Abstract

Increasing oxygen consumption during intensive physical exercise may increase production of free radicals, and if it exceeds physiological capacity may cause oxidative stress as shown as chance of MDA level. Buah merah oil contains high betacarotene and tocopherol, as well as fatty acid such as oleic acid, linoleic acid, linolenic acid and decanoic acid. Tocopherol is major biological lipid-soluble antioxidant, protecting structures and function of cell membranes from free radicals. The objective of this study was to know in vitro antioxidant activity and the effect of buah merah oil on MDA level in blood during maximum physical activity treatment. In vitro antioxidant activity test was conducted using DPPH method (2,2-diphenyl-1-picryl hydrazyl). In vivo antioxidant activity test was done by using 24 male Wistar rats in pre - post test control group design. The rats were grouped into 4 groups. The control group was given destillated water and three treatment groups were given buah merah oil in the dose of 0,15 ml/kgBW; 0,3 ml/kgBW; 0,6 ml/kgBW respectivelyfor 10 days. Before buah merah oil was given, level of MDA was measured. Ten days later, the four group were given maximum physical activity mean of swimming until the sign of fatigue occurred (nearly drowned) and the blood was taken for blood MDA examination. In vitro, the IC50 of buah merah oil was 451,51 μg/ml. In vivo test, dosage 0,15 ml/kgBW could decrease MDA 5,22% Dosage 0,3 ml/kgBW could decrease MDA 11,96% and dosage 0,6 ml/kgBW could decrease MDA 8,19% . Buah merah oil showed antioxidant activity in vitro. In vivo experiment, all dosage of buah merah oil decreased blood MDA level but in the dose of 0,3 ml/kgBW decreased MDA level more than dose of 0,15 ml/kgBW and 0,6 ml/kgBW.

Abstract

Antioxidants are compounds which have ability to inhibit free radical reactions in the human body. Objectives of this research are to evaluate the antioxidant activities of extract and fractions from Pandanus canoideus Lam. fruits, to determine total phenolic and total flavonoid contents, and to correlate the total phenolic and total flavonoid contents of those fractions with their antioxidant activities. The Pandanus conoideus Lam. fruits or locally known as “buah merah” were extracted by methanol, then evaporated and dissolved in aquadest. This solution is subsequently partitioned using chloroform and ethyl acetate to obtain chloroform, ethyl acetate, and water fractions. The antioxidant activities were determined by radical scavenging assay using DPPH (2,2-diphenyl-1-picrylhydrazyl) radical. The total phenolic and total flavonoid contents were determined by mean of spectrophotometry. The results showed that fractions of water, ethyl acetate, and chloroform revealed antioxidant activities with IC50 values of 399.06, 10.41, 1416.68 µg/mL respectively, whereas the methanolic extract of Pandanus conoideus Lam. fruits before partition has IC50 of 186.31 µg/mL. The antioxidant activities of these extract and fractions are lower than that of vitamin E (IC50 8.27 µg/ml). A linier positive relationship existed between the total phenolic contents of these extract and fractions with their antioxidant activities as shown by equation of y = 13.085x + 754.91; r2 = 0,3641, while the correlation between the total flavonoid contents and their antioxidant activities revealed a linier regression of y = -282.9x + 868.05; r2 = 0.4484.

Abstract

Antioxidants are substances that may inhibit free radical reactions in the body. The purpose of this research is to identify lethal dose, in vitro antioxidant activity and effect of kemboja merah flowers decoction (Plumeria rubra L.) on MDA level. Lethal dose was measured by Weil method. Antioxidant activity assays was performed in vitro by DPPH method and in vivo by measuring MDA plasma level. The level of MDA was conducted using the thiobarbituric acid reactivity test. In the test, 30 mices were divided into 6 groups: normal, negative control, positive control and 3 group treated with variation dose of kemboja merah flowers decoction. LD50 of decoction kemboja merah flowers is > 15 g/kg of body weight. In vitro, antioxidant activity showed the IC50 value of decoction kemboja merah flowers is 36.96 μg/mL.The antioxidant activity of decoction kemboja merah flowers is smaller than vitamin C (IC50 5.89 μg/mL). In vivo antioxidant activity showed levels of MDA on normal group is 1.66 nmol/mL, negative control group is 5.67 nmol/mL, positive control group is 1.45 nmol/mL, treated group which was given by decoction kemboja merah flowers at dose 0.39 g/kg of body weight is 4.32

