Abstract

Sumber daya perairan yaitu lintah laut (Discodoris sp.) dapat dibuat menjadi minuman fungsional. Lintah laut telah dilaporkan memiliki sifat antioksidan dan mengandung taurin. Tujuan dari penelitian ini adalah untuk mengetahui penerimaan konsumen pada minuman serbuk fungsional lintah laut. Tiga formula terbaik yang diterima secara organoleptik yaitu Formula T1 (Discodoris sp. 20%, jahe 40%, 20% curcuma, lemon 20%.), T2 Formula (Discodoris sp. 25%, jahe 40%, 15% curcuma, lemon 20%) dan T3 Formula (Discodoris sp. 30%,. jahe 40%, 10% curcuma, lemon 20%).

Abstract

Meretrix meretrix was one of gastropods seawater has been used by people as a food. These shells are still categorized of bycatch and have not explored the benefits. This study aimed to characterize and analyze the content of bioactive Meretrix meretrix. This research was conducted morphological characterization shellfish sensory, measurement of yield, the content of the proximate analysis, heavy metals, and analysis of bioactive compounds from the extracts of water and methanol. Observation found that the Meretrix meretrix has a yellowish white color with black bars at the end of the shell and the white meat and chewy with a yield of meat and shells of 11.09% and 69.85%. Analysis of water content, ash, fat, protein, and carbohydrates respectively by 79.99%, 1.50%, 0.22%, 9.42% and 8.81%. Heavy metal obtained  8x10-4 ppm Hg content, 13x10-4 ppm Pb, Cd, and Cu 45x10-4 and 88x10-4 ppm. Bioactive compounds in water and methanol extract of shellfish Lamis including alkaloids, steroids, and saponins.

Abstract

The Arrhenius model ASLT method is widely used to estimate the shelf life of food products that are easily damaged by the effects of chemical reactions, including fat oxidation, Maillard reactions, protein denaturation and so on. The aim of this study was to determine the best shelf life through stability testing. Testing shelf life was done by acceleration time or acceleration model using Arrhenius method. During the storage period, the product was stored in three different temperature conditions, ie, 30 ° C, 35 ° C and 45 ° C. Stability test performed on every 7 days observation of water content test (aw). Estimation of shelf life based on the critical parameter that is water activity value (aw) hence can be known save age of functional beverage powders sea slug of T2 have a longer shelf life when compared with  T1.

Abstract

 Metabolite profiles and antioxidant activity of Caulerpa racemosa extract with different handlings were investigated. Three different handlings during transportation were applied, namely samples chilled with ice, stored in liquid nitrogen and soaked in seawater. The different handling significantly affected the yield of ethanolic crude extracts and inorganic fractions but insignificantly to organic fractions. Different handlings resulted in differences of major fractions of C. racemosa extracts. Major fractions of the sample that was handled with chilling temperature contained low polar fractions (K10, K11, K12, and K13), while seawater handling extract contained very polar (K1, K2 and K3), polar (K6, K7, and K8) and low polar (K13) fractions. The extract of the sample handled in liquid nitrogen contained balanced fractions. Chilling temperature handling produced highest antioxidant activity (IC50 below 2,000 ppm) in ethanolic extract of C. racemosa. Abstract

Abstract

Since 500 years ago sea cucumber has been used as wound healing medicine. Sea cucumbers are thought to function as a cell growth factor and have the potential as an antibacterial agent. This study aims to determine whether the methanol extract of sea cucumber has antibacterial activity against Staphylococcus aureus and Pseudomonas aeruginosa so that it can be used to help diabetic wound healing of people infected with the same bacteria. The extraction method used was maceration with methanol to dried frozen simplicia. The simplicia was obtained by freeze drying to liquid preparation of heated fresh sea cucumber. The antibacterial activity test was performed using well diffusion method. There were no any antibacterial activity found in the two methanol extracts of sea cucumber. Abstract

Infectious diseases became more serious problem for public health in recent years. Although existing antibacterial drugs have been relatively effective, they do not rule out the emergence of resistance to the drug. Therefore, the intensive exploration of new bioactive compounds from natural, especially peptide compounds began in recent decades in order-handling infection. This study aimed to isolate, purify and test the potential application of Xylaria psidii KT30 extracellular protease as antibacterial agent against Gram-positive bacteria. X. psidii KT30, a marine fungus isolated from red seaweed Kappaphycus alvarezii showed antibacterial activity against Bacillus subtilisand Staphylococcus aureus. Antibacterial compounds of this fungus were predicted as a group of proteases. Extracellular protease exhibited an optimum activity when potato dextrose broth was used as cultivation medium. Furthermore, the highest activity of these proteases was found on fungal extract after day 15 of cultivation with value of 2.33 ± 0.19 U/mL. The partial purification of proteases using G-75 column chromatography resulted in 2 groups of fractions and showed protease activity based on zymogram assay. The extracellular proteases obtained from those fractions have 3 patterns of molecular mass based on sodium dodecyl sulfate–polyacrylamide gel electrophoresis which are 56.62, 89.12, 162.18 kDa.

One of the efforts of cancer treatment is the utilization of natural compounds. One of the microorganisms having the potential as anticancer agent is endophytic fungi. Endophytic fungi from the marine habitat can be isolated from sea weeds, sea grasses, sponges, and mangroves. Xylaria psidii KT30, a marine fungus used in this study was isolated from red seaweed Kappaphycus alvarezii. Xylaria psidii KT30 was cultivated in potato dextrose broth medium for nine days at room temperature 27-29°C in shaking condition. This study aimed to obtain protein fractions from X. psidii KT30 and determine their toxicity againt Chang and HeLa cells. The fractionation process was conducted using DEAE Sephadex A-50 column chromatography and the toxicity was determined by Brine Shrimp Lethality Test (BSLT). The metabolites excreted in the culture broth was extracted using 90% of ammonium sulphate. The extract was then tested for their toxicity against HeLa and Chang cells by Microculture Tetrazolium Technique (MTT) assay.The results revealed that LC50 of the protein extract of X. psidii KT30 was 104.95 ppm and IC50 was 69.9 ppm. Based on the National Cancer Institute (NCI), this value showed moderate cytotoxicity against HeLa cells.
