Abstract

Teknik kultur jaringan mamalia telah menjadi bagian tak terpisahkan dari bioteknologi modern yang diaplikasikan secara luas di lingkup biologi, kedokteran, dan farmasi. Di Madrasah Aliyah Negeri (MAN) 2 Surakarta, siswa jurusan IPA mendapatkan pengenalan bioteknologi secara teori di dalam mata pelajaran biologi, termasuk mengenai sejarah perkembangan dan aplikasi kultur jaringan. Karena kendala fasilitas dan sumber daya manusia yang terbatas, pembelajaran konsep-konsep kultur jaringan tidak pernah diimbangi dengan pengalaman praktek di laboratorium. Tujuan dari kegiatan pengabdian ini adalah: (i) meningkatkan wawasan umum mengenai bioteknologi, (ii) membantu pemahaman mengenai kultur jaringan mamalia yang tercakup dalam materi pembelajaran di sekolah. Metode pelaksanaan kegiatan adalah teori dan praktek di laboratorium Prodi Biologi UNS. Materi kultur jaringan difokuskan pada pengetahuan mengenai teknik aseptis, isolasi sel dari jaringan kulit, pemeliharaan kultur sel, dan pengamatan morfologi sel dengan mikroskop. Adapun selama KLB corona, praktek pengamatan jaringan hewan awetan dilakukan secara mandiri di rumah masing-masing menggunakan mikroskop kertas (foldscope). Dengan demikian, tiap siswa yang saat ini melaksanakan pembelajaran jarak jauh tetap mendapatkan kesempatan untuk praktikum. Hasil yang diperoleh dari kegiatan ini adalah tingkat pemahaman mengenai kultur sel dan jaringan yang lebih menyeluruh, serta pengalaman belajar yang lebih bervariasi untuk mengisi kekurangan pembelajaran jarak jauh yang minim praktek.

Abstract

Intense physical exercise can increase lactic acid levels in the blood as a result of the anaerobic metabolism that occurs at that time. The impact of increased levels of lactic acid includes the occurrence of acidosis, damage to body cells, and fatigue which reduces muscle performance. Antioxidants can play a role in preventing body fatigue caused by oxidative stress. Onion Dayak (Eleutherine palmifolia) is a type of medicinal plant that is useful for health. The high content of antioxidants in Dayak onions can be used as an alternative anti-fatigue ingredient. This study aimed to look at the effect of the dose and duration of Dayak onion extract on lactic acid levels and physical activity in mice forced swimming test models. Type of Research Randomized Pre and Post-Test Control Group Design. Male White Mice (Sprague Dawley) 2 months old (weight 150-200g). Divided into 5 groups K- (the experimental group was not given Dayak onion extract, they were still given FST), K+ (the experimental group was given xanthine and FST), P1 (given Dayak onion extract at a dose of 50mg/200gBW), P2 (given the extract onion Dayak dose of 100mg/200gBB), P3 (given onion extract dose of 200mg/200gBW). Dayak onion extract was given for 28 days and given the forced swimming test, then the changes in lactic acid and physical activity endurance in swimming were observed.

Abstract

Metabolic syndrome is a threat to health problems in the world, namely a group of metabolic disorders that occur simultaneously. Obesity is a risk factor for metabolic syndrome. Fat accumulation causes insulin resistance and leptin resistance resulting in weight gain. Tali bamboo leaf tea contains a natural source of antioxidants that can increase insulin sensitivity so that it can reduce leptin levels. This study aims to determine the effect of bamboo1tali leaf tea on leptin levels in rats with metabolic syndrome. This type of research is experimental laboratoric with a Pre-Post Test with a Control Group design. Wistar male white rats, 8 weeks old, and 150 – 250 grams in weight, were fed HFHF (High Fat High Fructose) for 14 days, induced by Nicotinamide and Streptozotocin. Then, the rats were divided into 5 groups, namely negative control, positive control, and 3 treatments of tali bamboo leaf tea (300, 600, and 900 mg/200 g BW) for 28 days. Measurement of leptin levels using the ELISA method.Bamboo leaf tea had a significant effect (p <0.005) on leptin levels in rats with metabolic syndrome. The largest decrease in leptin levels occurred in the TDB3 group which was 82.8% and the lowest decrease in leptin levels occurred in the TDB1 group which was 34.8%. Bamboo leaf tea doses of 300, 600, and 900 mg/200 g BW reduced leptin levels in rats with metabolic syndrome for 28 days.

