Abstract

Cassava (Manihot esculenta) is an important crop to food security under climate change due to its various tolerance mechanism under stress conditions. However, the sustainable growth of cassava in the field depends on many factors especially temperature. The objective of the research was to investigate the growth performances and proline contents of irradiated Ubi Kuning at dosage of 10 Gy, cultured in Murashige Skoog (MS) hormone-free solid medium for 4 weeks at three different temperature treatments i.e 25oC, 28oC and 30oC. Each treatment consisted of 3 clone explants with 5 replicates. Results show that growth performances of irradiated plantlets were better compared to that of non-irradiated plantlets in terms of plant height and number of leaves at all temperature tested. The best growth performances were obtained from irradiated plantlets grown under 30oC. The proline content of irradiated Ubi Kuning was high when they were grown under 25 oC and 30oC, implying that these plantlets had the possibility to tolerant to lower and higher-temperature condition. This study is initially useful to find out the growth ability of irradiated Ubi Kuning in response to lower and higher temperature.

Abstract

Cassava is an important carbohydrate source that provides food security and alternative renewable energy development. This plant is naturally drought tolerant, but there is a wide variation within cassava genotypes in their ability to maintain high yield and starch production under drought stress. It has been reported that cassava clones with leaf retention or stay green trait can produce more total fresh biomass and high root dry matter compared to drought susceptible cultivars. The genetic, biochemical and molecular bases of stay green trait need to be understood to develop drought resistant cassava cultivars since water stress limits yield and starch production. Differential Display (DD)RT-PCR is a powerful technique for analyzing differences in gene expression. The method is based on the detection of the differentially expressed cDNAs from two or more samples. The main objective of this study was to identify differentially expressed cDNA in cassava under drought stress by employing a cDNA-RAPD approach. In this study, we used cassava genotype Ubi Kuning which was considered the most responsive to water insufficiency (45 days without watering). Leaf samples were collected from water stressed and well-watered plants at day 45. Among 11 random primers, OPB03 and OPH17 have identified differentially expressed cDNA in Ubi Kuning. Further characterization of these PCR products of expressed cDNA under drought stress may open possibility the development of cassava with improved drought resistance through genetic engineering and/or marker assisted selection (MAS).

Abstract

ABSTRACTObjectives of the research were to determine the best source of explant origin, the optimum culture media and effects of various concentrations and types of growth regulators on the induction and multiplication as well as the growth of in vitro shoot of cassava Ubi Kuning genotype. The experiment was arranged in a complete randomized design with three factors, i.e the source of explant origin (from field and greenhouse), the growth media containing of 6-Benzyl Amino Purine (BAP) and Gibberellin (GA3) at the concentration level of 1-2 ppm, and multiplication medium supplemented with BAP, Naphtalenic acetic acid (NAA) and Kinetin at concentration level of 0.1 mg/l and 0.5 mg/l. The results showed that the cuttings explants from the field showed more shoots and the average shoot height of axenic culture in the fourth week ranging from 0.2-1.2 cm compared to those from the field (0.5- 1.3 cm). Axenic culture showed no significant growth in growth media (MS) supplemented with BAP and GA3 (1-2 ppm). In the multiplication medium, the highest number of roots were obtained when plantlets were grown in the treatment medium MS + BAP 0.5 mg / l followed by MS + NAA 0.1 mg / l. The largest number of nodes was produced on plantlets grown in MS + BAP media 0.1 mg / l and MS + NAA 0.5 mg / l (p <0.05). While the longest root growth was obtained in plantlets cultured on MS + Kinetin 0.1 mg / l medium. Plantlets showed high survival (100%) after they were acclimatizedfor 4 weeks in a greenhouse using a medium containing a mixture of compost, coco peat, and ground burn husk with a ratio of 1: 1: 1. Keywords: genotype, ubi kayu, ubi kuning  in vitro
