ENKAPSULASI PIRAZINAMID MENGGUNAKAN ALGINAT DAN KITOSANENCAPSULATION OF PYRAZINAMIDE USING ALGINATE AND CHITOSAN

Abstract

Abstrak. Telah dilakukan penelitian tentang enkapsulasi pirazinamid yang merupakan obat primerbagi penderita tuberkulosis dengan menggunakan polimer alginat dan kitosan. Penelitian inibertujuan untuk mengetahui pengaruh konsentrasi alginat terhadap efisiensi enkapsulasi danmengetahui karakteristik hasil enkapsulasi menggunakan spektrofotometer FT-IR dan PSA (ParticleSize Analyzer). Enkapsulasi pirazinamid dilakukan dengan menggunakan polimer alginat dan kitosandengan CaCl2 sebagai agen pengikat silang dengan memvariasi konsentrasi alginat. Konsentrasialginat yang digunakan yaitu 0,5%; 1,0%; 1,5%; 2,0%; 2,5%; dan 3,0% (b/v). Penelitian inimenggunakan teknik pembentukan droplet gel alginat-CaCl2 dengan metode ekstruksi. Pirazinamidterenkapsulasi alginat-kitosan diukur efisiensi enkapsulasinya dengan spektrofotometer UV, yangmenunjukkan nilai efisiensi enkapsulasi tertinggi pada penggunaan alginat 2,0% (b/v) dan terendahpada alginat 3,0% (b/v), masing-masing sebesar 82,51 % dan 58,94 %. Hasil enkapsulasi dengannilai efisiensi enkapsulasi tertinggi dikarakterisasi menggunakan spektrofotometer FT-IR dan PSA.Hasil analisis gugus fungsional menunjukkan bahwa terdapat interaksi polielektrolit antara guguskarboksil dari alginat dan gugus amina dari kitosan yang ditunjukkan adanya vibrasi pada bilangangelombang 1497,91 cm-1. Hasil analisis PSA menunjukkan bahwa pirazinamid terenkapsulasi alginatkitosanmempunyai distribusi ukuran partikel pada rentang 68,06 nm sampai 295,30 nm.Kata Kunci: Enkapsulasi, Pirazinamid, Kitosan, Alginat.Abstract. Has done research on encapsulation pyrazinamide which was the primary drug fortuberculosis patients by using alginate and chitosan polymers. This study aimed to determine effect ofalginate concentration on encapsulation efficiency and investigated the characteristics ofencapsulation results using FT-IR spectrophotometer and PSA (Particle Size Analyzer).Encapsulation pyrazinamide performed using alginate and chitosan polymers with CaCl2 as acrosslinking agent based on types of alginate concentrations. The variations of concentrations thatused were 0,5%; 1,0%; 1,5%; 2,0%; 2,5%; and 3,0% (w/v). This study used the technique of dropletformation of alginate-CaCl2 gel with extrusion method. Encapsulated pyrazinamide in alginatechitosanwere analyzed for efficiency encapsulation by UV spectrophotometer, which showed thehighest encapsulation efficiency values on the use of alginate 2,0% (w/v) and the lowest in thealginate 3,0% (w/v), respectively by 82,51% and 58,94%. Encapsulation resulted with the highestencapsulation efficiency values were characterized uses FT-IR spectrophotometer and PSA. Theanalysis resulted of functional group indicated that there is interaction between the carboxyl groupsof the polyelectrolyte alginate and the amine group of chitosan indicated the presence of vibration inwave number 1497,91 cm-1. PSA analysis results showed that pyrazinamide encapsulated in alginatechitosanhas a particle size distribution in the range of 68,06 nm to 295,30 nm.Keywords: Encapsulation, Pyrazinamide, Alginate, Chitosan.
