Abstract

Sisik naga (Drymoglossum piloselloides) has been used empirically for dysentry and candidiasis. This diseases were caused by bacteria and fungi. The objective of this research was to evaluate the antibacterial effect and to find out the antimicrobial compound of sisik naga. Leaf of sisik naga was extracted with ethanol 90% by using maseration. This extract were tested for antibacterial activity against Streptococcus mutans using agar difussion. TLC Bioautography methods was used to identify the active compound. Result of antibacterial test showedf ethanolic extract of sisik naga had antibacterial effect against S. mutans. Bioautography showed one spot on Rf =0 give clear zone.

Abstract

Aggressive Periodontitis is a form of periodontitis which is characterized with the presence of deep pocket, rapid loss of connective tissue attachment and alveolar bone. The rapid damage is caused by the dysfunction chemotaxis and phagocytosis from neutrophil.Calprotectin is a major cytosolic protein of neutrophils, monocytes and epithelial cells. The calprotectin level in gingival crevicular fluid is useful marker for evaluation the progression of inflammation in periodontitis disease. In this study, we investigated calprotectin level at different periodontal pocket depth from patient with aggressive periodontitis. This research involved twelve subjects who had been diagnosed as suffered from aggressive periodontitis. Their gingival crevicular fluid was taken from different periodontal pocket depths (slight ≤3mm, moderate 4-6mm, severe ≥ 7 mm), using sterile papperpoints. Level of calprotectin in gingival crevicular fluid were analysed by enzyme-linked immunosorbent assay (ELISA) procedures. The result showed that gingival crevicular fluids calprotectin level increased approximately in proportion to probing depth slight, moderate, and severe (p< 0,05), and showed a significant positive correlation with probing depth (R=0,776, p< 0,05). This research concluded that the calprotectin level in gingival crevicular fluid is positively related to clinical indicators (probing depth) and reflect the degree of gingival inflamation in patient with aggressive periodontitis

Abstract

The CPITN is a screening procedure for identifying actual and potential problems posed by periodontal disease. The aim of this study was to know periodontal status and periodontal treatment needs from patient that come to RSGM department periodontia faculty of dentistry Jember University in the year 2011. The study population comprised 191 adolescences (10 – 59 years). An observational study was conducted at RSGM of Jember University. The results of the study showed that individuals with a health periodontium is 1.05 %, score (1) with bleeding on probing is 3.14 %. Periodontal pocket (4-5mm) was recorded 17.27 %. The highest periodontal score is scores 2 (calculus) 78.53% of the population need dental scaling. We conclude that prevalence periodontal disease in patients that come to periodontics department is very high and periodontal treatment need with scaling.

Abstract

Porphyromonas gingivalis is a gram-negative anaerobic bacteria and has been shown previously to be one of the major pathogens in aggressive periodontitis. Outer membrane protein is the major virulence factor of P. gingivalis and plays role in the host immune response impair againts P. gingivalis, which in turns, causing tissue destruction and bone resorption. This study was aimed to investigate the isolation and characteristic outer membrane protein of P. gingivalis. Protein of OMP P. gingivalis ATCC 33277 was isolated using sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and than continued by hemaglutination test. The result showed that protein profil in SDSPAGE of OMP protein was 40 kDa molecular weight dan hemaglutination test was positive in titer of hemaglutination 1/8.In conclusion, outer membrane protein molecular mass of 40kDa produced by P. gingivalis is a key virulence factor involved in the co-aggregation activity of P. gingivalis

