Abstract

This study aimed to get to know the effect of different doses formulated Trichokompos doses on the growth and production of onion varieties in peat area, as well as to get a dose of Trichokompos formulated and the best interaction on several onion varieties. Research using split plot design draft with 12 combination and each treatment was repeated 3 times, analyzed in the laboratory and analysis of variance. The main plot treatments consisted of: (V1): Varieties Bauji, (V2): Varieties Maja Cipanas, (V3): Varieties Bima Brebes. Treatment subplot, consists of 4 levels, namely: T1: (5 ton / ha), T2 (10 tons / ha), T3 (15 ton / ha) and T4 (20 ton / ha). The data obtained were statistically analyzed using analysis of variance (ANOVA) followed by HSD test at 5% level. The results of the research showed that, the provision of oil palm empty bunches Trichokompos formulated doses of 10 ton / ha and a dose of 20 ton / ha on the varieties Bauji can increase the content of chlorophyll by 78.64% and K nutrient uptake value of 2.46 mg / plant. Provision of Oil Palm Empty Fruit Bunch Trichokompos formulated doses of 10 ton / ha on Maja Cipanas varieties could increase plant height of 24.93 cm and a dose of 5 tonnes / ha can increase the diameter of the bulbs by 24.70% and the provision of oil palm empty bunches Trichokompos formulated doses of 5, 10 , 15 and 20 ton / ha on Bima Brebes varieties can increase the number of tillers, the number of bulbs, tubers fresh weight, fresh weight of the plants, the rate of photosynthesis, transpiration rate, stomata conductivity, CO2 concentration, the value of N uptake and nutrient uptake value P. Increasing doses of oil palm empty bunches Trichokompos formulated from 5 to 20 ton / ha on the varieties Bauji, Maja Cipanas and Bima Brebes not affect all parameters observed.

Abstract

Cellulolytic bacteria are bacteria that have ability to hydrolize cellulose into smaller oligosaccharides and into glucose units. Glucose is used as a source of carbon and energy for bacterial growth. The aims of this study wasto determine the ability of three bacillus sp. bacteria from rhizosphere of mustard plants (Bacillus sp. B1), rhizosphere of rice plants (Bacillus sp. B2), and peat soil of Giam Siak Kecil (Bacillus sp. B3) to produce cellulase enzymes based on clear zone which is seen surrounding the colonies. Cellulolitic activity of the bacteria was determined by their ability bacteria to degrade carboxymethyl cellulose (CMC) substrate. The results showed that all isolates tested were able to produce cellulase. Bacillus sp. B1 isolate had the highest cellulolitic activity [(6.502 ± 0.462) x 10 -3 U/mL] than 2 isolates based on 24 hourproduction time and120 rpmspeed of agitation.

Abstract

The Diseases found in oil palm plantations is the Upper Stem Rot (BBA) caused by pathogens Ganoderma sp. The alternatives that can used in controlling the disease BBA is by use endophyte microorganisms derived from plant tissue one of the Trichoderma virens endophyte. This study aimed to determine the effect of the administration of the different material inducer T. virens endophyte and get the best inducers ingredient in promoting growth and controlling upper stem rot disease on the oil palm seedlings, This research has been conducted in the experimental garden of the Faculty of Agriculture, State University of Riau, Industrial Business Unit Biofertilizer and Biopesticides University of Riau, Laboratory of Soil Science, Faculty of Agriculture, University of Riau, the Laboratory photomicrography and Laboratory of Biochemistry, Faculty of Mathematics and Natural Sciences, University of Riau, on Campus Binawidya km 12.5 Baru Simpang Village District of Handsome Pekanbaru. This study was conducted in May 2016 to November 2016. This study was conducted experiments using completely randomized design with 3 treatments and 7 replications so that obtained 21 experimental units. Data were analyzed statistically by ANOVA and tested with LSD at 5% level. The results showed that treatment of inducing materials in the form of T. virens endophyte suspension showed is the best treatment to be better increase plant height, stem diameter, root volume, and tend to be higher in prolong the incubation period, reduce the intensity of the disease, and increases the concentration of salicylic acid in the leaves of oil palm seedlings. Abstract