Abstract

Metabolic syndrome is a metabolic disorder such as obesity, hyperglycemia, hypertension, and dyslipidemia that occur simultaneously. The prevalence of metabolic syndrome globally is increasing and the risk of T2DM, cardiovascular, stroke, and death. Metabolic syndrome is related to dyslipidemia due to excess energy intake which can cause oxidative stress and hypertriglyceridemia. Tali bamboo (Gigantochloa apus Kurz.) leaf tea contains antioxidants such as flavonoids, tannins, and saponins which have the potential to reduce BMI and triglyceride levels. This study aims to determine the potency of tali bamboo leaf tea on BMI and triglyceride levels in rats metabolic syndrome. This type of research is laboratory experimental with a control group pre-post test design. 30 male Wistar rats, aged 8 weeks, with a weight of 150 – 250 g, which were induced by High Fat High Fructose (HFHFr) for 14 days, then continued with STZ-NA. Rats were divided into 5 groups namely K- (aquades), K+ (metformin), and 3 doses steeping tali bamboo leaf (150, 300, and 450 mg/100 g BW) for 28 days. The BMI of rats was measured using the index Lee and triglyceride levels using the GPO-PAP method. Data were analyzed using paired t-test and One Way ANOVA. The results showed that there was a significant difference (p < 0,05) between BMI and triglycerise levels before and after treatment. The effective dose to reduce BMI and triglyceride levels is 300 and 450 mg/100 g BW. Tali bamboo leaf tea has the potential to improve BMI and triglyceride levels so that it can be developed as a therapy for metabolic syndrome.ABSTRAK Sindrom metabolik merupakan gangguan metabolisme tubuh seperti obesitas, hiperglikemia, hipertensi, dan dislipidemia yang terjadi secara bersamaan. Prevalensi sindrom metabolik secara global semakin meningkat dan berisiko terjadinya DMT2, kardiovaskular, stroke dan kematian. Sindrom metabolik berkaitan dengan dislipidemia akibat asupan energi berlebih yang menyebabkan stress oksidatif dan hipertrigliserida. Teh daun bambu tali (Gigantochloa apus Kurz.) mengandung antioksidan seperti flavonoid, tanin, dan saponin yang berpotensi dalam menurunkan IMT dan kadar trigliserida. Penelitian ini bertujuan untuk menganalisis potensi teh daun bambu tali terhadap IMT dan kadar trigliserida tikus sindrom metabolik. Jenis penelitian ini penelitian eksperimental laboratorik dengan rancangan pre-post test kelompok kontrol. Subjek penelitian ini adalah 30 ekor tikus Wistar, jantan, umur 8 minggu dengan BB 150 – 250 g yang diinduksi High Fat High Fructose (HFHFr) selama 14 hari, kemudian dilanjutkan STZ-NA. Tikus dibagi menjadi 5 kelompok yaitu K- (aquades), K+(metformin), dan 3 dosis seduhan teh daun bambu tali (150, 300, dan 450 mg/100 g BB) selama 28 hari. Indeks massa tubuh tikus diukur menggunakan rumus indeks Lee dan kadar trigliserida menggunakan metode GPO-PAP. Data dianalisis menggunakan paired t-test dan One Way ANOVA. Hasil penelitian menunjukkan bahwa terdapat perbedaan yang signifikan (p < 0,05) antara IMT dan kadar trigliserida sebelum dan setelah perlakuan selama 28 hari. Dosis efektif untuk menurunkan IMT dan kadar trigliserida adalah 300 dan 450 mg/100 g BB. Pemberian teh daun bambu tali berpotensi dalam memperbaiki IMT dan kadar trigliserida sehingga dapat dikembangkan sebagai terapi untuk sindrom metabolik.
