Abstract

The native plants of East Borneo the Costus specious (Koening) J.E. Smith stem, Lagerstroema spesiosa Pers leaf, Cerbera mangans L leaf, Vitis trifolia L fruit., Scurrula atropurpurea (Blume) Danser root, Brucea javanica (L.) Merr. leaf, Lygodium microphyllum, Bidens Chinensis Willd., Sonneratia caseolaris L. peel, Sonneratia caseolaris L. stem is almost under-explored for their potensial benefits. They were extracted by the solvents of increasing polarity (n-hexane, ethyl acetate, and n-butanol) were tested for their free radical activity against DPPH (2,2-diphenyl-1-picrylhydrazyl). The ethyl acetate extract of Costus spesiosus (Koening) J. E. Smith antioxidant were screnned concentration of 68 ppm, similarly athyl acetate extract of Vitis trivolia L., showed antioxidant activity at 64,30 ppm. As the ethyl acetate extract of Scurrulla atropurpurea (Blume), Brucea javanica (L.) Merr, Lygodium microphyllum and Sonneratia caseolaris L. stem, showed antioxidant activity at 273,52 ppm, 91,12 ppm, 17,39 ppm and 7,03 ppm. N-butanol extract of Lagerstroema spesiosa Pers, Cerbera mangans L, Bidens chinensis Willd, and Sonneratia caseolaris L. peel showed 8,37 ppm, 128,59 ppm, 18,17 ppm and 54,29 ppm antioxidant activity using DPPH model systems. Owing to the property, the studies can be further extended to exploit them for their possible application for preservation of food products as well as their use as health supplements.

Abstract

In an effort to find new sources of medicinal plants as antimalarials based on ethnobotany knowledge from generation to generation. Therefore, tested the inhibition of heme polymerization from 70% ethanol extract of papaya leaves (Carica papaya) and Guava (Psidium guajava) leaves. The data obtained were calculated by IC50 using SPSS 22 obtained that 70% ethanol extract of Pepaya leaves (Carica papaya) and Jambu Biji leaves (Psidium guajava) had antimalarial activities of 7,914 and 8,794 mg/ml respectively and their had the potential to be developed in more research further as antimalarial.

Abstract

The study aimed to determine the total alkaloid, phenolic, and flavonoid content of ethyl acetate extract of Peperomia pellucida (L.) Kunth herbs, especially the herb was obtained from oil palm plantations in Baras, Nort Mamuju District, West Sulawesi Province. The sample was macerated using n-hexane then was followed with ethyl acetate. Determination of total phenolic content was performed using Folin-Ciocalteu reagent, and its absorbance was measured at 750 nm wavelength with gallic acid as a standard. Determination of total flavonoid content was conducted using AlCl3 reagent, and the absorbance was calculated at 415 nm wavelength using quercetin as a standard. Determination of total alkaloid content was calculated against the equal piperine at a wavelength of 435 nm using Dragendorff reagent. Based on the results of determination on ethyl acetate extract of Suruhan herbs was obtained the total phenolic content of 178.768+10.63139 mg/gram, total flavonoid content of 1.397+0.004509 mg/gram, and total alkaloid content of 0.615+0.155885 mg/gram. These results are preliminary data of further studies to the isolation of the targeted compounds and the utilization purposes as a raw material of herbal medicine.

Abstract

The present study aims to optimize the natural deep eutectic solvent (NADES) as a green solvent-based microwave-assisted extraction (MAE) of total polyphenols content (TPC) from Mitragyna speciosa Korth Havil leaves using response surface methodology (RSM). Preparation of natural deep eutectic solvent (NADES) as a green solvent was performed by melting the two-component of malic acid and glucose using a magnetic stirrer. The leaves sample was extracted using the NADES-MAE method with various extraction conditions with four factors and three levels (Box Behnken Design) and optimized using RSM with licensed software of Design Expert V12. TPC was determined using a Folin-Ciocalteau reagent and absorbance was measured on a UV-VIS spectrophotometer at 770 nm and gallic acid as a standard. This study was obtained the optimum extraction conditions include: NADES ratio (malic acid: glucose) of 1:2 g/g, 50% microwave power, a solvent-sampel ratio of 12:1 mL/g, and extraction time for 7 minutes. The optimum conditions were obtained according to the equation formula: TPC = 165.17 – 33.97X1 + 38.36X2 – 6.08X3 + 16.12X4 – 62.77X1X2 + 79.61X12X2, where a R2 value = 0.7102 with a TPC prediction of 402.087±43.81 mg GAE/g sample. These conditions can be used to extract TPC from this plant efficiently, quickly, easily, and environmentally friendly.

