Abstract

Klebsiella pneumoniae as an opportunistic pathogen can cause nosocomial infection. The main concern on this bacterium is directed on the extended-spectrum β-Lactamase (ESBL)-producing Klebsiella pneumoniae. The therapy of ESBL-producing Klebsiella pneumoniae infections is very limited because of its multidrug resistance. It had been found new local isolates Streptomyces sp-MWS1, Streptomyces sp-MWS3, and Streptomyces sp-MWS6 in mangrove East Coast of Surabaya. These isolates have potential to produce antibiotics (Retnowati, 2008). This study was aimed to prove these isolates may inhibit the growth of ESBL-producing Klebsiella pneumonia. The test of antibacterial activity of Streptomyces sp-MWS1, Streptomyces sp-MWS3, and Streptomyces sp-MWS6 using the modification Agar print method against clinical isolates of ESBL-producing Klebsiella pneumoniae. The diameter of inhibition zone (mm) formed shows activity of these isolates. The profiles of antibacterial activity of Streptomyces sp-MWS1, Streptomyces sp-MWS3, and Streptomyces sp-MWS6 were different in terms of periode required to reach peak activity, duration of activity and inhibition zone diameter produced during 10 days of observation. Streptomyces sp-MWS3 reached the peak activity most rapidly on day 3 with the largest inhibition zone diameter of 9 mm in ESBL-producing Klebsiella pneumoniae. There were significant differences in inhibition zone diameter between Streptomyces sp-MWS1, Streptomyces sp-MWS3, and Streptomyces sp-MWS6 against ESBL-producing Klebsiella pneumoniae.

Abstract

Drinking water is both a basic human need and a strong determinant of health. In Indonesia, there are drinking water refill stations (DWRS) which conveniently offer refill drinking water at affordable price. This study aims to determine whether station management amd raw water source influence the quality of drinking water. This cross-sectional study used primary data from questionnaires filled by DWRS owners and secondary data about the quality of refill drinking water from the Surabaya City Health Department. Data was then analyzed by using Chi-Square and Mann-Whitney tests. From 33 registered DWRSs, only 18 stations fulfilled all quality standards of drinking water. Some DWRSs failed to pass microbiological (n=13) and chemical (n=2) standards. All DWRS passed the physical standard. There was no significant difference found in duration of operation (pmicro = 0.956, pchemical = 0.382), production capacity (pmicro = 0.384; pchemical = 0.495). There was no significant difference found in terms of station management and raw water sources on the quality of refill drinking water. Keywords: drinking water refill station (DWRS); duration of operation; production capacity; raw water source

Abstract

Abstract. Staphylococcus aureus is a normal human skin flora that can cause infection when human immune system is weak. Staphylococcus aureus treatment is difficult because Staphylococcus aureus is easily resistant to some antibiotics so several terms such as Methicillin-resistant Staphylococcus aureus (MRSA), Vancomycin-intermediate Staphylococcus aureus (VISA), and Vancomycin-resistant Staphylococcus aureus (VRSA) are known to indicate the resistance. The growth of Staphylococcus aureus can be inhibited by Leucaena leucocephala leaf extract. Leucaena leucocephala is used by Indonesian people as traditional medicine to heal wounds, treat worm infection, and treat acne. The objective of this study was to identify the antibacterial activity of Leucaena leucocephala seed extract against Staphylococcus aureus. This was a laboratory experimental study using dilution method as many as five replications. Each replication used Leucaena leucocephala seed extract in concentrations of 3.125%, 6.25%, 12.5%, 25%, 50%, and 100% to determine the minimal inhibitory concentration (MIC) and the minimal bactericidal concentration (MBC) against Staphylococcus aureus. The results were not able to determine the minimal inhibitory concentration (MIC) of Leucaena leucocephala seed extract against Staphylococcus aureus due to dark color of the extract. While the minimal bactericidal concentration (MBC) of Leucaena leucocephala seed extract against Staphylococcus aureus was 50%. This showed that the concentration of 50% was the lowest concentration where there was no growth of the bacteria Staphylococcus aureus. In conclusion, the extract of Leucaena leucocephala seed has antibacterial activity against Staphylococcus aureus. Keywords: Staphylococcus aureus, Leucaena leucocephala, dilution method

Abstract

Introduction: Methicillin-Resistant Staphylococcus aureus (MRSA) bacterial infections may cause poor manifestations and even increase patients’ morbidity and mortality. The use of antibiotics in the management of infections remains the main therapy. However, ever-increasing antibacterial resistance has prompted researchers to find the solution. This study aimed to determine the effect of a combination of red coconut coir (Cocos nucifera L. var rubescens) in various concentrations and linezolid 10 µ/ml on the growth of MRSA bacteria in vitro. Methods: The antibacterial activity test method was well-diffusion test. Bacteria was plated in mueller hinton agar for 24 hours at 37oC. The treatment groups were red coconut coir extract and its combination with linezolid 10 µg/ml.  The well-diffusion test results were statistically analyzed with the One-Way ANOVA and Tukey HSD post-hoc tests. Results: Mean inhibitory zone diameter formed in the C. nucifera L. var rubescens extract test in various concentrations was 12.5 ± 0.36 - 16.2 ± 0.79 mm, while its combination with 10 µg/ml linezolid produced mean inhibitory zone of 15.1 ± 1.31-18.4 ± 0.46 mm. There was a significant difference between groups (p<0.05).Conclusion: Increasing concentration of C. nucifera L. var rubescens extract was in line with the increase of mean diameter of the inhibitory zone. Extract and antibiotic combination tests showed higher mean diameter zones than the single test of materials. The combination of extract and linezolid has the potential to synergistically prevent antibacterial resistance.

Abstract

Jalur Lingkar Wilis is a road which is connecting six cities in East Java Province, throughout Nganjuk, Ponorogo, Madiun, Tulungagung, Trenggalek, and Kediri. This road was placed along the foothill of Wilis Mountain which is this location was part of a protected forest and located in Geger Village, Sendang Sub – District. Therefore, some parts of protected forests face deforestation as the consequence of the Jalur Lingkar Wilis construction. This deforestation made the total of trees in Wilis Mountain decrease to worried conditions and cause the water absorption area in Tulungagung also fall on decreation. According to this condition, it takes awareness from all of the stakeholders to maintain the water absorption area through reforestation. This activity is in collaboration between Universitas Airlangga students with Perhutani, KPHLS Sendang, and Tulungagung Society to prepare and execute this reforestation. By the reforestation 220 trees were planted along the Jalur Lingkar Wilis. The reforestation is expected to prevent the protected forest area in Wilis Mountain from environmental damage as the consequence of Jalur Lingkar Wilis construction and also protect the water availability in Tulungagung.abstrakJalur Lingkar Wilis adalah jalur yang menghubungkan enam kabupaten di Jawa Timur, mulai dari Kabupaten Nganjuk, Ponorogo, Kabupaten Madiun, Tulungagung, Trenggalek, dan Kediri.
