Abstract

 The strain LBF-005 from marine bacteria have already isolated and screened for mannanase degrading enzyme in submerged fermentation process. This strain was further identified by using 16S rRNA showed that bacterium is belong to Bacillus subtilis that could produce mannanase with activity around 9.5 U/mL. The optimum pH and temperature for the activity of crude enzyme for mannanase was 6.0 and 50 oC. Cloning of mannanase gene from B. subtilis was conducted using six primers set designed based on the homology analysis conserve region several mannanase from bacteria (Bacillus sp.) glycosyl hydrolase (GH) family 26. Optimization of PCR conditions was performed by gradient PCR to obtained PCR product of b-mannanase gene. The PCR product was obtained by third primer combination and was estimated to be around 972-bp. Analysis of the nucleotide sequence showed that sequences has similarity with mananase gene from other Bacillus sp., such as the B. subtilis strain WLY-12, B. subtilis strain WL-8, B. subtilis strain CICC 10260, B.subtilis strain CD-25, B.subtilis strain G1, and Bacillus sp. SWU60 were about 98%, 98%, 98%, 98%, 97% and 95%, respectively. The next research will to obtain a whole gene encoding β-mannanase by using in vitro cloning method, characterization of recombinant mannanase and application this enzyme for mannooligosaccharides production.

Abstract

Poultry farming businesses such as UD Mahakarya Farm always have internal and external problems, so the desire to develop the business has not been achieved. UD Mahakarya Farm needs to do creative and innovative things with its business models. The purpose of this research is to determine the business model canvas and business strategy. Research methods using qualitative and quantitative. The results of the research show that business development with the Business Model Canvas is still using conventional models. The results of the SWOT analysis showed an IFAS value of 2.95 and an EFAS value of 3.12. In the SWOT analysis, there are eight alternative strategies that can be recommended for business development at UD. Mahakarya Farm.

Abstract

Penelitian ini bertujuan untuk menguji faktor-faktor yang berpengaruh terhadap kepuasan kerja karyawan dirumah makan ayam betutu Mbak Timah di Kecamatan Kalipuro Kabupaten Banyuwangi. Penelitian ini menggunakan tiga variabel independent yaitu kompensasi, lingkungan kerja dan motivasi. Populasi pada penelitian ini adalah semua karyawan rumah makan ayam betutu Mbak Timah, sampel yang diambil sebanyak 35 responden dengan teknik pengambilan sampel yaitu sampling jenuh. Analisis data yang digunakan adalah Analisis Regresi Linier Berganda. Analisis datanya meliputi uji validitas dan uji reliabilitas, uji asumsi klasik, analisis regresi linier berganda, pengujian hipotesis melalui uji T dan uji F, serta uji koefisien determinasi (R2). Hasil penelitian berdasarkan uji F menunjukkan bahwa nilai Fhitung = 5,515 > nilai Ftabel = 2,91 yang berarti variabel kompensasi dan variabel lingkungan kerja serta variabel motivasi secara simultan berpengaruh signifikan terhadap variabel kepuasan kerja karyawan. Hasil uji T menunjukkan bahwa variabel kompensasi berpengaruh signifikan dan memiliki pengaruh paling dominan terhadap kepuasan kerja karyawan di rumah makan ayam betutu Mbak Timah.
