Abstract

This research aimed to study the effect of kinds of culture media composition added with organic matter (soya bean extract, corn extract and fish emulsion) and PGR concentration to the growth of each cross breed orchid explants. Research was conducted in Plant Physiology and Biotechnology Laboratory, Agriculture Faculty, Sebelas Maret University of Surakarta from March 2009 until the end. This research was done in Randomized Completely Design with two factors of treatment and four replications. The first factor was media and organic matter, consist of 3 levels: soya bean extract, corn extract and fish emulsion. The second factor was atonik concentration, consist of 3 levels: 0 cc/l, 0.5 cc/l and 1 cc/l. The research concluded that for the explants of crossing (Phalaeonopsis pinlong cinderela ♀ >< Phalaeonopsis joanekileup June” ♂) two treatment of corn extract affected significantly to the root emergence time (21.33 ) and member of root (1,92 day after planting) The treatment of fish emulsion affected significantly to the root length (2,82 cm), number of root (1,92) and produced the longest leaf. The treatment of Atonik increased the number of leaf and root.

Abstract

Character was very important cytological nature orchids studied to support the success of breeding orchids. The purpose of the research to study the cytological characters orchid number, size) and chromosome karyotype in order to conserve germplasm. The study was conducted at the Laboratory of Biotechnology Faculty of Agriculture University Sebelas Maret Surakarta. Materials research Paphiophedilum glaucophylum, Coelogyne spesiosa, Dendrobium crumenantum, Dendrobium mutabile,, Bulbophyllum. Blumei dan Bulbophyllum beflorum. The method uses squashing methods include : fixation : Carnoy solution of 2 (6 ethanol : 3 chloroform : 1 glacial acetic acid 45 %), hydrolysis (1 N HCl) and staining chromosomes (aceto - orcein solution 2 %). Observations (number, size, shape) chromosomes with a light microscope. The results showed that the studied semuia orchids have the same chromosome number is 2n = 38, medium size chromosomes Paphiopedilum glaucophyllum 4.26 ± 0.14 to 9.73 ± 0.19, Coelogyne spesiosa 0.35 ± 0.03 to 6.30 ± 0.84, Dendrobium crumenantum 2.46 ± 0.20 until 5.33 ± 0.02 Dendrobium mutabile 2.56 ± 0.72 until 6.54 ± 0.73, Bulbophyllum blume 2.76 ± 0.09 until 6.07 ± 0.43 and Bulbophyllum beflorum 1.04 ± 0.07 sampai 1.35 ± 0.16. Karyotype patterns all orchids studied 2n = 38 m except Coelogyne spesiosa 2n = 37 m+ 1 ak and Dendrobium mutabile 2n = 37 m+ 1 sm.

Abstract

Efforts to improve the genetic quality of orchids have constraints on seed propagation techniques crosses and from crosses. The study aims to: (1) obtain a cross technique that can produce seeds with high fertility rates, (2) to obtain a new hybrid character which has advantages in flower. The study was conducted at the Center for Plant Conservation Kebun Raya Bogor LIPI. Research was an interspecific crosses that ♀ Vanda celebica x ♂ Vanda tricolor, ♀ Vanda celebica x ♂ Vanda dearei, ♀ Vanda celebica x ♂ Vanda insignis. And crosses the reciprocal is ♀ Vanda tricolor x ♂ Vanda celebica, ♀ Vanda dearei x ♂Vanda celebica, ♀ Vanda insignis x ♂Vanda celebica. Crosses performed by crossing parent elected as male or female parent. Activities include characterization of elders, crosses, harvesting, seed dispersal, seed Deployment and transfer done aseptically in tissue culture laboratory. Observations included the successful crossing, when formed fruit, ripe fruit, and formed protokorm. Of research produced 5 series hybrid to form protokorm is Vanda dearei x Vanda Celebica, Vanda celebica x Vanda dearei, Vanda insignis x Vanda celebica, Vanda celebica x Vanda insignis, Vanda celebica x Vanda tricolo. The success of crosses 33%- 100%, time required for seed maturation varies between 122-262 days, 16-23 days old germinated.

