Abstract

This research aims to find out the most effective concentration (LC50) and the most effective time (LT50)  and  to select a biological agent netamoda of nematodes Steinernema sp. dan Heterorhabditis sp. as the environmentally friendly biological control agents on the ground termite Coptotermes sp. and   Microtermes sp. in Lumajang. All the data were analyzed by using the termite mortality percentage variance analysis. The LC50 and LT50 were tested by using the probit analysis. Therefore, the empirical probit values were obtained from the percentage of mortality after they were calculated by the Abbot formulation. The biological agents netamoda entomopathogenic Heterorhabditis sp. and Steinernema sp. had the high value on the pathogenicity of Coptotermes sp. and Microtermes sp., the LC50 values on Coptotermes sp. of nematodes Heterorhabditis sp. was 14.94 IJ/ml and Steinernema sp. was 15.22 IJ/ml., the LC50 values on Microtermes sp. of nematodes Heterorhabditis sp. was 16.54 IJ/ml and  Steinernema sp. was 20.39 IJ/ml., the LT50 values on Coptotermes sp. of nematodes Heterorhabditis sp. was 1.29 hours and Steinernema sp. was 1.35 hours, and the LT50 values on Microtermes sp. of nematodes Heterorhabditis sp. was 2.14 hours and Steinernema sp. was 1.82 hours. Through pathogenicity test, the entomopathogenic nematodes Heterorhabditis sp. is more effective to control subterranean termites compared with the nematode Steinernema sp. This happens because Heterorhabditis sp. are actively engaged and looking for a host so that there are a large number of possibilities of contacting them with a very large subterranean termites.Keywords: Entomopathogenic nematodes,   Heterorhabditis sp., Steinernema sp., LC50, LT50

Abstract

Corn is the important food and crop for Indonesian. Recently, production of corn decreased because ofthe pest (Helicoverpa sp.). Until now, the control of Helicoverpa sp. depend on the using of chemicalelement. However, effect of synthetic chemical element usage was the appearance of secondary pest,the death of useful insect and the high accumulation of pesticide residue both in biotic and abioticagroecosystem. The residue of pesticide should effect on human health and environment balance.Furthermore, the control method using biological agents that have pathogenicity to host was done..One of biological agents is entomopathogenic nematodes. Entomopathogenic nematodes can kill it’shost at 24-72 hours, have wide host range such as Lepidoptera, Coleoptera, Diptera and Hymenopteraorder, and have no negatif effect on environment. The aim of the research is to find nematodes asbiological agents that effective against pest of corn (Helicoverpa sp.). The research was done inlaboratory of HPT UPN ”Veteran” Jawa Timur and HPT University of Jember. Methods of theresearch were : 1) Isolating entomopathogenic nematodes from many endemic area of Helicoverpaspp. In East Java; 2) Identificating Entomopatogenic Nematodes; 3) Screening IsolatesEntomopathogenic nematodes from East Java. The result indicated that there was anEntomopathogenic Nematodes Isolate from Tulungagung that has the highest pathogenicity (100%).