Abstract

Coronavirus Diseases 2019 (COVID-19) merupakan pandemic global yang disebabkan oleh severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Berbagai negara terdampak akibat wabah ini, termasuk Indonesia. Di Indonesia, perubahan kebijakan dari pembatasan sosial berskala besar (PSBB) ke new normal menjadi dilematis, mengingat kasus COVID-19 belum ada gejala kecenderungan mengalami penurunan. Kebijakan tersebut berdampak pula pada kebijakan di lingkungan kampus Universitas Mulawarman. Penelitian ini bertujuan untuk mengetahui hubungan tingkat pengetahuan dengan perilaku mahasiswa Universitas Mulawarman Samarinda terhadap wabah pandemic COVID-19. Penelitian ini dilakukan dengan pendekatan observasional analitik dengan rancangan cross sectional. Responden penelitian ini adalah 304 mahasiswa UNMUL yang dipilih secara acak dengan mengisi kuisioner dengan google form. Analisis dilakukan menggunakan uji univariat untuk memperoleh gambaran distribusi dan frekuensinya, sedangkan uji bivariat untuk memperoleh tingkat kemaknaan 95%. Hasil penelitian menunjukkan adanya hubungan antara pengetahuan dengan perilaku (kewaspadaan) responden terhadap wabah pandemi COVID-19 terbukti dengan nilai signifikansi sebesar 0,002 (p<0,005) dan tidak ada hubungan antara pengetahuan dengan perilaku (kepedulian) responden terhadap wabah pandemi COVID-19 berdasarkan nilai signifikansi sebesar 0,821 (p>0,005). Kesimpulan yaitu mahasiswa Universitas Mulawarman memiliki pengetahuan yang baik mengenai pandemi COVID-19 akan tetapi masih memiliki perilaku yang kurang dalam memutus penyebaran virus ini dan tidak terdapat hubungan yang signifikan pengetahuan dengan Perilaku (Kepedulian) Terhadap Wabah Pandemi COVID 19 Mahasiswa Universitas Mulawarman

Abstract

The previous research showed that the fraction C from active extract of Garcinia latissima Miq. fruit was active against Bacillus subtilis. This study aims to isolate and identify the active compound as an antibacterial agent from the fraction C. Fraction C was purified by recrystallization using chloroform and n-hexane solvents and then isolated using preparative-thin layer chromatography-silica gel 60 GF254 to give a yellow compound. The antibacterial activity was determined using microdilution with thiazolyl blue tetrazolium bromide indicator against B. subtilis American Type Culture Collection 6633. The isolate was identified using UV-Vis, IR, MS, Proton Nuclear Magnetic Resonance (1H-NMR) and carbon NMR (13C-NMR), and NMR-2D techniques including HMQC and HMBC. Based on the spectroscopic analysis and literature review, the compound was identified as 6-deoxyjacareubin, which is a new compound from Garcinia latissima Miq. The 6-deoxyjacareubin showed antibacterial activity with MIC value of 156.25 ppm and was categorized as a weak antibacterial agent because the MIC value was more than 100 ppm. According to in silico approach to the docking study, 6-deoxyjacareubin showed similar hydrophobic interaction with several amino acid residues including C2565, C2589, G2484, U2590, and U5588 between a native ligand.

