Abstract

Aktivitas fisik berat meningkatkan senyawa oksigen reaktif dalam tubuh yang diketahui dengan mengukur kadar malondialdehid (MDA) dari hasil proses lipid peroksidase yaitu kerusakan oksidatif pada biomolekul lipid akibat reaktivitas senyawa oksigen reaktif (SOR), namun pola perubahan kadar MDA plasma seseorang setelah beraktivitas fisik berat masih belum dipahami. Antioksidan potensial seperti astaxanthin dan latihan teratur diduga dapat memengaruhi pola perubahan kadar MDA tersebut. Untuk itu dilakukan penelitian eksperimental dengan subjek 15 orang pria terlatih dan 15 orang pria tidak terlatih anggota sebuah pusat kebugaran di Bandung, usia 18–25 tahun yang terbagi menjadi kelompok yang mendapatkan suplemen astaxhantin atau plasebo selama satu minggu secara buta ganda. Setelah pemberian suplemen atau plasebo, setiap kelompok melakukan tes aktivitas anaerobik berat. Dilakukan pengukuran MDA rerata (mmol/mL) sebelum dilakukan tes, langsung setelah tes, 6 jam setelah tes, dan 24 jam setelah tes. Data yang dianalisis menggunakan uji ANOVA diikuti uji Duncan menunjukkan bahwa sebelum tes keempat kelompok mempunyai rerata yang hampir sama, sementara rerata MDA yang diperiksa pada setelah tes pada kelompok pria terlatih dengan suplementasi astaxanthin memiliki rerata MDA terendah, sementara yang tertinggi pada kelompok pria tak terlatih dengan plasebo (p<0,05). Perubahan rerata MDA pada setiap kelompok menunjukkan pola dinamis sama yaitu meningkat tajam langsung setelah tes latihan fisik, mulai mengalami penurunan pada jam ke–6 dan kembali ke rerata awal pada jam ke–24 (p<0,05), kecuali pada kelompok pria tak terlatih dengan plasebo meningkat 2 kali dari nilai awal. Hal ini menunjukkan astaxanthin dan latihan mempunyai efek positif terhadap peningkatan kadar MDA namun tidak memengaruhi pola perubahannya setelah aktivitas fisik berat.

Abstract

High intensity exercise (HI) induces oxidative stress. Running for 2400 meters is one of the simple exercise form which can be counted as HI. During HI, increase of oxygen consumption happened in muscle cell resulted in an increase of oxidant level. The aim of the study was to measure the effects of Indonesian green tea supplementation, before and after HI to the level of lipid peroxidation (TBARSMalonyldialdehyde (MDA)). The subjects of study were 42 male students from Faculty of Medicine, Universitas Padjadjaran (FMUP) and Faculty of Physical Exercise, Indonesia University of Education (FPOK-UPI). The subjects were divided into trained and untrained subject. Subjects were divided into groups using Astrand Harvard step test. The purpose of this test was measuring the level of VO2 max. Blood samples for MDA level were collected at 3 hours before and after high intensity aerobic  exercise. Oxidative stress was induced by 2400 meters run. The result showed that green tea supplement-ation was effective to decrease MDA plasma level especially after exercise. Green tea could serve as natural antioxidant resources, which potentially can be used as potential sport supplemental drink.Key words: Aerobic exercise, Malonyldialdehyde (MDA), pre and post exerciseStudi Pra dan Pasca Pemberian Suplemen Ekstrak Teh Hijauterhadap Kadar MDAHigh intensity exercise (HI) dapat menginduksi stres oksidatif. Subjek penelitian ini adalah 42 mahasiswa laki-laki dari Fakultas Kedokteran Universitas Padjadjaran dan Fakultas Pendidikan Olahraga dan Kesehatan,Universitas Pendidikan Indonesia. Subjek penelitian dibagi ke dalam kelompok terlatih dan tidak terlatih dengan menggunakan cara pengujian Astrand Harvard. Tes ini bertujuan untuk mengukur kadar VO2 max. Sampel darah untuk kadar MDA diambil 3 jam sebelum dan setelah high intensity aerobic exercise. Stres oksidatif diinduksi dengan berlari sejauh 2400 meter. Hasil penelitian menunjukkan bahwa suplemen teh hijau efektif dalam menurunkan kadar MDA dalam plasma khususnya setelah berlari. Abstract

