Abstract

Rheumatoid Arthritis (RA) is autoimmune disease that attacks joints and synovial tissues. Rheumatoid Arthritis is characterized by inflammation that involves production of cytokines such as Interleukin 6 (IL-6). Increasing of interleukin 6 has correlation with activity and progressivity of the disease. This research was designed to know whether IL- 6 could be used as indicator progressivity of RA. The method that used for measuring IL-6 level is ELISA (Enzyme Linked Immunosorbent Assay), using the principle antigen-antibody binding. In early stage there were 9 persons (S01, S02, S03, S04, S07, S08, S09, S10 and S14) with of IL-6 level (mean : 3.640 pg/mL), intermediate stage there were 3 persons (S06, S13 and S15) with IL-6 level (mean : 12.453 pg/mL), late stage : 3 persons (S05, S11 and S12) with IL-6 level (mean : 59.349 pg/mL).

Abstract

The research was designed to find out the toxicity of 2-methoxyethanol (2-ME) to the number of spermatogenic cells. Ten mice divided into 2 groups, control group was administered by 0.5 % CMC (negative control) and treatment group were administered by 2-ME dose 100 mg/kg body weight. Days thirteenth until sixty thirth, the mice were administered by aquabides orally, andat  the end of the treatment, the mice were sacrified.  The number of spermatogenic cells were observated histologically with Hematoxylin-Eosin staining. The observation shows that 2-ME decreases the number of spermatogenic cells, especially spermatocyte I. Exposure of 2-ME destructs anatomy of tubulus seminiferusin testicular, which is affects degradation of ephitellium and atopy of testicular.

Abstract

Insiden kandidiasis sangat tinggi di dunia yang disebabkan Candida albicans. Agen antifungi telah banyak dikembangkan, namun resistensi terhadap antifungi masih menjadi suatu permasalahan. Resistensi antifungi ini diakibatkan oleh kemampuan C. albicans dalam membentuk biofilm. Terbentuknya biofilm dari infeksi C. albicans diawali oleh proses inflamasi. Proses inflamasi oleh Candida albicans terjadi pada beberapa tahap yang salah satu indikatornya adalah dilepaskannya sitokin proinflamasi (IL-6) dan anti inflamasi (IL-10). Penelitian ini bertujuan untuk mengetahui kadar IL-6 dan IL-10 pada tiap tahapan inflamasi C. albicans pada intestinal pada tikus putih. Penelitian ini menggunakan 32 ekor tikus wistar (Rattus norvegicus) yang dibagi menjadi 2 kelompok, yaitu kelompok kontrol dan perlakuan. Kelompok perlakuan diinokulasi dengan C. albicans. Variabel yang diukur adalah kadar IL-6 dan IL-10 pada hari ke-7, 14, 21, 28 dan 35 setelah inokulasi C. albicans. pengambilan data sebanyak 5 kali dilakukan untuk mewakili tahapan pembentukan biofilm candida. Respon imun pada setiap tahap pembentukan biofilm ditunjukkan dengan kadar sitokin IL-6 dan IL-10 serum darah dengan metode ELISA. Hasil penelitian menunjukkan bahwa kadar IL-6 dan IL-10 pada hari ke-7 dan 14 sebesar 0pg/mL. Pada hari ke 28 telah terjadi proses inflamasi akut  pelepasan IL-6 dan IL-10 dengan kadar tertinggi masing-masing 31,75±9,99pg/mLdan 757,94±576,73pg/mL. Selanjutnya pada hari ke-35, kadar sitokin IL-6 dan IL-10 masing-masing mulai menurun menjadi 28±11,53pg/mL dan 349,5±188,48pg/mL. Respon imun tubuh mulai terjadi pada tahap awal pembentukan biofilm C. albicans intestinal tikus, sehingga kedepannya dapat dikembangkan terapi imunomodulator terhadap kandidiasis sehingga dapat menghambat pembentukan biofilm C. albicans.

Abstract

High doses of paracetamol create necrosis in the liver and produce free radicals. When liver function decreased in a long time, it will lead to severe liver damage and it will be irreversible. Rhizome of Curcuma zedoaria has the potential effect as an antioxidant, it is assumed that  its properties inhibit the formation of free radicals which formed from toxic doses of paracetamol. The aim of this study was to examine the histological structure of the liver and to determine MDA levels in the administration of C. zedoaria toxic dose and paracetamol on the Rattus norvegicus. The study was used twenty-four rats divided into four groups (positive control: CMC 0.5%; negative control: paracetamol 1.35g/kg body weight; treatment group 1 (T1): C. zedoaria 105 mg/200g and paracetamol 1.35g/kg Body weight 2 hours later, and treatment group 2 (T2): paracetamol 1.35g/ kg Body weight and C. zedoaria 105 mg/200g 2 hours later).  The Kruskall-Wallis test results showed MDA level did not significantly different between groups (p = 0.087). Hepatocellular changes were observed descriptively with Hematoxylin-eosin staining. Positive control showed greater hepatocellular changes rather than other groups, hepatocyte cells were enlarged with cytoplasm showing eosinophilic granules infiltrates, enlarged irregular nuclei, nucleolus prominent, there are many necrosis cells.