Abstract

This study was an experimental laboratory. The purpose of this study was to determine the role of probiotics in inhibitingthe activity of pathogenic bacterial oral cavity. The result of study is expected to be used for prevention and treatment ofperiodontal disease which is still complex and derived products with high quality probiotics. The main product is thedevelopment potential of probiotics in maintaining oral health, especially periodontal disease is effectively and efficiently.This study used rats and divided into 4 groups: Group I, control without treatment; group II, induction of LPS E. coli for 5days, Group III, LPS induction of E. coli + Probiotic Bacterial Lactobacillus casei injection for 5 days together, and groupIV, induction of LPS E. coli for 5 days + 5 days injection of Lactobacillus casei. Immunohistochemical examination wasdone to evaluate the activity of osteoclasts in alveolar bone resorption to detect TRAP (tartarate-resistant acidphosphatase) and Carboxyterminal Telopeptide of Type 1 Collagen (1CTP) and subsequently analyzed the data collectedstatistically. The results of this study showed that induction of LPS could trigger the occurrence of alveolar boneresorption mediated by osteoclast and Lactobacillus casei administration both at the same time with the LPS induction orafter LPS induction can inhibit osteoclastic alveolar bone resorption. In conclusion, clinically it can be interpreted that theadministration of probiotic can be given at the beginning of periodontal disease occurs before or at the time of periodontaldisease is underway.

Abstract

Oral health among elderly people needs a specially attention because it will affect their general health. A good knowledge of oral hygiene is essential to prevent oral disease. Therefor maintenance of oral hygiene was the best way to maintain the health condition of the elderly people. The purpose of this research was to determine the correlation between oral health knowledge of elderly people with oral health status in the working area ofPuskesmas Kalisat Jember. This study was observasional research on 80 elderly people aged 50-70 years old who came to 12 elderly posyandu of Puskesmas Kalisat Jember. Data were collected by structured questionnaires about oral health knowledge and clinical examination to determine oral hygiene status based on Oral Hygiene Index-simplified (OHIS). Pearson correlation was used for statistical analysis. Most of participant demontratedoral health knowledge score as sufficient category (52,5%) and the oral hygiene status was moderate (57,5%). The correlation between the level of oral health knowledge with OHI-S index was significant (p< 0,05). Among elderly people, the knowledge of oral health correlated with the oral hygiene.Keywords: elderly, dental health, Index OHI-S, Puskesmas Kalisat Jember

Abstract

Background: Probiotics are microbes derived from the group of lactic acid bacteria that work to maintain the health of hosts. Probiotics can also be used to improve oral health. Periodontal disease is usually marked with gingival inflammation and alveolar bone resorption. Gram negative anaerobic bacteria that play important role in human periodontal disease are Porphyromonas gingivalis. (P. gingivalis). P. gingivalis is a virulent bacteria in vivo or in vitro, and mostly found in subgingival plaque of periodontitis patients. Purpose: This study is aimed to know the role of probiotics to inhibit the resorption of alveolar bone induced with P. gingivalis. Methods: This study used male wistar rats divided into 4 groups. Group I was control group (without treatment); group II was induced with P. gingivalis ATCC 33277 for 5 days; group III was induced with P. gingivalis ATCC 33277 and also injected with probiotics (Lactobacillus casei ATCC 4224) for 5 days simultaneously; and group IV was induced with P. gingivalis ATCC 33277 for 5 days and also injected by probiotics (Lactobacillus casei ATCC 4224) in the next 5 days. After that, the samples were decapitated, taken their alveolar bone, and then were examined by immunohistochemistry to observe osteoclast activity in alveolar bone resorption by using tartrate-resistant acid phosphatase (TRAP) expression. All data were then analyzed statistically. Results: It is known that there were significant differences of TRAP expression among all those treatment groups (p < 0.05). Conclusion: It then can be concluded that probiotics can decrease osteoclast activity in periodontal tissue of wistar rats, so it can inhibit alveolar bone resorption.Latar belakang: Probiotik adalah mikroba dari golongan bakteri asam laktat yang bekerja mempertahankan kesehatan host dan probiotik dapat digunakan untuk meningkatkan kesehatan rongga mulut. Penyakit periodontal ditandai dengan adanya keradangan pada gingiva dan resobsi tulang alveolar. Bakteri Gram negatif anaerob yang sangat berperan dengan penyakit periodontal pada manusia adalah Porphyromonas gingivalis (P. gingivalis). P. gingivalis merupakan bakteri yang virulen, baik diuji secara in vivo maupun in vitro, dan banyak ditemukan pada plak subgingiva penderita periodontitis. Tujuan: Penelitian ini bertujuan untuk mengetahui peran probiotik dalam menghambat resorbsi tulang alveolar yang diinduksi P.gingivalis. Metode: Penelitian ini memakai tikus jenis wistar jantan sebagai sampel dan dibagi menjadi 4 kelompok: Kelompok I yaitu kontrol tanpa perlakuan; kelompok II di induksi P.gingivalis ATCC 33277 selama 5 hari; Kelompok III di induksi P.gingivalis ATCC 33277 ditambah suntikan probiotik (Lactobacillus casei ATCC 4224) selama 5 hari secara bersamaan; dan Kelompok IV di induksi P.gingivalis ATCC 33277 selama 5 hari ditambah suntikan probiotik (Lactobacillus casei ATCC 4224) 5 hari selanjutnya. Setelah itu sampel didekaputasi, diambil tulang alveolar dilakukan pemeriksaan imunohistokimia untuk melihat aktivitas osteoklas dalam resopsi tulang alveolar dengan mendeteksi tartrate-resistant acid phosphatase (TRAP). Data yang dikumpulkan dianalisis secara statistik. Hasil: Terdapat perbedaan yang bermakna antar kelompok untuk ekspresi TRAP (p < 0.05). Kesimpulan: Dapat disimpulkan bahwa probiotik dapat menurunkan aktivitas osteoklas pada wistar tikus yang mengalami periodontitis sehingga dapat mencegah resorbsi tulang alveol.