Proses penuaan kulit terjadi karena kombinasi penurunan kapasitas proliferasi sel-sel kulit, berkurangnya sintesis matriks dermis, dan peningkatan ekspresi enzim yang mendegradasi matriks kolagen. Penelitian ini merupakan eksperimental laboratorik, menggunakan 24 ekor tikus jantan galur Sprague-Dawley sebagai objek penelitian. Pada kelompok perakuan, kulit dorsal kedua kelompok dicukur, kemudian diberikan paparan sinar UVB seminggu tiga kali selama 4 minggu dengan dosis total penyinaran 840 mJ/ cm2. Kulit kelompok perlakuan diolesi krim Lactobacillus plantarum FNCC 0020 yang tidak bereplikasi, sehari 2 kali dengan komposisi aplikasi topikal koloni 247,27x107, sedangkan kelompok kontrol tidak diolesi apapun. Hasil penelitian diperoleh, baik intensitas, distribusi, maupun histoskor MMP-13 antara kelompok perlakuan dan kelompok kontrol tidak menunjukkan ada perbedaan yang bermakna (p>0,05). Intensitas kolagen III baik pada kelompok perlakuan maupun kontrol semua mempunyai derajat kuat, sedangkan distribusi kepadatan kolagennya paling rendah 20–50% dan tertingginya di atas 80%. Perbandingan distribusi kepadatan kolagen III secara statistik sangat bermakna (p<0,01). Pemberian krim topikal L. plantarum FNCC 0020 meningkatkan ekspresi distribusi kepadatan kolagen III namun tidak menurunkan ekspresi MMP-13. Kata kunci: Lactobacillus plantarum, kolagen, (matrix metalloproteinases) MMP13 Effect of Topical Probiotic on MMP-13 and Collagen III Expression in The Dermis Layer of Male Rats Irradiated with Ultraviolet-B Abstract Nowadays, there is a big interest in the use of topical probiotic preparations for skin health. One of the probiotics therurapeutic benefits is used as anti-aging. During aging, there is stimulation of activator protein-1 (AP-1) which is a transcription factor that inhibits the production of collagen and AP-1 supports the breakdown of collagen by enzymes called matrix metalloproteinases (MMPs). As administration of oral Lactobacillus plantarum could inhibit skin aging by lowering the activity of MMP, so the collagen degradation can be derived so probably topical use of Lactobacillus plantarum may give more prominent effects. We used 24 male rats Sprague-Dawley strain as research objects. This study was divided into two groups, the treatment and control groups. The shaved dorsal skin of rats were irradiated with UVB three times a week for 4 weeks with total irradiation dose of 840 mJ/cm2. Skin cream, containing 247.27x107 CFU non-replicating Lactobacillus plantarum FNCC 0020, was smeared on the treatment group, two times daily, whereas the control group did not receive any treatment. Skin biopsies were done at the end of the study for examination of MMP-13 and collagen III expressions. Intensity, distribution, and histoscore of MMP-13 between the treatment and the control group showed no significant difference (p>0.05). The treatment group showed a significant different in the intensity of collagen III with the density distribution of 20–50% and the highest density was 80% (p<0.01). Administration of topical cream L. plantarum FNCC 0020 increased the expression of collagen III density distribution, but not the MMP-13 expression.Keywords: Collagen, Lactobacillus plantarum, (matrix metalloproteinases) MMP13

Abstract

Moderate and high aerobic exercises are the activities that can induce high oxidant. Demandof endogen and exogen antioxidant for neutralizing oxidant induced by exercise will playimportant role in maintaining body performance. Microtrace elements, such as selenium,may able to balance high oxidant formation. However, limited research and data have beencollected with regards to the exercise and selenium. Therefore, this research was conductedto investigate the effect of aerobic exercise on selenium level in plasma. An analytic studywith experimental method of data collection was performed from October -November 2016.17 male students with the age between 19-25 years old and without medically compromiseddisease from Universitas Padjadjaran, Jatinangor, were selected as subjects. Blood sampleswere taken pre- and post-aerobic exercise, i.e., brisk walking with the intensity of 4.5 mi/h.Selenium level in plasma was then examined by enzyme linked immunosorbent assay (ELISA)kit as selenium binding protein (SELENBP1). This study found that 11 (65%) subjectshad a decrease in their selenium plasma after exercise (3.1 ± 2.58 ng/ml), 5 (29%) subjectshad an increase in their selenium plasma after exercise (2.08 ± 2.8 ng/ml) and 1 (6%) subjecthad unchanged selenium (SELENBP1) plasma (1.4 ± 1.4 ng/ml) level. Abstract

