Abstract

Study on isolation of catechin from gambier (Uncaria gambier (Hunter). Roxb) for pharmaceutical and cosmetic uses has been conducted.Raw materials of gambier were collected from three different areas including Andalas University Medicinal Plant Garden, Lima Puluh Kotaand Siguntur Pesisir Selatan, West Sumatera. Catechin was isolated from gambier using two methods, namely purification and pre nonpurification. Several evalutions on quality of the obtained catechin was done including descriptions of the catechins, solubility, colorreactions, thin layer chromatography, yield, drying shrinkage, melting point and maximum uptake levels of catechins. The best results forpharmaceutical and cosmetic uses are gambier originated from Siguntur with pre purification method which obtained 56.3% yield and 96.17%levels of catechins.

Abstract

The qualitative and quantitative analysis of coumarin in cinnamon (Cinnamomum burmanii Bl.) which are qualified in to AA, KA and KB has been done by gas chromatography and gas chromatography-mass spectrometry. Extraction of quality KB coumarin was carried out by soxchletation in hexane, dichloromethane and methanol. Meanwhile, quality AA and KA coumarins are soxchletated in methanol. The qualitative analysis using gas chromatography-mass spectrometry with column BP-5 showed that hexane, dichloromethane and methanol extracts of qualified KB coumarin, as well as methanol extracts of qualified AA and KB only contain coumarin compounds, no other coumarin groups were found. From coumarin quantitative analysis by gas chromatography using BP-624 column and FID detector of methanol extracts obtained 1855.51 ppm (0.186%), 1915.75 ppm (0.192%), 9177.36 ppm (0,918%), for qualified KB, AA, and KA respectively.

Abstract

Indonesia is one of the largest tea producer and exporter in the world. However, the European Union has issued Commission Regulation (EU) No.1146/2014 by setting the MRL (maximum residue limit) of anthraquinone in tea at 20 μg/kg, significantly limiting the amount of tea export from Indonesia to the European Union. Viewed from the side of the danger, especially for synthetic anthraquinone that is potential effect of human health then performed an analysis to determine the presence of anthraquinone in Indonesian tea. To find out anthraquinone concentrations in various tea products, anthraquinone was developed and validated analysis method using LC-MS/MS. anthraquinone was extracted from tea sample using dichlormethane with a soxhlet. Black tea and green tea samples were obtained from one of the tea plantation companies.The validated method was used for the determination of anthraquinone residues in black tea and green tea samples. Anthraquinone residues was detect in all of the test samples.

Abstract

Optimization studies out on isolation of catechin from gambier and pasta for the pharmaceutical and marker compounds have been carried. Gambier gambier and paste were obtained from the Medicinal Plant Garden Andalas University, Siguntur and Lima Puluh Kota. Isolation method used was non-purification method, pre-purification for gambier and fractination for pasta. The analysis performed included catechin solubility, melting point, maximum absorption, thin-layer chromatography, loss on drying, ash content, yield and determination of levels of catechin. Product which has the highest levels of catechins, the determination was continued by analysis of UV spectra and NMR. The best results for pharmaceuticals derived from Siguntur with pre-purification methods which resulted in yield 56.3% with 96.17% catechin content, whereas for a marker compound was obtained from gambier Siguntur pasta with fractionation method which yielded 12.13% with 97.96% of catechin level.

Abstract

Peroxidase enzyme activity of the introduced shallots with endophytic bacteria and resistant to bacterial leaf blight (Xanthomonas axonopodis pv. allii). Bacterial leaf blight caused by Xanthomonas axonopodis pv. allii is an important disease in shallots. We have earned six isolates of endophytic bacteria, which have the ability to induce systemically resistance to shallots. One mechanism in induce resistance in plants is a change in the plant defense enzyme activity such as peroxidase. The purpose of this study was to calculate the peroxidase enzyme activity of shallots crop is being introduced with endophytic bacteria and is able to induce resistance to Xanthomonas axonopodis pv. allii. This research was conducted by introducing six isolates of endophytic bacteria on shallot bulbs and planted in greenhouse. Shallots crop that was 14 days old then inoculated with the bacterium Xanthomonas axonopodis pv. allii and incubated until symptoms appear. Peroxidase enzyme activity was calculated on the roots and leaves of shallots are 0, 1, 2, 4, 6, 8, 10, 15, and 30 days after inoculation (dai). The results showed an increase in the peroxidase enzyme activity of shallots crop is being introduced by endophytic bacteria compared to the control. Isolates ULG1E2 (Serratia marcescens PPM4) was isolate with the highest peroxidase enzyme activity both in the roots and leaves are 0,051 ìm/ ml.

