Abstract

Transtuzumab has proven to be a great improvement in the treatment of HER2 + of breast cancer patients, but it is associated with relevant adverse cardiac events and with significantly elevated cost of treatment.  One of the risk factors for cardiotoxicity due to transtuzumab is the I655V HER2 polymorphism (GTC> ATC mutation) in which the allele mutant (Ile val or val / val) has a higher risk than the wild type (Ile/Ile). The detection of specific alleles is very important for therapeutic decision-making. In this study, our group have developed a HRM with EvaGreen dye method to discriminate specific allele of I655V HER2 (wild type, heterozygote mutant or homozygote mutant) in 66 frozen section samples of HER2+ of breast cancer patients. Our group reported the wild type is the most prevalent allele (77,27%), whereas heterozygous mutation is significantly present in this research (21,21%) and only 1,52% of sample detected as minor allele.  It showed that only one sample detected as a minor allele (val/val) and may have relatively low abundance in the population. This method has been compared with sanger sequencing and giving 100% of validity.

Abstract

Human Papilloma Virus  (HPV) has an out standing feature for its vast intraspecies variability. Of all known 100 types or  more, 15 t ypes of them are classified as high risk  because of their occurrence  in  more than 95% of cervical cancer cases. Among all genes in their genome, E6 and E7 genes are considered oncogenes and have close relevance with their pathogenicity, whilst L1 and L2 ge nes produce capsid proteins that directly interact with their  host  receptors.  Considering  the  importance  of  L1  and  L2  in  host-receptor  relationship,  we  tried  to investigate their molecular variability thereby uncover their specificity as Indonesian variant s. Here we reported about the conservation of L1 minor capsid protein and variability of L2 capsid protein among high-risk types Human Papilloma Virus (HPV). The results indicated that L1 DNA was relatively more conserved than its L2 counterpart Also it was indicated that the middle part of either L1 or L2 CDS‟ showed more DNA variability than those  at  their  upstream  sequences.  It  is  concluded  that  L2  middle  sequences  are  important  factors  for intraspecific variations found in HPV of Indonesian variants.   Keywords: Human Papilloma Virus (HPV), L1 gene, L2 gene, cervical cancer, high risk HPV

Abstract

Background: E6 and E7 are oncoproteins of HPV16. Natural amino acid variation in HPV16 E6 can alter its carcinogenic potential. The aim of this study was to analyze phylogenetically E6 and E7 genes and proteins of HPV16 from Indonesia and predict the effects of single amino acid substitution on protein function. This analysis could be used to reduce time, effort, and research cost as initial screening in selection of protein or isolates to be tested in vitro or in vivo. Methods: In this study, E6 and E7 gene sequences were obtained from 12 samples of  Indonesian isolates, which  were compared with HPV16R (prototype) and 6 standard isolates in the category of European (E), Asian (As), Asian-American (AA), African-1 (Af-1), African-2 (Af-2), and North American (NA) branch from Genbank. Bioedit v.7.0.0 was used to analyze the composition and substitution of single amino acids. Phylogenetic analysis of E6 and E7 genes and proteins was performed using Clustal X (1.81) and NJPLOT softwares. Effects of single amino acid substitutions on protein function of E6 and E7 were analysed by SNAP.Results: Java variants and isolate ui66* belonged to European branch, while the others belonged to Asian and African branches. Twelve changes of amino acids were found in E6 and one in E7 proteins. SNAP analysis showed two non neutral mutations, i.e. R10I and C63G in E6 proteins. R10I mutations were found in Af-2 genotype (AF472509) and Indonesian isolates (Af2*), while C63G mutation was found only in Af2*.Conclusion: E6 proteins of HPV16 variants were more variable than E7. SNAP analysis showed that only E6 protein of African-2 branch had functional differences compared to HPV16R.

Abstract

This study is aimed at finding out what students need in taking English as required course, what students want to learn, and how they want to learn. The data were taken from 500 questionnaires distributed to students. The data revealed that most of the students do have positive attitude in learning English, and because of this the students have more chances to take the best of their learning. However, as most of the students also take English because they are required to do so, there are also some factors that may discourage learners’ motivation in learning, i.e. materials selection, and teaching methods.
