FRAKSI ETIL ASETAT DAUN SERNAI (Wedelia biflora) SEBAGAI ANALGESIK DENGAN METODE WRITHING ABDOMINAL PADA MENCIT (Mus musculus)

Abstract

ABSTRAK Penelitian ini bertujuan mengetahui khasiat fraksi etil asetat daun sernai sebagai analgesik yang diberikan secara oral pada mencit dan melihat perbandingan dengan ibuprofen. Penelitian ini menggunakan 25 ekor mencit yang berumur 2,0-4,0 bulan dengan berat badan 25-30 g. Penelitian ini menggunakan rancangan acak lengkap (RAL) pola split plot, terdiri atas 5 kelompok, P0 diberi akuades (kontrol negatif), P1 diberikan  ibuprofen 0,25 mg/kg bb (kontrol positif), P2, P3, P4, masing-masing diberi fraksi etil asetat daun sernai dengan dosis 30, 45, dan 60 mg/kg bb. Uji analgesik dengan metode whriting abdominal test. Hasil penelitian menunjukkan bahwa kelompok perlakuan dan waktu perlakuan fraksi etil asetat daun sernai sebagai analgesik pada mencit berpengaruh secara nyata (P0,05). Berdasarkan hasil uji Duncan memperlihatkan penurunan geliat mencit pada kelompok P1, P2, P3, dan P4 secara nyata (P0,05) dibandingkan kelompok P0. Sedangkan kelompok P2, P3, P4 tidak menunjukkan perbedaan nyata (P0,05)  dibandingkan dengan kelompok P1. Berdasarkan hasil penelitian rataan jumlah geliat mencit stiap 5 menit dalam 1 jam pada kelompok P3=6,8 kali/jam lebih baik dibandingkan dengan kelompok P2=7,7 kali/jam dan P4=10,3 kali/jam, sehingga dapat disimpulkan bahwa fraksi etil asetat daun  sernai dengan dosis 45, mg/kg bb memiliki aktivitas analgesik yang lebih baik dibandingakan dengan dosis 30 dan 60 mg/kg bb yang diinduksi secara kimiawi dan cenderung setara dengan ibuprofen.Kata kunci: Wedelia biflora, analgesik, ibuprofen. ABSTRACT                 The objective of this study was to determine the efficacy of ethyl acetate fraction of sernai leaf as an analgesic was given orally in mice and see comparisons with ibuprofen. This study used 25 mices aged 2.0 to 4.0 months with body weight of 25-30 g. Using a completely randomized design (CRD) pattern split plot, consists of 5 groups, P0 was given distilled water (negative control), P1 was given ibuprofen 0.25 mg / kg bw (positive control), P2, P3, P4, were given fraction sernai leaf ethyl acetate with dose  30, 45, and 60 mg / kg bw. Test analgesic with abdominal whriting test method. The results showed that the treatment group and the treatment time ethyl acetate fraction sernai leaf as an analgesic in mice affect significantly (P 0.05). Based on the results of Duncan test showed a decline in the movement of mice in group P1, P2, P3, and P4 significanyt (P 0.05) differences  than group P0. While groups of P2, P3, P4 was not showed the difference significant (P 0.05) compared with the group P1. Based on this study the avarege number of wrhiting in mice every 5 minutes in 1 hour at group of P3=6,8 times/hour is better then the group P2=7,7 times/hour and P4=10,3 times/hour, so it was concluded that fraction of ethyl acetate of sernai leaves  by doses of 45 mg/kg bw capable of showing analgesic activity better than doses of 30 and 60 mg/kg bw, those which induced chemically tend to be similar to ibuprofen. Keyword: Wedelia biflora, analgesic, ibuprofen.
