Abstract

Buah delima dilaporkan mengandung senyawa antioksidan yang tinggi dan berfungsi sebagai hepatoprotektor. Penelitian ini bertujuan menguji aktivitas ekstrak polifenol buah delima sebagai hepatoprotektor dan mengetahui pengaruhnya terhadap gambaran histopatologi hati tikus yang diinduksi parasetamol. Ekstraksi polifenol dilakukan dengan menggunakan pelarut aseton, air, asam asetat (90:9.5:0.5). Sebanyak 22 ekor tikus dibagi menjadi 7 kelompok, yaitu kelompok I diberi pakan standar dan akuades, kelompok II diberi pakan standar dan parasetamol 500 mg/BB, kelompok III diberi pakan standar, parasetamol 500 mg/BB, dan Cursil-70 13.3 mg/BB, kelompok IV, V, VI dan VII diberi pakan standar, parasetamol 500 mg/BB, serta ekstrak polifenol buah delima dengan konsentrasi 13.3 mg/BB, 100 mg/kg BB, 250 mg/kg BB dan 500 mg/kg BB untuk masing-masing kelompok. Perlakuan dilakukan selama 34 hari, pada hari terakhir dilakukan analisis histopatologi hati dengan mengamati kerusakan hepatosit yang meliputi jumlah sel nekrotik, sel apoptosis, sel kupffer, dan degenerasi lemak. Hasil penelitian menunjukkan bahwa ekstrak polifenol buah delima dengan dosis 500 mg/BB dapat menghambat kerusakan hati (p < 0.05) karena pada konsentrasi tersebut jumlah sel nerkotik rendah, tidak ada sel apoptosis, jumlah sel kuffer rendah, dan jumlah degenerasi lemak rendah.

Abstract

The present study aimed to evaluate different agro-morphological traits among twenty genotypes of Curcuma aeruginosa Roxb. and three varieties of Curcuma zanthorrhiza Roxb. Agro-morphological data traits were investigated based on qualitative and quantitative parameters from PPVFRA descriptors with modification. All the recorded data was analyzed through SPSS 16.0 and R 3.4.2 for ANOVA and similarity analysis, respectively. Significant differences (P <0.05) were observed in the traits studied of the habit of the rhizome, the shape of the rhizome, length of primary rhizome, number of mother rhizome, plant height, pseudostem diameter, number of leaves, leaf length, leaf width, and number of shoots. Hierarchical cluster analysis (HCA) classified the genotypes into three groups. The principal component analysis (PCA) were showed consistent with results of the HCA. These genotypes could be used as novel genotype in agricultural biochemistry for breeding programs.

Abstract

Torbangun (Plectranthus amboinicus) merupakan tanaman obat dari keluarga Lamiaceae. Pada masyarakat Batak, daun torbangun digunakan untuk meningkatkan produksi air susu. Keragaman kandungan fitokimia dapat ditingkatkan salah satunya dengan teknik iradiasi sinar gamma untuk program pemuliaan tanaman. Pada penelitian ini, metode metabolomik yang tidak tertarget digunakan untuk mengevaluasi profil senyawa pada bagian daun tanaman mutan dari iradiasi sinar gamma dan kontrol. Kandungan senyawa planlet mutan dan kontrol dianalisis dengan GC-MS. Data GC-MS dianalisis dengan kemometrik dengan menggunakan hierarchical cluster analysis (HCA). Keragaman kandungan kimia ditunjukkan oleh planlet mutan dan kontrol daun torbangun. Hasil analisa planlet kontrol torbangun menunjukkan terdapat lima senyawa utama diantaranya hydroxymethylfurfurole (35,15%), brevifolin (7,69%), 3-Dihydro-3,5-Dihydroxy-6-Methyl-4H-Pyran-4-one (DDMP) (13,13%), stigmasterol (6,51%) dan ferruginol (8.63%). Hasil analisa mutan torbangun memiliki kandungan senyawa DDMP (19,57%), neophytadiene (8,47%), linolenic acid (9,10%), ferruginol (7,61%), stigmasterol (14,14%) dan gamma.-sitosterol (7,08%). HCA menunjukkan tiga komponen senyawa yang berbeda antara planlet kontrol dan mutan yaitu kenaikan kandungan senyawa DDMP dan stigmasterol, serta penurunan kandungan hydroxymethylfurfurole pada mutan torbangun.

