Abstract

ABSTRAKDaun tenggulun (Protium javanicum Burm. F.) secara tradisional telah dimanfaatkan sebagai obat dan insektisida seperti repelan dan antimakan terhadap larva Epilachna sparsa suatu bioindikator universal terhadap hama ulat sehingga dapat dikembangkan untuk ulat lain seperti ulat kubis. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi golongan senyawa aktif antimakan daun tenggulun (Protium Javanicum Burm. F.) terhadap ulat kubis Plutella xylostella. Hasil maserasi 1000 g serbuk kering daun tenggulun dengan metanol menghasilkan 42,7218 g ekstrak kental metanol. Ekstrak ini menunjukkan aktivitas antimakan 71,01% pada konsentrasi 0,1% (b/v) yang selanjutnya dipisahkan dengan n-heksana, etil asetat, dan air. Ekstrak n-heksana, etil asetat, dan air pada konsentrasi 0,1% (b/v) menunjukkan aktivitas antimakan berturut-turut 30,56%, 84,24%, dan 5,05%. Pemurnian ekstrak aktif etil asetat dilakukan menggunakan kromatografi kolom dengan fase diam silika gel dan fase gerak etil asetat-n-heksana (1:1) menghasilkan tiga fraksi. Fraksi B yang menunjukkan aktivitas antimakan paling tinggi yaitu 87,26% pada konsentrasi 100 ppm dan relatif murni secara KLT. Hasil identifikasi menunjukkan isolat merupakan senyawa golongan triterpenoid alkohol dengan gugus fungsi OH, CH3, CH2, CH, C=O, -Vis pada 282,5 nm (???*), 403 nm (n??*), dan 437,5 nm (n??*).Kata kunci: Protium javanicum Burm. F.) have been used traditionally as a medicine and insecticide such as repellant and antifeedant for Epilachna sparsa larva a universal bioindicator to larva that can be developed for other larva such as cabbage larva. This research aimed to isolate and identify antifeedant active compounds from tenggulun leaf (Protium javanicum Burm. F.) to cabbage larva Plutella xylostella. Dry powder of tenggulun leaves extracted with methanol was obtained 42,7218 g of methanol crude extract. This extract showed 71,01% antifeedant activity at 0,1% (w/v) subsequently separated by partition to obtain n-hexane, ethyl acetate, and water extracts. Extract of n-hexane, ethyl acetate, and water at concentration 0,1% (w/v), showed the antifeedant activity of 30,56%, 84,24%, and 5,05%. Purification of etil acetat extract using silica gel column chromatography and mobile phase ethyl acetate-n-hexane (1:1) yielded three fractions. Fraction B is the most active which showed 87,26% antifeedant activity at 100 ppm was found relatively pure with thin layer chromatography. The antifeedant active isolate was identified as triterpenoid alcohol group of compounds which have functional group OH, CH3, CH2and gives UV-Vis rays at 282,5 nm (???*), 403 nm (???*), and 437,5 nm (n??*).Keywords: Protium javanicum Burm. Abstract

This research aimed to identify steroid compound from leaf of gayam (Inocarpus fagiferus Fosb) and test its antioxidant activity. Dried leaf powder of gayam was extracted with 5000 mL of 96% ethanol to yield 28,05 g concentrated extract. This extract was then fractionated into n-hexane and choloform to yield 14,18 g n-hexane and 2,21 g choloform fractions respectively. The test using Lieberman Burchard reagent showed that both fractions contain steroid, but n-hexane fraction contained more steroid than chloroform fraction. Separation of n-hexane fraction was done using silica gel column chromatography with n-hexane : ethanol (7:3) as eluent and four groups of fraction were obtained. Fraction F1 contained steroid and was relatively pure therefore it was then identified. The infrared and ultraviolet-visible spectra suggest that the isolate (F1 fraction) has functional groups such as O-H, C=O, C-H, and unconjugated C=C and showed lmax at 279,407, and 669 nm. The antioxidant test showed that the isolate had antioxidant activity toward DPPH with IC50 of 4 ppm.

