Abstract

Sea cucumbers are one of Indonesia’s marine organism with the potential to be developed as possible herbal medicines. In a preliminary study of cytotoxic activity of ethanol extracts of 14Indonesian sea cucumber species, the most active extract came from the Holothuria sp. Thecurrent research aimed to identify the major cytotoxic component in the sea cucumber Holothuriasp., that yielded the most active extract. The samples were collected from South Lampung beach.Isolation of the cytotoxic component was done employing liquid flash and preparative reversedphase (C 18) chromatography. Cytotoxic ity evalu atio n of fractio ns collected durin g thechromatography was conducted using the MCF-7 tumor cell line. Elucidation of the structure ofmost active isolate was done by NMR (Nuclear Magnetic Resonance) spectroscopy and GC-FID(Gas-Chromatography-Flame Ionisation Detector) analysis. The results of these analyses showedthe most active compound to be stearic acid; IC 50towards MCF-7 cells 10.32 ppm.

Abstract

Teknik zymogram merupakan pengembangan dari teknik elektroforesis yang di dalam gel elektroforesis disisipkan substrat sehingga protein/enzim yang telah dipisahkan sekaligus dapat dipelajari kemampuannya dalam mendegradasi substrat tersebut. Teknik elektroforesis sendiri merupakan teknik yang biasa digunakan dalam bidang biokimia untuk memisahkan campuran yang berisi biomolekul seperti enzim sehingga enzim yang ada akan terpisah berdasarkan muatan dan berat molekulnya. Teknik zymogram telah luas digunakan, diantaranya  untuk deteksi jenis-jenis enzim yang disekresikan oleh mikroba dan mempelajari karakteristiknya. Dengan semakin berkembangannya peran protein/enzim dalam berbagai bidang kesehatan, teknik zymogram semakin populer dan diperluas aplikasinya.

Abstract

Indonesia has a great sea cucumber resource which is useful as functional food. Even though much information about the biopotency of sea cucumbers have been published, however, studies on nutrition value of Indonesian sea cucumbers are still beneficial due to the great diversity and uniqueness of each species. The present work was  intended to obtain information about chemical composition and fatty acid profile of 4 (four) types of sea cucumbers which are frequently found in Halmahera water, North Maluku (Molucca),  one  of  sea  cucumbers  producing area  in Indonesia. The samples coded as  H-03, H-04, H-05 and H-10, were identified as Bohadschia  argus, Holothuria  fuscogilva, Thelenota  ananas, and Actinophyga lecanora. All sea cucumbers showed high protein content, more than 60% (drybase/db), except for T. ananas which had the lowest protein content (48.26% db). As for fats, T. ananas showed the highest amount (2.35%db) with dominated saturated fatty acids (SFA). On the other side, A. lecanora showed higher value of PUFA compared to SFA and MUFA.  Palmitic acid (C16:0) was the most abundant SFA in the most samples with the amount of 0.49–4.9 mg/g sample. Among PUFA, eicosapentaenoic acid/EPA (C20:5n3) was detected, and eicosatrienoic acid (C20:3n6) was relatively higher than the others. The ratio of w6/w3 fatty acid was 1.2–3.2 showing that total w6 fatty acid was higher than total w3 fatty acid. However, this value is safe enough to protect against chronic and degenerative diseases.

Abstract

Spons merupakan hewan multiseluler yang paling sederhana sekaligus merupakan sumber senyawa bioaktif yang sangat berguna di dalam bidang farmasi. Senyawa-senyawa baru yang sangat beragam sudah banyak yang diisolasi dari spons. Kesulitan utama dalam memproduksi senyawa obat baru dari spons adalah kurang mencukupinya jumlah senyawa bioaktif yang dihasilkan spons untuk keperluan uji preklinis dan klinis karena rendemen yang dihasilkan sangat rendah. Artikel ini merupakan tinjauan terhadap metode kultur sel spons untuk memproduksi senyawa bioaktif  dalam rangka mengatasi masalah suplai senyawa bioaktif yang diuji.  Produksi senyawa avarol dari kultur primmorph spons Dysidea avara dibahas dalam review ini.  Avarol dilaporkan  memiliki bioaktivitas yang kuat sebagai substansi antitumor, antibakteri, dan antivirus. Model pendekatan melalui kultur primmorph dapat digunakan sebagai cara yang menjanjikan  untuk memproduksi senyawa bioaktif dari spons.

Abstract

This research was aimed to isolate marine derived-fungi which associated with macro algae and sponge from Indrayanti Beach, Jogjakarta and to screen the antitumor (T47D and HeLa) bioactivity of the fungi extracts. Three solid media of MEA (malt extract agar), GPY (glucose peptone yeast) and MFM (minimal fungi medium) were used as isolation medium. Each of the pure fungi isolates was then cultivated in 100 ml of liquid medium for 4 weeks at room temperature (27-28°C) in static conditions. The antitumor activity of the fungi extracts were tested against breast tumor cells (T47D) and cervical cancer cells (HeLa) using Thiazolyl Blue Tetrazolium Blue (MTT) assay method. A number of 21 isolates of fungi were isolated from 4 macro algae and 1 sponge samples. The identification of fungi isolate was conducted using combination of molecular approach (ITS1-5.8S-ITS4 DNA regions) and macro-micro morphological characteristics. Among those 21 marine fungi species isolated, MFGK-21 extract showed the best anti-servical tumor (HeLa) with an IC50 value of 240.1 µg/ml and MFGK-27 extract showed the best anti-breast tumor (T47D) with an IC50 value of 59.6 µg/ml. The MFGK-21 fungi isolate was identified as Penicillium steckii, while the MFGK-27 fungi isolate was identified as Aspergillus sydowii.