Abstract

Phosphate solubilizing bacteria (Pseudomonas putida 27.4B) is effective in dissolving undissolve P;humik compound can decrease Al exchange in acid soil and can be used as microorganism liquidmedia; Zeolite can improve CEC and bases concentration; and Trichorderma sp. can control diseaseand increase crop yield. Therefore, aim of this research is to make product having multifunction bothenvironment friendly soil amandement (fertilizer) and biopesticide, combination Pseudomonas putida27.4B, humic compound, zeolite, and Trichorderma sp.Potetoes Dextro Agar (PDA) media used to see resistance and growth, continued humic compoundliquid media added zeolite. Research design is completele randomized design with three factor are: 1)Trichorderma sp. concentration are: 0 (control); 104 CFU.ml-1; 105 CFU.ml-1; dan 106 CFU.ml-1; 2)good combination obtained last research are: a). 1000 ppm humic compound; 0,1% zeolit; P. putida27.4B 1012 CFU.ml-1 and b). 5000 ppm humic compound; 0,1% zeolit; P. putida 27.4B 1012 CFU.ml-1; and 3) incubation time are: 2 and 4 weeks.Result of research indicate that combination Pseudomonas putida 27.4B and Trichorderma sp. inPDA media can grow together, with visually speed of Trichorderma sp. growth faster thanPseudomonas putida 27.4B. Pseudomonas putida 27.4B proven can live on, even very good, in humiccompound liquid media added zeolite and Trichorderma sp. Its best growth is at humic compoundconcentration 1000 ppm. Besides that is tendency of combination Pseudomonas putida 27.4B andTrichorderma sp. exactly can increase growth of Pseudomonas putida 27.4B both at humic compoundconcentration 1000 ppm and without addition of humic compound. While at humic compoundconcentration 5000 ppm, Trichorderma sp. inokulasi exactly decrease Pseudomonas putida 27.4B.

Abstract

Its well known, That Entomopathogenic Nematods (EPNs) used as biological agentswhich effective to control pest of Spodoptera litura (Noctuidae: Lepidoptera).The aims of thisresearch were to find out the multifungsion product as well as a biofeltilizer and biopesticide whichsuitable for agriculture sustainability, secondly the materials were to mixed the humic compound andbiological agent Entomopathogenic Nematodes (EPNs). Combination of biopesticide with EPNs asactive ingredient and humic compound as biofertilizer to find out a good product combination whichindicated the highest viability and activity of EPNs Steinernema spp on humic compound, in orderapplication of these meterial more effective and efficient to prepare the soil fertility and pest problem.This research was done with inoculation of EPNs on the medium/culture e.c: phosphate solublebacteria Pseudomonas putida 27.4B, zeolit and the humic compound (liquid culture). Pathogenecitytest of EPNs were done after incubation on the humic compound to the larvae of S. litura.The result of this research showed that long time of viability EPNs only for fourth weeks on the humicculture at (4000 ppm). Actually the EPNs from this incubation could 100 % mortality of the S. lituralarvae after 72 hours inoculation. The infection rate of EPNs on the larvae S. litura was 30,3 tailsduring 24 hours incubation. The viability of EPNs Steinernema spp on the medium with 1000 ppmhumic compound was 20 % after 120 hours and after 4 weeks later all EPNs could not survive on thismedium. It seems, the medium containing zeolith, P. putida 27.4B and humic compound at 1000 ppmwas not suitable medium neither for nematods growth or larvae S. litura survival.

Abstract

Onion is one of lowland vegetables commodity that has been long cultivated intensively by farmers in Indonesia. Probolinggo is one of the regions which give considerable contribution of producing onion to the national needs. In an effort to increase production, onion farmers in Probolinggo mostly relies on external supply in the form of artificial chemicals (fertilizers and pesticides). However, the using of artificial chemicals can cause pollution and damage the environment. So that utilization of biological agents and organic fertilizers which are environmentally friendly can become other solutions to increase the yield of onion production. Based on this case, it is very important in efforts to increase production of onion by combining organic fertilizers to enrich the content of organic matter in the soil with biological agents in tackling pests attack. Based on the research results in application of combining organic fertilizers granules plus NEP and biological agents Heterorhabditis sp. in District Gending Probolinggo shows that : 1. reducing the population of pests Spodoptera exigua up to 72.260%. 2. improving onion plant growth more optimal. From the result, it shows that the combination of organic fertilizer granule plus biological agents and NEP Heterorhabditis sp. (P2A1) is more effective than the combination of the others. Abstract