Abstract

Black pepper (Piperis nigri fructus) has been empirically used to eliminate fatigue and to increase stamina or as stimulant. The aim of this study was to determine the efficacy of black pepper infuse as stimulant on 30 male DDY mice using swimming and rotarod methods. The mice were divided into five groups. First group was given distilled water as control, second group was given caffeine of 0.36 mg/20 g bw. The other three groups were given orally with black pepper infuse of 0.32 mg/20 g bw, 0.65 mg/20 g bw, or 1.3 mg/20 g bw. Observations on mice swimming endurance and mice endurance on a rotarod, were carried out 60 minutes after oral administration of the solutions. Analysis using Kruskal-Wallis showed that there was significant difference in the stimulant efficacy between the black pepper infuse and the negative control (p<0.05). There was no significant difference in stimulant effect between the 0.32 mg/20 g bw of black pepper infuse and the caffeine positive control, however, the infuse dose of 0.65 mg/20 g bw and 1.3 mg/20 g bw have demonstrated stronger stimulant effects (p>0.05) as compared to the caffeine positive control.

Abstract

Biji pepaya mengandung vitamin E dan flavonoid yang diketahui memiliki aktivitas sebagai antioksidan. Tujuan penelitian ini untuk mengetahui efek antioksidan ekstrak etanol biji pepaya (Carica papaya L.) terhadap aktivitas SOD sel darah merah dengan metode adenokrom dan kadar MDA plasma dengan metode TBARs pada mencit yang diberi aktivitas fisik perenangan. Mencit diberi ekstrak etanol biji pepaya selama 7 hari dan perenangan selama 65 menit pada hari ke-7. Pengujian ini menggunakan 36 ekor mencit yang dibagi 6 kelompok, yaitu kelompok I (normal), kelompok II (kontrol negatif), kelompok III (kontrol positif vitamin E), kelompok IV (dosis 0,105 g/kg bb), kelompok V (dosis 0,210 g/kg bb) dan kelompok VI (dosis 0,420 g/kg bb). Hasil penelitian kelompok I, II, III, IV, V, VI berturut-turut untuk aktivitas SOD sel darah merah adalah 89±6,4329; 47,37±3,8225; 85,56±3,9210; 46,35±10,0929; 58,89±3,8271 dan 81,59±5,7700 U/mL, kadar MDA plasma berturut-turut adalah 1,6831±0,0942; 6,3567±0,1046; 1,3697±0,2254; 4,7914±0,2139; 3,8231±0,3574 dan 1,7560±0,1215 nmol/mL. Ekstrak etanol biji pepaya dosis 0,420 g/kg bb memberikan efek antioksidan paling tinggi.

Abstract

Atherosclerosis is chronic inflammatory disease that can be prevented with anti-inflammatoryactivity agents and reduce thrombus formation in blood vessels. Tembelekan leaves (Lantana camaraLinn.) contain fl avonoids, saponins, and essential oils which potential to inhibit platelet aggregation andbe anti-inflammatory. This study aimed to test the in vivo anti-platelet aggregation and anti-inflammatoryactivity of tembelekan leaves. Platelet anti-aggregation test was carried out on mice by assessing thedecrease in plasma absorbance after addition of ADP. Mice were divided into normal group, positivecontrol group which was given clopidogrel and 3 test groups which were given Tembelekan leaf extractat a dose of 2.5, 5, 10 g/kgBW. In vivo anti-inflammatory test used the Winter method with intraplantar1% carrageenan as inducer. There was a signifi cant decrease in plasma absorbance after addition ofADP at doses of 2.5 and 5 g/kgBW of tembelekan leaves i.e 5.47% and 12.28% compared to thenormal group. Positive control and dose of 10 g/kgBW gave the same eff ect. In anti-inflammatorytest, the doses of 3.2 and 6.4 g/kgBW of tembelekan leaves gave percentage value of inhibition whichwas not signifi cantly diff erent with diclofenac sodium. Tembelekan leaves potentially as anti-plateletaggregation and anti-inflammatory properties.

