Abstract

The study was conducted to evaluate the effect of intervention of skim milk based functional drink supplemented with 100 mg of soy isoflavone and 8 mg of Zn-sulfate (susumeno) on serum estradiol and thymulin hormones levels of permenopausal women. After 2 month of intervention, the estradiol levels did not change, and the levels were in the range of 39,16 – 41,99 pg/ml. However the thymulin hormone levels increased significantly from 2,37 µg/ml to 3,31 µg/ml. Increase in thymulin levels shows that the disfuction of immune system of premenopausal could be improved by the functional drink. Abstract

The research was conducted to find out the effect of Zn supplement to immune status of premenopausal women intervented with isoflavoned drinking. Respondents were 22 women, more than 40 year of age. They were divided into two groups, i.e. 11 women intervented with isoflavone, and other 11 women intervented with isoflavone and 8 mg Zn. The activities of SOD, catalase and GPX were determined by spectrophotometer, thymulin levels by ELISA, whereas Zn levels by AAS. Result showed that Zn had significantly increased SOD lymphocyte activities (p=0.002) and thymulin plasma (p=0.011). Zn had increased catalase (p=0.103) and GPX (p=0.322) as well, but Zn plasma had decreased (0.163). It was indicated that Zn had improved the immune status by increasing lymphocyte and thymus cells activities.

Abstract

Phytosterol may reduce the absorption of cholesterol, and used for preventing atherosclerosis. It is limited in soybean, but potentially abundant in wheat germ. Research on the utilization of wheat germ sterol had not been reported so far. Many aspects of germ sterol extraction from wheat germ and its characteristics were still unknown. In this research, the best extraction method, kinds and content of phytosterol from wheat germ were investigated. This research consisted of two steps: (1) extraction of phytosterol directly form whole germ and ground germ using hexane, and indirect extraction through germ oil using hexane and mixed solvent of hexane and ethanol, and direct extraction from ground germ using ethanol; (2) analysis of the type and content of phytosterol in the crude extract through the following steps: preparation of crude extract, fractionation, and analysis. Results showed that indirect extraction through germ oil was considered as the best method which yielded 1.37% of phytosterol. The highest yield was obtained when extracted using a mixed solvent of hexane – ethanol 82:18. However, the odor of ethanol and hexane (gasoline like odor) was still detected. The solvent’s ratio of hexane to ethanol at 1:2 resulted better odor of the extract. Extraction of sterol using ethanol yielded 18.39% of sterol when the ratio of germ to ethanol at 1:10 (w/v) was applied. Results of quantitative analysis on the main component of crude extract of wheat germ sterol showed that the total content of sterol extracted with mixed solvent was higher than those extracted with ethanol. The ratio of hexane to ethanol at 1:1 (v/v) gave higher content of total sterol, stigmasterol and campesterol, whereas higher content of -sitosterol was produced at the solvent’s ratio of hexane to ethanol at 1:2 (v/v).

Abstract

Numerous studies showed that chlorophyll and its derivatives had antioxidant activity. This research was conducted to obtain the chlorophyll-rich suji leaf liquid extract (SLE) and then to investigate the oral administration this extract on the antioxidant status by means of animal study. The use of Tween 80 0.75%  in sodium citrate 12 mM solution as an extraction solution combined with 30 minutes incubation time at 70-75oC increased significantly the total chlorophylls content, the water-soluble chlorophylls content and the antioxidant activity of the yielded extract, compared to the suji leaf water-only extract.  Two-month oral administration of SLE  to male Sprague Dawley rats showed that the liver MDA level significantly decreased, and the liver catalase and superoxide dismutase antioxidant enzyme activities significantly increased compared to the control group. It was suggested that the chlorophyll content in SLE might increase the antioxidant status of animal tested.

