Abstract

Decline quality of Tuna as a result of abuse temperature and long shelf life time. Histamine, microbial and sensory odours become one of the global problems that can cause health problems. Large number of bacteria dominated in viscera area rather than other fish parts. This study was conducted to determine the effect of storage temperature on histamine, microbial and sensory odours on middle of Yellowfin Tuna. This study used descriptive explorative method, sampel were periodically taken for analyses at intervals 48 hours (0ºC), 8 hours (10ºC) and 4 hours (25ºC). Yellowfin Tuna was rejected earlier by the sensory odours than TPC and histamine. During storage at 0ºC for 720 hours histamine levels still acceptable for consumption however TPC already exceeded the limit after 672 hours and rejected by sensory odours for 624 hours storage. During storage at temperature 10ºC did not reached the limit for 120 hours while TPC value already reached 2x106 cfu/g for 88 hours storage and rejected by sensory odours for 72 hours storage. During storage at temperature 25ºC histamine reached 67.3 ppm for 32 hours, ALT reached 5.5x105 cfu/g for 24 hours and rejected by sensory odour for 20 hours of storage. Histamine formation correlates with the growth of microbial counts and decrease of sensory odours value. Suhu yang tinggi dalam waktu simpan lama berpengaruh terhadap penurunan kualitas ikan tuna. Histamin, mikroba dan bau busuk menjadi salah satu permasalahan global yang dapat menyebabkan masalah kesehatan. Bagian tengah ikan menjadi sumber bakteri paling tinggi dibandingkan bagian lainnya. Penelitian ini bertujuan untuk mengkaji perkembangan histamin, ALT dan sensori bau pada bagian tengah Tuna sirip kuning dalam penyimpanan suhu yang berbeda. Penelitian ini menggunakan metode deskriptif eksploratif, sample di analisis secara berkala pada suhu 0ºC (48 jam), 10ºC (8 jam) dan 25ºC (4 jam). Tuna sirip kuning mengalami pembusukan lebih awal berdasarkan parameter sensori bau, kemudian disusul ALT dan Histamin. Kandungan histamine pada suhu 0ºC masih layak untuk dikonsumsi setelah 720 jam, namun nilai ALT melebihi batas aman setelah 672 jam penyimpanan dan nilai sensori bau setelah 624 jam penyimpanan. Penyimpanan suhu 10ºC tidak menyebabkan peningkatan histamin melebihi batas limit setelah penyimpanan selama 120 jam, sedangkan nilai ALT mencapai 2x106 cfu/g setelah 88 jam dan nilai sensori bau menyatakan ikan busuk setelah penyimpanan selama 72 jam. Penyimpanan suhu 25ºC histamin mencapai 67,3 ppm setelah penyimpanan 32 jam dan nilai ALT 5,5x105 cfu/g pada penyimpanan ke 24 jam serta ikan dinyatakan busuk berdasarkan sensori bau setelah 20 jam penyimpanan. Peningkatan histamin berkorelasi dengan pertumbuhan jumlah mikroba dan penurunan nilai sensori bau.

Abstract

Information about oceanographic conditions is important to determine the fertility level of waters. Oceanographic parameters in water can be influenced by global climate factors, one of them is the ENSO (El Niño Southern Oscillation) phenomenon. There have been many studies on the effect of ENSO phenomenon on oceanographic variability, but no studies have been carried out in the waters of North Sulawesi. This study aims to determine the effect of the ENSO phenomenon on the variability distribution of oceanographic parameters in North Sulawesi waters. The data used for this study were Sea Surface Temperature (SST) and chlorophyll-a from the AQUA-MODIS imaging results, wind reanalysis results from ECMWF, and Nino 3.4 index as an indicator of ENSO from NOAA. The data were processed and analyzed using quantitative analysis methods in the form of graphics. The results showed an indirect effect of the ENSO phenomenon on SST parameters and chlorophyll-a. This is because the effect of the ENSO phenomenon occurred in a certain period:  when strong El Niño triggered low temperatures of sea surface and high chlorophyll-a, and when La Niña was strong it triggered high temperatures of sea surface and low chlorophyll-a. Meanwhile, the wind speed pattern showed an insignificant effect because the wind speed was still dominated by the influence of the monsoon pattern.Indonesian title: Hubungan antara El Niño Southern Oscillation (ENSO) dan parameter oseanografi di perairan Sulawesi Utara

Abstract

Nine unidentified marine sponges from Bunaken Island waters in Manado Indonesia were observed on anti-tuberculosis (TB) against non-pathogenic Mycobacterium smegmatis NBRC 3207 and it was found that the ethanol crude extracts and ODS fractions of these sponges inhibited the growth of bacteria M. smegmatis. Sponge SP.323 is the most inhibiting and showed Intermediate (I) activity with inhibition zones of 9, 11, 11, 8 mm/disk at 10µg/disk., for fraction 3,4,5,6 respectively, and need further experimental analysis to justify the antimicrobial activity, NMR analysis and structure elucidation.