Abstract

Background: Calprotectin is a calcium-binding protein expressed by neutrophil, monocytes, gingival keratinocytes, and oral epithelial cells. The concentrations of calprotectin increase in plasma, urine and synovial fluid of patients with inflammatory diseases. This protein is known as a marker for periodontal diseases and is detected in gingival crevicular fluids. Purpose: This study was aimed to investigate the detection of inflammation on the aggressive periodontitis by calprotectin expression. Method: The gingival crevicular fluids were taken from five aggressive periodontitis patients and five healthy subjects by using sterile paper points. Calprotectin expression was analyzed by ELISA technique. Result: The results showed the significant difference in calprotectin expression between subject with aggressive periodontitis and healthy subjects p = 0.002 (p < 0.05). Conclusion: It was concluded that the calprotectin expression on the aggressive periodontitis patients may be useful for evaluation the progression of inflammation in periodontitis.

Abstract

Background: Demineralized dentin matrix (DDM) scaffold is a substitute material for the bone contained in human teeth. DDM is a scaffold-derived tooth dentine containing type I collagen and bone morphogenetic protein (BMP). While DDM possesses the ability to perform osteoinductive and osteoconductive roles, a cytotoxicity test of DDM scaffold remains extremely important in evaluating the level of toxicity of a material if cultured in cells. Adipose-derived mesenchymal stem cells (ADMSCs) are multipotent in nature because they contain progenitor cells and have the potential for differentiation via adipogenic, osteogenic and chondrogenic pathways. ADMSCs are also known to have high biocompatibility and the ability to combine with other bone material. Purpose: The purpose of this study was to determine the cytotoxicity and characteristics of DDM scaffolds derived from bovine teeth in the ADMSCs of rats cultured in vitro. Methods: This research constituted an experimental study. ADMSCs were isolated from the inguinal fat of rats. Thereafter, DDM was extracted from bovine teeth and formed 355-710 μm-sized particles. DDM scaffolds were assessed using SEM and the effects of DDM scaffolds on the cell viability of ADMSCs at concentrations of 10%, 50%, and 100% analyzed by means of 3-4,5’dimethylihiazol-2-yl,2.5-di-phenyl-tetrazolium bromide (MTT) assay. The results obtained were then analyzed by an ANOVA to establish the difference between the groups. Results: SEM results showed the diameter sizes of the dental tubulis DDM scaffolds to be approximately 4.429 μm and 7.519 μm. The highest cell viability (97.08%) was found by means of an MTT test to be in ADMSCs at a concentration of 10% compared to those at concentrations of 50% and 100%. Conclusion: In conclusion, DDM scaffold derived from bovine teeth with a particle size of 355-710 μm produces a low cytotoxicity effect on ADMSCs.