Abstract

Calophyllum nagassarium Burm.f. or nagasari is a kind of plant that often be used as medical plant such as for antiseptic, laxative, blood purifier, helminthic, and tonic. This study aimed to isolate and identify cytotoxic compounds of nagasari leaves by extracting 1000 g of leaf powder with 10 L of 96% ethanol  (5L x 2) to produce 66.81 g of ethanol extract (LC50 = 19.06 ppm). Fractionation of the ethanol-water extract (7: 3) in a row with 300 mL of n-hexane, chloroform 400 mL, 400 mL of n-butanol, and water and toxicity tests against larvae of Artemia salina Leach generate 2.52 g of n-hexane extracts (LC50 = 275.42 ppm), 7.06 g chloroform extract (LC50 = 63.10 ppm), 9.54 g of n-butanol extract (LC50 = 27.54 ppm) and 25.38 g aqueous extract (LC50 = 478 , 63 ppm). N-buthanol extract which was the most toxic was then separated, purified and identified. The separation of 2.0 g of n-butanol extract using column chromatography techniques with silica gel 60 as the stationary phase and chloroform- n-butanol (5: 3) as the mobile phase resulted in 5 fractions (F1, F2, F3, F4, and F5). Fraction F1 which was the most toxic (LC50 = 91.20 ppm) was then separated by preparative Thin Layer Chromatography and resulted two fractions (FA and FB). Fraction FB was the most toxic with LC50 = 83.18 ppm. UV-Vis and IR spectrophotometric analysis of FB identified a compound suspected to be terpenoides that has the characteristics of functional groups such as O-H, O = C-OH, aliphatic C-H.

Abstract

The isolation and repellent activity of essential oil extracted from fruit of Vitex trifolia against Aedes aegypti has been done. Distillation of 15 kg samples produced 7,6 mL essential oils with 0,819 ± 0,05 g/mL of density and 0,0418% ± 0,01% (b/b) of yields. Identification with Gas Chromatography-Mass Spectrometry results showed that the essential oil composed of 16 compounds  and was a rich source of ?-Pinene, Sabinene, 1,8-Cineol, and ?-Charyophyllene. Varied concentrations of the essential oil of 7,5%, 15%, and 20% solution in 96% ethanol and DEET 15% as positive control were tested for their repellent activities against Aedes aegypti. The result of ANOVA test showed that there was significant difference in protection abilities amongst the essential oil solutions for 6 hour test. The result of Tukey/HSD test showed that 15% and 20% of essential oils solutions possesed repellent activity as strong as positive control at first test and the 20% of essential oils solution possesed repellent activity as strong as positive control at first hour. Based on the above results it can be concluded that the essential oil of Vitex trifolia fruit posseses repellent activity against Aedes aegypti.

Abstract

Isolation of antifeedant active compound from sarcocarp of Calophyllum inophyllum L. with  Epilachna sparsa larvae were use as bioindicator was conducted. The antifeedant assay was performed on crude extract, fraction, and isolate.  Dried sarsocarp powder of nyamplung was extraction with 6L metanol to yield 186,38 g dark brown extract.  This Active extract was then fractionated into n-hexane to yield 19,27 g yellow oil.. Separation of 2 g oil was done using silica gel column chromatography with chloroform n-hexane (2:1) as eluent and five group of fraction were obtained with F5 the most active isolat wich showed 27,89% antifeedant activity at 200 ppm was found and relatively pure . Base on analysis of Gas chromatography-mass spectroscopy and data base NISTO2.L the antifeedant active isolat were identify as methyl-14-methyl-pentadecanoid acid, n-hexanedecanoid acid, methyl-9,12-octadecadienoic, methyl-9-octadecenoic acid, methyl-octadecanoic, 9-octadecenoic acid, octadecanoic acid, and the last peak has a moleculer ion at 341.

Abstract

Preparation of ZnO-activated carbon composite with a mass ratio of ZnO : activated charcoal of 5:2; 5:1 and 15:2 have been carried out using the sol-gel method The characterisation of  ZnO-activated carbon composite was done by using  X-Ray Diffraction (X-RD) and Fourier Transform Infra Red (FT-IR). The characterization using X-RD showed that the ZnO has an hexagonal crystal structure  with crystal size of 23.13 nm; 23.38 nm and 40.25 nm, while FT-IR spectra showed that there were Zn-O strain, C-O aliphatic, C-H bending, C-C aliphatic  and O-H bending vibrations. ZnO-activated carbon composite was applicated as photocatalyst to degradate  methylene blue by irradiatting  the compound with UV light for 5 hours. The  results showed that the ZnO-activated carbon composite at the ratio of 5:1 at optimum pH of 11 gave the highest percentage of methylene blue degradation which was of 99,97±0,02 %.