Abstract

Scope of pharmacy practices as well as the role of pharmacist have been improving recently. Pharmacists have responsibility for doing the pharmacy practices, including pharmacist it self and also the technicians. According to PP 51 of the year 2009 it was known that pharmaceutical practices have vary of choice of carreer-field. This present study was a descriptive study which was conducted by survey method using stratified random sampling. The respondents of this study were 1st, 2nd, 3rd and 4th grade students from Faculty of Pharmacy, Andalas University. The result showed that, in term of the profile of respondent, it was known that most of the students chose to work at industry of pharmacy of 48.2%, followed by hospital of  35.3%, University/Research Institute of 4.7%, pharmacy of 3.5%, pharmacy retail of 2.4% and others career-field (such as BPOM, civil servant at ministry of health and enterpreneur) up to 5.9%. Based on descriptive analysis, it was described that field of work was the most important criteria of affecting factors for pharmacy student in choosing their future career. The others important criterias those also afforded as the affecting factors were geographis, payment/income, flexibility of work-schedule, possibility of career-development and benefit, successively. As for result of independent sample t-test in term of gender, both of men and women chose same three main affecting factors in choosing career-field (including field of work, geographis as well as payment/income). The different result showed for 4th, 5th and 6th factors.

Abstract

The quality control of herbal medicine should be started from the determination of the active substance at harvest time. The Raman spectrometry has been used for this propose. The aim of this study is to determine the quantification of curcumin in turmeric rhizome (Curcuma longa Linn.) using Raman spectroscopy combined with multivariate analysis of PLS-R that are expected to provide reference method for quality control in turmeric rhizome, especially for raw materials of Herbal drugs. Parameters that can be used for analysis of curcumin levels on turmeric rhizome obtained using intensity of data Raman and the data obtained from the standard method will be processed with multivariate analysis methods PLS-R. The validation value of quantification result using Raman-PLSR is seen from R2 value of 0.957, RMSEC value of 0,199 and p-value of 0.00. The study showed the developed method could be implemented on to determine the prediction quantification of raw material herbal medicine.Keyword : Curcumin,  PLS-R, Turmeric, Raman Spectroscopy

Abstract

Odour and frequently visited plants by Bactocera sp. were screened for fruit fly attractants. The attractiveness of hexane extracts for B. dorsalis were determined by Y-tube method. Of 20 species treated, 5 species such as Cymbopogon nardus, Ocimum basillicum, Cymbopogon citratus, Cananga odorata, and Hyptis suaveolens showed the attractiveness and the presence of methyl eugenol (ME). These plants could be used as the source of ME for controlling fruit flies infestation. It was also found that Ruta graveolens (Rutaceae) revealed the absence of ME peak by GC-spectrum but responded to fruit flies B. carambolae. The results reveal that the chemical compounds existing in the extracts of R. graveolens may have great potential for development as attractants of the Oriental fruit fly. However, further isolation, identification, and evaluation of active compounds are needed in future studies.

Abstract

Characterization of Endophytic Bacillus Isolated from Shallot Root as Biocontrol of Bacterial Leaf Blight Disease. Endophytic Bacillus isolated from the roots of healthy shallots has potential as biological control agent of bacterial leaf blight (BLB) disease. Based on the in planta screening, four endophytic bacteria that capable of controlling BLB diseases from the group of Bacillus were obtained, such as Bacillus cereus P14, Bacillus cereus Se07, Bacillus sp. HI, and Bacillus sp. SJI. The aims of this study were to investigate the characteristics of endophytic Bacillus that capable of controlling BLB disease and to determine the characteristic that most responsible in the disease control. This study method was descriptive. The characteristics observed were production of antibiotics by paper disc method, production of salicylic acid by capillary electrophoresis, and root colonization by in planta method. Linear regression analysis was used to determine the characteristic most responsible in the disease control. The results showed that four endophytic Bacillus were capable of producing salicylic acid and colonizing the root, and three of them were also capable of producing antibiotic. Salicylic acid production varied ranging from 13.96 to 14.72 ppm mL-1. Three endophytic Bacillus were capable of producing antibiotic with inhibition zone of 16.25 to 20.25 mm. Endophytic Bacillus was able to colonize the shallot roots with a population of bacteria ranged from 3.20 × 105-6, 20 × 105 CFU g-1 root. Based on the correlation coefficient of linear regression analysis, the root colonization of endophytic bacteria played the role in decreasing BLB disease.
