minutes-1), K12 (minutes-1) and K21 (minutes-1) were obtained at 18.10-3 ± 1.10-2, 14.10-3 ± 1.10-2 and 1.10-3 ± 1.10-3. The mean values of β (minutes-1) and K13 (minutes-1) were obtained at 1.4.10-4 ± 4. 10-5 and 3.10-3 ± 2.10-3. The mean values of Tmax (minutes) and Cmax (µg.ml-1) were obtained at 53.71 ± 30.61 and 13.40 ± 8.13. The mean values of Vds (L), Cl (ml.minutes-1), T1/2β(hours-1) and AUC0→∞ (µg.L-1.minutes) were obtained at 4.91 ± 3.12, 14.29 ± 4.08, 94.91 ± 33.23 and 12.680 ± 2.722. Abstract

Penelitian ini dilakukan untuk menemukan profil kinetik suramin sebagai suatu kemoterapi tripanosidal menggunakan 3 sapi peranakan Ongole betina sehat dan 3 sapi Madura sehat. Tiap sapi mendapatkan dosis tunggal 10 mg/kg berat badan. Peparasi sampel dilakukan secara hidrolisis menggunakan HCL 2 M HCL selama 18 jam. Kadar suramin pada plasma darah diukur dengan metode sensitif melalui spectrofotometer Ultra Lembayung-Tampak pada X, 535,6 nm. Hasil-hasil menunjukkan sebagai berikut : Terdapat hampir 94 % perolehan kembali analit dalam plasma setelah dilakukan deproteinasi setiap sampel. Rerata (± SD) eliminasi obat pada plasma sapi peranakan Ongole dilakukan secara bieksponensial dengan waktu paruh 11,579 ± 8,608 dan 288,75 ±100,026 jam untuk fase distribusi dan elimination pasca injeksi intravena. Karakteristik eliminasi sapi madura menunjukkan bieksponensial dengan waktu paruh distribusi dan eliminasi (± SD) 9,1 ± 6,389 jam dan 200,2 ± 53,347 jam pasca diberikan intravena. Rerata waktu keberadaan obat pada sapi peranakan Ongole pada 384,792 ± 141,882 jam, dan sapi Madura diperoleh pada 274,209 ± 76,417 jam. Abstract

Herb medicine have an active substances that can dissolved on polar, semi polar and non polar liquid extract. The methanol and ethyl acetate as a polar and semi polar extract liquid were used to study of herb medicine such as Dendrophthoe species. Diethyl ether as a non polar liquid extract was never use to study the Dendrophthoe species. The aim of this study was to investigate the anti proliferation activity of Dendrophthoe species to myeloma culture cells after extracted by non-polar extract solution (diethyl ether). The post test only control group design was used for this research. A thirty six of microtiterplates wells were used for myeloma culture cells in RPMI medium. The wells were devided placing in two groups: treatment groups and controls groups. Each three wells of six treatment subgroups added with 100 µl of 1.1; 5.5; 11; 22; 33 and 44 µg/ml crude diethyl ether extract series. A RPMI solution at similar method and volume were used as control substances. The cells were assessed by inverted microscope in 200x magnified two days after. The cells were quantified analyzed for anti proliferation activity by using 1:1 methylene blue solution. The results showed that started from 11.0 µg/ml of crude diethyl ether extract of Dendrophthoe species have been anti proliferation abilities of myeloma culture cells (p<0.05). In conclusion, actives substances of Dendrophthoe species where dissolved in non polar liquid as diethyl ether has anti proliferation activities to cancer cell in vitro.

