Abstract

ABSTRACTSimilarity Level of Patchouli oil from Protoplast Fusion based on Morphology andAnatomy Leaves. Protoplast fusion between Aceh patchouli (Pogostenton cablin Benth)Tapak Tuan 75 clone and java patchouli (Pogostemon heyneanus Benth) Girilaya clone gonegenotypes variation. The aim of this experiment was to examine the similarities ofmorphology and anatomy of leaves. The experiment was inducted at laboratory and greenhouse of the Indonesian Spice and Medicinal Crops Research Institute from February to July2001. The parameters observed were the number of leaves length, leaves width and leafthickness, the number of oil cell, stomata and trachoma index. Analysis cluster was used toanalysis the data with single linkage method and Euclidean distance concept. The resultshowed that the leaves length, width, and thickness have close similarity with Tapak Tuan 75.The similarity level of genotype from protoplast fusion was 62.68 to 83.27, the parent TapakTuan 75 was 61.53 and the parent Girilaya 58.69. The highest numbers of oil cell was showedby clone 9 I1 4 (10.55) and the lowest was clone ( IV 14 (2.93).Key words: Pogostemon sp., similarity, protoplast fusion, morphology, anatomy, leaves.

Abstract

Nilam (Pogostemon cablin) merupakan sa-lah satu tanaman penghasil minyak atsiri yang mempunyai peranan penting, baik sebagai sumber devisa negara maupun se-bagai pendapatan petani. Tanaman nilam telah lama dibudidayakan di Indonesia dengan daerah sentra produksi Aceh, Su-matera Utara dan Bengkulu yang meng-alami perkembangan cukup pesat. Minyak nilam digunakan dalam industri parfum, pembuatan sabun kosmetik, antiseptik dan insektisida. Produksi minyak nilam ditentu-kan oleh varietas. Untuk mendapatkan produksi secara kualitas dan kuantitas yang tinggi diperlukan varietas unggul. Ta-hapan penelitian untuk mendapatkan va-rietas unggul dimulai dari eksplorasi ka-rakterisasi morfologi, anatomi dan pro-duksi terna. Penelitian dilakukan di Rumah Kaca Balai Penelitian Tanaman Obat dan Aromatik sejak Januari sampai Desember 2009 dengan menggunakan Rancangan Acak Kelompok (RAK), bertujuan untuk mengetahui karakter morfologi, anatomi dan produksi 10 aksesi nilam asal Aceh dan Sumatera Utara. Parameter yang di-amati meliputi karakter morfologi, anatomi dan produksi terna serta minyak. Berda-sarkan karakter bentuk daun dan batang tidak banyak ditemukan variasi, namun bi-la dilihat dari karakter bentuk pangkal dan ujung daun terdapat variasi, diantaranya berbentuk tumpul, rata, gasal dan runcing. Karakter jumlah sel/kelenjar minyak yang terletak pada sel palisade maupun sel bu-nga karang sangat bervariasi. Potensi pro-duksi terna segar berkisar antara 96,0-319,1 g/phn, tertinggi terdapat pada ak-sesi TM-3 (319,1 g) terendah pada aksesi SK (96,0 g), produksi terna kering berki-sar antara 35,6-97,9 g/phn, tertinggi pa-da aksesi TM-3 (97,9 g) terendah pada aksesi SK (35,6 g). Kadar minyak atsiri berkisar antara 2,52-4,15% per pohon, tertinggi pada aksesi SK (4,15%) dan terendah pada aksesi Sipede 4 (2,52%).