Low plasma selenium level was associated with the increasing risk of death in geriatric patients, particularly those with multiple comorbidities. The sufficient level of selenium intake as an antioxidant is necessary for this population. This study aimed to investigate the plasma selenium level based on dietary intake in geriatric clinic population at Dr. Hasan Sadikin General Hospital Bandung.This study was a descriptive study using cross-sectional method. Fourteen geriatric patients were selected by consecutive sampling technique. Semiquantitative food frequency questionnaire (SQ-FFQ) was used as a tool to assess dietary intake. Plasma selenium level was measured as selenium binding protein 1 (SELENBP1) using enzyme-linked immunosorbent assay kit. Overall, mean of plasma selenium level of the subjects was 2,68 µg/L and mean of selenium intake was 62,85 µg/day. Selenium level of the subjects with sufficient selenium intake (85,7%) was 2,62 µg/L and selenium level of the subjects with deficient selenium intake (14,3%) was 3,05 µg/L. In conclusion, plasma selenium level among geriatric patients was varied and not dependent on dietary intake.Keywords: antioxidant, dietary intake, geriatric, plasma selenium level

Abstract

Geriatric physiologically undergoes aging process that can cause decreasing cell functionand increasing risk of degenerative diseases caused by the accumulation of reactive oxygenspecies (ROS) in the body. Selenium (Se) is an antioxidant, which is needed for maintainingthe balance of ROS. The aim of this study was to observe the selenium-binding protein(SELENBP1) level in relation with the geriatric patients charcateristics including sex, age,body mass index (BMI), activity of daily living (ADL), instrumental activity of daily living(IADL), cognitive function, nutrition, depression, and insomnia status. The study usedcross-sectional quantitative descriptive study design on 14 geriatric patients in Geriatric OutpatientsClinics, at a hospital in Bandung. The data was obtained by interviewing the patientsand then blood samples were taken. The analysis of SELENBP1 was done using ELISA kit.The average level (SD) of SELENBP1 from all of the characteristics group was 2.68 (0.69)ng/ml. The highest SELENBP1 level was identified in female geriatric patients and followedby male and pre-obese groups. The lowest SELENBP1 level was identified in geriatric patientsaged 70-79 years.Keywords: elderly, SELENBP1, selenium.

Abstract

Aerobic physical activity is known to induce skeletal muscle adaptation. Some genes, including MyoD, are known to have a major role in the process of muscle adaptation. Several studies has stated that the expression of MyoD increases during the aerobic activity process; however, the studies were carried out in a single bout. Studies on the effects of acute phase exercise (<2 weeks) are still rare, especially those regarding the difference effect of acute training phase at different times. This study was performed in 2018 at the Central Laboratory and Animal Physiology Laboratory, Faculty of Medicine, Universitas Padjadjaran. The aim of this study was to determine the different effects of MyoD gene expression in a two-week acute aerobic physical activity. The effects were measured in the skeletal muscle in day 3, 7, and 14 of the activity. Twenty four male rats (rattus novergicus) were divided into 4 groups. Those rats in the treatment groups were run on an animal treadmill (P1=3 days, P2=7 days, and P3=14 days). On the last day of ran, rats were sacrificed and the soleus and gastrocnemius muscles were dissected. The expression of the MyoD gene in both muscles was then amplified using reversed PCR and detected by agarose gel electrophoresis. This study showed that there were differencesin MyoD gene expression levels in both muscles and that the level increased with days of treatment, although it was statistically insignificant. This study concludes that the MyoD gene expression level is not significantly affected by an acute aerobic physical activity in certain periods (3, 7 and 14 days).Key words: Acute, Aerobic physical activity, gastrocnemius, MyoD, rats, soleus Perbedaan Pengaruh Aktivitas Aerobik Akut Intensitas Sedang terhadap Ekspresi Gen MyoD pada Otot Rangka Tikus Galur WistarAktivitas fisik aerobik diketahui menginduksi adaptasi otot rangka. Beberapa gen diketahui memiliki peran utama dalam proses adaptasi otot termasuk MyoD. Dalam beberapa penelitian diketahui bahwa ekspresi MyoD meningkat selama proses aktivitas aerobik, tetapi penelitian dilakukan dalam pertarungan tunggal. Penelitian tentang efek latihan fase akut (<2 minggu) masih belum banyak dilakukan, terutama mengenai perbedaan efek fase latihan akut pada periode tertentu (3, 7, dan 14 hari). Penelitian ini dilakukan pada tahun 2018 di Laboratorium Sentral dan Laboratorium Fisiologi Hewan Fakultas Kedokteran Universitas Padjadjaran. Tujuan penelitian ini adalah mengetahui perbedaan efek ekspresi gen MyoD dalam aktivitas fisik aerobik akut pada berbagai hari (3, 7, 14 hari) pada otot rangka. 24 tikus jantan Rattus novergicus dibagi menjadi 4 kelompok. Kelompok perlakuan dijalankan dengan treadmill hewan (P1=3 hari, P2=7 hari, dan P3=14 hari). Pada hari terakhir, tikus dikorbankan dengan soleus dan otot gastroknemius dibedah. Ekspresi gen MyoD pada kedua otot kemudian diamplifikasi menggunakan PCR terbalik dan dideteksi dengan elektroforesis gel agarosa. Penelitian ini menunjukkan perbedaan level ekspresi gen MyoD pada kedua otot, hasil ini menunjukkan bahwa ada peningkatan level perawatan per hari, walaupun secara statistik tidak signifikan. Penelitian ini menyimpulkan bahwa pada otot gastrocnemius tidak terlihat efek yang berbeda dari durasi aktivitas fisik aerobik terhadap tingkat ekspresi gen MyoD, hanya otot soleus menunjukkan perubahan dan peningkatan ekspresi konsisten terlihat pada hari ke 14 (3, 7, dan 14 hari).Kata kunci: Aktivitas fisik aerobik, akut, gastroknemius, MyoD, soleus, tikus