Abstract

Although heavy metals have relatively low levels in water column and sediments, they will not be degraded and can even be absorbed and accumulated by marine algae. Research on accumulation of heavy metals on algae was done in Kima Bajo Waters, North Minahasa Regency. The research objective was to evaluate the accumulation of four heavy metals, i.e. arsenic (As), cadmium (Cd), plumbum (Pb), and mercury (Hg), on brown macroalgae Padina australis. The alga was introduced and cultivated in the research area using bottom method. The same method was also used in the experiment. One-week acclimatization process was done prior the experiment. Sampling of alga’s thallus was carried out every two weeks to measure the concentration of metals. The experiment was terminated after six weeks. Bottom sediment was also collected at the beginning of the experiment to measure the metal concentration. The results showed that the heavy metals (As, Cd, Pb, and Hg) were accumulated in the algae. The highest concentration of Cd, Pb, and Hg accumulated in the algae was 0.15 mg/kg wet weight, 3.5 mg/kg wet weight, and 0.009 mg/kg wet weight, respectively. All the concentrations were higher than the initial concentration of each metal. Accumulation of As was also occurred with the highest concentration (1,9 mg/kg wet weigh) occurred at fourth weeks; however, it was lower than the initial concentration. The accumulation varied according to type of metals and time; accumulation of Cd, Pb, and Hg were occurred on second weeks of cultivations and As on the fourth weeks. Abstract

There are a large variety and quite abundant types of small pelagic fish that have high economic value in the Sulawesi Sea. One of which is the blue scad fish or commonly known as malalugis (Decapterus macarellus). This study aims to analyze and determine the status of scad fisheries management and to develop recommendations in the management of scad fisheries in the waters of Sulawesi Sea, North Sulawesi Province. The evaluation of fishery management status is carried out using multi-criteria analysis (MCA) through the development of composite index of each indicator of Ecosystem Approach to Fisheries Management (EAFM). The results showed that, in general, the status of scad fisheries management in North Sulawesi Province, was in ‘good’ category, specifically reviewed from the domain of fish resources fall into the category of “medium”, habitat and ecosystem “good”, fishing techniques “good”, economy “not good”, social “medium” and institutional “good”. Management actions take precedence over domains that have “poor” indicator values. Abstract

Ascidian is a benthic invertebrate that produces secondary metabolites. The substances were produced by the ascidian as self-defense from many environmental factors. Several studies report the similar structure between the substances extracted from ascidian and symbiotic fungi with biological activities. This study aims to isolate fungi associated with ascidian Eudistoma sp., then observe its potency on inhibiting bacteria S. aureus dan A. hydrophila. PDA was used to isolate and culture fungal isolate, then extraction using solvent ethyl acetate. Antibacterial activity of this symbiotic fungi isolates extract was performed using the Kirby-Bauer disc diffusion method. Molecular identification of fungi results that the symbiotic fungi were Schizophyllum commune. The observing result showed that fungal extract has the ability on inhibiting the growth of S. aureus and with the inhibition zone 8 mm and A. hydrophila 8,5 mm.Keywords: Ascidian, Eudistoma sp., Schizophyllum commune, Symbiotic, Antibacterial

Abstract

Nudibranch are part of a class og Gastropods that do not have a shell and has a bright and striking colour. Nudibranch has different of bacteria and bioactive compounds contained therein for protect theselves from predators.The aim of this study were to isolate symbiotic bacterial drived from Thurudilla lineolata and Phyllidiella pustulosa, and to observe the antibacterial activity of bacterial isolate against Escherichia coli and Staphylococcus aureus. Isolation and culture of the symbiotic bacterial were made on Nutrient Agar. The antibacterial screening showed that Thrudilla lineolata and Phyllidiella pustulosasymbiotic bacteria were able to inhibit the growth of against Escherichia coli and Staphylococcus aureus Nudibranch merupakan bagian dari kelas Gastropoda yang tidak memiliki cangkang dan memiliki warna cerah dan mencolok. Nudibranch memiliki berbagai jenis bakteri dan senyawa bioaktif yang terkandung di dalamnya untuk melidungi diri dari predator. Tujuan dari penelitian ini yaitu untuk mendapatkan isolat bakteri yang bersimbion dengan Thrudilla lineolata dan Phyllidiella pustulosa, kemudian mengamati aktivitas antibakteri dari isolat bakteri tersebut terhadap Escherichia coli, dan Staphylococcus aureus. Isolasi dan kultur bakteri yang bersimbion dengan Thrudilla lineolata dan Phyllidiella pustulosa, dibuat pada media Nutrient Agar. Skrining aktivitas antibakteri menunjukkan isolat bakteri yang bersimbion dengan Thrudilla lineolata dan Phyllidiella pustulosa mampu menghambat pertumbuhan organisme uji Staphylococcus aureus dan Escherichia coli. Abstract