Abstract

Tenggulun leaves (Protium javanicum Burm.F.) have been used traditionally as a medicine and insecticide. Activity test of essential oils from Tenggulun leaves as repellent against Aedes aegypti was done at concentrations of 7.5%, 15%, 20%, and 40% in 96% ethanol to determine its effectiveness. The essential oils were obtained by steam distillation methods, while the repellent activity test was conducted using Aedes aegypti adult females. Isolation of 12 kg fresh tenggulun leaves using steam distillation yielded 0.07% yellowish oils (13.7 mL) with the density of 0.8774 g/mL and sour sting smell. Gas chromatogram of the essential oils indicated that there were 13 compounds and identification of each compounds based on their mass spectra suggested that the compounds were ?-pinene (16.85%), myrcene (1.53%), ?-phellandrene (45.34%), p-cymene (5.60%), limonene (15.70%), ?-ocimen (0.34%), bicyclogermacrene (1.61%), ?-elemene (2.27%), ?-caryophyllene (7.90%), ?-humulene (0.88%), germacrene (1.50%), spathulenol (0.23%), caryophyllene oxide (0.24%). The repellent test result analyzed by One Way ANOVA indicated that at each concentration of the essential oils given showed significant differences during 6 hours treatment. Furthermore Tukey HSD test result showed that the essential oils at  concentration of 40%  revealed repellent activity which was comparable to positive control at the start of the test until the fifth hour of testing. Four of the compounds which are ?-pinene, myrcene, limonene and ?-caryophyllene have been known to have activity as repellent against mosquitoes.

Abstract

Activated clay can be used as an adsorbent to adsorb phosphate ions in solution. The aims of this research are to study the characteristics of the clay before and after activated (covering the surface acidity, surface area and basal spacing) and to study the adsorption capacity of activated clay to phosphate solution. The results showed, the activated clay gave surface acidity of 0.7202 mmol/g and 2.0018 mmol/g, surface area of 12.6602 m2/g dan 25.6101 m2/g, and d-spacing of montmorillonite was d (Å) = 0.12. The optimum condition for phosphate adsorption by this clay was found at 15 minutes contact time, pH 3, and  concentration of 70 ppm yielding phosphate adsorption capacity of 3.0349 mg/g following the Langmuir isotherm.

Abstract

Isolation and identification of active antifeedant compounds from gadung tuber (Dioscorea hispida Dennst)has been carried out. As much as 12.03 g of concentrated methanol extract was resulted from 5.0 kg dry gadung tuberthat was macerated using methanol. Further, this extract was partitioned with n-hexane to obtain 2 concentratedextracts in n-hexane and methanol. The result of antifeedant test showed that the methanol extract was more activethan the n-hexane extract.Separations of the methanol extract using column chromatography (stationary phase : silica gel 60, mobilephase : methanol-chloroform 6:5) resulted four fractions. The most active fraction, FA , showed 2 spots on the TLC,so it was separated using preparative TLC which resulted in 2 fractions (FA.1 and FA.2). The antifeedant activity ofFA.1 fraction was higher than FA.2 fraction and it gave only 1 spot on the TLCThe result of identification using phytochemical test and spectrophotometer analysis suggested that FA.1fraction was alkaloid compound whose maximum wave length was 277.1 nm and it contained N-H (3425.3 cm-1), CHaliphatic (2927.7 and 2854.5 cm-1), C=O (1735.8 cm-1), C-N (1110.9 cm-1) and N-C=O (621.9 cm-1).

Abstract

Isolation, identification, and antibacterial activity examination have been conducted for triterpenoidscompounds of carica papaya L. Maseration of 500 g pawpaw seed powder in n-hexane gave 21.66 g extract whichreacts with Liebermann-Buched reagent resulting in colour change (yellow to purple). Column chromatographyseparation of the extract resulted in 0.05 g yellow isolate (F3) which contains aldehyde triterpenoids. Infrared spectraindicated aliphatic –C-H stretching, -CH2 bending, -CH3 bending, and C=O. Ultra violet – visible spectra showedabsorption at 228.5 nm and proad peak at 287.7 nm. Antibacterial axamination showed that 1000 ppm at the isolateinhibits the growth of Escherichia coli and Staphylococcus auerus.