Abstract

Optimization of Western Blot Technique for Protein Expression of Rice Plant (Oryza sativa L.) Western blot (WB) technique has been widely used for analyzing protein expression and for identifying specific proteins derived from animals, plants, and microorganisms. During the use of WB, especially in agricultural studies, some difficulties are encountered such as unclear or unspecific protein bands, the presence of bubbles, and the absence of protein bands on membrane. This study aims to determine the WB conditions appropriate for the protein expression of rice plants (Oryza sativa L.). Protein from rice plant was extracted and the obtained protein lysate was then used for proteomic analysis using western blot with β-actin antibody. Our experiment showed that some optimized parameters like blocking buffers, the concentration of primary antibody and the ratio of secondary antibody determined the clarity of the results. β-actin was used as internal control that measured the success of the WB technique. Results showed that lysis process was important in determining good WB results in addition to the optimal blocking solution using a BSA of 0.2%, a primary antibody concentration of 1 μg mL–1, and a secondary antibody of 1:10,000. Optimizing techniques during extraction, incubation, and documentation facilitated good WB results.Keywords: β-actin; optimization; protein; rice plant; western blotABSTRAKTeknik western blot (WB) telah banyak digunakan untuk analisis ekspresi protein dan mengidentifikasi protein spesifik dari hewan, tumbuhan dan mikroorganisme. Dalam implementasi teknik WB, khususnya studi dalam bidang pertanian, beberapa kesulitan ditemui seperti pita protein tidak jelas, tidak spesifik, adanya gelembung, hingga tidak munculnya pita protein pada membran. Penelitian ini bertujuan untuk mengetahui kondisi WB yang tepat untuk deteksi protein tanaman padi (Oryza sativa L.). Protein tanaman padi diekstraksi, kemudian lysate protein yang didapat dianalisis dengan metode westernblot menggunakan antibody β-actin. Penelitian kami menunjukkan bahwa beberapa parameter yang dioptimasi seperti larutan blocking, konsentrasi antibodi primer dan rasio antibodi sekunder akan menentukan hasil yang jelas. β-actin digunakan sebagai kontrol internal yang menjadi tolok ukur keberhasilan teknik WB. Hasil menunjukkan bahwa proses lisis menjadi hal penting dalam menentukan hasil WB yang baik disamping larutan blocking yang optimal menggunakan BSA 0,2%, konsentrasi antibodi primer 1 µg mL–1 dan antibodi sekunder 1:10.000. Mengoptimalkan teknik selama ekstraksi, inkubasi dan dokumentasi membantu mendapatkan hasil WB yang baik.

