Abstract

Kesejahteraan hewan coba mencakup dua masalah utama yaitu pemeliharaan umum dan penanganan selama prosedur eksperimental. Kedua masalah utama tersebut berkaitan dengan perlakuan yang diberikan oleh peneliti sejak awal hewan coba diterima hingga penelitian berakhir. Penelitian yang menggunakan hewan percobaan terutama secara etis harus menerapkan prinsip replacement, reduction, dan refinement (3R), serta prinsip lima kebebasan hewan yaitu (1) bebas dari rasa lapar dan haus, (2) bebas dari rasa tidak nyaman, (3) bebas dari rasa nyeri, luka dan penyakit, (4) bebas dari rasa takut dan stres, dan (5) bebas untuk mengekspresikan tingkah laku alamiah. Penerapan kaidah kesejahteraan hewan dalam penelitian ini bertujuan meminimalisir stres pada hewan dan diharapkan dapat memberikan hasil penelitian yang lebih akurat.

Abstract

Exercise-induced hearth muscle adaptation is important for physiological process after exercise. This adaptation will ensure basal mitochondrial homeostasis and as a part of the mitochondria quality control. This process is reflected by equal level of biogenesis stimulation and as well as the selective degradation of old and undesirable mitochondria through fusion or fission cycle and Mitophagy. There is limited information about genetic regulation stimulated by training in cardiomyocytes. We believe there is a specific myokines or protein release in the serum and initiate cardiac muscle adaptation process. In the present study, twelve male wistar rats were appointed to two group: sedentary control and aerobic-intensity (AE, 15m/minute). Rats were trained for running with specific protocol as follows: 30 minutes/day with a 5 times/week interval for 8 weeks. On the last day, serum form control and exercise groups were taken via retro-orbital sinus. Then, 3.105  H9C2 cells (Rat cardiomyocytes cell line)  were cultured and incubated by this serum for 24 hours. After treatment, cell were extracted using trisure for RNA purification and continue with reverse transcriptase PCR. Our data showed that expression of the Pgc-1α, Mfn1, Mfn2, Opa1, Drp1, Pink, and Parkin genes were altered and modulated. Specifically, Mfn1, Mfn2, and Opa1 gene expression levels significantly increased. Interestingly, we did not find significant modulation for  Pgc-1α, Drp1, Pink, and Parkin. Taken together, serum of exercise rats might be contained with myokines or specific protein which was released during training and it altered mitochondrial genes expression in cardiomyocytes culture cell. We believe that myokines release in the serum had a contribution in cardiacmyocyte adaptation.

Abstract

The prevention of liver disease could be conducted through preventive strategies, such as antioxidants and exercise. Moringa leaf extract has substances that could act as antioxidant, while exercise is also known protecting the liver from disease by changing hepatic metabolism and autophagy. This study aimed to investigate the effect of moringa leaf extract and moderate intensity exercise on histopathological appearance and autophagy gene expression of wistar rat liver. The method used in this study was animal experiment using 24 male wistar rats divided into 4 groups, including control group, moringa group, exercise group, and moringa + exercise group. Moringa leaf extract was given in low dose (5.7 mg/kgW) per oral, 5 times a week, for 4 weeks. Meanwhile, the 20 m/minute treadmill exercise was given for 30 minutes per day, 5 times a week, for 4 weeks. Results showed a significant change on histopathological scoring in exercise group (p=0.011) compared to control group. Increased autophagy gene expression was found in moringa + exercise group compared to control group (LC3 0.90 fold; p62 0.87 fold). In summary, this study presented that moderate intensity exercise induced changes on histopathological appearance of wistar rat liver that might be associated with physiological inflammation. Moringa, with its antioxidant properties, combined with increased autophagy might improve histopathological changes in moringa + exercise group.

Abstract

“Bihormonal hypothesis” is disturbances of both insulin and glucagon in diabetes mellitus. It resulted in a high blood glucose level. Interestingly, as one of Diabetes Mellitus’s risk factors and metabolic disorders, obesity may also play role in altering homeostasis and regulation of glucose metabolism in blood and its utilization in tissues. Unfortunately, there is limited information about the alteration of glucagon levels in various degrees of obesity. This research objective is to learn the plasma glucagon levels alteration in pre-obese, obese, and very obese persons in Jatinangor in 2015. We had observed 31 obese female subjects in one village. This study was conducted using descriptive quantitative with cross-sectional design. Blood vein samples from the left arm were collected, stored, and transferred to Dr.Hasan Sadikin General Hospital. Glucagon plasma was measured by the ELISA method. We discovered an interesting pattern that showed a correlation between glucose level and the glucagon level in a very obese group. We observed average glucose level is declined and linearly associate with the glucagon level from pre obese to obese and to very obese group. The average level of glucagon in the pre-obese group is 158.62 pg/mL, the obese group is 149.99 pg/mL, and the very obese group is 111.98 pg/mL.