Kecap ikan adalah salah satu produk hasil fermentasi dengan bahan baku ikan. Ikan yang digunakan dalam penelitian ini adalah ikan tongkol ( Euthynnus Affinis C. ) yang merupakan hasil tertinggi komoditi perikanan di Sulawesi Utara yang memiliki kandungan protein, asam lemak omega-3, vitamin dan mineral yang tinggi dan juga sangat kaya gizi. Penelitian ini bertujuan untuk mendapatkan konsentrasi terbaik dari buah nenas yang ditambahkan dan meningkatkan pengaruh konsentrasi buah nenas terhadap lama fermentasi dan profil asam amino. Penelitian menggunakan penambahan sari buah nanas dengan konsentrasi (0%, 10%, 15% dan 20%) dan garam 10% dengan lama penyimpanan selama 10 hari dalam inkubator.Variabel yang diamati dalam penelitian ini adalah Asam Amino, Kadar Protein, Kadar Karbohidrat, Kadar Lemak, Kadar Nitrogen, Kadar Abu, Kadar Air, dan pH. Hasil penelitian menunjukkan bahwa sampel kecap ikan sari buah nanas 15% dengan garam 10% dapat mempercepat proses hidrolisis pada hari ke-10. Dari mutu kimiawi, hasil laporan asam amino LC / MS didapatkan 9 kandungan utama yang ada pada kecap ikan tongkol 15% yaitu: L-Leucine, L-Valine, L-Tryosine, L-Cystine, AABA, L-Alanine, L-Glutamic Asam, Glisin, dan L-Serin.Hasil analisis kandungan proksimat (kadar air 61,3%, kadar abu 9,22%, kadar lemak 1,52%, kadar protein 3,98%, kadar karbohidrat 3,11%) pada kecap ikan tongkol 15% masih dalam standar mutu kecap ikan yang baik, dan kandungan nitrogen kecap ikan tongkol 15% dapat memenuhi Standar Industri Thailand Grade 1.

Abstract

Glutathion is a tripeptides produced by almost all living creatures, including marine fauna such as rotifers and shrimps. Judging from the important molecular role of glutathion in living cell,  the ability of glutathion as antiviral compound, has been assed by application of the AutodockTools to predict the ability of glutathion as ligand to bind receptors of several viral diseases such as  covid-19, malaria, and hepatitis B. Docking results of glutathion toward target receptor were shown by the parameter, Gibbs energy value, and supported by other data such as the amount of hidrogen bond, amino acid residues, and bond distance. Evidently, the glutathion could bind to  each receptor of covid-19, malaria and hepatitis B, with values of  Gibbs energy (∆G), -7,9, 7,1 and -7,1 kcal/mol, respectively. This proves that glutathion has ability to bind covid-19 receptor, even stronger than its ability to bind receptors of other viral diseases, and to the remdesevir. Between glutathion and covid-19 receptor, there were 11 hidrogen bonds, with distance ranged from 2,76 to 3,25 Å. These were stronger than those between glutathion and receptors of malaria and hepatitis B, only  8 and 4 Å hidrogen bonds, respectively.  To develop glutathion as antiviral drug, it is necessary to carry out in vitro and in vivo analyses, prior to clinical test. Therefore, it requires to explore the marine fauna species that produce glutathion and their cultivation.Keywords: molecular docking, glutathion, marine fauna, autodock vina, viral diseases, binding affinity

Abstract

This research aims to study the anatomical structure of macroalgae. Sample of macroalga was collected by using Line Intercept Transect (LIT) method with sampling quadrat. Macroalgae samples were dried and separated from the roots, stems, leaves and receptacles (if present). Samples were cut transversely and longitudinally at each section and observed under binoculared microscope of Olympus CX with monitor. Based on the histology, anatomical structure of macroalgae species can be divided into two tissues from outside to inside, namely cortex and medullary cells. The cortex is composed of one layer or more. The cortex is the area between the epidermis and the central column. The medulla cells only have one layer which is the largest layer. Based on the observation result of cells type from observed spesies Eucheuma denticilatum, Gracilaria arcuata, Hydropuntia edulis, G. salicornia, Hypnea valentiae, Turbinaria deccurens, and T. ornata, it shows that cells of medulla become small toward cortex.Indonesian title:  Karakteristik anatomi jenis makroalga dari Pulau Bombuyanoi, Kabupaten Bolaang Mongondow Timur, Sulawesi Utara