Abstract

Curcuma aeruginosa Roxb. rhizome is an essential herb material with significant antimicrobial, anticancer, and antioxidant activities. Thus, it is important to search for C. aeruginosa plant that has the rhizome yield trait with more excellent stability in changing environmental situations. This research aimed to evaluate the genotype x environment interaction using additive main effects and multiple interaction (AMMI) analysis for photosynthetic rate and rhizome yield of C. aeruginosa genotypes. Twenty clones of C. aeruginosa plus three control varieties of C. zanthorrhiza Roxb. were evaluated in three environments (Bogor, Cianjur, and Sukabumi) using a randomized complete block design with three replicates. The photosynthetic rate and rhizome yield showed significantly (p < 0.01) genotype x environment interaction. The AMMI2 explained 100% variability of the photosynthetic rate and rhizome yield parameters. G2, G10, G12, G13, G14, G16, G17, G19, and G20 were stable genotypes for rhizome yield based on AMMI analyses. Abstract

Xylocarpus granatum is mangrove plant that traditionally used as face powder in Central Sulawesi, Indonesia which related to antioxidant, antiglycation and tyrosinase inhibition activities. This study aimed to evaluate the potency of X. granatum as a tyrosinase inhibitor, antiglycation, and antioxidant. The leaves, stem, stem bark, fruit flesh, fruit peel, and kernel of X. granatum were extracted using ethanol then their tyrosinase inhibition, antiglycation, and antioxidant were evaluated. Tyrosinase inhibition activity was evaluated using in vitro assay with L-tyrosine and L-DOPA as the substrate of monophenolase and diphenolase. Antiglycation activity was studied by measuring the excitation and emission fluorescence from glucose and fructose reaction with Bovine Serum Albumin. Antioxidant activity was measured using the DPPH radical scavenging assay. The result showed that the ethanolic extract of fruit flesh has higher potency as tyrosinase inhibitor (IC50 of 393.8 mg/L and IC50 of 448 mg/L, respectively for monophenolase and diphenolase). Antiglycation assay showed that the ethanolic extract of stem bark provides the strongest antiglycation activity with an IC50 of 118.1 mg/L. Meanwhile, fruit peel provides the strongest antioxidant activity with an IC50 of 5.5 mg/L. Fractionation of ethanolic extracts of each part of X. granatum tree yield fractions with lower bioactivity compared to the crude extract. Moreover, stem extract and fractions from two different locations (Tarakan and Kendari) tend to have different bioactivities strengths.  The stem part of X granatum could be developed as new raw material of cosmetic product in Indonesia, while ethanol as the solvent for extraction, and the different bioactivity of stem extract from different location can be the consideration for the industry to standardize the extract prior to production of final product.

Abstract

Penelitian mengenai potensi ekstrak air dan etanol 30%, 70%, serta 96% dari kulit batang  kayu  manis Padang (Cinnamomum burmannii) terhadap aktivitas enzim a-glukosidase secara in vitro telah dilakukan. Penelitian ini bertujuan untuk menguji aktivitas inhibisi ekstrak air dan etanol (30%, 70% dan 96%) dari kulit batang kayu manis Padang terhadap aktivitas enzim a-glukosidase (dengan akarbosa sebagai kontrol positif), identifikasi senyawa pada ekstrak yang mempunyai daya inhibisi tertinggi dengan GC-MS pyrolisis (Py-GC-MS) serta uji fitokimianya. Kulit batang kayu manis di ekstraksi dengan metode maserasi 3 kali 24 jam.  Hasil penelitian menunjukkan bahwa ekstrak air dan etanol 30% kayu manis Padang mempunyai daya inhibisi berturut-turut sebesar 94.88% dan 94.51% yang tidak berbeda nyata dengan daya inhibisi a-glukosidase dari akarbosa 1%. Hasil fitokimia menunjukkan adanya kandungan flavonoid, tanin, senyawa fenolik dan karbohidrat, pada kedua ekstrak tersebut.Sedangkan hasil analisis kualitatif pada ketiga ekstrak dengan Py-GC-MS menunjukkan adanya senyawa fenolik-fenolik sederhana seperti pyrocatechol, catechol, guaiacol,dan hidroquinone yang diduga merupakan hasil penguraian senyawa golongan polifenol dan diduga sebagai agen antidiabetik oral. Selain dari itu, kedua ekstrak mengandung 1,6-anhidro-beta-D-glukosapiranosa (Levo glukosan) yang tidak menyebabkan peningkatan kadar gula darah.Research about inhibition activity of aqueous and 30%, 70% and 96% ethanolic bark extracts of Cinnamon Padang (Cinnamomum burmannii) against a-Glukosidase enzyme activities throught in vitro assay has been done. The aim of this research are to determine activity of aqueous and ethanolic (30% 70%, and 96%) bark extracts of Cinnamon Padang against activity of a-glucosidase enzyme (with acorbose as a positive control), identification compunds by GC-MS pyrolysis from those extract that the result showed have the highest inhibition activity and its phytochemical assay. Cinnamon was extracted by maseration method 3 times 24 hours. The result showed that the aqueous and 30% ethanolic extract of cinnamon Padang inhibited a-Glukosidase enzyme on 94.88% and 94.51% respectively, but not significantly different with the inhibition to a-Glucosidase from 1% of acarbose. Those extracts contain flavonoids, tannin, phenolic and carbohydrate compounds, while the qualitative analyzed from those extracts by Py-GC-MS, presence the phenolic compounds such as pyrocatechol, catechol, guaiacol, and hydroquinone from polyphenol compound that act as antidiabetic oral agent. Instead of that, those extracts contain 1,6?anhydro? beta ? D ? glucopyranose (Levoglucosan), but its compound could not increase blood glucose level.

