Abstract

This research aimed to determine the effect of Effective Microorganisms-4 (EM-4) concentrations and its application frequency and their interactions on growth and yield of green mustard plant. The experiment was conducted from November 2014 until January 2015 at Mantikulore Sub District, Palu, at an altitude about ± 50 m above sea level. This research was carried out using a Randomized Block Design (RBD) with two factors and three replications. The first factor was the concentration of EM-4 (A) with 3 (three) levels : K1 (0,5 %), K2 (1,0 %) and K3 (1,5 %), and the second factor was the application frequency of EM-4 also with 3 (three) levels : A1 (1 time; 1 week after planting-WAP), A2 (3 times; 1, 3, and 5 WAP) and A3 (5 times; 1, 2, 3, 4 and 5 WAP). The experimental data were analyzed using analysis of variance method followed by HSD (Honestly Significant Difference) Test at 5%. The results of the research showed that the application of various concentrations of EM-4 had a significant effect on plant height (cm) at age 4, 5, and 6 WAP, number of leaves at age 5 WAP, root fresh weight (g, and gave a very significant effect to root dry weight (g). The application frequency of EM-4 gave an effect to plant height at age 5 WAP, number of leaves at age 5 and 6 WAP, and root length (cm). Whereas the interaction between both factors only had a significant effect on number of leaves at the age of 5 WAP.

Abstract

The research was conducted at Bulu Pounto Jaya, Biromaru sub District, Sigi District, Central Sulawesi Province from August to November 2014. This research aimed to determine the effect of mulch and liquid organic fertilizer types on growth and yield of shallot (Allium ascalonicum L.) Lembah Palu varieties. The design used in this research was a two factorial randomized block design (RAK).  The first factor was application of mulch (M) i.e. M0 = no mulching, M1 = silver black plastic mulching, M2 = rice straw mulch and the second factor was the application of liquid organic fertilizer i.e. F1 = once application (7 days after planting-WAP), F2 = twice applications (7, 14 DAP), F3 = thrice applications (7, 14, 21 DAP). Therefore, there were nine combinations of treatments and replicated three times. The results showed that the type of mulch (M) affected significantly the components of plant height, number of leaves, leaf area, plant fresh weight, and tuber fresh weight. The liquid organic fertilizer treatment (F) had a significant effect on the components of leaf area and plant fresh weight. The rice straw mulch (M2) produced higher growth than the silver black plastic mulch (M1) and without mulch (M0). While the liquid organic fertilizer applied three times (F3) produced higher yield than the other applications. There was significant interaction effect between the two factors on leaf area components.

Abstract

The research aimed to gain the appropriate NAA concentration in performing tissue culture of dragon fruit plants. The research was compiled by using completely randomized design consisting of 4 treatments and each treatment was repeated 6 times that 24 experimental units were done. The variables observed include the number of shoots, the number of roots, the number of segments, and the length of shoots observed at the age of 4 weeks and 8 weeks after planting. The research findings reveal that on the observation at 4 weeks after planting the NAA concentration has effect on the number of segments and the elongation of shoots, yet it does not have effect on the number of shoots and roots, while at 8 weeks after planting the NAA concentration has effect on the number of shoots, the number of roots, the number of segments, and the length of shoots. The research findings also show that the treatment of N3 (0.3 ppm NAA) gives better result compared with other treatments in which the average number of shoots formed is 5.50 per explant, the number of roots formed is 3.50 per explant, the number of segments formed is 8.83 per explant and the average length of shoots formed is 2.13 cm per explant. Key Words : Dragon Fruit, NAA Concentration, In Vitro.

Abstract

Several studies have revealed a wide range of beneficial effects of seaweed products applications on plants growth and development. Seaweeds may cause different effect in plant responses because there are a range possible factors acting in concert. These factors may include to the amount of plant growth regulator (PGR) and nutrients present in the products. PGR activity in two seaweed products (Maxicrop® and Seasol®) was investigated using Radish (Raphanus sativus). Treatments included a control, standard concentrations of the seaweed Products (SS1 and MC1), 2 and 4 times the recommended rates (SS2, SS4, MC2 and MC4) ashed seaweed products (SS-ash and MC-ash), and equivalent amounts of N, P and K to that found in seaweed products (SS-NPK and MC-NPK). The results obtained show that both Maxicrop® and Seasol® can significantly increase crop performance. Maxicrop® increased chlorophyll content and root and shoot growth, while Seasol® only increased the chlorophyll content

Abstract

The most crucial step during the development of tissue culture is the method of explant sterilization. Especially, explants is sourced directly from fields that were more susceptible to microbial contaminations. This study aimed to obtain sterilization technique  from several sterilant for reduced contamination and support growth of shoots grape in vitro. This research was conducted at the Laboratory of Plant Biotechnology, Faculty of Agriculture, University of Tadulako, during January to April 2018. The study used a factorial completely randomized design, one factor was tested i.e S1 = Taft 8,3 g/L (0,1% carbendazim), S2 =  taft 25 g/L (0,3% carbendazim),  S3 = taft 41,67 g/L (0,5% carbendazim), S4 = Taft 8,3 g/L (0,1% carbendazim) + HgCl2 0,1%, S5  = Taft 25 gr/L (0,3% carbendazim) + HgCl2 0,1% dan S6 = 41,67 gr/L  (0,5% carbendazim) + HgCl2 0,1 %. There were six treatment and each treatment was replicated three times to obtain 18 experimental units. Each experimental unit using two explant. Data were analyzed using analysis of variance and followed by Honestly Significant Difference test at level of 5% if the treatment effects were significant. The results showed sterilization technique using taft 41,67 gram / L containing 0.5% carbendazim for 30 minutes, followed by 70% alcohol for 30 seconds and HgCl2 0,1% for 10 minutes gave free contamination for explant.whereas, Taft 8.3 g / L (Carb 0.1%) for 30 min, followed by 70% alcohol for 30 seconds is a sterilization technique that does not inhibit explant growth as indicated from the most callus formation.