Abstract

Indonesia sebagai negara dengan penduduk berusia 15-44 tahun sebanyak 46,87% dari total penduduk, terdapat lonjakan kasus positif COVID-19 yang mencapai 797.723 kasus per 7 Januari 2021. Kelompok usia muda diduga menjadi kelompok penyumbang kasus infeksi tanpa gejala atau asimtomatis. Pasien asimtomatis umumnya tidak menyadari bahwa mereka berpotensi untuk menularkan infeksi dari SARS-CoV-2 penyebab COVID-19. Penulisan literatur review ini menggunakan metode pencarian literatur dari Pubmed dengan kata kunci asymptomatic, COVID-19, cytokine, inflammation. Studi ini bertujuan untuk menelaah sistem imun dari pasien asimomatis dan mengapa pasien tersebut menunjukkan gejala yang lebih ringan.

Abstract

Fast food consumption including used of repeated heating-oil, causing various of health problems, especially the formation of free radicals characterized by elevated MDA levels. Pomegranates have many therapeutic functions, especially as antioxidants that scavenges free radical of oxidative stress thus improving the quality of spermatozoa. The aim of this study was to see the effect of pomegranate juice to reduce oxidative stress and improve sperm motility. This study used 24 rats divided into 4 groups, namely KN, KP, JD1 and JD2. The mean MDA content (nmol / ml) in KN, KP, JD1 and JD2 was 23.35; 8,53; 12.15; and 12,83 followed by Kruskal-Wallis test obtained p value 0,021 (p <0,05). The quality of spermatozoa includes the motility of spermatozoa which distinguished 4 criteria: fast moving spermatozoa and straight forward (criteria 1), spermatozoa move slowly / difficult to go straight (criteria 2), street spermatozoa (criteria 3) and immobile spermatozoa (criteria 4). Mean percentage (%) number of spermatozoa criteria 1 on KN, KP, JD1, and JD2 among others 41,6; 18.2; 38; and 62.2. Mean percentage (%) number of spermatozoa criteria 2 on KN, KP, JD1, and JD2 among others 4,4; 21.8; 15.6 and 14.2. Mean of percentage (%) motility of spermatozoa criteria 3 on KN, KP, JD1, and JD2 among others 10.6; 24.8; 11.2 and 15.2, while for criteria 4 is 21.8; 37.2; 40.4; and 13.8. Statistical test for sperm motility using ANOVA one way, on criteria 1, 2, 3 and 4 each got p value 0,000; 0.010; 0,000; and 0.000, which means the administration of pomegranate juice shows the effect on the motility of spermatozoa.

Abstract

Abstract: Chronic inflammation contributes to the occurrence of various metabolic diseases and even cancer. Chronic inflammation results from excessive response of free radicals. Free radicals are triggered by various agents and oxidation processes in the body as Reactive Oxygen Stress (ROS). The high level of free radicals (oxidants), without adequate intake of antioxidants will lead to oxidative stress conditions. Oxidative stress triggers persistent of inflammation. The formation of ROS is difficult to avoid because it can be generated from cellular metabolic processes. Therefore, we should intake adequate of antioxidants and avoid the various agents induces ROS in everyday life. Keywords : ROS, Stress Oxidative, Inflammation