The research aims to observe the effect of some formulation of bio-fertilizer containing Bacillus sp. endophytes and to obtain the best formulation to increase the growth and production of tomatoes plant. The research has been conducted at the experimental farm, Faculty of Agriculture, University of Riau in March to August 2016. The research was arranged in a completely randomized design (CRD) and consisted of 5 treatments : Bs0 (Bacillus sp. without formulation), BsAk (Bacillus sp.+Coconut Water+Rice hulle Ash 1%+10% molasses), BsLs (Bacillus sp.+Palm Oil Mill Effluent+Rice hulle Ash 1%+10% molasses), BsLt (Bacillus sp.+Tofu Mill Effluent+Rice hulle Ash 1%+10% molasses) BsUs (Bacillus sp.+Cow Urine+Rice hulle Ash 1%+10% molasses). Data were analyzed statistically by analysis of variance and continued with DNMRT at 5% confident level. The result of the research showed that the formulation of Bacillus sp. was significantly affected on the plant height, fruit weight per fruit and fresh cenopy plant weight, but not significantly effected on stem diameter, flowering age, harvesting age, the amount of fruits, fruits diameter, fruits weight per plant and roots volume. Formulation of Bacillus sp. with coconut water is the best formulation in increasing the growth of tomatoes plant while the formulation of Bacillus sp. with cow urine is the best formulation in increasing the production of tomatoes plant.Keywords: Biofertilizer, Bacillus sp., tomatoes, liquid waste.

Abstract

This study aims to determine the effect and get the best microwave radiation period to control the pathogenic fungus of rice seeds while maintaining viability and seed vigor. This research was conducted in Plant Disease Laboratory, Plant Ecophysical Laboratory of Agriculture Faculty and Synthesis Organic Laboratory Faculty of Mathematics and Natural Sciences University of Riau, Pekanbaru in February 2018. The study was using complete randomized design (RAL). The parameters observed were kind of seed born pathogenic fungus, incidence of seed born pathogenic fungus, seed viability and vigor through seed germination test, first count test and index value test. The detection results indicate that the isolate of the suspected pathogenic fungus of rice seed was Rhizoctonia sp. (isolate PK-1 and PK-7), Pyricularia sp. (isolate PK-3), Curvularia sp. (isolate PK-5 and PK-9) and unidentified fungus (isolate PK-2, PK-4, PK-6 and PK-8). The total incidence of fungi from rice seed on 60 seconds of radiation effectively reduced the incidence percentage to 38% lower than the duration of other radiation and without radiation. Radiation treatment for 15 seconds was the best treatment obtained because it was able to suppress the incidence percentage by 84.67% with a 92% seed germination, 49.67% first count sprout and 1.5%/etmal speed sprout.Keywords: microwaves,radiation period, rice seeds, seed born pathogenic fungus, seed germination

Abstract

Kabupaten Kampar merupakan satu diantara beberapa sentra penanaman padi di Provinsi Riau. Kendala dalam melaksanakan budidaya tanaman padi yang masih ditemukan diantaranya adalah kurangnya pengetahuan petani tentang teknik budidaya dan pengendalian penyakit pada tanaman padi. Tujuan kegiatan ini ialah untuk mendorong petani agar dapat membuat biofungisida berbahan aktif konsorsium Trichoderma virens dan Pseudomonad berflourescens dalam formulasi Biofungisida Plus untuk pencegahan dan pengendalian penyakit yang menyerang tanaman padi. Kegiatan pemberdayaan petani di Desa Sawah melalui penerapan teknologi pengendalian penyakit tanaman padi dengan biofungisida plus dilaksanakan dengan menerapkan Model Community Development. Pada metode ini secara langsung dilibatkan sebagai subyek dan obyek dalam pelaksanaan kegiatan. Hasil yang diperoleh dari kegiatan pemberdayaan masyarakat ini diantaranya ialah, masyarakat bersedia untuk menerapkan teknologi Biofungisida Plus di lahan sawahnya. Berdasarkan hasil kuisioner juga diketahui pemahaman dan wawasan petani bertambah setelah diperkenalkan dengan Teknologi Biofungisida Plus ini. Aplikasi Biofungisida Plus di lahan petani menunjukkan kondisi tanaman yang lebih baik dibandingkan dengan teknik budidaya yang biasa digunakan oleh petani.