Abstract

Coronary heart disease is caused among others by atherosclerosis, which is the result of oxidized low density lipoprotein (LDL) and cholesterol accumulation on the macrophage. This were reported to be inhibited by temulawak (Curcuma xanthorizza Roxb). The objective of this study was to find out the types and consentrations of temulawak extract which could inhibit LDL oxidation, and to find out the effect of temulawak extract on the accumulation of cholesterol on macrophage. Temulawak was extracted by water, ethanol, aceton and dichlorometane. Inhibition of LDL oxidation was analyzed by measuring the level of malonaldehyde content of the oxidized LDL-CuSO4 which were given water extract, ethanol extract, aceton extract and dichlorometane extract. of temulawak at concentrations of 43 ?g, 430 ?g, and 4300 ?g per ml of LDL. The percentage of malonaldehyde reduction due to addition of water, ethanol, acetone and dichloromethane extract were 44.27; 47.68; 51.83 and 61.2 respectively. The inhibition of LDL oxidation by temulawak extract depends on the concentrations. The percentage of malonaldehyde reduction due to addition of temulawak extract of 43 µg, 430 µg, and 4300 µg per ml of LDL were 43.63; 56.72; and 53.89 Concentrations of temulawak extract resulting in the highest inhibition of LDL oxidation was 430 µg/ml LDL. Temulawak extract tends to inhibit cholesterol accumulation on the macrophage. There is a correlation between the inhibition of cholesterol accumulation on the macrophage and the inhibition of LDL oxidation by temulawak extract. Abstract

Stress condition has beeb reported to decrease intracellular antioxidant-superoxide dismutase(SOD) in the liver and kidneyof rats. This study was conducted to evaluate the antioxidant activies of ginger oleoresin on the profile of superoxide dismutase(SOD) in the kidney of rats under stress condition. The stress condition was achieved by five days of fasting together with swimming for 5 min/day. Ginger oleoresin was orally administrated in a dose of 60 mg/KgBW/day for seven days. Drinking water was provided ad libitum to all groups. The treatment of ginger oleoresin significantly decreased malondialdehyde (MDA) levels and increased SOD activity, as well as immunohistochemicall, increased the content of copper, zinc-SOD (Cu, Zn-SOD) in the kidney tissues compared to that of untreated group. The antioxidant content in ginger oleoresin such as shogaol, zingeron, and gingerol, etc. were shownto have activities in the kidney tissues of rats under stress condition that is increasunf the profile of SOD. Ginger oleoresin treatment in combination both before and after stress gave the best result. Key words : antioxidant, ginger oleoresin, kidney, stress, superoxide dismutase (SOD)

Abstract

The aim of this study was to evaluate acute toxicity of methanol extract of Sargassum echinocarpum on male and female of mice (Mus musculus) strain BALB/c two month age. S. echinocarpum was obtained from Talango Island in April 2008. The powder of S. echinocarpum was macerated by methanol (1:3) at 4 °C for 3 x 24 hours, then concentrated, dried, and extracted. The extract was screened to observe the phytochemical compounds. For acute toxicity extracts was given orally and once on one hour before feeding. The dose treatments were 0; 625; 1250; 2500; and 5000 mg/kg body weight (BW). Parameters measured were phytochemical contain, body weight during the study, 50 percent of lethal doses, and histology score of liver and kidney. The phytochemical study showed that methanol extract of S. echinocarpum contained tannins, polyphenols, saponins, glycosides, and steroids. On 1250 mg/kg BW or more, methanol extract of S. echinocarpum inhibited the increasing of body weight on mice, but, until 5000 mg/kg BW, it did not cause mortality on mice. On 1250 mg/kg BW or more, methanol extracts caused necrosis on hepatocyt and tubulus.Key words: acute toxicity, methanol extract, Sargassum echinocarpum