Abstract

Research purposes to determine the response of soybean varieties Kaba under protected forest tree stands besides soybean crop response to P fertilizer and manure fertilizer micro coupled with organic fertilizer uptake and utilization of P element to the low light under protected forest tree stands. The research was conducted in JulyDecember 2011. Experiments using Randomized Complete (RAKL) factorial consists of 3 factors, namely fertilization with manure P 1, factor 2 micro fertilization with manure, and a factor of 3 fertilizing with organic fertilizers. Each treatment combination was repeated 3 kali.Faktor first fertilizer P fertilizer P The first factor consists of three levels, namely P1 (50 kg / ha), P2 (75 kg / ha), and P3 (100 kg / ha). The second factor consists of 2 fertilizer micro level, ie M1 (0.5 l / ha) and M2 (1.5 l / ha). The third factor of organic fertilizer consisting of 2 levels, ie O0 (0 tonnes / ha) and O1 (5 tonnes / ha). Data analysis using the F test level of 5% followed by comparison test (DMRT) level of 5%. The results show the interaction between P fertilizer, micro fertilizer and organic fertilizer effect on soil pH, P uptake, and the number of pods. P fertilizer on fertilizer and organic fertilizer micro effects and no significant effect on available P and heavy pod soybeans (Glycine max L.).

Abstract

The research aimed to study the influence of the application of NAA concentration, the addition of organic materials and a combination of both on media Knudson C towards the growth of orchid plantlet (♀) Dendrobium liniale >< (♂) Dendrobium biggibum. The purpose of using media Knudson C in culture in vitro is to optimize the growth of orchid plantlet Dendrobium liniale and Dendrobium biggibum. The experiments were carried out in the Laboratory of Plant Physiology and Biotechnology Faculty of Agriculture, Sebelas Maret University Surakarta. The research used Complete Randomized Design (CRD) with 2 treatment factors, factor I was the concentration of NAA (0 ppm, 1 ppm, 3 ppm and 5 ppm); and factor II was the organic materials (coconut water, banana, potatoes and sweet potatoes). In total there were 20 treatment combinations . Each combination was repeated four times. The results showed that the combination of sweet potato and NAA 5 ppm produced the following characteristics: the fastest root growth, most root length, and the largest number of leaves; while the combination of banana extracts and NAA 5 ppm produced a number of roots, the shoots appear most rapidly, and the highest number of shootsKeywords: genetic diversity; tissue culture; medium;

Abstract

Biduri (Calotropis gigantea) was a wild plant that had many benefits in the health and textile industries. In the health sector, biduri was used as a herbal plant, while in the textile industry, used as the main raw material. Biduri could be used as a substitute for kapok as the main raw material, therefore it was necessary to develop cultivation technology to met the needs. This research was conducted at the Jumantono Field Laboratory of Faculty of Agriculture UNS, Karanganyar using a factorial Randomized Complete Block Design (RCBD), which consisted of 2 factors, namely cow dung fertilizer, and mycorrhizae. Cow dung fertilizer consists of 0 ton/ha, 10 ton/ha, and 20 ton/ha. Mycorrhizae consisted of 0 grams/plant, 5 grams/plant, 10gram/plant, and 15gram/plant. There were 36 treatment combination that consist of 12 treatment combination repeated 3 times. The interaction between cow dung did not significantly affect all physiological observation variables. The results showed that the application of 20 ton/ha of cow dung fertilizer gave the best results on the width of stomata openings, the rate of photosynthesis, and the rate of transpiration. Abstract

The phalaenopsis hybrid has become an economically important flower in the horticultural sector. Phalaenopsis Fuller's Pink Stripe Orchid is a hybrid orchid resulting from a cross between Phalaenopsis Fuller's Pink Swallow and Phalaenopsis Fullers Black Stripe. Rice washing water (leri) is a residue that accumulates when washing rice. This residue is usually thrown away when it is actually rich in various organic and mineral compounds. Waste that is wasted in vain can be used as nutrients for the growth of orchid plants. This research was conducted to study the effect of dose and frequency of leri water spraying on the growth of Phalaenopsis Fuller's Pink Stripe orchid at the vegetative stage. This study consisted of 10 treatments, namely giving 5 ml of water every 2 days (L1), 10 ml every 2 days (L2), 15 ml every 2 days (L3), 5 ml every 4 days (L4). a dose of 10 ml every 4 days (L5), a dose of 15 ml every 4 days (L6), a dose of 5 ml every 6 days (L7), a dose of 10 ml every 6 days (L8), a dose of 15 ml every 6 days (L9), and control treatment (L0). Each treatment was repeated 3 times. Based on the results of the study, it are often all over that there's no important interaction between the dose and also the frequency of administration of Leri water on the growth and performance of the Phalaenopsis Fuller's Pink Stripe.Keywords: Rice Wash Water, Phalaenopsis Fuller's Pink Stripe, vegetative growth
