Abstract

Penelitian ini bertujuan untuk menemukan isolate bakteri dari ornament gua (stalaktit, stalagmit, limestone, and flowstone) yang memiliki aktivitas urease sebagai dasar kajian biogrouting. Metode penelitian adalah survei dengan teknik purposive sampling. Medium yang digunakan adalah medium B-4. Hasil penelitian ditemukan sebanyak 42 isolat bakteri. Masing-masing 21 isolat dari stalaktit, 9 isolat dari batu gamping, dan 12 isolat dari flowstone. Aktivitas urease hanya diujikan terhadap isolat dari ornamen flowstone. Dari 12 isolat yang diujikan 10 isolat diantaranya positif memiliki aktivitas enzim urease. Kata Kunci : bakteri, ornament, gua, biogrouting, aktivitas urease

Abstract

This activity is motivated by concern for the facts of climate change occurring globally and regionally. The agreement of many countries to address this change problem has been initiated since 1990, until the most recent high-level meeting "climate adaptation summit" in 2020. The Intergovernmental Panel on Climate Change (IPCC) warned that there are only 12 years left or until 2030 to prevent extreme climate disasters that occur due to global warming with a maximum temperature increase of 1.5°C. In response to this, the Indonesian government has also taken a number of concrete steps in all aspects of climate change adaptation and mitigation, both at the local, regional and national levels. One of the national programs that directly touch local communities is the climate village program or known as "PROKLIM". The city of Bukittinggi has actively responded to this national program, so that two of the three climate villages developed by the Environmental Service of the city of Bukittinggi have received national awards for the achievements of their community group activities. This activity is one of the activities that is of interest to community groups in one of the climate villages in Bukittinggi City, because it sees the potential to overcome the problem of relatively high organic waste generation both in the village itself and in the nearest market location, with the use of larvae (maggot) from Black Soldier Fly which can be done in groups. It is hoped that organic waste management with the use of larvae (maggot) will not only overcome the problem of organic waste, but also at the same time increase people's income, both from selling the maggot products to fish/poultry farmers, as well as being able to be used by group members for their own business. fisheries and livestock.

Abstract

Isolation and Characterization of Bacteria from nest sand and egg shell of Olive Ridley (Lepidochelys olivacea L.) which hatched and failed to hatch in Pariaman turtle conservation site, West Sumatra were conducted from December 2012 to September 2013 in Microbiology Laboratory, Andalas University and Laboratory of Provincial Health Department, West Sumatra. The aim of this study was to identify species of bacteria found at egg shells and nest sand of Olive Ridley in Pariaman. The result found 4 species of bacteria: Bacillus cereus and Klebsiella sp. from sand nest that hatched; while Bacillus cereus, Klebsiella sp. and Shigella sp. were identified from sand nest that failed to hatch. Bacteria isolates from egg shell that hatched were identified as Bacillus cereus, Klebsiella sp. While Bacillus cereus, Klebsiella sp., Salmonella sp. and Shigella sp. were identified from egg shell that failed to hatch. Keywords: isolation, bacteria, nest sand, egg shell, Olive Ridley

Abstract

A study on the characterization of thermophilic bacteria in producing neutral protease enzyme has been conducted in the Laboratory of Microbiology, Biology Department, Andalas University from October 2013 to March 2014. The bacteria were isolated from a hot spring at Sungai Medang, Kerinci. Water samples  were collected purposively based on gradient temperature at the hot spring which ranged from 450C to 880C. Colony of the bacteria were inoculated using nutrient agar (NA). We obtained 10 isolates of neutral protease enzyme from the thermophilic bacteria with proteolytic index ranged from 0.27 to 3.89. The MIV-14 isolate has the highest proteolytic index which collected from 830C of the hot spring. Characterization of macroscopic, microscopic and biochemical tests were reported. Generally, the colonies have round shape, wavy edge, white, producing endospores and motile. Keywords: characterization, neutral protease, thermophilic bacteria.

Abstract

The district of Pariaman has a conservation site for sea turtles, including olive ridley (Lepidochelys olivacea L.). Several reports have stated that fungus infect egg of the turtles and caused the failure of hatching. However, there is no study on this site yet. The aim of this study was to identify fungus on turtle's egg, hatched and inhatched nest sand. Collection and identification were conducted from December 2012 until June 2013 in turtles’ breeding in Pariaman and Microbiology Laboratory of Biology Department of Mathematic and Natural Sience, Andalas University. Result showed that Fusarium sp. 1, Fusarium sp. 2, Fusarium sp. 3, Fusarium sp. 4, Fusarium sp. 5, Fusarium sp. 6, Fusarium sp. 7 dominated among identified fungus, other were Aspergillus sp. 1 and Trichoderma sp. 1 and than there was no fungus found on hatched egg. Abstract

The research on isolation and characterization of amylothermophilic bacteria has been done in Research Laboratory of Microbiology, Biology Department, Mathematics and Natural Sciences Faculty, Andalas University, Padang. The aimed to obtain amylothermophilic isolate and describe their characteristic. The study used purposive sampling method and descriptive analysis. The study results obtained four amylothermophilic isolates with amylolitic index ranged from 2.23 until 3.51 were different macroscopic characters, rod shape, motile and produce catalase enzyme, three isolates  were  Gram positive and spore forming, one isolate was  Gram negative.