This study aims to identify entomopathogenic nematodes symbiotic bacteria phenotypically and based on the gene encoding 16S rRNA sequences. Bacterial symbionts of entomopathogenic nematodes, obtained from isolates from the area Wonokerto (WN01) and isolates Sukapura (SP01), Bromo, Probolinggo, two symbiont bacteria was found in entomopathogenic nematodes Steinernema sp. The method used in this study are: the isolation of entomopathogenic nematodes Steinernema sp. and bacterial symbionts conventionally for the identification of phenotypically, after the characterization of bacterial isolates, the isolation of genomic DNA, 16S rRNA PCR, DNA purification and DNA sequence analysis. The results based on phenotypic characterization showed that isolates WN01 and SP01, yellowish white, gram positive, negative bioluminenscene, catalase positive, can not hydrolyze urea, and also can not produce H2S. The results of the gene encoding 16S rRNA sequence can be deduced WN01 isolates have in common with the bacteria Bacillus strain toyonensis BCT 7112, while the SP01 isolates have in common with the bacteria Bacillus strain cereus ATCC 14 579.

Abstract

Effect of Humic Compounds and CaCO3 Combination on Aluminium and Phosphate of Typic Paleudult Kentrong Banten (S. Winarso, E. Handayanto, Syekhfani and D. Sulistyanto): Decomposition of organic matter releases humic compounds, that can chelate metals include Al in acidic soils. This chelation of Al is important for decreasing of Al activity and P fixation in acidic soils. This study was aimed to test the effect of humic compounds extracted from rice-straw compost and CaCO3 combination on aluminium activity and phosphate (available P) on an Ultisol (Typic Paleudult) collected from Kentrong Banten.  A laboratory study was conducted by series consentration of CaCO3 (0, 500, 1.000, 2.000, dan 3.000 ppm) in 8 grams of acidic soil. The acidic soil had previously been subjected to continuously addition of humic compounds and none. Then, these mix of soil and CaCO3 were added 40 ml contained 5.000 ppm humic compounds to obtain 1:5 soil:suspension ratio and  control was made by using aquadest. They were shaked for 2 hours every day and pH was measured. At 11st days incubation  Alexch (N KCl) and soluble of P (PB and PC) were measured.  The results  showed that liming or addition of CaCO3 to the acidic soils decreased Alexch (precipitated to be Al(OH)3) linearly with formula y = -0.778x + 6.108; y = Alexch (Cmol kg-1), x = CaCO3 (M); R² = 0,916. Combination addition of humic compounds and CaCO3 was able to increase pH and Alexch up to not detected. The increased of P-soluble or desorption P untill 384% took place at addition of 0.0016 M CaCO3 and 5000 ppm humic compounds. More desorption of P was observed if the acidic soils has previously been subjected to continuously addition of humic compounds, such us the increased of soluble P up to 739% at addition of CaCO3 0.0008 M.