Abstract

Optimization of Western Blot Technique for Protein Expression of Rice Plant (Oryza sativa L.) Western blot (WB) technique has been widely used for analyzing protein expression and for identifying specific proteins derived from animals, plants, and microorganisms. During the use of WB, especially in agricultural studies, some difficulties are encountered such as unclear or unspecific protein bands, the presence of bubbles, and the absence of protein bands on membrane. This study aims to determine the WB conditions appropriate for the protein expression of rice plants (Oryza sativa L.). Protein from rice plant was extracted and the obtained protein lysate was then used for proteomic analysis using western blot with β-actin antibody. Our experiment showed that some optimized parameters like blocking buffers, the concentration of primary antibody and the ratio of secondary antibody determined the clarity of the results. β-actin was used as internal control that measured the success of the WB technique. Results showed that lysis process was important in determining good WB results in addition to the optimal blocking solution using a BSA of 0.2%, a primary antibody concentration of 1 μg mL–1, and a secondary antibody of 1:10,000. Optimizing techniques during extraction, incubation, and documentation facilitated good WB results.Keywords: β-actin; optimization; protein; rice plant; western blotABSTRAKTeknik western blot (WB) telah banyak digunakan untuk analisis ekspresi protein dan mengidentifikasi protein spesifik dari hewan, tumbuhan dan mikroorganisme. Dalam implementasi teknik WB, khususnya studi dalam bidang pertanian, beberapa kesulitan ditemui seperti pita protein tidak jelas, tidak spesifik, adanya gelembung, hingga tidak munculnya pita protein pada membran. Penelitian ini bertujuan untuk mengetahui kondisi WB yang tepat untuk deteksi protein tanaman padi (Oryza sativa L.). Protein tanaman padi diekstraksi, kemudian lysate protein yang didapat dianalisis dengan metode westernblot menggunakan antibody β-actin. Penelitian kami menunjukkan bahwa beberapa parameter yang dioptimasi seperti larutan blocking, konsentrasi antibodi primer dan rasio antibodi sekunder akan menentukan hasil yang jelas. β-actin digunakan sebagai kontrol internal yang menjadi tolok ukur keberhasilan teknik WB. Hasil menunjukkan bahwa proses lisis menjadi hal penting dalam menentukan hasil WB yang baik disamping larutan blocking yang optimal menggunakan BSA 0,2%, konsentrasi antibodi primer 1 µg mL–1 dan antibodi sekunder 1:10.000. Mengoptimalkan teknik selama ekstraksi, inkubasi dan dokumentasi membantu mendapatkan hasil WB yang baik.

Abstract

Based on data Riset Kesehatan Dasar (Riskesdas) in 2007, the number of people who had no or low physical activity reached 48.2 %. This condition contributes to the increasing number of metabolic disease cases such as hypertension, heart disease, stroke, and diabetes mellitus. Optimizing training approaches especially the aerobic type helps to counter metabolic diseases. Unfortunately, the benefits of resistance training (RT) are less understood. RT improves muscle strength, induces muscle hypertrophy, improvement of local muscular performance, and also helps to strengthen body balance and coordination. There is an interplay between training and hormone in muscle adaptation during resistance training. The hormone plays an important role in the regeneration of muscle after resistance training. The changes in hormone level cause hypertrophy. Regeneration and muscle hypertrophy are mediated by activation, proliferation, and differentiation of satellite cells. It is regulated by mitotic and myogenic activity, namely insulin-like growth factor-1 (IGF-1), which serves as a paracrine or autocrine. A better understanding of homeostasis hormone during training in skeletal muscle and its ultimate purpose to counter metabolic disease will lead us to a better treatment approach for the patient.

