Abstract

Dedak padi dan ampas kelapa dicampur dengan perbandingan tertentu dan kemudian difermentasi menggunakan Rhizopus oryzae untuk pakan ikan. Uji pemberian pakan lalu dilakukan untuk mengetahui pengaruh pakan terhadap pertumbuhan ikan mas (Cyprinus carpio L.). Dalam penelitian ini digunakan 5 perlakuan: satu perlakuan pakan tanpa fermentasi (pakan komersial 100%), dan empat perlakuan pakan fermentasi substrat campuran bekatul dan ampas kelapa dengan empat perbandingan yang berbeda, yakni 75%:25%, 50%:50%, 25%:75%, dan 0%:100%. Hasil penelitian menunjukkan bahwa pemberian pakan komersial 100% (protein sejati 15,25% dan serat kasar 6,27%) memperlihatkan hasil terbaik terhadap pertumbuhan ikan mas dengan pertambahan bobot badan 2,56 g dan rasio konversi pakan 1,95. Sementara itu pemberian pakan fermentasi (protein sejati berkisar 4,89-9,97% dan serat kasar 22,87-25,70%) hanya menghasilkan pertambahan bobot badan ikan pada kisaran 0,47-0,64 g dengan rasio konversi pakan 2,50-2,64. Dengan demikian pakan fermentasi tersebut mampu mendorong pertumbuhan ikan mas meskipun masih kurang optimal dibandingkan dengan pakan komersial.Kata Kunci: Rhizopus oryzae, Cyprinus carpio, rice bran, coconut bagasse, fermentation  ABSTRACTRice bran and coconut bagasse were mixed and then fermented using Rhizopus oryzae for preparing aquafeed. Subsequent feeding test was carried out to determine the effect on the growth of carps (Cyprinus carpio L). Five feeding treatments were employed, one unfermented feed (commercial feed 100%), and the other four feeds produced by fermentation using substrate mixture of rice bran and coconut pulp in four different ratios, namely 75%:25%, 50%:50%, 25%:75%, and 0%:100%. The results showed that feeding 100% commercial feed (true protein 15.25% and crude fibre 6.27%) showed the best results on the fish growth with body weight gain of 2.56 g and feed conversion ratio of 1.95. Meanwhile, feeding fermented feeds (true protein 4.89-9.97% and crude fiber 22.87-25.70%) only resulted in body weight gain in the range of 0.47 to 0.64 g with feed conversion ratio of 2.50 to 2.64. Abstract

In detergent industry, enzymes are used enormously in terms of quantity and economic value. Lipase catalyzes the hydrolysis of triglycerides into diglycerides and monoglycerides by releasing fatty acids. Lipase is produced by bacteria, fungi, and yeasts. This study aims to determine the potential of Alcaligenes faecalis lipase  for its application  as biodetergen, through stability testing of its lipase activity against detergent components by exposing the enzyme to the commercial detergents, as well as performance testing through washing. Alcaligenes faecalis lipase was produced using Luria Bertani (LB) culture medium supplemented with 1% olive oil inducer. Production is carried out for 24 hours, and the enzyme was harvested at the 18th hour. The harvested enzyme was tested for their stability after being exposed to commercial detergents at a concentration of 1-5%. Results showed that the exposure to the detergents decreased the enzyme activity to 22, 38, 48, 68 and 90%. Performance test showed that the olive oil impurity removal from the fabric was 29%.Keywords: Alcaligenes faecalis Lipase, biodetergent, lipase activities, washing test AbstrakPada industri detergen penggunaan enzim sangatlah besar baik secara jumlah maupun nilai ekonomi. Lipase mengkatalis hidrolisis trigliserida menjadi digliserida dan monogliserida dengan membebaskan asam lemak. Lipase dihasilkan oleh bakteri, jamur, dan yeast. Penelitian ini bertujuan untuk mengetahui potensi lipase Alcaligenes faecalis untuk aplikasi biodeterjen, melalui uji stabilitas aktivitas lipase terhadap komponen deterjen dengan memaparkan terhadap deterjen komersial serta uji kinerja melalui washing test. Lipase Alcaligenes faecalis diproduksi menggunakan media Luria Bertani (LB) dengan penambahan induser minyak zaitun 1%. Produksi dilakukan selama 24 jam dengan waktu pemanenan enzim pada jam ke-18. Enzim yang sudah dipanen diuji stabilitasnya setelah dipapar dengan deterjen komersial pada konsentrasi 1-5%. Berdasarkan hasil pengujian aktivitas setelah dipapar terlihat penurunan aktivitas berturut-turut sebesar 22, 38, 48, 68 dan 90%. Hasil uji kinerja menunjukkan bahwa noda minyak zaitun yang hilang dari kain sebesar 29%.Kata kunci: Lipase Alcaligenes faecalis, biodeterjen, aktivitas lipase, washing test