Abstract

Curcumin has various activities such as antioxidant, anti-inflammatory, antitumor, and antibacterial. The bioavailability of curcumin is low and needs to be optimized. The manufacture of curcumin nanoparticles with several solvents is one way to increase the bioavailability of curcumin. This study aims to examine the safety of curcumin nanoparticles through acute and subchronic toxicity tests and the antioxidant activity of the formula. Acute toxicity test was performed by OECD 425 method, using AOT425StatPgm device for the LD50. Subchronic toxicity test was carried out by OECD 407 method using 4 groups: normal, a dose of 50;100; and 200 mg/kg BW for 28 days, then evaluate body weight, clinical biochemistry, and hematology. An antioxidant activity test was carried out using DPPH. The results showed were the LD50 value of curcumin nanoparticles was>5000 mg/kg BW and the results of the subchronic toxicity test (p>0.05) did not show any significant differences in AST, ALT, BUN, creatinine, albumin, and hematology compare to the normal group. The IC50 of curcumin and curcumin nanoparticles were 118.76 µg/mL and 90.87 µg/mL, respectively. It can be concluded that curcumin nanoparticles are safe and do not affect kidney and liver function and the antioxidant activity of the curcumin nanoparticle formulation is greater than that of curcumin.

Abstract

Diabetes mellitus characterized by hyperglycemia whereas the condition for high blood glucose. InIndonesia, there are a lot of medicinal plants that used as antidiabetic treatment. Medicinal plants are known have relatively few side eff ects compared to chemical based drugs. Asparagus (Asparagus offi cinalis L.) was reported to have fl avonoids and saponins compounds that could be functioned as antidiabetic. The aim of this research was to fi nd out the eff ect of boiled asparagus on blood glucose level in mice induced by Alloxan. This research used 30 male DDY strained mice, which were classifi ed into 6 groups, and each group consisted of 5 mice. These 6 groups were: normal control, negative control, positive control (Acarbose), the boiled asparagus with dose of 280, 560, and 840 mg/kgBW for 14 days. Blood glucose level of mice were observed on day-0, 8, 15, and 22. The results showed that Alloxan injection increased the level of blood glucose. Another result showed that boiled asparagus with dose of 280, 560 and 840 mg/kgBW reduced the level of blood glucose and the eff ect showed treatment groups doses of 560 dan 840 mg/kgBW had no signifi cantly diff erent with positive control group (Acarbose).

Abstract

Bamboo Kuning (Bambusa vulgaris Shard) leaves its efficacy as antioxidant, anti-inflammation, antidiabetic and antiulcer. The study aims to know volume edema decrease of rats foot as anti-inflammatory effect and the amount of strectching mice an analgesic effect the decoction of Bamboo Kuning. Anti-inflammatory test used Winter methods. Rats were be diveded into 5 groups: negative control be given aquadest, positive control be given sodium diclofenac 4,367 mg/200g BB, the test groups were given the decoction of Bamboo Kuning leaves 2,8 mL/200g BB with a concentration of 10%, 20% and 40%. Analgesic testing used methods Sigmund. Mice were divided into 5 groups: negative control be given aquadest, positive control were given sodium diclofenak 0,26 mg/20g BB, the test group was given the boiled of bamboo kuning leaves 0,4 mL/20g BB with concentration 10%, 20% and 40%. Statistic result decoction of leaves Bamboo Kuning dose 2,8 mL/200g BB with concentration 10%, 20% and 40% have anti-inflammatory and analgesic effect. Decoction of Bamboo Kuning leaves dose 2,8 mL/200g BB with concentration of 10%, 20% and 40% have anti-inflammatory and analgesic effect are lower than sodium diclofenac. Increased dose boiled of Bamboo Kuning leaves no provide increased effect as an anti-inflammatory and analgesic.

Abstract

Treatment for ovarian carcinoma is still far from optimal, animal models are still needed to study human epithelial ovarian cancer. Animal models of ovarian cancer are very important for understanding the pathogenesis of the disease and for testing new treatment strategies. Ovarian carcinogenesis models in mice have been modified and repaired to produce preneoplastic lesions and neoplastic ovaries that are pathogens resembling human ovarian cancer. Although spontaneous ovarian tumors in mice have been reported, some of the shortcomings of existing studies preclude their use as animal models of ovarian cancer. Because of this, many efforts have been made to develop animal models that are relevant for ovarian cancer. Experimental animal models are developed accurately to represent cellular and molecular changes associated with the initiation and development of human ovarian cancer. Accurate experimental models have significant potential in facilitating the development of better methods for early detection and treatment of ovarian cancer. Several animal models of ovarian cancer have been reported, including manipulation of various reproductive factors or exposure to carcinogens. The latest advance in ovarian cancer modeling is using genetically engineered mice.