Abstract

Endophytic bacteria are bacteria that live in plant tissue, including in dragon fruit plant tissue. There are many species of endophytic bacteria living inside tissue dragon fruit plant wich play a beneficial role for plant. Endophytic bacteria can be used as a biological fungicide and biological fertilizer which are useful for plants. The purpose of the research is to observe the characteristics of the endophytic bacteria. Plants samples was taken by using purposive sampling method. The location is at Balai Pelatihan dan Pengembangan Masyarakat PT. Arara Abadi the district of Siak Riau. Plant tissues were taken from roots and shoots of the dragon fruit plants. The data were analyzed descriptively. Results showed that the morphological characterization of endophytic bacteria such as colonies color, colonies form and colonies elevation had some similarities each others, but there was a difference on the edges of colonymorphology. Two isolates had a flat edges and three isolates have wavy edges. Physiologycal assays such as catalase assay, oxidase assay and starch hydrolysis assay of the bacteria showed a positive on the each isolate, but there was a difference on the motility assay and indole assay. Two isolates showed that they were motile and positive indole assay,while the three other isolates showed that they were non-motile and indol negative assay.Based on the morphological and physiological similarities of the endophytic bacteria, it can be assumed the bacteria belong to Genera of Bacillus and  Corynebacterium. Keyword :      Isolation and characterization, endophytic bacteria, and dragon fruit plant

Abstract

White root fungus (Rigidoporus microporus) is a major pathogen in the rubber plant that is difficult to control because it has a structure that survives in plant tissue.  Biological control of endophytic fungi is very potential to control  Mushrooms JAP.  This research was conducted at Plant Disease Laboratory of Agriculture Faculty of Riau University and Photomikografi Laboratory, FMIPA Organic Chemistry Laboratory Department of Biology, University of Riau, Simpang Baru Campus Tampan District, Pekanbaru.  Starting from March to June 2017 with the aim of Identification of morphological cultivation of endophytic fungi and In-Vitro microbial test on Rigidoporus microporus in rubber plant.  The study consisted of site survey, sampling, tool sterilization, PDA & PDB media manufacture, fungi isolation, rubber plant tissue incubation, purification and observation. Based on the results of isolation and identification of endophytic fungi from plant tissue obtained 3 isolates where there are 2 types of Aspergilus SP fungus and 1 type of Trichoderma sp.  All three isolates are potential endophytic fungi to control JAP disease in rubber. Hypovirulensi test results did not produce symptoms of chlorosis or necrosis after inoculated Trichoderma 48 hours after incubation. Observation of secondary metabolite Trichoderma sp endophytes after 14 days incubated in a petridish showed metabolite results with a mean of 0.75 cm clear zone. Keywords: Rigidoporus microporus, Identification, Endophytic Fungi

Abstract

Development of cocoa (Theobroma cacao L.) requires high-quality seeds. Adding growth substance from Bacillus sp. endophytes is expected to increase the growth of cocoa seedlings. The purpose of this research was to evaluate the ability of Bacillus sp. endophytes to produce Indole Acetic Acid (IAA) hormone, evaluate the effects of Bacillus sp. endophytes application on growth of cocoa seedling, and obtain the best concentration to increase growth of cocoa seedling. The experiments were conducted in the laboratory and in the field from June to October 2015. The study was conducted in two stages: in vitro and in vivo.The first experiment tested the ability of Bacillus sp. endophytes to produce IAA using tryptophan enriched picovskaya liquid medium and non-enriched tryptophan. The results from the first experiment showed that all Bacillus sp. endophytes produce IAA hormones. The second experiment tested the concentrations of Bacillus sp. endophytes to improve the growth of cocoa seedlings. The experiment was conducted using a completely randomized design consisting of 5 treatments and 4 replications, i.e., without treatment of Bacillus sp., and concentrations of 1011, 1012, 1013, and 1014 cfu/mL. The parameters observed were the number of bacterial colonies of Bacillus sp. endophytes, cocoa seed height, stem diameter, number of leaves and planting area of 4 month cocoa seedlings. The results from the second experiment showed that all concentrations of Bacillus sp. endophytes increase the growth of cocoa seedlings. The concentration of 1011 cfu/mL produced more colony in planting medium, increased height, stem diameter, leaf number and leaf area in cocoa seedlings.

Abstract

Proteolytic bacteria are bacteria that are able to hydrolize proteins into smaller peptides or completely into amino acid units. This study was carried out to determine the proteolytic activity of Bacillus sp. B1 isolated from rhizosphere of mustard plants. The proteolytic activity of the enzymes were determined based on the amount of tyrosin liberated in Unit/mL, while the specific activity of the enzymes were indicated by activity per unit weight of proteins. The protein concentration of enzyme  extracts were determined by the Lowry method. The activity of enzyme was determined at various production times (6, 12, 18, 24, 30, and 36 hours). The results showed that the Bacillus sp. B1 isolate was able to produce protease. Bacillus sp. B1 isolate obtained its maximum activity in 30 hours of production time. The specific activity of protease Bacillus sp.