Abstract

Application of natural deep eutectic solvent (NADES) citric acid-glucose-based microwave-assisted extraction (MAE) method for total polyphenol content (TPC) enriches the use of Eleutherine bulbosa (Mill.) bulb more widely and commercially. Therefore, this study aims to optimize NADES citric acid-glucose-based MAE on the TPC from E. bulbosa bulbs using response surface methodology (RSM). In the present study, the dried sample of E. bulbosa bulb was extracted using the NADES based MAE. Determination of TPC was conducted using Folin-Ciocalteau reagent and standard gallic acid, then measured using a UV-Vis spectrophotometer at 761 nm. Extraction condition optimization of the NADES based MAE method was performed using RSM with Box Behnken Design (four-factors-three-levels and 29 runs) on Design Expert v12. Based on the results study, the optimum condition extraction on the TPC enrichment was obtained at the NADES ratio (citric acid: glucose) 1:1 g/g; solid-liquid ratio 1:8 g/mL; extraction time of 15 min; and 270 Watt microwave power. The confirmation test and scale-up (50 g samples) were obtained with a 61.63 ± 2.23 mg GA/g sample.

Abstract

Kadamba (Mitragyna speciosa Korth Havil) merupakan salah satu tumbuhan endemik di Asia Tenggara, di Indonesia, tumbuhan ini banyak di jumpai di pulau Kalimantan. Tumbuhan ini dipercaya oleh masyarakat dapat mengobati berbagai macam penyakit. Namun, karena kandungan alkaloidnya, tumbuhan ini dilarang digunakan sebagai bahan baku obat herbal oleh Badan Pengawas Obat dan Makanan. Oleh karena itu, perlu dilakukan pengembangan metode ekstraksi untuk menarik senyawa metabolit sekunder target dan meminimalkan senyawa yang tidak diinginkan yaitu dengan menggunakan metode ekstraksi berbantu mikrowave dengan pelarut hijau (Natural Deep Eutectic Solvent) yang dioptimasi menggunakan response surface methodology (RSM). Simplisia kering daun Kadamba diekstraksi menggunakan metode ekstraksi berbantu mikrowave dengan pelarut hijau (asam sitrat-glukosa) dengan berbagai kondisi ekstraksi dengan empat faktor dan tiga level (Box Behnken Design) yang dioptimasi menggunakan RSM dengan aplikasi perangkat lunak Design Expert versi 12 berlisensi. Penetapan kadar polifenol total dilakukan menggunakan reagen Folin-Ciocalteau dan diukur absorbansinya pada spektrofotometer UV-Vis pada panjang gelombang 770 nm, serta asam gallat sebagai standar. Berdasarkan hasil penelitian, diperoleh kondisi optimum pada perbandingan rasio pelarut NADES (asam sitrat : glukosa) 5:1 g/g, rasio pelarut-sampel sebesar 1:20 g/mL, waktu ekstraksi selama 20 menit dan kekuatan gelombang mikro sebesar 30% dengan kadar polifenol total prediksi sebesar 314,924 ±35,95 mg GAE/g sampel, sedangkan dari hasil proses verifikasi (scale-up confirmation) yaitu dengan meningkatkan jumlah sampel yang digunakan sebanyak sepuluh kali lipat dengan Kadar polifenol total diperoleh  sebesar 427,12 ±35,95 mg GAE/g. Penelitian ini merupakan langkah awal dalam pengembangan metode ekstraksi untuk memperoleh senyawa target secara cepat, mudah, efisien, dan ramah lingkungan.

