Abstract

The objectives of this study were to observe effect and  the effective dose of cinnamon (Cinnamomum burmanii) aqueous extract on pancreas structure of strain Balb-C mice (Mus musculus) diabetic after alloxan exposure. A total of 30 strains Balb-C mice (Mus musculus L.) were divided into 5 groups. Negative control group was not induced  by alloxan and a positive control group was induced alloxan 0,15 mg/g bw intraperitoneally for 9 days. The treatment group was divided into 3 groups was induced alloxan 0,15 mg/g bw and given an aqueous  extract of cinnamon at a dose of 0,73 mg/g bw; 1,09 mg/g bw and 1,45 mg/g bw orally for 7 days. Mice were dissected pancreas histhological taken using paraffin method and Haematoxylin-eosin (HE) stain. The result showed giving aqueous extract of cinnamon gave a strong effect in the repairing of pancreas structure after giving the alloxsan and the most effective aqueouse xtract of cinnamon with dose of 0,73 mg/g bb was able to repair the structure of mice's pancreas after giving the alloxan. Abstract

Streptozotocin cause hyperglycemic condition in mice. The objective of this study was to determine whether the hyperglycemic conditioned by streptozotocin (STZ) has an impact at folicullar atretic and estrous cycle prolonged. Mice were injected by STZ until hyperglycemic condition was reached. The vaginal smear was recorded daily for twice estrous cycle. Mice were sacrificed soon after the last vaginal smear observation. The ovary was removed, prepared by parafin method and stained by hematoxylin-eosin.The result showed that hyperglycemic causes increasing the number of atretic follicles, decreasing the number of corpus luteum and prolonged of estrous cycle.

Abstract

The objectives of this study were to observe the effect of synthetic estrogen (diethylstilbestrol/DES) on hepar structure and level of SGOT as well as SGPT of female mice (Mus musculus). Mice were gavage by DES dose 0.084 μg/g bb, 0.168 μg/g bb, 0.336 μg/g body weight, sesame oil and control. DES was treated by gavage for 15 days. Observation were performed liver structure contain of number of normal hepatocyte, and hepatocyte disruption (picnosis, vacuole cell, and necrosis), also SGOT-SGPT level. The result showed that DES effected to the disruption of hepatocyte particularly necrosis was relativity high in 0,168 μg/g bb DES treatment. SGOT and SGPT level on blood increased after DES treatment.

Abstract

Estrogen deficiency can changes the reproductive system especially in menopausal women, such as a decreasing of smooth muscle contraction, a thinning of vaginal structure, epithelial atrophy and a collagen decreasing. The disorders can be overcome by phytoestrogens treatment. Tempeh is a fermented soy product contains isoflavones that has estrogen-like structure with 17β-estradiol so that it can be able to bind estrogen receptors. Ovariectomized animal as animal test model has estrogen deficiency. Therefore, the purpose of this study to determine the effect of tempeh soy flour extract on vagina histological structure of Swiss Webster ovariectomized mice. The method used animals tested by treating 45 female Swiss Webster mice that were divided into five groups: negative control group (normal mice had not ovariectomized was not given tempeh flour extract), positive control (ovariectomized mice was not given tempeh flour extract), the treatment group (ovariectomized mice and was given tempeh flour extract D1 (0.21 g/ml / day), D2 (0.42 g/ml/day), and D3 (0,63 g/ml/day). Soy tempeh flour extract was given for 10, 20, and 30 days by gavage. The histological parameter were observed include tunica mucosa epithelium and tunica muscularis thickness of vagina. The result showed that tempeh flour extract increase epithelium thickness and tunica muscularis of the vagina ovariectomized mice. Treatment by taking longer time and giving higher dosage of tempeh flour extract could increase the ephitelium thickness of tunica mucosa and tunica muscularis vagina. The treatment of 0.63 g/ml/day dosage during 30 days showed the highest thickness average values of epithelium and tunica muscularis. The ephitelium thickness was 33.90 μm and the tunica muscularis dense was 90.20 μm.

