Abstract

Karawang is a rice center in West Java and National, rice straw potentially used as a media for merang mushroom to grow. In research, tofu pulp planting media was carried out and tested the productivity of the merang mushroom F3 seedlings from Cilamaya. The purpose of the research to obtain the composition of the tofu pulp growing media which gave the highest result towards merang mushroom growth. The research was conducted in Plant Biotechnology Laboratory, Faculty of Agriculture, Singaperbangsa Karawang University and in Kumbung (Mushroom House), Pasirmulya Village, Majalaya, Karawang. The method that is used was experimental, single factor randomized block design (RBD) of 6 treatments in 4 replications, the treatments were: A (Control + Straw 100%), B (Tofu pulp 5% + straw 95%), C (Tofu plus 15% + straw 85%), D ( tofu pulp  25% + straw 75%), E (tofu pulp 35% + straw 65%) and F (tofu pulp 45% + straw 55%). The results showed that treatment C (15% tofu pulp + 85% straw) has the best value, there is a significant effect on the parameters of fruit body length of 6,98 cm, fruit body diameter with a value of 18,22 mm, number of fruit bodies with a value of 5,80, fruit body weight 10,05 g.

Abstract

The experiments begin at Lebak Sari village of Sukaluyu the East Telukjambe sub-districts of Karawang districts from March 2021 until April 2021. The research method that used in this experiments was the experimental method using singular factor Randomized Block Design (RBD). The factor that would be tested was rice rice cleanse water. The design to be done was 7 level treatments that repeated 4 times. The concentration treatment that done in this experiments were A (0% Rice Cleanse Water Concentration), B (20% Rice Cleanse Water Concentration), C (40% Rice Cleanse water Concentration), D (60% Rice Cleanse Water Concentration), E (80% Rice Cleanse Water Concentration), F (100% Rice Cleanse Water Concentration), G (4 gram NPK Fertilizer as a Comparison). Overall, concentration giving of rice cleanse fermentation water into caisim crop (Brassica juncea L.) not yet could gave better significant growth doesn’t really reach maximum for the caisim was because the contents of nutrition that inside of the rice cleanse fermentation water was classified as low. Abstract

Globba leucantha var. bicolor Holttum is a part of Zingiberaceae plant that is not yet widely known but can be used as a medicinal material as well as an ornamental plant. So far Globba plant propagation has not been widely done and has relied solely on from its habitat. Research aims at acquiring Naphthalene Acetic Acid (NAA) concentrations and optimal kinetin for the multiplication of ginger plant shoots (Globba leucantha var. bicolor Holttum) on in vitro culture. The research method uses experimental methods with non-parametric statistics and 24 repeated treatments 3 times as well as being analyzed descriptively using the Kruskal Wallis Test. The treatment used NAA with concentrations of 0 mg/l, 0.1 mg/l, 0.5 mg/l, 1 mg/l, 1.5 mg/l, and 2 mg/l combined with Kinetin with concentrations of 0 mg/l, 0.1 mg/l, 1 mg/l and 10 mg/l. The results showed that the growth of Globba plant's best shoots at N3K3 treatment (NAA 1 mg/l + Kinetin 10 mg/l) with a shoot emergent time of 21.58 HSI, a percentage of shoot forming 66.67%, a shot count of 0.89 buds per explant, and a shoot length of 0.18 cm. Additionally at the same time also formed Globba plant roots with the best treatment of N5K2 (NAA 1 mg/l + Kinetin 10 mg/l) with a time of appearing rooted 25.67 HSI and a root count of 7.78 roots per explant.

Abstract

The experiment was carried out in kumbung mushroom, Gadog Village, Tamelang Village, Purwasari District, Karawang Regency, West Java Province from May to June 2021. The experimental method used was a single factor Randomized Block Design (RBD) with 4 replications. There were 7 treatments, namely A (Control without treatment), B (LOF 30 ml), C (LOF 40 ml), D (LOF 50 ml), E (Coconut Water 250 ml), F (Coconut Water 350 ml), and G (Coconut Water 450 ml). If the data were analyzed by means of variance and the F test at 5% level showed significant results, then the Duncan Multiple Range Test (DMRT) test at 5% level was carried out to determine the best treatment. The results showed that the response was not significantly different from the application of liquid organic fertilizer nutrition and coconut water nutrition to all treatment parameters. The highest average fruit body length was found in treatment G (450 ml Coconut Water) which was 3,30 cm and was not significantly different from the others.