Abstract

ABSTRACT Quantitative Structure-Activity Relationship (QSAR) study have been conducted to antimalarial active compounds of cassane-type diterpenes isolated from seed kernels of Bagore (Caesalpinia crista Linn.), using the atomic net charges data resulted from AM1 method of semi-empirical calculations. The aim of this research is to determine the mathematic equation model of quantitative structure-activity relationship from antimalarial active compounds of cassane-type diterpenes isolated from seed kernels of Bagore (Caesalpinia crista Linn.) using the electronic parameter of AM1 method. The best QSAR equation model was determined by multiple linier regression analysis and tested by leave-one-out type of cross validation methods. The result indicates that the biological activity is represented by a linier function of activity versus atomic net charges of C1, C2, C3, C4, and C6 which is expressed by : Log (1/C) = -7,362(+ 4,251) qC1 +3,258 (+ 4,590) qC2 + 11,038 (+4,485) qC3 + 29,421 (+20,542) qC4 + 19,849 (+6,171) qC6 + 8,541 (+2,256) The equation is significant at 95% level. The cross validation analysis give minimal value of PRESS=64,372 and SEP = 2,93; where Log (1/IC50) = biological activity and q = atomic net charges Key words : QSAR, Atomic Net Charges, Cross Validation Methods ABSTRAK Studi hubungan kuantitatif struktur-aktivitas (HKSA) telah dilakukan terhadap 42 senyawa aktif antimalaria turunan diterpen kassan hasil isolasi dari biji Bagore (Caesalpinia crista Linn.) menggunakan data muatan bersih atom hasil perhitungan semiempiris metode (Austin Model 1) AM1. Tujuan penelitian ini adalah menentukan model persamaan matematis hubungan kuantitatif struktur-aktivitas dari senyawa aktif antimalaria turunan diterpen kassan furano hasil isolasi dari biji Bagore (Caesalpinia crista Linn.) dengan parameter elektronik menggunakan metode AM1. Pemilihan model persamaan HKSA terbaik ditentukan dengan analisis regresi multilinier dan diuji dengan metode validasi silang leave-one-out. Hasil penelitian menunjukkan bahwa aktivitas biologis merupakan fungsi linier dari muatan bersih atom C1, C2, C3, C4, dan C6 yang dinyatakan dalam persamaan : Log (1/C) = -7,362(+ 4,251) qC1 +3,258 (+ 4,590) qC2 + 11,038 (+4,485) qC3 + 29,421 (+20,542) qC4 + 19,849 (+6,171) qC6 + 8,541 (+2,256) Persamaan signifikan pada tingkat kepercayaan 95%. Hasil analisis validasi silang memberikan data parameter pengujian relatif minimal yaitu PRESS = 9,645; dimana Log (1/IC50) = aktivitas biologis dan q = muatan bersih atom Kata kunci : HKSA, Muatan Bersih Atom, Metode Validasi Silang

Abstract

ABSTRACT Research about active antioxidant compounds isolated from plants ant nests (Myrmecodia pendens Merr. & Perry) from Papua by the method of column chromatography has been done. This study aimed to explore the isolation of the active class of compounds as antioxidants from ant nest plant. Isolates obtained using the method of column chromatography and antioxidant activity of the ant nest plants isolates tested used spraying method with free radical DPPH (2,2-diphenyl-1-picrylhydrazyl) after eluted on thin-layer chromatography. At separation by column chromatography, eluate collected in vials with a volume of 10 ml. Based on the results of fractionation, separation of the components obtained by 74 vials. Identification using TLC by determining the Rf value of each component. The same Rf value is considered as a single fraction. Based on the results obtained seven fractions TLC. Active isolates (F6) obtained by spraying. The results of fraction of spot with DPPH staining showed that the isolates was considered the most active isolates as seen from the formation of a yellow color that is lighter and more dominant than the other fractions, this indicates that the fraction (F6) binds more free radicals (DPPH). ABSTRAK Telah dilakukan penelitian isolasi senyawa aktif antioksidan dari tumbuhan sarang semut (Myrmecodia pendens Merr. & Perry) asal Papua dengan metode kromatografi kolom. Penelitian ini bertujuan untuk mencari isolat golongan senyawa yang aktif sebagai antioksidan dari tumbuhan sarang semut. Isolat diperoleh dengan menggunakan metode kromatografi kolom dan aktivitas antioksidan isolate sarang semut diuji menggunakan metode penyemprotan dengan radikal bebas DPPH (2,2-diphenyl-1-picrylhydrazyl) setelah dielusi pada kromatografi lapis tipis. Pada pemisahan dengan kromatografi kolom, eluat ditampung dalam vial dengan volume 10 ml. Berdasarkan hasil fraksinasi, diperoleh komponen hasil pemisahan sebanyak 74 vial. Identifikasi menggunakan KLT dengan cara menentukan nilai Rf masing-masing komponen. Nilai Rf yang sama dianggap sebagai satu fraksi. Berdasarkan hasil KLT di dapatkan 7 fraksi. Isolat aktif (F6) yang diperoleh dengan penyemprotan. Hasil pewarnaan spot fraksi dengan DPPH menunjukan bahwa isolat yang dianggap paling aktif adalah isolat yang dilihat dari terbentuknya warna kuning yang lebih terang dan lebih dominan dibanding fraksi yang lainnya, ini menandakan bahwa fraksi (F6) lebih banyak mengikat radikal bebas (DPPH).
