Abstract

Availability of drugs in Indonesia is still limited by the high prices of drugs due to on the imported raw materials that reaches 95%. Developing antibiotic raw materials can be achieved by increasing of penicillin G production, which is the raw material for the formation of semisynthetic penicillin derivatives through the production of 6-aminopenisillanic acid (6-APA). One of the important enzyme in the penicillin G biosynthesis is Isopenisilin N Synthase (IPNS) that encodes by pcbC gene on Penicillium chrysogenum. This study aimed to obtain a recombinant of pcbC gene fragments that is inserted into pPICZA plasmid. Amplification of pcbC gene used pcbC-F and pcbC-R primers. The pcbC gene fragment was inserted into pPICZA vector and then transformed into TOP 10 F’. The results showed that the recombinant of the pcbC gene fragment from P. chrysogenum has been obtained. Analysis of DNA sequences using the BLAST program showed that the pcbC gene fragment has high homology (99%) with the  pcbC gene from P. chrysogenum Wisconsin 54-1255 and P. chrysogenum AS-P-78 which encodes IPNS   Keywords: pcbC Gene, Penicillium chrysogenum, cloning, penicillin G

Abstract

Motivation to become a teacher is important information to be used in preservice teacher education programs. The purpose of this study was to determine the relationship between demographic factors and motivation to choose teachers as career choices in teacher education students. The type of research used in this research is survey. The research subjects were students of the second year at the Department of Biology Education, Universitas Negeri Yogyakarta. The results showed that hometown of students had a relationship with motivation to become a teacher, students from city areas had higher scores. The results of the analysis on each aspect indicate that hometown of students has a relationship with the aspect of job security (Sig = 0.036) and gender has a relationship with the aspect of work with children and adolescents (Sig = 0.042). In addition, gender and hometown of students have a relationship with aspects of enhancing social equality (Sig = 0.015; Sig = 0.021). The conclusion in this study is that the student's hometown has a relationship with their motivation to become a teacher as a career choice, while gender and family background have no relationship.

Abstract

Compost is one of the potential microbial habitats for the discovery of a wide range of novel biocatalysts. Compost sample originally made of plant litter, were collected at the Expo Park (Osaka, Japan). The temperature of composts is around 50 °C. Metagenomic DNA was isolated from compost and was used to amplify partial 16S rRNA genes (rDNAs) by polymerase chain reaction for investigation of the microbial diversity in the compost. Molecular microbial identification was carried out by metagenome analysis of 16S rRNA using MinION Next Generation Sequencing (NGS). The results indicated that branch-leaf compost sample from Expo Park is dominated by thermophilic microorganism. The majority of bacteria are Caldicoprobacter, Hungateiclostridium, and Thermoclostridium. To conclude, thermophilic microorganism is not only found in extreme environment, they also presence even in moderate temperature.

Abstract

Purpose: The aim of this study was to obtain a primer design capable of amplifying the PhtD gene from various S. pneumoniae bacterial serotypes as a protein-based pneumonia vaccine candidate that does not depend on bacterial serotypes.Methods: Primary preparation was carried out through a bioinformatics approach using several software followed by testing the use of primers in the amplification process which was carried out in silico.Results: The results of this study were a pair of primers capable of amplifying the PhtD gene from several S. pneumoniae serotypes.Conclusion: The PhtD gene of the 6 serotypes of S. pneumoniae has high similarities, making it suitable for use as a protein-based vaccine candidate. Based on the consensus of the six serotypes, a pair of primers was obtained that were able to amplify 17 S. pneumoniae strains from several serotypes contained in the reference sequence in silico.