Abstract

The aims of this research are to analyze the phytochemical contents and to know the antibacterial potential of medicines herbs of Maek Society in controlling S. aureus and Methicillin Resistant Staphylococcus aureus (MRSA). This research is conducted on March – July 2017 in Biota Sumatera Laboratory and Microbiology Research Laboratory of Biology Departement, Andalas University. The method is experiment with five treatments of concentration against two bacterias test. The result showed that phytochemical test of medicine herbs extract contains alcaloids and phenol. All of the extracts concentration able to control S. aureus and MRSA. Antibacteria activity of the medicine herbs against S. aureus is stronger than againts MRSA. The highest diameter of the inhibitory zones extract against S. aureus is 12,98 ± 1,29 mm and against MRSA is 12,56 ± 0,38 mm. Abstract

Staphylococcus aureus is an opportunistic pathogenic bacteria. These bacteria can cause infections of the skin, bones, lungs, heart or systemic infections. Giving antibiotics does not fit the recommendations result in the emergence of resistant strains. Methicillin Resistant Staphylococcus aureus (MRSA) is one of the bacteria beta-lactam antibiotic resistant. Pakih Sipasan (Blechnum orientale) is a low level plants that is widely used Simanau villagers at Solok District in overcoming skin infections. The aims of this research are to determine the phytochemical content of B. orientale and to know antibacterial potential against S. aureus and MRSA. This research was conducted in March-July 2017 at Biota Laboratory of Sumatera and Microbiology Research Laboratory of Biology Department, Andalas University. Research method is experiment. The extracts were prepared with concentrations of 20 mg/ml, 40 mg/ml, 60 mg/ml, 80 mg/ml and 100 mg/ml. The data obtained are analyzed and presented in the form of tables and drawings are presented descriptively. The results showed that the B. orientale contain chemical compounds class of alkaloids, phenols and flavonoids. All treatments have potential as antibacterial to S. aureus and MRSA. The antibacterial activity of the B. orientale against S. aureus is stronger than against MRSA. The average diameter of inhibitory zone formed from concentrations of 20 mg/ml to 100 mg/ml of S. aureus ranged from 13.00 mm - 14.95 mm and to against MRSA between 11.31 mm - 14.48 mm. Abstract

The city of Bukittinggi has responded actively to this national program and has received national awards for the achievements of its activities in two community groups. The purpose of community service activities in the Climate Village Program is to increase the group's capacity in processing household organic waste through the application of Maggot cultivation from black soldier fly (BSF). The main methods of implementing the activities are training, providing assistance and companiment the target group. The activity results showed that the target group had the knowledge and skills to cultivate BSF and produce larvae/maggots. The target group had been able to produce fish/livestock feed from the larvae and developed plans to use the business results for the commercial market. One thing that becomes the next challenge is how to build continuity of feed availability for the maggot caterpillars, especially from household organic food leftovers in the group members' environment. In conclusion, the application of organic waste processing technology with maggot caterpillar cultivation is very potential to overcome the problem of waste in their neighborhood so that it can then be applied more widely in the city of Bukittinggi, to be able to overcome the problem of urban organic waste.

Abstract

Abstrak Pengelolaan limbah medis padat merupakan tahapan dimulai dari pemilahan hingga pemusnahan. Pada pelaksanaan pengelolaan limbah medis padat masih ditemukan beberapa tahapan yang belum sesuai dengan kebijakan seperti penggunaan kantong plastik belum sesuai dengan jenis dan karakteristik limbah, penggunaan desinfeksi terhadap wadah limbah medis yang telah digunakan, masih minimnya penggunaan Alat Pelindung Diri. Disamping itu pengetahuan dan sikap petugas pengelola limbah medis masih rendah, sehingga diperlukan dukungan pimpinan. Tujuan penelitian ini adalah menganalisis pengelolaan limbah medis padat dan mengetahui pengaruh tingkat pengetahuan, sikap dan dukungan pimpinan terhadap pengelolaan limbah medis padat di Fasilitas Pelayanan Kesehatan Tingkat Pertama (FKTP) Kota Padang. Penelitian ini menggunakan metode deskriptif kuantitatif dengan sampel sebesar 58 sampel dengan analisis Regresi Berganda. Untuk FKTP sebagian besar telah melaksanakan pengelolaan limbah medis sesuai dengan peraturan yang telah. Hasil regresi berganda bahwa pengetahuan, sikap dan dukungan pimpinan berpengaruh nyata terhadap pengelolaan limbah medis padat. Untuk FKTP seperti Puskesmas dan Klinik sudah melaksanakan pengelolaan limbah medis sesuai standar yang ditetapkan namun untuk praktek dokter gigi masih rendah dalam penerapan peraturan pengelolaan limbah medis padat. Diharapkan untuk seluruh FKTP mempunyai komitmen dalam pengelolaan limbah medis padat serta adanya konstribusi dari Dinas Lingkungan Hidup, Dinas Kesehatan, Manajemen dari FKTP serta stake holder lainnya. Kata Kunci: Pengelolaan limbah medis, FKTP, pengetahuan, sikap, dukungan pimpinan  AbstractSolid medical waste management is a stage starting from sorting to destruction. In the implementation of solid medical waste management, several stages were found that were not following the policy. The purpose of this study was to analyze the management of solid medical waste and determine the effect of the level of knowledge, attitude and leadership support on the management of solid medical waste in the First Level Health Service Facility (FKTP) Padang City. This study uses a quantitative descriptive method with a sample of 58 samples with multiple regression analysis. Most of the FKTP have implemented medical waste management following existing regulations. Multiple regression results show that knowledge, attitudes and leadership support have a significant effect on solid medical waste management. For FKTPs such as Puskesmas and Clinics, they have implemented medical waste management according to the standards set, but for dentistry, the implementation of solid medical waste management regulations is still low. It is hoped that all FKTPs commit to solid medical waste management as well as contributions from the Environment Service, Health Service, Management from FKTP and other stakeholders. Keywords: Medical waste management, FKTP, knowledge, attitude, leadership support