Abstract

Jamur merang merupakan salah satu jamur konsumsi yang sangat diminati, sehingga kebutuhan jamur merang semakin meningkat. Namun untuk memenuhi kebutuhan tersebut banyak ditemukan kendala, salah satunya dalam penyediaan biakan murni terkait jenis dan konsentrasi media biakan murni. Penelitian ini bertujuan untuk mendapatkan jenis dan konsentrasi media biakan murni yang memberikan pertumbuhan koloni miselia jamur merang tertinggi. Penelitian dilaksanakan di Laboratorium Bioteknologi Tanaman Fakultas Pertanian UNSIKA dari Maret sampai Agustus 2017, menggunakan rancangan acak kelompok faktorial dengan 3 ulangan. Faktor pertama : jenis media dengan 4 taraf  m1 = PDA, m2 = arang sekam padi, m3 = jerami, m4 = campuran arang sekam padi dan jerami. Faktor kedua : konsentrasi media biakan murni dengan 3 taraf : k1 = 200 g l-1, k2 = 250 g l-1, k3 = 300 g l-1. Hasil penelitian ini menunjukkan bahwa, media arang sekam padi dan PDA dengan konsentrasi media biakan murni 200 g l-1 memberikan pengaruh yang sama baiknya bagi pertumbuhan miselium jamur merang dengan diameter sebesar 8 cm.  Media arang sekam padi dapat dijadikan sebagai alternatif pengganti media PDA. Straw mushroom is one of the most popular mushroom. So needs of straw mushroom more  increasing. However, to meet those needs had been founded obstscles, one of them is type and consentration of straw mushroom pure culture media. The aims of this research was to find out the type and consentration of pure culture media which could give the best growth of straw mushroom mycelia. The research was conducted in laboratory of plant bioctehnology, faculty of Agriculture, University of Singaperbangsa Karawang from March until August 2017. The experiment method used the factorial randomized block design with 3 replications. The first factor was the type of media, consisted of 4 levels m1 = Potato dextrose agar (PDA), m2 = rice husks m3= Rice Straw m4 = Mixture of rice husk and straw. The second factor was the concentration of media, consisted of 3 levels k1 = 200 g l-1, k2 = 250 g l-1, k3 = 300 g l-1. The results of the research showed that rice husk media and PDA with consentration’s 200 g l-1 gave good affect to mycelia colony diameter of straw mushroom by 8 cm. The rice huskwas an alternative as pure culture media for straw mushroom, substitute PDA.

Abstract

Salah satu permasalahan utama dalam upaya peningkatan produksi padi adalah terjadinya serangan penyakit blas yang disebabkan oleh jamur Magnaphorte oryzae B. Couch (anamorf Pyricularia oryzae Cavara). Penyakit blas merupakan penyakit penting pada pertanaman padi. Patogen penyakit blas memiliki keragaman genetik dan kemampuan beradaptasi yang tinggi sehingga dapat dengan cepat mematahkan ketahanan varietas yang baru ditanam. Penanaman varietas tahan harus disesuaikan dengan keberadaan populasi ras blas pada lokasi tersebut. Untuk itu informasi hasil monitoring keberadaan ras blas di suatu agroekositem sangat diperlukan. Penelitian ini dilakukan tahun 2009 sampai 2010, di BB-Biogen, penelitian bertujuan untuk 1) memperoleh isolat blas dengan frekuensi intensitas serangan dan tingkat virulensi tertinggi, 2) melakukan pengelompokan ras dominan per daerah asal isolat, serta 3) mendapatkan informasi keragaman genetik cendawan blas dari empat daerah berbeda yaitu : Sukabumi, Lampung, Jasinga, dan Sitiung berdasarkan tingkat virulensinya. Analisis keragaman fenotipik dilakukan dengan cara menguji satu set varietas diferensial Indonesia terhadap 100 isolat blas daun dan leher malai koleksi BB-Biogen. Berdasarkan hasil pengujian fenotipe telah teridentifikasi bahwa 100 isolat blas terdiri dari 19 ras yang berbeda. Banyaknya macam ras dalam satu lokasi berkisar antara 4-12 macam. Ras 001 merupakan ras yang dominan dengan frekuensi virulensi tertinggi, yaitu 35 % (35 isolat) dari total populasi. Ras 001 terdapat di seluruh lokasi sampling yaitu Sukabumi, Lampung, Jasinga, dan Sitiung. Terdapat pula ras spesifik lokasi, yaitu 1) ras 153, 235, dan ras 313 yang merupakan ras spesifik di Sukabumi, 2) ras 060, 061, dan ras 101 di Jasinga, dan 3) ras 021, 023, 033, dan 103 di Sitiung. Nilai keragaman genetik (Hi) pada tiap lokasi (Lampung, Sukabumi, Jasinga dan Sitiung) berkisar antara 0.79 (Lampung) sampai 0.82 (Sukabumi, Jasinga, dan Sitiung). Hasil perhitungan nilai koefisien diferensiasi genetik (GST) menunjukkan nilai GST yang cukup tinggi yaitu 0.76. Kata Kunci: Virulensi, blast, Pyricularia, Ras

