PENENTUAN KONDISI OPTIMUM PADA PEMBENTUKAN SENYAWAN-ASETIL-D-GLUKOSAMIN HASIL HIDROLISIS KITIN NON ENZIMATIS

Abstract

Abstrak. Kitin dapat dihidrolisis dengan menggunakan asam klorida (HCl) menghasilkan N-asetil-D-glukosamin (GlcNAc). Tujuan penelitian ini adalah untuk mengetahui konsentrasi HCl dan waktu inkubasi optimum pada pembentukan N-Asetil-D-Glukosamin (GlcNAc) hasil hidrolisis kitin. Identifikasi dan Jumlah GlcNAc ditentukan dengan menggunakan High Performance Liquid Chromatography (HPLC). Waktu retensi senyawa GlcNAc pada HPLC ditentukan dengan menggunakan GlcNAc standar. Penentuan konsentrasi HCl optimum dilakukan dengan menghidrolisis kitin dengan variasi konsentrasi HCl  1N – 5N selama 24 jam. Dari hasil penelitian diperoleh konsentrasi HCl optimum adalah 4N. Penentuan waktu inkubasi optimum dilakukan dengan menghidrolisis kitin menggunakan konsentrasi HCl optimum dengan variasi waktu inkubasi (0, 12, 24, 36, 48) jam. Waktu inkubasi optimum yang diperlukan untuk menghasilkan senyawa GlcNAc adalah 36 jam.   Kata kunci: Kitin, N-Asetil-D-Glukosamin, hidrolisis Asam, HPLC   Abstract. Chitin can be hydrolized by hydrochloric acid produced N-Acetyl-D-Glukosamine (GlcNAc). The aim of this experiment were to know the concentration of hydrochloric acid and optimum incubation time on formation of N-Acetyl-D-Glukosamin (GlcNAc) from hydrolysis of chitin. The identification and amount of GlcNAc was determined using High Performance Liquid Chromatography (HPLC) analysis. The HPLC retention time of GlcNAc was determined using GlcNAc standard. Determination of concentration of hydrochloric acid to hydrolize chitin was done at variation of concentration HCl (1N- 5N) during 24 hours. The result showed that the optimum concentration of hydrochloric acid to hyrolized chitin is 4N. Determination of incubation time to hydrolize chitin was done at variation of incubation time (0, 12, 24, 36, 48) hours.