Abstract

ABSTRAKStaphylococcus aureus adalah bakteri Gram positif yang merupakan  patogen penyebab peningkatan jumlah penyakit dan kematian setiap tahun. Bakteri ini berkolonisasi di hidung dan kulit manusia sehingga menyebabkan berbagai penyakit. Resistensi bakteri patogen terhadap antibiotik merupakan satu permasalahan besar yang membutuhkan strategi khusus dalam penanganannya. Salah satunya dengan penggunaan medan listrik AC digital sebagai pengendali pertumbuhan bakteri patogen S. aureus dan E. coli. Penelitian merupakan penelitian eksperimental dengan biakan bakteri S. aureus dan E. coli yang masing-masing dibagi menjadi 4 kelompok yaitu K0, K1, K2 dan K3 berdasarkan variasi kuat medan listrik AC yaitu 200 V/m, 400 V/m dan 600 V/m dengan waktu paparan 5 menit. Data diambil berupa jumlah koloni bakteri. Hasil penelitian menunjukkan adanya pengaruh kuat medan listrik terhadap pertumbuhan bakteri S. aureus dan E. coli. Semakin besar kuat medan magnet, maka pertumbuhan kedua bakteri semakin terhambat ditandai dengan kecilnya jumlah koloni yang hidup. Hasil uji statistik menunjukkan nilai p < 0,001 dengan uji ANOVA pada E. coli dan p < 0,001 menggunakan uji Kruskal-Wallis pada S. aureus. Kata Kunci: Escherichia coli; jumlah koloni; medan listrik AC; Staphylococcus                        aureus  ABSTRACTStrength of AC electricity for inhibiting of  Staphylococcus aureus and Escherichia coli colony growth. Staphylococcus aureus is a gram-positive bacteria as human pathogen, causing an increase in the number of diseases and deaths every year. The bacteria growing in the nose and skin of humans and can cause some illnesses. Resistance of pathogenic bacteria to antibiotics is one of the biggest problems and the typhical strategies are necessary, such as the use of AC electricity to inhibit the growth of S. aureus and E. coli. This research was an experimental study with S. aureus and E. coli bacteria is divided into 4 groups, each, there are K0, K1, K2 and K3 based on variations in the AC electric field, 200 V/m, 400 V/m and 600 V/m for 5 minutes. The study showed that there is effect of AC electricity to S. aureus and E. coli growth. The stronger AC electricity, the growth of those two bacteria is more inhibited. It is marked with smaller viable colony. Statistical test results obtained p value < 0.001 with ANOVA test on Escherichia coli and p < 0.001 using Kruskal-Wallis test on Staphylococcus aureus. Keywords : AC electricity; colony number; Escherichia coli; Staphylococcus aureus

Abstract

One of the efforts to optimize the utilization of natural materials that is used them as medicinal plants, including Dlingu (Acorus calamus L.) and Garlic (Allium sativum) which have the potential as antioxidants. This study aims to determine the effectiveness of a combination extract of Acorus calamus L. (Dlingu) and Allium sativum (Garlic) on antioxidant activity. This research is an experimental research which is the test of antioxidant content and antioxidant activity test with 5 series of concentrations and 4 times of repetition. The research materials, namely dlingu stem and single clove garlic tubers were extracted, phytochemical screening was carried out and then antiradical free IC50 was determined. Based on the test results of the chemical extract of dligu and single clove garlic, the highest content was alkaloid 5.16%, tannin 4.05%, saponin 3.01%, and flavonoid 2.18%. The test results of the chemical antioxidant activity of extracts dligu and single clove garlic contain IC50 values of 17.062 ppm, including a very strong class of antioxidants.Keywords: antioxidant, Dlingu, Single Clove (Male) Garlic.

Abstract

Penggunaan herbal belakangan banyak ditemukan, terutama sebagai tanaman obat. Daun salam (Eugenia polyantha) adalah herbal yang banyak digunakan sebagai obat. Daun salam memiliki bahan aktif flavonoid, tannin dan vitamin B3 (niasin). Banyak penelitian menyatakan bahwa daun salam bermanfaat sebagai antikolesterol. Hiperkolesterol dapat terjadi dari asupan makanan tinggi lemak yang berkorelasi terhadap terjadinya aterosklerosis dan bermacam masalah kesehatan lainnya. Aterosklerosis sangat terkait dengan peningkatan kadar LDL. Tingginya kadar LDL sebagai faktor risiko aterosklerosis. oleh karena itu penelitian ini bertujuan untuk mengetahui korelasi antara kolesterol dan LDL pada tikus hiperkolesterol yang diobati dengan ekstrak daun salam. Desain penelitian ini adalah penelitian eksperimental dengan hewan coba ditikus jantan yang dibagi mejadi 4 kelompok yaitu KP, KN, P1, dan P2 semua tikus dikondisikan hiperkolesterolemia dengan pemberian diet tinggi lemak. P1 dan P2 adalah kelompok tkus hiperkolesterolemia yang diberikan ekstrak daun salam masing-masing dosis 0,72 dan 1,80 gram. Pada akhir perlakuan semua kelompok diterminasi dan diisolasi darahnya untuk dihitung kadar kolesterol total dan LDL pada tiap ekor tikus. Pada KP; KN; P1 dan P2 didapatkan rerata kadar kolesterol total adalah 37,4; 36,6; 44,8; dan 32,4 mg/dL, sedangka kadar LDL serum adalah 27,8; 23,8; 31,8 dan 25. Hasil uji statistik dengan korelasi Pearson menunjukkan signifikansi (2-tailed) sebesar 0,007 dan Pearson Correation sebesar 1. Dapat disimpulkan bahawa ada korelasi yang kuat antara kadar kolesterol total dan kadar LDL pada serum.
