Abstract

ABSTRACTNitrification rate in a closed recirculating water system with different stocking density of common carp (Cyprinus carpio L.) had been observed in the Laboratory of Aquaculture, Faculty of Fisheries and Marine Science, Mulawarman University.   Fish were cultivated  in rounded tanks with four level of stocking densities i.e. 10, 20, 30 and 40 fish/100 l in a closed recirculting water system, and fed on a commercial pellet given thrice a day at a quantity of 5% from the total body weight of fish per day.  The experiment was designed completely randomized with three replications. Nitrification rate was calculated stoichiometrically through the determination of  ammonia and nitrite oxidation rate. Results showed that in this experiment the ammonia and nitrite oxidation rate, and nitrification rate in a closed recirculating water system was increased with the increasing stocking density of fish.   On the contrary, the growth of fish was decreased with the increasing of stocking density.Key words :  Nitrification rate, stocking density, recirculation system, common carp (Cyprinus carpio L.). ABSTRAKLaju nitrifikasi dalam budidaya sistem resirkulasi air tertutup dengan padat penebaran ikan mas (Cyprinus carpio L.) yang berbeda telah diamati di Laboratorium Budidaya Perairan, Fakultas Perikanan dan Ilmu Kelautan Universitas Mulawarman.  Ikan dipelihara dalam tong plastik dengan empat tingkat padat penebaran, yaitu 10, 20, 30 dan 40 ekor/100 l, dan diberi makan pelet komersial tiga kali sehari dengan jumlah 5 % dari berat badan ikan per hari.  Penelitian ini dirancang secara acak lengkap dengan tiga ulangan.  Laju nitrifikasi dihitung secara stoikiometrik melalui penentuan laju oksidasi amoniak dan nitrit. Hasil penelitian menunjukkan bahwa laju oksidasi amoniak dan nitrit serta laju nitrifikasi dalam sistem resirkulasi air tertutup meningkat dengan meningkatnya padat penebaran.  Sebaliknya pertumbuhan ikan menurun dengan meningkatnya padat penebaran.Kata kunci :  Laju nitrifikasi, padat penebaran, sistem resirkulasi, ikan mas (Cyprinus carpio L.).

Abstract

This study aimed to determine the effect of Pseudomonas ECP and ICP injection incubated on TSA and TSB media with incubation periad of 72 hours on the nila tilapia (O. niloticus) haematology. Parameters observed were hemoglobin, hematocrit, erythrocyte total and of leukocyte total. The ECP or ICP ware injected into intraperitial nila tilapia as much as 0.2 ml/fish. Observations were made on 6, 12, 24, 48, 72, 96, 120, 144, 168 hour after injection. Extracellular products were obtained by centifuging Pseudomonas suspension grown on TSA and TSB media (incubation time 72 h + 30oC) using a 10.000 g centrifuge for 30 min at 4oC. Supernatant was filtered using filter paper 0.45 μm. To get the ICP, supernatant was centrifuged in room temperature. The results revealed that ICP injection caused the decrease of haematology faster than ECP injection. This indicates that the ICP Pseudomonas more virulent than ECP.

Abstract

This study was conducted to determine the effects of different levels of vitamin C (ascorbic acid) supplementation on NVC (Nutrition Value Coeficient), clinical signs, pathological change, phagocytic index, antibody, and survival rate of fingerlings common carp (Cyprinus carpio L.). Three diets were formulated incorporating vitamin C at levels of 0 (control), 250 and 500 mg/ kg. Fish were randomly fed with the experimental diets for 35 days. On 15th day fish were injected intra muscular with 0.1 ml/fish, 106 CFU/ml concentration of A. hydrophila. The present study suggest that vitamin C concentration 250 mg/kg of diet most appropriate concentration, can enhance fish health (NVC), immune system (phagocytic index and antibody), survival rate, normal clinical signs and pathological change after infected by A. hydrophila.

Abstract

Tilapia is one type of freshwater fish that has high economic value. With the increasing demand by the community, tilapia is cultivated in a semi-intensive system so that bacterial diseases caused by Aeromonas hydrophila arise. One way to treat disease in fish safely is the use of lactic acid bacteria. The purpose of this study was to examine the isolates of lactic acid bacteria in controlling diseases caused by Aeromonas hydrophila in tilapia. This study used a completely randomized design experimental method with 4 treatments and 3 replications. The treatments consisted of feed mixed with Phosphate Buffered Saline (PBS), Enterococcus faecalis isolate, Lactobacillus plantarum isolate and Lactococcus lactis isolate at a dose of 0.1 mL/g of feed at a concentration of 106 CFU/mL and mixed with 0.1 mL of egg white. The results showed that the treatment with lactic acid bacteria affected the condition of clinical symptoms and anatomical pathology of tilapia, but was better than the control treatment. The feed mixed with lactic acid bacteria also had an effect on increasing the immune response and showed higher survival than the control treatment. The condition of water quality in the rearing media showed the normal range for the life of tilapia.

Abstract

This research was conducted to determine the effect of inulin on the efficiency of feed utilization and the hematological parameters of red tilapia. This study used a completely randomized design (CRD), with 4 treatments and 3 replications. The doses of inulin used in the treatment were 0,00 g/kg, 1,00 g/kg, 2,00 g/kg and 4,00 g/kg. Tilapia with body length of 6,00-6,50 cm and weight of 4,00-4,50 g were reared in a plastic container with a volume of 40 l of water in a semi-closed water recirculation system for 30 days. Fish were fed with treatment using the at satiation method with a frequency of 3 times a day. The results showed that the administration of inulin had no effect on the efficiency of feed utilization (p>0,05), but gave the best value on hemoglobin levels, hematocrit levels, total erythrocytes, total leukocytes, lymphocyte levels, monocyte levels, and neutrophil levels at dose 2,00 g/kg (p<0,05).

This research was conducted from March to April 2022 at the Laboratory of Microbiology and Aquaculture Biotechnology, Faculty of Fisheries and Marine Sciences, Mulawarman University. This study used a completely randomized design (CRD) with 4 treatments with different lactic acid bacteria and 3 replications of the same lactic acid bacteria. used, namely: P0 (PBS), P1 (Enterococcus feacalis), P2 (Lactobacillus plantarum), P3 (Lactococcus lactis) with a dose of 106 CFU/mL and mixed with 2% egg white given for 14 days. The results showed that lactic acid bacteria supplementation able to relieve clinical symptoms and anatomical pathology, increase survival compared to the control treatment in this study, the highest survival rate was 100% in the Enterococcus feacalis treatment. (P>0.05) and the lowest total colony average was 7.45 x 103 CFU/mL, namely the treatment with Enterococcus faecalis and the water quality parameters were still within normal limits.