Abstract

[ENGLISH]One cause of the pest was found to be associated with the presence of cropping systems is monoculture. So, It required an effective alternative to control in suppressing the development of pest. The research was conducted in the village of Lempeni, District of Lumajang, with the aim to determine the benefits of intercrops and effect on pests in maize and legumes in intercropping cropping systems. This study uses a design Plots Divided, with intercrops as main plot treatments consisted of three levels (mung beans, peanuts, and soybeans), and the number of lines of treatment legumes as subplot (1 line, 2 line, 3 line legumes between two rows of maize and Control). Each treatment was repeated three times. The results showed that the highest populations of Heliothis armigera at age 60 hst is ktb4 treatment with the average value and the lowest 4.00 bugs khb1 treatment with an average value of 2.67 bugs. Intensity of the treatment the number of rows planted 3 rows provide significant effect on the level of damage to the maize, namely the attack Atherigona exigua with an average of 16.84%, Oxya chinensis with an average of 12:06%, leaf rollers attack with an average of 7.63% and Ostrinia furnacalis with an average of 53.33%. However, the results do not lead to the level of the damage corn plants die.Keywords: Atherigona exigua; Oxya chinensis; Heliothis armigera; Ostrinia furnacalis; Integrated Pest Management.[INDONESIAN]Salah satu penyebab munculnya hama ternyata dapat dikaitkan dengan adanya sistem pertanaman yang bersifat monokultur. Untuk itu diperlukan suatu alternatif pengendalian yang efektif dalam menekan perkembangan hama. Penelitian ini dilaksanakan di Desa Lempeni, Kabupaten Lumajang, dengan tujuan untuk mengetahui manfaat tanaman sela dan pengaruhnya terhadap hama dipertanaman jagung dan tanaman kacang-kacangan dengan sistem tanam tumpangsari. Penelitian ini menggunakan Rancangan Petak Terbagi, dengan tanaman sela sebagai perlakuan petak utama terdiri dari tiga taraf (Kacang hijau, Kacang tanah, Kedelai), dan perlakuan jumlah baris kacang-kacangan sebagai anak petak (1 Baris, 2 Baris, 3 baris kacang-kacangan diantara dua baris tanaman jagung dan Kontrol). Masing-masing perlakuan diulang tiga kali. Hasil penelitian menunjukkan bahwa pada populasi Heliothis armigera tertinggi pada umur 60 hst yaitu perlakuan ktb4 dengan nilai rata-rata 4.00 ekor dan terendah perlakuan khb1 dengan nilai rata-rata 2.67 ekor. Intensitas serangan pada perlakuan jumlah baris tanam 3 baris memberikan pengaruh yang nyata terhadap tingkat kerusakan pada tanaman jagung, yaitu pada serangan Atherigona exigua dengan rata-rata 16.84 %, Oxya chinensis dengan rata-rata 12.06 %, serangan penggulung daun dengan rata-rata 7.63 % dan Ostrinia furnacalis dengan rata-rata 53.33 %. Abstract

Local isolates of entomopathogenic nematodes are symbiotically associated with bacteria of the genus Xenorhabdus or Photorhabdus. Symbiont bacteria isolated from local isolates of entomopathogenic nematode (Pujon, Cemoro Lawang and X. nematophilus) were identical with Xenorhabdus spp and isolate of Ngadas was identical with Photorhabdus luminescens. Some isolates, such as Xenorhabdus (isolate of Pujon) and P. luminescens (isolate of Ngadas) has a high virulence when the bacteria was injected into insect haemocoel. High virulence was indicated by Xenorhabdus nematophilus when it was applied orally.

Abstract

Salah satu kendala produksi padi menurun adalah serangan hama Penggerek Batang Padi Kuning (Shcirpophaga incertulas). Aplikasi insektisida akan lebih tepat jika berdasarkan pemantauan ngengat dan juga tingkat kerusakan. Kerusakan tanaman padi yang di sebabkan hama Penggerek Batang Padi Kuning (Shcirpophaga incertulas) dapat dilakukan degan memanfaatkan feromoid baik untuk pemantauan maupun penangkapan massal ngengat jantan. Penelitian ini bertujuan mengkaji Pengaruh desain dan warna perangkap feromon sebagai alat pemantau Shcirpophaga incertulas pada pertanaman padi (Oryza sativa L.). Penelitian dilakukan di Lahan Padi Kecamatan Tanggul, Kabupaten Jember pada bulan Desember 2017 sampai selesai. Penelitian ini dilakukan dengan menggunakan Rancangan Acak Lengkap (RAL) Faktorial dengan 3x3 perlakuan dan 4 ulangan. Faktor 1). Desain perangkap yaitu Ukiriguru ukuran 1,5 L, Aqua ukuran 1,5 L, dan Stoples ukuran 1,5 L. Faktor 2). Warna perangkap yaitu transparan, kuning dan hijau. Parameter penelitian meliputi jumlah ngengat yang tertangkap, intensitas serangan hama penggerek batang padi kuning. Data yang diperoleh dianalisis berdasarkan analisis sidik ragam atau anova, kemudian dilanjutkan dengan uji Duncan pada taraf 5% bila perlakuan berbeda nyata. Hasil penelitian yang dilaksanakan tidak terdapat interaksi antara desain perangkap dan warna perangkap terhadap tangkapan ngengat jantan. Perangkap desain aqua dan warna transparan paling efektif menarik ngengat jantan.