Abstract

The determining of Phenazone to human blood plasma from healthy man after separated by solid phase extraction (SPE) and spectroscopic measurements has been investigated. The objective of that research was to obtain system suitable test for determine the Phenazone level in biological fluids (human blood plasma), for new performed dosage regimented in clinical dentistry. The method can be divided into the following four steps. 1. Centrifugation the blood sample, 2. Extraction from blood plasma and, 3. Separation by SPE with manual pressured, 4. Elution to SPE followed by the measurement on a spectrophotometer in the ultra violet region. The critical value of  │t │at the 5% confidence level indicates that there is no systematic error in the linearity proposed method. Recoveries for this research were obtained at ranging 93.460 to 95.598%. The coefficient variation precision of this procedure was clearly good at smallest than 2%. The analytical procedure can be carried out in one working operation as a monitored therapeutic activity.

Abstract

BACKGROUNDHuman infertiliy cases in Indonesia have tended to increase at about 2-5%annually since 2000. Many tropical plants in Indonesia are potential sources of novel anti-infertility compounds, e.g. Dendrophthoe pentandra L. Miq. (benalu duku), a parasitic plant growing on Lansium domesticum. The objective of this study was to identify the effect of crude methanolic Dendrophthoe pentandra leaf extract on follicle stimulating hormone (FSH) and progesterone levels in female rats.METHODSFourteen Rattus norvegicus Wistar strain female rats were divided into anintervention group and a control group, and synchronized to estrus via thepheromone synchronizing method. The intervention group was given daily single intramuscular injections of crude methanolic Dendrophthoe pentandra leaf extract at 100 mg/kgBW for 4 days, while the control group was given daily single intramuscular injections of 1 mL of distilled water. Determination of FSH and progesterone levels in whole blood was done using the Randox Evidence Investigator analyzer. The data were subjected to the t test for two independent samples.RESULTSMean FSH in the intervention group was 9.28 ± 6.72 mIU/mL, which was lower than mean FSH of 24.80 ±16.35 mIU/mL in controls (p<0.05). Mean progesterone level in the intervention group was 33.55±13.96 nmol/L, twice as high as that in the control group, which was 18.47± 06.47 nmol/L (p<0.05).CONCLUSIONSCrude methanolic Dendrophthoe pentandra leaf extract reduces FSH andincreases progesterone levels in female rats, but cannot yet be recommended for use as fertility hormone inhibitor or stimulator in rats.

Abstract

Food safety and the phenomenon of antimicrobial resistance have caused world problems since the beginning of 2019, while the abuse and use of antibiotic problems in fresh animal origin products (PSAH) are still common. The purpose of this training activity is to provide direct competency strengthening for farmers who are closely related to the use of antibiotics where PSAH obtained will be directly related to human health. A total of 50 dairy farmers were conducted training using the method of learning by doing and doing by learning using 3 stages of activities, namely (1) strengthening competencies face to face, (2) continued training in counting downtime of veterinary medicines using props, ending the application of tetracycline residue examination at PSAH and examples of milk residues. The results of the training show that there is mastery of training materials by the average farmer farmers controlled by 85%.AbstrakKeamanan pangan dan fenomena antimicroba resistance menjadikan persoalan dunia sejak awal tahun 2019, sementara penyalahgunaan dan penggunasalahan antibiotika pada produk segar asal hewan (PSAH) masih sering terjadi. Tujuan kegiatan pelatihan ini adalah untuk memberikan penguatan kompetensi langsung terhadap peternak yang erat kedekatannya dengan penggunaan antibiotika dimana PSAH yang diperoleh akan langsung berhubungan dengan kesehatan manusia. Sejumlah 50 peternak sapi perah dilakukan pelatihan menggunakan metode learning by doing dan doing by learning menggunakan 3 tahapan kegiatan yaitu (1) penguatan kompetensi secara tatap muka, (2) dilanjutkan pelatihan penghitungan waktu henti obat hewan menggunakan alat peraga, diakhiri penerapan pemeriksaan residu tetrasiklin pada PSAH dan contoh adanya residu apada susu. Hasil pelatihan menunjukkan terdapat penguasaan materi pelatihan oleh petani peternak rerata dikuasai 85%.