Abstract

Tempe merupakan produk olahan fermentasi kedelai asli Indonesia. Telah diketahui bahwa kandungan gizi tempe lebih baik dibandingkan kedelai yang tidak difermentasi. Masalah utama tempe adalah umur simpan yang relatif rendah. Salah satu alternatif pengolahan tempe adalah tepung tempe. Tujuan penelitian ini adalah untuk mengevaluasi kualitas protein tepung tempe yang terbuat dari kedelai impor dan lokal. Penelitian ini menggunakan tikus putih Sprague-Dawley sebagai hewan model. Tikus dibagi 4 kelompok berdasarkan sumber protein dalam ransum, yaitu tepung tempe kedelai impor Genetically Modified Organism (GMO)/hasil rekayasa genetika, tepung tempe kedelai impor non-GMO, tepung tempe kedelai lokal Grobogan, dan kasein sebagai kontrol. Parameter kualitas protein diukur berdasarkan metode pertumbuhan dan metode keseimbangan nitrogen. Hasil analisis menunjukkan tidak terdapat perbedaan yang nyata pada nilai food convertion efficiency (FCE), protein efficiency ratio (PER), dan net protein ratio (NPR) dari semua jenis tepung tempe. Nilai true protein digestibility (TPD) tepung tempe kedelai grobogan dan non-GMO tidak berbeda nyata, namun nyata lebih tinggi dari tepung tempe kedelai GMO, dan lebih rendah dari kasein. Tidak terdapat perbedaan yang nyata pada nilai biological value (BV) dan net protein utilization (NPU) semua sampel. Dari penelitian ini dapat disimpulkan bahwa secara umum nilai gizi protein tepung tempe kedelai lokal Grobogan tidak berbeda dengan tepung tempe kedelai impor non-GMO.Tempe is a fermented soybean product from Indonesia. It has been known that nutritional values of tempe are better than unfermented soybean. The main problem of tempe product is its short shelf life. An alternative way to solve this problem is tempe flour. The objective of this research was to evaluate the protein nutritional quality of tempe flours made from imported and local soybeans. This research used albino Sprague-Dawley rats as an animal model. The rats were divided into 4 groups based on protein source of the diet, namely: tempe flour of imported Genetically Modified Organism (GMO) soybean, tempe flour of imported non-GMO soybean, tempe flour of local Grobogan soybean, and casein as a control. Protein nutritional parameters were observed based on growth of rats and nitrogen balance methods. The results showed that there were no significant different of food conversion efficiency (FCE), protein efficiency ratio (PER), and net protein ratio (NPR) values from all of tempe flours. True protein digestibility (TPD) value of Grobogan and non-GMO tempe flours was not different, but higher than GMO soybean tempe flour, and lower than casein. There were no significant different of biological value (BV) and net protein utilization (NPU) values from all samples. This research concluded that generally the nutritional quality of protein of tempe flour from local Grobogan soybean tempe flour was not different from the protein quality of import non-GMOZT.

Abstract

Margarine is a water in oil (w/o) emulsion product which is widely used for household cooking and baking industry. Consuming of margarine, which contains trans fatty acid may cause health problem due to the increase of LDL cholesterol. Since margarine is also a good carrier of phytosterol which prevent the absorption of cholesterol, there is a possibility to formulate a healthier margarine. In this research formulation and characteristics of products was investigated. The research work consisted of two steps: (1) supplementation of wheat germ sterol into margarine (two methods) and  (2) analysis of physical, chemical characteristics and hedonic score. Parameters of physical characteristics were melting point and emulsion stability, whereas chemical characteristics were water and oil contents. The hedonic test was carried out based on product’s color, odor, taste, texture, and spreadability. Results showed that method II of supplementation produced better margarine than method I, in which the concentration of sterol in the margarine was higher with a melting point similar to that of control, better emulsion stability, and higher hedonic score. Supplementation process was carried out by mixing sterol into fat phase melted at 50 0C, followed by mixing with aqueous phase at 4 0C. Sterol used for method II was extracted using mixed solvent of hexane and ethanol at the ratio of 1:2 (v/v), which was resulted from previous experimentation.

Abstract

Premenopause is part of natural aging process in women indicated by the decrease of ovarial estrogen production, and usually followed by development of menopausal syndrome. Susumeno, skim milk fortified with 100 mg of soy isoflavone and 8 mg of Zn was given to premenopausal women for decreasing menopausal syndrome. This research aimed to evaluate the effect of susumeno on menopausal syndrome in premenopausal women. About 33 premenopausal women aged 43-52 years old in Purwokerto were choosen as respondent. Their menopausal syndrome were identified by using questioner. The answers were scored from 1 to 5 ; i.e., 1 = not detected; 2 = less detected; 3 = mildly detected; 4 = detected; 5 = much detected. It was revealed that some of menopausal syndromes had significantly decreased, especially vaginal dryness from less detected to not detected (p=0,01); fatigue from mildly detected to not detected (p=4,07E-05); mindless from mildly detected to not detected (p=0,022); and dyspareunia from less detected to not detected (p=0,036).