Abstract

Physical fitness is the ability of an athlete to fulfill the physical activities that are needed for the sport without becoming weary. This is a very important thing that determines an athlete’s performance. The predominant component of physical fitness for rhythmic gymnastics are muscular endurance, muscle strength, power, and flexibility. The study aimed to gather information regarding the physical fitness profile of Bandung City rhythmic gymnastic athletes before competing in Pekan Olah Raga Daerah Jawa Barat (PORDA Jabar) XI event in 2010. The descriptive studywas conducted using the survey method. Subjects consist of 9 female Bandung City rhythmic gymnastic athletes. Muscle endurance, muscle strength, power, and flexibility were measured using appropriate tests and tools and then grouped according to Standard KONI Pusat. The results of this study showed that most of the rhythmic gymnastic athletes were in this categories: perfect for arms and shoulders muscle endurance, the average for stomach muscle endurance, below standard formuscle strength of the hands, the average for muscle strength of arms and shoulders, good and very good for muscle strength of the back, the average for muscle strength of the legs, below average and average for arms and shoulders power, very good for legs power, and perfect for flexibility. In a conclusion, the performance of Bandung City rhythmic gymnastic athletes can be improved byincreasing the training that is specific for each of the physical fitness components.

Abstract

Kegiatan Kuliah Kerja Nyata Pengabdian Pada Masyarakat (KKN-PPM) merupakan kesempatan kepada mahasiswa untuk serta berinovasidan mengembahkan pemikiran agar dapat membantu mengatasi masalah dimasyarakat. Pada periode ini KKN dilaksanakan secara daring akibat adanya kondisi khusus darurat yakni pandemi Covid-19. Tema seminar online KKN-PPM “Potensi Manggis dalam Sediaan Sabun Antiseptik dan Olahan Pangan Fungsional Sebagai Upaya Pencegahan Covid-19 di Masa Adaptasi Baru”. Seminar online ini dilakukan pada tanggal 25 Juli 2020, pukul 10.00 WIB hingga 12.05 WIB. Seminar dan workshop virtual ini mengedukasi perilaku hidup sehat dan bersih saat masa Pandemi Covid-19, termasuk bagaimana kebiasaan mencuci tangan dan penggunaan masker sebagai upaya menekan penularan Covid-1, serta manfaat kulit manggis sebagai antioksidan. pemaparan beragam manfaat antioksidan pada kulit manggis salah satunya sebagai pengatur berbagai aktivitas inflamasi dan memberi signaling pathways dalam sel imun, terutama makrofag sehingga dapat mengoptimalkan kerja sistem imun. Peran bahan aktif Xanthones pada kulit manggis sebagai anti-bakteri, anti-jamur, anti-virus dan sumber antioksidan serta pengolahan produk kulit manggis menjadi fokus edukasi KKN-PPM. Kulit manggis dapat diolah menjadi sedian antiseptik dan olahan pangan fungsional seperti pempek, yogurt, dan wedang. Evaluasi kegiatan menggunakan One-Group Pretest-Posttest Design tentang PHBS dan potensi kulit manggis Berdasarkan hasil analisa data, terdapat peningkatan pengetahuan tentang upaya perilaku hidup sehat sebagai pencegahan Covid-19 dan manfaat buah manggis sebesar 24.09 % dari data pra dan pascakegiatan. kegiatan ini berkontribusi memberikan pengetahuan pada partisipan dalam mendukung penguatan sosialisasi perilaku hidup sehat saat masa Covid-19 dan manfaat kesehatan dari kulit manggis.Dari Penelitian Xanthonespada kulit manggis berpotensi sebagai anti-virus yang mungkin dapat dikembangkan lebih lanjut sebagai sumber zat aktif untuk terapi Covid-19.

Abstract

Selenium is a trace element that has antioxidant properties and is a component of selenoproteins. In certain circumstances, such as injury, selenium supplementation is required to regulate the inflammatory response. Contusion injury increases the inflammatory response to a muscle or tissue that begins with vasodilation, increases blood flow, increases vascular permeability, and leads to edema formation at the site of injury. Objectives: To determine the effect of selenium supplementation on changes in blood markers in rats treated with contusion injury. Methods: Experimental design, using experimental male rats (Rattus norwegius), eight weeks old, weighing 200-220 grams, divided into three groups, namely: The Control group, the contusion group, the contusion + Selenium group. Selenium supplementation was given orally on days 1-3 after injury at a dose of 0.0153 mg into 2% PGA. Examination of markers such as White Blood cells (WBC), lymphocytes, monocytes, and neutrophils was carried out on day three after treatment. Results: This study found that the administration of post-muscular selenium supplementation based on examination of blood markers such as WBC, Lymphocytes, Monocytes was higher in the contusion group and lower in the selenium group. Neutrophils were higher in the contusion and selenium groups. This was not significantly different from the control group. The result of blood marker examination tended to change, but it was not statistically significant. Conclusion: Selenium supplementation in post-contusive injury mice did not provide significant changes in blood markers such as WBC, lymphocytes, monocytes, and neutrophils.Keywords:  antioxidant, blood marker, contusion, selenium