Abstract

Training on non-alcohol based hand sanitizer (HS) was conducted for assisted youth in five districts, namely Gresik, Bojonegoro, Lamongan, Probolinggo and Nganjuk for three days. In the training, competency strengthening was carried out, namely theory and practicum, ending with evaluation of activities. During the implementation of theory and practicum, training materials were provided using social media facilities in the form of WhatsApp and e-mail. Specifically for coordination with participants, a training group was created using social media. The results of the training evaluation turned out that in terms of strengthening the competence of knowledge in making HS and working mechanisms of HS, it was increasing. In terms of straightening the meaning of HS production to be more focused according to the basis of science or good manufacturing practices (GMP). Thus, youth assisted with criteria include youth who are able to pass on knowledge to all young people in their respective districts. Furthermore, the knowledge and skills of the assisted youth in making HS based on non-alcoholic substances will be more agile and targeted. Thus, in reality, the training on making non-alcohol based HS has made the assisted youths become pioneers at the end of the line to pass on their knowledge. The conclusion in the implementation of this training is that the assisted youth are capable and ready to make non-alcohol-based HS as well as being ready to produce for a small scale while showing real work to be transmitted (98%). abstrakTelah dilakukan pelatihan pembuatan hand sanitizer (HS) berbasis non-alkohol terhadap pemuda binaan di lima kabupaten yaitu Gresik, Bojonegoro, Lamongan, Probolinggo dan Nganjuk selama tiga hari. Dalam Pelatihan tersebut dilakukan pemberian penguatan kompetensi yaitu teori dan praktikum diakhiri dengan evaluasi kegiatan. Selama berlangsung pelaksanaan teori dan praktikum, materi pelatihan diberikan menggunakan fasilitas media sosial berupa whatsapp dan e-mail. Khusus koordinasi dengan peserta dilakukan pembuatan grup pelatihan menggunakan media sosial. Hasil evaluasi pelatihan ternyata dari segi penguatan kompetensi pengetahuan pembuatan HS dan mekanisme kerja HS, makin meningkat. Segi pelurusan pengertian produksi HS menjadi makin terarah sesuai basis ilmu pengetahuan atau cara pembuatan obat yang baik (CPOB). Dengan demikian pemuda binaan dengan kritetria termasuk pemuda yang sanggup menularkan ilmu ke seluruh masyarakat muda di kabupaten masing-masing. Lebihlanjut pengetahuan dan ketrampilan pemuda binaan dalam membuat HS berbasis bahan non-alkohol makin cekatan dan terarah. Dengan demikian seacara nyata pelatihan pembuatan HS berbasis non-alkohol telah menjadikan pemuda binaan sebagai pelopor di ujung garis untuk menularkan ilmunya. Kesimpulan dalam pelaksanaan pelatihan ini adalah para pemuda binaan telah sanggup dan siap membuat HS berbasis non-alkohol sekaligus siap berproduksi untuk skala kecil sambil memperlihatkan karya nyata untuk ditularkan (98 %).

Abstract

This study was aimed to identify the ultra structural morphology of Toxocara cati First Stage Larvae and Second Stage Larvae using Light Microscope. Toxocara cati larvae were obtained from adult worm eggs then were treated in phospat buffer saline with comparasion 1 : 1 until it reached the stage of larvae 1 and 2. The shell of egg Toxocara cati were thick and usually consist of three layers. The first layer was inner membrane, the second layer was middle layer and the third was outer layer. The results of larvae morphology that were identified using light microscope showed that the anterior part of Toxocara cati first stage larvae has a dorsal lip and esophagus and intestine on the posterior part but it could not be identified perfectly. The anterior part of Toxocara cati second stage larvae that were identified has three lips that leads directly into the oesophagus. Three lips on the anterior part of Toxocara cati larvae 2 consist of a dorsal lip and two subventral lips. Morphological of Toxocara cati larvae similar with the Toxocara cati adult worms because morphology of larvae and adult stage was difficult to differentiated.