Abstract

Heavy metals contamination is a main issue which has negative impacts to environment and organisms. Various methods have been developed to reduce such pollutants, including utilization of organisms’ capability in order to minimize the contamination. Mycoremediation is one of remediation process in contaminated environment using fungi and its reduction mechanisms, involving intracellular, as well as extracellular system. There are some species of fungi that are frequently used as remediator agents, for example Aspergillus sp., Fusarium sp., Penicillium sp., Phanerochaete sp., Trichoderma sp. There are some methods that have been used for heavy metal reduction mechanisms such as biosorption, bioaccumulation, bioprecipitation, bioreduction, and bioleaching.Keywords: Mycoremediation, fungi, heavy metal, biosorption, bioaccumulation ABSTRAK Kontaminasi logam berat adalah suatu permasalahan utama yang berdampak negatif bagi lingkungan dan juga makhluk hidup. Berbagai metode telah dikembangkan untuk mereduksi cemaran, termasuk memanfaatkan kemampuan organisme untuk meminimalkan kontaminan tersebut. Mikoredemiasi adalah salah satu proses remediasi cemaran di lingkungan dengan melibatkan fungi beserta mekanisme reduksinya, baik secara intraselular maupun ekstraselular. Beberapa jenis fungi yang sering dijadikan agen remediator antara lain Aspergillus sp., Fusarium sp., Penicillium sp., Phanerochaete sp., Trichoderma sp. Abstract

EFFECT OF AUXIN AND CYTOKININ ON MICROPROPAGATION OF BINAHONG (Anredera cordifolia (Tenore) Steenis)Binahong (Anredera cordifolia (Tenore) Steenis) is known as miraculous medicinal plant for its potential to cure for various kinds of diseases such as diabetes, stabilizing blood pressure and circulation, accelerating wound healing, and preventing stroke. In order to provide high quality seedlings of this medicinal plant continuously in large amount, the study on binahong micropropagation was performed. Plant growth regulators of auxins and cytokinins were applied in single or in combination so as to observe their effect on the growth of binahong explants. The results showed that 2,4-D induced callus formation in large diameter on all treatments. Nevertheless, this plant growth regulator had a negative effect on growth and development of shoot and leaves. In the combination treatments between IAA and BAP, it revealed that the higher the concentration of BAP in the media, the lower the number of leaves initiated on shoot explants. Increasing the concentration of IAA upto 1.5 ppm influenced the increasing of shoot tallness and the number of internodes. Our results can be useful for improving the binahong shoot propagation efficiency, as well as callus culture studies.Keywords: Auxin, cytokinin, callus, micropropagation, medicinal plantABSTRAKBinahong (Anredera cordifolia (Tenore) Steenis) dikenal sebagai tanaman obat ajaib karena dapat digunakan untuk pengobatan berbagai macam penyakit seperti diabetes, melancarkan peredaran dan tekanan darah, mempercepat penyembuhan luka, mencegah stroke. Dalam mendukung ketersediaan bibit tanaman obat herbal yang berkualitas secara berkelanjutan maka dilakukan kajian tentang perbanyakan bibit tanaman binahong. Zat pengatur tumbuh auksin dan sitokinin dalam bentuk tunggal maupun kombinasi diaplikasikan pada penelitian ini untuk melihat pengaruhnya terhadap berbagai eksplan binahong. Hasilnya menunjukkan bahwa 2,4-D merangsang pembentukan kalus dengan ukuran yang besar pada semua perlakuan. Namun demikian zat pengatur tumbuh ini memberikan pengaruh negatif terhadap pertumbuhan dan perkembangan tunas dan daun. Dari perlakuan kombinasi zat pengatur tumbuh IAA dan BAP, pertambahan konsentrasi BAP di dalam media menurunkan jumlah daun yang terbentuk pada eksplan pucuk binahong. Demikian pula dengan pertambahan konsentrasi IAA hingga 1,5 ppm sangat berpengaruh terhadap pertumbuhan meninggi tunas dan pertambahan jumlah ruas. Hasil dari studi ini dapat dimanfaatkan untuk studi lanjutan dalam meningkatkan efisiensi perbanyakan tunas serta kultur kalus binahong.Kata kunci: Auksin, sitokinin, kalus, perbanyakan mikro, tanaman obat herbal