Abstract

Temulawak (Curcuma  xanthorrhizaRoxb.) belongs to the family Zingiberaceae, has been empirically used as herbal medicines. The research was aimed to evaluate three promising lines of Temulawak based on their high bioactive contents (xanthorrhizol and curcuminoid) and its in vitro bioactivity (antioxidant and toxicity), and to obtain information on agrobiophysic environmental condition which produced high bioactive compounds. The xanthorrhizol and curcuminoid contents were measured by HPLC. In vitro antioxidant and toxicity were determined by DPPH (1,1-diphenyl-2-picryl-hydrazyl) method and BSLT (Brine Shrimp Lethality Test). The result showed that promising line A produced the highest yield of bioactive and bioactivity, i.e. 0.157 and 0.056 g plant-1of xanthorrizol and curcuminoid respectively. The IC50 of antioxidant activity was 65.09 mg L-1and LC50of toxicity was 69.05 mg L-1. In this study, Cipenjo had the best temulawak performance than two other locations. According to the agrobiophysic parameters, Cipenjo environmental condition was suitable for temulawak cultivation with temperature 28-34 ºC, rainfall ± 223.97 mm year-1 and sandy clay soil. Abstract

ABSTRACTCurcuma aeruginosa Roxb., popularly known as “temu ireng”, is considered as a potential source of medicinal plant for pharmacological activities. However, varieties of C. aeruginosa are still limited in Indonesia so it needs more accessions for improvement and development of new varieties. Rhizome colors, phytochemical contents and extract yield from 20 promising lines of C. aeruginosa were investigated by qualitative method for rhizome colors and phytochemical contents, and maceration method using 70% ethanol for yield extract. Similarity analysis was used for cluster analysis based on rhizome colors, phytochemical contents and yield extract. Blue was the color characterization of rhizome C. aeruginosa. The extract yield for 20 promising lines of C. aeruginosa varied from 7.92 to 19.71%, with KN and BH promising lines having the lowest and highest value, respectively. All promising lines of C. aeruginosa contain saponin and triterpenoid. Based on similarity analysis, all promising lines could be divided into 3 clusters. Cluster I consisted of 14 promising lines i.e. WG, SH, KA, GD, BH, KP, NW, PW, MB, PR, PT, KN, MD, and PK. Cluster II consisted of 4 promising lines i.e. LC, CB, KL, and GK. Abstract

Celosia cristata L., namely jengger ayam, is an ornamental plant with a unique structure. The flower of C. cristata is shaped grains with small size; thus, the hybridization for increasing the diversity in this plant is challenging. One alternate technique for improving diversity is mutation induction by gamma rays. This study aimed to evaluate the variability of agro-morphological traits in seventeen M3 mutant C. cristata genotypes at the Sabisa Farm garden. Agro-morphological characteristics were evaluated based on descriptors of the International Union for the Protection of New Varieties of Plants (UPOV) with modification. Analysis of variance was performed using SPPS 20.0, whereas a similarity analysis was performed using PBSTAT-CL 2.1. Significant variability among C. cristata genotypes (p < 0.05) was found for plant height and flower length. Three clusters based on agro-morphological traits were obtained from similarity analysis with the dissimilarity distance of 32%. The results indicated that the agro-morphology traits were able to differentiate the 17 genotypes of C. cristata studied, and therefore it can be used for further research development in plant breeding programs. Abstract

INTRODUCTIONTumor  is  a  degenerative disease  as  the second agent cause of death in human. This disease is caused by disturbances of cell’s growth that show alteration and uncontrolled of cells proliferation. The uncontrolled of cells proliferation is also accompanied by penetration of that cells into others tissue and develop on it. Removed of tumor is usually done by operation or chemotherapy. Alternative treatment that can be used is herbal treatment. Typhonium flagelliforme known as “Keladi Tikus” in Indonesia; is known has a chemopreventive effect (2). Other researchers also show flavonoid glucoside of T. flagelliforme (1) can induce the activity of apoptosis in colon cancer cells (5). The aim of this study is to examine the in vitro anti-proliferation activity of Typhonium flagelliforme leaves ethanol extract on MCA- B1and MCM-B2 cell line.
