Abstract

Telah dilakukan penelitian tentang kultur tunas Scoparia dulcis, Lindernia anagalis, Lindernia ciliata untuk mengetahui kemampuan penyerapan logam berat Pb, Cr dan Cd sebagai proses bioremediasi terhadap polusi logam berat.Tunas-tunas tersebut ditumbuhkan pada media Murashige-Skoog (MS) padat yang ditambah kinetin 2 ppm. Subkultur dilakukan terhadap semua jenis tunas pada umur 21 hari  kemudian dikultivasi pada media MS padat ditambah logam berat 0, 2 dan 5 ppm selama 30 hari. Semua tunas mampu menyerap Cr tetapi tidak mampu menyerap Cd, tunas Lindernia anagalis dan Lindernia ciliata mampu menyerap Pb.

Abstract

One of the natural materials as potentially efficacious chemopreventive agents are Ciplukan (Physalis angulata L.). Several previous studies reported that Physalis angulata L. herbs ethanolic extract (PEE) has cytotoxic activity and induction of apoptosis in breast cancer cells MCF-7 and HeLa cervical cancer cells. This study aims to determine the effects of  PEE as an chemopreventive agent on rat liver cells induced 7,12-dimetilbenz[a]anthracene (DMBA). This study used Sprague Dawley strain female rats aged 40-50 days were divided into 5 groups : (1) DMBA control group, mice were induced with DMBA in per oral dose of 20 mg/kg; (2) DMBA + PEE dose 750 mg/kgBW group ; (3) DMBA + PEE dose 1500 mg/kgBW group ; (4) solvent control group of CMC-Na 0,5%; (5) PEE dose 1500 mg/kgBW control group. PEE was dissolved in CMC-Na 0,5% and administered daily, starting the seventh week after administration of DMBA. At the beginning of  the tenth week of the study, rats were necropted and liver organs were isolated and stored in buffered formalin 4%. Qualitative analysis to determine the histopathology of liver cells through staining method of Hematoxyllin Eosin (HE), while quantitative analysis to determine the level of liver cell proliferation by AgNOR staining method. The results showed in the DMBA control group that liver cell morphology changes that hiperproliferation leading to carcinogenesis. In DMBA + PEE dose of  1500 mg/kgBW group improved the situation of DMBA-induced liver cells histopathology and antiproliferation activity better than DMBA + PEE dose of 750 mg/kgBW on DMBA-induced rat liver cells. The results showed that Physalis angulata L. herbs ethanolic extract can inhibit cell proliferation in rat liver caused by DMBA administration through antiproliferation mechanism and have potential for the development as chemoprevention material on liver cancer

Abstract

The research about shoot culture of Scoparia dulcis, Lindernia anagalis, Lindernia ciliata has beeen done. The aim of this research is to know ability of shoot culture of Scoparia dulcis, Lindernia anagalis, Lindernia ciliata to adsorb heavy-metal Pb, Cr and Cd as bioremediation process for heavy metal pollutant. Scoparia dulcis, Lindernia anagalis, and Lindernia ciliata shoots are inoculated in Murashige-Skoog (MS) semi-solid medium with added 2 ppm of kinetin as growth factor. Shoots are subcultured at 21 days then they are cultivated in MS semi-solid medium with added heavy-metal Pb, Cr and Cd during 30 days. Scoparia dulcis, Lindernia anagalis, Lindernia ciliata able to adsorb Cr but they can not adsorb Cd. Scoparia dulcis is not able to adsorb Pb.

Abstract

Active chemical compounds from medicinal plants are mostly secondary metabolites that can be histochemically tested. It enables the identification and localization of diverse secondary metabolites within the tissue, which is not possible with screening phytochemicals. In this study, the histochemistry of the leaf blades of Ericales plants i.e., cantigi (Vaccinium varingiaefolium (Blume) Miq.), persimmon (Diospyros kaki Thunb.), sapodilla (Manilkara zapota (L.) van Royen), and star apple (Chrysophyllum cainito L.) were evaluated. These species have been reported as folk medicines that have secondary metabolites. Each leaf tissue was observed under a microscope, and phytochemical compounds presence and location were detected by using specific reagents. The results showed that those four leaves contained phenols, flavonoids, and terpenes, while quinones were only found in cantigi leaves. The secondary metabolites were present in the idioblast of parenchymal and epidermal tissues. The persimmon and star apple leaves had secretory cavities in trichomes with terpenes and phenolic compounds.

Abstract

Cumin leaf are medicinal plants that are used as treatment and useful for health. Cumin leaf contain secondary metabolites, one of which is flavonoids. Flavonoids are the largest phenolic compounds found in plants and function as antioxidant, antibacterial, antiviral, anti-inflammatory, antiallergic, and anticancer activities. The flavonoid content of a plant can be affected by the drying temperature in the manufacture of simplicia. The purpose of this study was to determine the total flavonoid content in cumin leaf if dried by several drying methods. In this study, cumin leaf simplicia was used with different drying methods, namely sun drying, wind drying, and oven drying. The simplicia powder was extracted using ethanol and then tested quantitatively using a UV-Visible Spectrophotometer at a wavelength of 440 nm. Total flavonoid content by sun drying, oven drying, and wind drying was 3,38 mg QE/g extract, 3,58 mg QE/g extract, and 5,83 mg QE/g extract respectively. Based on the research on the flavonoid test on cumin leaves with different drying methods, the lowest average total flavonoid content was obtained on Sun drying was 3.38 mg QE/g extract and the highest was wind drying with 5.83 mg QE/g extract while oven drying was 3.58 mg QE/g extract.