Abstract

The mustard plant (Brassica juncea L.) is one of the vegetable commodities which very potential to be developing, because this comodities high economic value. The aims of this experiment was to fine out the concentration of coconut water fermentation that is most suitable for the growth and yield of mustard. This research has been conducted at Teluk Jambe Timur, Karawang district, West Java in March to April 2021. This research used experimental method with randomized block design (RBD) consisting of 6 treatments in 4 replication. The treatments were , treatment (NPK), treatment B (25 %), treatment C (50 %), treatment D (75 %) and treatment E (100 %). The data of research are analyzed by the analysis of variance at 5 % significance level and if the real effect is done by further test with DMRT test at 5 % significance level. The results showed that the treatment of concentration of water coconut fermentation had no effect on plant height age 14 day after planting (DAP), the number of leaves age 21 DAP, and significant effect on plant height age 7, 21, and 28 DAP, the number of leaves age 7, 14, and 28 DAP. The best treatment of coconut water fermentation was found on treatment C and D.

Abstract

Merang mushroom (Volvariella Volvaceae) is one of the commodities of agricultural horticulture crop yield that has quite high economic value. Planting media become the most important thing for the farmer. Planting media that usually used by farmer is just the rice hay. The purpose of this research is to obtain plant media subtitution of dry banana leaf (klaras) and husk charcoal that most effective, so it would produce optimal growth and yield of Merang Mushroom. This experiment was implemented at Pasirmulya Village, Majalaya region, Karawang District at May untill july 2021. The research method that used is the experiment method with Randomized block design (RBK) single factor. There were 9 treatment l, which A (without treatment), B (Klaras 15%), C (Klaras 17%), D (Klaras 19%), E(Klaras 21%), F (Husk charcoal 15%), G (Husk charcoal 17%), H (Husk Charcoal 19%), I (Husk Charcoal 21%). Each of the treatment were repeated 3 times, so there were 27 Experiment units. If the data analyzed with analyze of variance and The test F rate at 5% showed significant results, then conducted to the continue test of Duncan Multiple Range Test (DMRT) at rate of 5% for acknowledging the best treatment. The experiment results showed that there were subtitution real effect of media dry banana leaf (Klaras) and husk charcoal towards the diameter of the crop body, and the total weight of yield by plots. Treatment I (Husk charcoal 21%) gave the highest results towards the diameter of crop body (2,40 mm), the total weight by plot (159,88 g).

Abstract

Karawang have been some of area become center of edible mashroom production, the problem in the production of mushroom is the farmers can’t to preparation of mushroom inoculant. The aim of this research is to get the best inoculant of edible mushroom whom use for spawn from Karawang. This research conducted by explorative design used four kind of edible mushroom source; Cilamaya, Purwasari, Lamaran and Pacing. Edible mushroom isolation doing by dilution methode until 10-7 by Saraswati (2008). Constanted purity of isolat from macro and microskopist analize in PDA media by place count. The last phase in this reaserchis to get the best spawn of edible mushroom based on direction radial growht rate and interaction betweet all kind of edible mushroom. The best kind of edible musroom inoculated to grain carrier. The result showed that the best radial direction growth rate is edible mushroom from Cilamaya (64,17 mm) compre with Lamaran (61,18 mm), Purwasari (57,82) and the lower is Pacing (43,31). The same result in colony area as long as interaction growth test, edible mushroom from Cilamaya give the best performance compare with the other. Edible mushroom from Cilamaya is the best based on growt rate colony radial test and interaction in PDA media with other all of kind.Keywords: Edible mushroom, isolation, characterization, Karawang

Abstract

Sugarcane (Saccharum officinarum L.) is one of the plantation commodities that have high economic value because it is the main raw material in the manufacture of sugar. Generally, sugarcane propagation was done by vegetative cuttings, but this method has drawbacks. In vitro plant, culture is an alternative technique for producing quality sugarcane seed, a high number of seeds with shoot multiplication. This research aims to review the influence of concentration NAA and BAP on shoot multiplication of sugarcane (Saccharum officinarum L.). The study was conducted in the Laboratory of Superior Farm Seed Management Unit, Indonesian Agency for Agricultural Research and Development from June until August 2021. Explant material used was young rolled leaves collected from AMS variety sugarcane. The experiment was arranged in Factorial Completely Randomized Design (RAL), with 12 treatments, and five replication. Explants were cultured during 8 weeks in MS media with concentration NAA ( 0 ppm, 0.5 ppm, and 1 ppm), and BAP (0 ppm, 1 ppm, and 2 ppm, 3 ppm). The parameters measured were the date of shoot initiation, number of shoots, length of shoot, number of leaves, date of root initiation, number of the root, length of root, and color of the shoot. The data were analyzed using ANOVA at the confidence level of 95%. The results showed that interaction between NAA and BAP in promoting Saccharum officinarum L. Micro cutting growth was observed. The combination between NAA of 0 ppm and BAP of 2 ppm was found to be the best in date of shoot initiation, number of shoots, and number of the leaf. The combination between NAA 1 ppm and BAP 0 ppm was recommended to promote the root number and length of the root.