Abstract

Uterine is one of organs that affected by estrogen hormone. This hormone can influence to increase uterus cells proliferation. Decreasing ovum production cause decreasing of estrogen level. This situation is faced of menopause that affect on uterine development. Phytoestrogens can bind to estrogen receptor and produce estrogenic affects like endogeneous estrogen. Tempeh is known as soybean fermented and consists of high level of isoflavon as phytoestrogens. Phytoestrogens have structure like mammalian estrogen 17β-estradiol. The aim of this study was to know effects of tempeh soy fluor extracts on mice uterine strain swiss webster ovariectomized.. Dosages that used on this study are 0.21g/mL, 0.42 g/mL and 0.63g/mL. The dosages are treated by gavage during 10 and 20 days after ovariectomy.The result showed that tempeh soy fluor extracts can increase the average of uterine mass, endometrial and myometrium thickness during treatment on 10 and 20 days. The 0,63g/mL dosage increase the highest of uterine mass (0.043g), endometrium thickness (389.46μm) and myometrium thickness ( 71.30μm.).

Abstract

Methoxychlor (MXC) is an insecticide (DDT derivates) that has the potential for bioaccumulation in mammal and causes a disruptive effect on the hepar and reproductive system. This study was done to find out the benefits of curcumin as a natural ingredient to overcome the negative impact of Methoxychlor (MXC) on hepar and male reproductive organ of Balb’C mice (Mus musculus L). The study was carried out in a Completely Randomized Design (CRD) Posttest Only Control Group Design used four treatments and six replications. The curcumin treatment after administration of MXC was carried out by gavage with curcumin doses: 0.05; 0,1; and 0.2 mg/g body weight, every day for two weeks, respectively. Histological observations of the liver, and testis was performed using the paraffin method and Hematoxylin Eosin stained. The results showed that MXC exposure caused liver disruption by increasing the number of pycnotic necrotic hepatocytes and hydrophic degeneration hepatocytes. On the male reproductive organ, MXC caused testis impairment by reducing the number of Sertoli cells and Leydig cells, spermatogenic cell counts, and the diameter of seminiferous tubules. The administration of curcumin at doses of 0.1 mg/g bw in mice exposed to methoxychlor can reduce the number of hydrophic degeneration hepatocytes and tend to reduce the number of pycnotic hepatocytes; and also increase the number of Sertoli cells, the number of spermatogenic cells, and the diameter of the seminiferous tubules, and tend to reduce the amount of Leydig cells. Curcumin treatment tends to recover hepar dan testis disruption of mice that were exposed by MXC.

Abstract

Rokok elektrik merupakan salah satu jenis rokok yang banyak digemari dewasa ini. Hal ini dikarenakan kandungan nikotin dalam cairan rokok elektrik yang dapat diatur oleh penggunanya sehingga dianggap aman. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pepaparan asap rokok elektrik dengan volume cairan yang berbeda terhadap profil hematologi mencit (Mus musculus L) Strain Balb’C jantan. Pada penelitian ini digunakan 24 ekor mencit jantan yang dibagi menjadi 4 kelompok. Kelompok 1 merupakan kelompok kontrol yang tidak dipapar asap rokok elektrik. Kelompok 2,3, dan 4 dipapar asap rokok elektrik dengan volume cairan sebanyak 1, 2, dan 4 ml. Dalam penelitian ini digunakan cairan rokok lektrik yang mengandung nikotin 6 mg/ml. Pemaparan asap rokok elektrik dilakukan selama 4 minggu. Setelah pemaparan dilakukan pengamatan profil hematologi meliputi kadar hemoglobin, jumlah eritrosit, jumlah dan jenis leukosit. Hasil menunjukkan bahwa setelah paparan asap rokok elektrik dengan volume yang berbeda terjadi peningkatan kadar hemoglobin dan jumlah eritrosit, penurunan jumlah leukosit dan limfosit serta tidak terjadi perubahan jumlah granulosit dan MID. Kesimpulan penelitian ini paparan asap rokok elektrik selama 4 minggu dengan volume cairan yang berbeda berpengaruh terhadap profil hematologi mencit (Mus musculus L) strain Balb’C jantan.

