Abstract

This experiment was performed to examine the effects of black cumin (Nigella sativa) seed oil extract onreproduction performance and fetus development if given during organogenesis period. Forty pregnantmice were randomly divided into four groups. Extract was treated orally by gavage with doses 0%(control), 0.1 ml, 0.2 ml, and 0.3 ml since gestation day 6 to 15. Caesarean section were performed topregnant mice on day 18 to remove fetuses. Observation covered reproduction performance i.e. the foetusnumber (reabsorptions, dead/ alive foetus), and morphological of the foetus (weight and length of thelitter, and malformations). Statistical analysis was performed using Anova and Duncans Multiple RangeTest. Teratogenic effects of black cumin oil extract caused decreasing of reproductive performance of themother (alive foetus decreased, dead and resorbed foetus increased) and an increased incidence of birthdefects (fetal weight and length and fetal hemorrhage).Keywords: Nigella sativa, congenital malformations

Abstract

Anjing kintamani bali (AKB) adalah kelompok anjing lokal jenis pegunungan yang hidup di sekitar Desa Sukawana, Kecamatan Kintamani, Kabupaten Bangli, Bali. Anjing kintamani bali merupakan plasma nutfah asli Indonesia yang sangat perlu dijaga kelestarian dan kemurniannya. Peripheral blood mononuclear cell (PBMC) sebagai komponen penting sistem kekebalan tubuh yang terlibat dalam imunitas humoral dan seluler, dapat digunakan untuk memonitor perkembangan penyakit atau respon terhadap pengobatan serta menemukan biomarker yang valid dalam kompartemen sel darah. Penelitian ini bertujuan untuk mengetahui ukuran sel limfosit dan sel monosit pada AKB sehat yang dievaluasi berdasarkan pemeriksaan cytomorphometry. Penelitian ini merupakan penelitian eksploratif menggunakan Rancangan Acak Lengkap (RAL) pola faktorial AxB. Data dianalisis dengan analisis ragam dan disajikan dalam bentuk tabel, grafik, dan gambar. Berdasarkan hasil penelitian, AKB memiliki nilai cytomorphometry yaitu diameter sel limfosit (7,312-8,761µm), diameter nukleus limfosit (5,786-7,138 µm), diameter sel monosit (7,785-8,883 µm), diameter nukleus monosit (6,099-7,457 µm), circumference sel limfosit (23,131-27,737 µm), circumference nukleus limfosit (18,160-22,402 µm), circumference sel monosit (24,514-27,966 µm), circumference nukleus monosit (19,093-23,442 µm), surface area sel limfosit (42,318-60,140 µm2), surface area nukleus limfosit (27,331-40,308 µm2), surface area sel monosit (48,313-62,334 µm2), surface area nukleus monosit(28,979-43,736 µm2), luas sitoplasma sel limfosit (8,863-27,509 µm2), dan luas sitoplasma sel monosit (12,230-25,953 µm2).

Abstract

Calliandra calothyrsus Meissn. leaf extract is potential as phytoestrogens. It influenced male mice reproduction, rat estrous cycle, and ovarian histology in previous study. This research aimed to prove the C. calothyrsus leaf extract potential as phytoestrogen source and the effect on endometrial thickness where the embryo implantation take place in early pregnancy. This study used a Completely Randomized Design used 54 days old female rats (Rattus norvegicus). Rats were divided into K as control group (treated with 0.5% Na-CMC as placebo) and P1, P2, and P3 as three groups with C. calothyrsus leaf extract administration with doses of 17.5; 35; and 70 mg/ kg bw respectively. Treatments were given 1 ml/rat/day orally for 20 days. At day 21st, animals were euthanized to collect blood samples for estrogen hormone analysis. After the dissection, all uterus were collected and weighed. Histological preparation was done with paraffin method and Hematoxylin-Eosin staining. The effective dose was 70 mg/ kg bw that did not decrease the weight of the uterus and the body. This dose even maintained the normal diameter and thickness of uterine walls (endometrium, myometrium, and perimetrium layers) like control rats. The extract in this study could increase estrogen concentration in female rats. This research novelty is that C. calothyrsus leaf extract (70 mg/ kg bw) can be used as an alternative herbal suplement to maintain uterine wall thickness and estrogen concentration in productive women. With further clinical research, this extract is a good candidate as potential estrogen source to overcome women infertility or pregnancy difficulties due to problem of endometrial thickness and lack of endogenous estrogen.
