Abstract

Background: Gracilaria Seaweed extracts are known to have an antibacterial content thatcan inhibit the growth of Porphyromonas gingivalis bacteria. P.Gingivalis are dominant inchronic periodontitis as a cause of periodontal tissue damage that can be inhibited by usingmaterials that contain antibacterial effect. Terms of materials to be applied in the oral cavitymust be biocompatible, through toxicity tests, one with bioviability test. In this studybioviability test performed on media mesenchymal stem cells. Purpose: To determinebioviability Gracilaria seaweed extract against mesenchymal stem cells as adjuvant therapyof periodontitis. Materials and Methods: study design was post test only control groupdesign. Mesenchymal stem cells in 96 wells were divided into a control group of cells (n=6),media controls (n=6), and treatment (n=6). The treatment group was given Gracilariaseaweed extract with a concentration of 2.5%, 5%, 10%, 20% and 40%. Stem cells wereincubated 24 hours before and after treatment. Once given MTT, optical density read with anELISA reader then calculated the percentage of living cells. Toxic material when living cells<50% and not toxic when living cells >50%. Living cell data were analyzed by statistical testKruskal Wallis and Man Whitney. Result: Viability cell at 2.5% concentration is 68.59%, 5%concentration = 29.67%, 10% concentration = 4.43%, 20% concentration = 2.5% and 40%concentration = 0.96 %. Conclusion: Extract of seaweed Gracilaria proven not toxic at 2.5%concentration and proved toxic at 5%, 10%, 20% and 40% concentration

Abstract

Background: The prevalence of diabetes mellitus (DM) is quite high in the world and Indonesia. Diabetes mellitus with poor glycemic control periodontitis as a complication the largest in the oral cavity which causes damage to the alveolar bone. Osteoblasts synthesize osteoid collagen to form bone as raw material. Oxygenation high pressure is able to increase the amount of alveolar bone osteoblasts in patients with DM. Sea cucumber contains growth factors that play a role in the differentiation of osteoblasts to bone formation. Purpose: To determine the effectiveness of combination hyperbaric oxygen therapy 2.4 ATA and sea cucumber gel 3% in the increasing of osteoblasts in rats with DM induced P.gingivalis. Materials and Methods: The study design was factorial design group. Rats are inducted with STZ single dose 65 mg/kg BB to make DM and are inducted with P.gingivalis 1x109 sel/ml to make them suffer periodontitis. Twenty male Wistar rats were divided into five groups. K-(without treatment); K1 (Streptozotocin + P.gingivalis); K2(Streptozotocin + P.gingivalis + sea cucumber gel); K3 (Streptozotocin+ P.gingivalis + HBO); K4 (Streptozotocin +P.gingivalis +HBO + sea cucumber gel). Result: One-way ANOVA here were significant differences (p<0.05) in K2 (24.97 ± 4.99), K3 (22.80 ± 6.45), K4 (28.47 ± 3.20) that compared with K1 (9.22 ± 4.95). Conclusion: Each hyperbaric oxygen therapy doses of 2.4 ATA 100% O2 3x30 minutes with 5 minute intervals for 7 days and sea cucumber gel (Stichopus hermanii) concentration of 3% and a combination of both therapies showed an increase of osteoblasts significantly in rats with diabetes mellitus induced P.gingivalis.

Abstract

Background: Bacterial mixed periodontopatogen dominated by gram negative. Eucheuma cottonii seaweed as one of the plant species have the antibacterial effect againts gram positive and gram negative bacteria.thus potentially developed as antibacterial agent in periodontal disease. Purpose: This study aimed to determine the inhibition effect of Eucheuma cottonii seaweed extract to the growth of mixed periodontopathogen bacteria concentration of 0,25%, 0,5%, 1% and 2%. Materials and Methods: Eucheuma cottonii seaweed extracted with 95% etanol by soxhlet method. Mixed periodontopathogen bacteria inoculated in BHI medium. Antibacterial effects of extracts of Eucheuma cottonii seaweed againts bacterial mixed periodontopathogen was tested by the diffusion method with the concentrations of 0,25%, 0,5%, 1%, 2%, aquadest and tetracycline, where each group consisted of 5 experiment unit. The effect of the inhibition observed by measuring the diameter of clear area on the disk using digital calipers stated in millimeters. Result: There were inhibition effect at each concentrations of treatment. The diameter zone were concentration of 0.25% (6.184), the concentration of 0.5% (6.356), the concentration of 1% (6.62), the concentration of 2% (7.148) and a positive control with tetracycline (39.336) Conclusion: Eucheuma cottonii seaweed extract has inhibition effect to the growth ofbacteria mixed periodontopatogen.