Abstract

Diabetes mellitus is a metabolic disease which indicated with increasing blood glucose level. Carbohidrate, protein, mineral and secondary metabolite (alkaloid, tannin and saponin) can be benefical to treat diabetes mellitus. Rats were randomly divided into three groups. First, control group. Second, STZ group, untreated diabetic. Third, STZ+GA 15% group, diabetic treated with GA 15%. Result showed that blood glucose level before STZ induction on control and STZ group within normal range 78,57±11,90 mg/dL and 74,85±6,86 mg/dL. Blood glucose level significantly increase after STZ induction on STZ and STZ+GA 15% group become 375±6,53 mg/dL and 346,42±50,23 mg/dL. Diabetic rat treated with GA 15% revealed decrease in blood glucose level compared to untreated diabetic rat. Blood glucose level on STZ+GA 15% group continuously decrease become 96,42±13,45 mg/dL and 82,14±9,19 mg/dL. In conclusion, GA 15% could reduce blood glucose level on diabetic rat.

Abstract

Bone is a dynamic structure and has the process to remodelling or regeneration. Bone remodelling includes the process of formation by osteoblasts and the process of resorption by osteoclasts. The balancing between formation and resorption are assisted by the estrogen hormone.Estrogen deficiency can lead the bone resorption. Ovariectomy is a model for estrogen deficiency by removing the ovaries. One way to overcome estrogen deficiency can be solved by offering phytoestrogens. This research have purpose to determine the effect of black soybean extract (Glycine soja) on the histology of the femur mice (Mus musculus) ovariectomized. In this research used a dose of black soybean extract as many as 0.42 g /ml/day and 0.63 g/ml/day, which are given orally for 30 days. The research is conducted with a completele randomized design with 4 treatments and 6 replications. Observations are made on the number of osteoblasts and the number of osteoclasts on the histological of the femur. The results showed that offering black soybean extract at a dose of 0.42 g/ml/day for 30 days have significantly increased the number of osteoblasts and have decreased the number of osteoclasts in the femur of ovariectomized mice.

Abstract

Estrogen production can be reduced due to menopause and ovariectomy. Decreased estrogen levels in the body cause physiological changes in the female reproductive system. Therefore, the intake of estrogen from outside the body is needed, in this case phytoestrogens from soy bean extract can be used to replace the role of endogenous estrogen. The study used 30 Swiss Webster mice (Mus musculus) aged 90 days weighing 35 grams which were unilateral ovariectomy. Mice are ready to be used as test animals after an estrogen deficiency period of 60 days. Mice are ready to be used as test animals after an estrogen deficiency period of 60 days. Mice were divided into five groups: negative control, positive control, dose 0.21, 0.42, and 0.63 g/mL/day . Data obtained from the results of the study were analyzed using the One Way ANOVA test with a confidence level of 99% or α = 0.01. Based on the results of the study, the administration of black soybean flour extract in unilateral ovariectomy mice can increase the average number of primordial follicles at doses of 0.42 g/ml/day and dosage 0.63 g/ml/day, primary follicles at a dose of 0.42 g/ml/day and doses of 0.63 g/ml/day, and secondary follicles at a dose of 0.63 g / ml / day, and tend to increase the average number of primordial follicles at doses of 0.21 g/ml/day, follicles primary at doses of 0.21 g/ml/day, secondary follicles at doses of 0.21 g/ml/day and 0.42 g/ml/day, de Graaf follicles at doses of 0.21, 0. 42 and 0.63 g/ml/day.