Abstract

Ethanol is an important product for biotechnology-based industries. Ethanol can be produced from various raw materials and some types of microbes. Determination of the best combination of galactose with nitrogen sources on ethanol production using Saccharomyces cerevisiae has been done. Combination of galactose at the concentration of 3 g/L and 20 g/L with nitrogen sources (casein, peptone, and urea, each at the concentration of 10g/L) was used to obtain the best composition of fermentation medium. Fermentation was carried out for 60 hours at 30°C, 250 rpm, and working volume of 50 mL in a 250 mL erlenmeyer. The results showed that the galactose concentration of 20 g/L was able to improve the productivity of ethanol and the growth of S. cerevisiae cells. The combination of 20g/L galactose and 10 g/L casein produced the highest ethanol concentration (6% v/v), whereas 20 g/L galactose-10 g/L peptone and 20 g/L galactose-10 g/L urea combinations produced 2.5% and 0.58% (v/v) ethanol, respectively. The use of 3 g/L galactose mixed with several nitrogen sources produced ethanol below 0.7% (v/v). Keywords: Ethanol, galactose, peptone, casein, Saccharomyces cerevisiaeABSTRAKEtanol merupakan salah satu produk penting bagi industri yang berbasis bioteknologi. Etanol dapat dihasilkan dari berbagai macam bahan baku dan beberapa jenis mikroba. Penentuan kombinasi terbaik antara galaktosa dengan sumber nitrogen pada produksi etanol menggunakan Saccharomyces cerevisiae telah dilakukan. Konsentrasi galaktosa 3 g/L dan galaktosa 20 g/L yang dikombinasikan dengan sumber nitrogen dengan konsentrasi 10 g/L dalam hal ini kasein, pepton, dan urea digunakan sebagai perlakuan untuk mendapatkan kombinasi medium sumber karbon dan sumber nitrogen terbaik. Fermentasi untuk menghasilkan etanol dilakukan selama 60 jam pada suhu 30°C, agitasi 250 rpm dengan volume kerja 50 mL dalam erlenmeyer 250 mL. Hasil penelitian menunjukkan penambahan galaktosa dengan konsentrasi sampai dengan 20 g/L mampu memperbaiki produktivitas etanol dan pertumbuhan sel S. cerevisiae. Konsentrasi 20 g/L galaktosa dengan 10 g/L kasein menghasilkan produktivitas etanol paling tinggi yaitu 6%(v/v), konsentrasi galaktosa 20 g/L dengan 10 g/L pepton menghasilkan 2,5% (v/v) etanol dan konsentrasi galaktosa 20 g/L dengan 10 g/L urea menghasilkan 0,58%(v/v) etanol. Abstract

