Abstract

Telah dilakukan penelitian penapisan bakteri penghasil antimikroba pada pasir Pantai Sialang Buah Kecamatan Teluk Mengkudu Kabupaten Serdang bedagai dalam menghambat beberapa mikroba patogen. Tujuan dari penelitian ini untuk mengetahui apakah terdapat isolat penghasil antimikroba, untuk mengetahui aktivitas antimikroba dari isolat bakteri terhadap mikroba patogen dan melihat bagaimana karakterisasi bakteri pada pasir Pantai Sialang Buah Kecamatan Teluk Mengkudu Kabupaten Serdang Bedagai. Penelitian ini dilaksanakan pada bulan Oktober sampai Desember 2020 di Pantai Sialang Buah Kecamatan Teluk Mengkudu Kabupaten Serdang Bedagai. Penelitian ini dilakukan dengan pengujian biokimia sederhana, pengujian fisiologis serta pengujian aktivitas antimikroba. Hasil dari penelitian ini diperolehi 4 isolat sebagai penghasil antimikroba, dengan nama isolat Sb1a (1,70 mm), Sb1b (1,23 mm), Sb2a (0,63 mm), dan Sb4a (0,93 mm). Karakterisasi bakteri isolat pasir Pantai Sialang Buah Kecamatan Teluk Mengkudu Kabupaten Serdang Bedagai pada isolat Sb1a, Sb1b, dan Sb4a ialah bakteri gram positif, dan Sb2a ialah bakteri gram negatif. Semua isolat berbentuk Basil, dapat menghasilkan enzim katalase, menggunakan sitrat sebagai satu-satunya karbon dan energi, bergerak serta dapat memfermentasikan laktosa dan sukrosa. Research has been done to screen for antimicrobial-producing bacteria on the sand of Sialang Buah Beach, Teluk Mengkudu District, Serdang Bedagai Regency in inhibiting several pathogenic microbes. The purpose of this study was to determine whether there were antimicrobial-producing isolates, to determine the antimicrobial activity of bacterial isolates against microbial pathogens and to see how the characterization of bacteria in the sand of Sialang Buah Beach, Teluk Mengkudu District, Serdang Bedagai Regency. This research was conducted from October to December 2020 at Sialang Buah Beach, Teluk Mengkudu District, Serdang Bedagai Regency. This research was conducted by simple biochemical testing, physiological testing and antimicrobial activity testing. The results of this study obtained 4 isolates as producers of antimicrobials, with the names of isolates Sb1a (1.70 mm), Sb1b (1.23 mm), Sb2a (0.63 mm), and Sb4a (0.93 mm). Bacterial characterization of sand isolates from Sialang Buah Beach, Teluk Mengkudu District, Serdang Bedagai Regency, on isolates Sb1a, Sb1b, and Sb4a were gram-positive bacteria, and Sb2a were gram-negative bacteria. All isolates are bacilli, can produce catalase enzyme, use citrate as the only carbon and energy, are mobile and can ferment lactose and sucrose.

Abstract

Tartrazine is a yellow synthetic dye with aromatic and chromo groups. It is widely used in food colouring products and cosmetics, and prolonged consumption can cause kidney damage. Andaliman (Zanthoxylum acanthopodium DC.) is a North Sumatra-native spice plant which reported as antioxidant to repair kidney damage. This study was conducted to determine the effect of ethanol extract of andaliman fruit on the morphology, ureum levels, histology, and glomerular diameter of tartrazine-induced rats. This study employed a completely randomised design (CRD). The ethanol extract of andaliman fruit was dissolved in CMC-Na 1%. Twenty-five rats were divided into 5 groups: negative control (KN), positive control (tartrazine indcution/PN), and treated groups (P1, P2, and P3 induced tartazine and treated with andaliman extract at dose 150 mg/kg BW, 300 mg/kg BW, and 450 mg/kg BW, respectively). After 30 days treatment, kidney damage was observed based on colour and surface of kidneys, kidney index, ureum level, kidney damage score, and glomerular diameter. There are no significant different in the kidney index among groups (p > 0.05). Tartrazin at a dose of 15 mg/kg BW was able to induce the kidney damage as indicated by the increase ureum levels, kidney damage score, and the decrease of glomerular diameter in the negative control. The treatment of andaliman fruit ethanol extract was able to repair the tartazine-induced kidney damage as indicated by the decrease of ureum level and kidney damage score and incread the glomerular diameter.