Abstract

Fitness becomes an important benchmark for public health in every age group. Activities with poor adverse fitness are at risk of injury. Good fitness is gained from a balance in everyday physical activity with conditions that are free of metabolic syndrome. This study investigated the relationship of physical activity, energy balance, and metabolic syndrome to the fitness of 166 professors of Universitas Padjadjaran who underwent a 1-month fitness checkup at BAPELKES Bandung in the period of 2017. Based on the regression analysis, the results showed a significant relationship between physical activity, energy balance, and metabolic syndrome on the physical fitness of the elderly group (F = 25.9290> F table = 2.427) with normal data distribution (Kolmogorov-Smirnov test). The results also showed a moderate relationship (R = 0.569 and R2 = 0.324). It also partially showed significant influence between physical activity and fitness (t = 8.541> t table = 1.975), significant influence between energy balance and physical fitness (t = 2.248> t table = 1.975), and no significant partial influence between metabolic syndrome and physical fitness of the elderly group (t = -0.029 <t table = 1.975). Hence, there is a relationship between physical activity, energy balance, metabolic syndrome and physical fitness  of Professors in Universitas Padjadjaran.Key words: Energy balance, metabolic syndrome, physical fitness  Hubungan Aktivitas Fisik, Keseimbangan Energi, dan Sindrom Metabolik terhadap Kebugaran Fisik pada Kelompok Usia LanjutKebugaran menjadi tolak ukur penting bagi kesehatan masyarakat pada setiap kelompok usia. Aktivitas dengan kebugaran usia lanjut yang kurang baik berisiko terjadi cedera. Kebugaran yang baik diperoleh dari keseimbangan dalam aktivitas fisik sehari-hari dengan kondisi bebas dari sindrom metabolik. Penelitian ini mempelajari hubungan aktivitas fisik, keseimbangan energi, dan sindrom metabolik dengan kebugaran 166 orang Guru Besar di Universitas Padjadjaran yang dilakukan pemeriksaan kebugaran selama 1 bulan di BAPELKES Bandung periode tahun 2017. Berdasar atas analisis regresi hasil penelitian menunjukkan hubungan yang signifikan antara aktivitas fisik, keseimbangan energi, dan sindrom metabolik dengan kebugaran fisik kelompok usia lanjut (F=25,9290 > Ftable=2,427) dengan distribusi data normal (tes Kolmogorov-Smirnov). Hasil juga menunjukkan hubungan yang moderat (R= 0,569 dan R2= 0,324). Secara parsial juga menunjukkan pengaruh yang signfikan aktivitas fisik dengan kebugaran (t= 8.541> t tabel= 1.975), pengaruh signifikan keseimbangan energi dengan kebugaran fisik (t= 2,248> t tabel= 1,975), dan tidak ada pengaruh signifikan secara parsial terhadap sindrom metabolik dengan kebugaran fisik kelompok usia lanjut (t= -0,029 < t tabel= 1,975). Berdasar atas hasil penelitian dapat disimpulkan hubungan = aktivitas fisik, keseimbangan energi, dan sindrom metabolik dengan kebugaran fisik guru besar Universitas Padjadjaran.Kata kunci: Kebugaran, keseimbangan energi, sindrom metabolik

Abstract

ABSTRAK Sarkopenia adalah penurunan massa dan fungsi otot rangka yang disebabkan oleh penuaan. Sarkopenia dapat meningkatkan morbiditas dan mortalitas pada lansia. Penyebab dan patofisiologi sarkopenia bersifat kompleks dan belum diketahui secara pasti hingga saat ini. Autophagy diketahui mempunyai peran penting untuk proses eliminasi dan daur ulang protein dan organel yang rusak sehingga sementara pada penuaan proses akumulasi berlebihan organel dan protein yang rusak sebagai konsekuensi metabolisme merupakan salah satu penyebab terjadinya penyakit karena penuaan. Kondisi autophagy basal yang baik mempunyai efek protektif terhadap pemeliharaan massa otot dan stimulasi sel satelit untuk proses regenerasi otot. Sementara di sisi lain, inhibisi autophagy menyebabkan penurunan massa otot dan berhubungan dengan atrofi serat otot pada proses penuaan. Inhibisi autophagy diperlukan di awal fase regenerasi myogenesis otot sarkopenia, namun kemudian diduga melalui peran FoXO,autophagy diaktivasi untuk mengeliminasi sel yang rusak dan biogenesis mitokondria. Keseimbangan dinamis dari autophagy merupakan salah satu faktor kunci peranautophagy yang berperan positif pada pencegahan sarkopenia dan bahkan prosesmyogenesis sebagai tatalaksana sarkopenia.