Taro or Satoimo (Colocasia esculenta (L) Schott var antiquorum) is an alternative of non-rice food. To support saitomo mass cultivation in several regions in Indonesia, shoot multiplication and induction of satoimo microtuber through in vitro technique is amongst the stage to be undertaken. The aims of this study were to determine the effect of BAP (benzylaminopurine) and sucrose for shoot multiplication and microtuber induction of in vitro culture of satoimo. The experiment was arranged in two factors: BAP (0, 1, 2 and 3 mg/L) and sucrose (30, 60, 90 and 120 g/L). The result showed that the single effect of BAP or sucrose and interaction of both significantly increased the number of shoots. The effect of 2 mg/L BAP was more homogeneous than that of 1 and 3 mg/L BAP. Sucrose with the concentration of 30 g/L was the best concentration for shoot multiplication. The highest number of microtuber was achieved with 2 mg/L BAP + 30 g/L sucrose treatments, but tended to decrease due to increasing sucrose concentration. In 2 and 3 mg/L BAP treatments, the number of microtuber increased along with the increasing sucrose concentration.Keywords: satoimo, in vitro shoot, microtuber, benzylaminopurine, sucrose ABSTRAKSatoimo (Colocasia esculenta (L) Schott var antiquorum) merupakan bahan pangan alternatif non-beras. Untuk mendukung produksi massal satoimo di beberapa wilayah di Indonesia, multiplikasi tunas dan induksi umbi mikro secara in vitro merupakan tahapan yang harus dilakukan. Tujuan penelitian ini adalah untuk mengetahui pengaruh BAP dan sukrosa terhadap multiplikasi tunas dan induksi umbi mikro satoimo dalam kultur in vitro. Perlakuan terdiri dari 4 taraf konsentrasi BAP (0, 1, 2 dan 3 mg/L) dan 4 taraf konsentrasi sukrosa (30, 60, 90 dan 120 g/L). Hasil penelitian menunjukkan bahwa BAP dan sukrosa secara tunggal serta interaksinya berpengaruh nyata terhadap multiplikasi tunas in vitro. Pengaruh konsentrasi BAP 2 mg/L lebih homogen dibandingkan perlakuan BAP 1 dan 3 mg/L. Sukrosa 30 g/L merupakan konsentrasi terbaik untuk multiplikasi tunas. Umbi mikro terbanyak terdapat pada perlakuan BAP 1 mg/L + sukrosa 30 g/L tetapi cenderung mengalami penurunan jika konsentrasi sukrosa dinaikkan pada konsentrasi BAP tetap. Pada perlakuan BAP 2 dan 3 mg/L jumlah umbi mikro yang terbentuk cenderung meningkat seiring dengan peningkatan konsentrasi sukrosa.Kata kunci: satoimo, tunas in vitro, umbi mikro, benzilaminopurin, sukrosa

Abstract

Existence of heavy metals in industrial waste is gaining global attention since their negative impact to environment. One of the efforts to solve the problem was to use plant to absorb metal in liquid medium, known as rhizofiltration. This research was aimed to select aquatic plants which showed relative resistantce and susceptibility to chromium. Four species of aquatic plants (Pistia stratiotes, Eichhornia crassipes, Lemna minor and Salvinia sp.) were grown in artificial medium (Hoagland) supplemented with 0, 1, 2, 4 and 8 ppm chromium. The plants resistance and absorption toward chromium was observed based on the morphology and chromium content in their biomass. Based on their resistance to and absorption of chromium, the selected plants were tested further in liquid waste of tanning industry. In Hoagland medium, Salvinia sp. demonstrated 67.2% higher resistance and absorption toward chromium while that of P. stratiotes 20.3% lower compared to other plants which were tested. This result could be applicable in reducing such environmental pollutant as the heavy metal chromium from industrial waste. Salah satu upaya untuk mengatasi masalah tersebut adalah memanfaatkan tanaman untuk menyerap logam dalam medium cair atau dikenal sebagai fitoremediasi. Penelitian ini bertujuan untuk mengetahui tanaman air lokal yang tahan dan peka secara relatif terhadap kromium. Empat spesies tanaman air (Pistia stratiotes, Eichhornia crassipes, Lemna minor, dan Salvinia sp.) ditumbuhkan pada medium buatan (Hoagland) yang dipasok kromium 0, 1, 2, 4, dan 8 ppm. Pengujian toleransi tanaman dan serapan terhadap kromium dilakukan berdasarkan pengamatan morfologis serta analisis kadar kromium dalam biomasa. Berdasarkan daya tahan dan serapan kromium, tanaman terseleksi diujikan lebih lanjut dalam limbah cair industri penyamakan kulit. Dalam medium Hoagland, Salvinia sp. mempunyai ketahanan dan serapan kromium lebih tinggi sebesar 67,2% sedangkan P. stratiotes mempunyai ketahanan dan serapan kromium lebih rendah sebesar 20,3% dibandingkan tanaman lain yang diujikan. Hasil penelitian ini dapat diterapkan untuk mengurangi bahan pencemar lingkungan berupa logam berat kromium dari limbah industri.Kata Kunci: Fitoremediasi, kromium, medium Hoagland, tanaman air, limbah cair
