Squalen publishes original and innovative research to provide readers with the latest research, knowledge, emerging technologies, postharvest, processing and preservation, food safety and environment, biotechnology and bio-discovery of marine and fisheries. The key focus of the research should be on marine and fishery and the manuscript should include a fundamental discussion of the research findings and their significance. Manuscripts that simply report data without providing a detailed interpretation of the results are unlikely to be accepted for publication in the journal.

Abstract

The objective of this research was to determine the saccharification and the fermentation efficiency of seaweed solid waste hydrolysate by Saccharomyces cerevisiae (Accession number 3012.254) in anaerobic condition. The optimum saccharification yield of acid pretreated waste (40.93±1.72)% was obtained after 48 hours with  saccharification rate of (0.51±0.02) g/l.h. Higher yield was shown by NaOH pretreated waste (67.29±1.24)% after 24 hours with saccharification rate of (0.81±0.06) g/l.h. The fermentation of enzymatic hydrolysates of acid and alkali pretreated samples by S. cerevisiae produced a maximum of 7.52±0.24 g/l and 14.5±0.54 g/l ethanol respectively after 72 hours fermentation. Maximum ethanol yield was 0.31±0.03 g/g and 0.40±0.02 g/g sugar respectively for acid and alkali pretreated samples. The ethanol yields showed that alkali pretreated sample produces higher conversion substrate ratio (80% of theoretical yield) compared to acid pretreated sample (62% of theoretical yield).

Abstract

Transglutaminase (TGase), an enzyme that catalyzes the formation of inter- and intra-molecular e-(l-glutamyl) lysine (GL) crosslinks, plays an important role in surimi-based products production. The development and the diversification of surimi-based products have recently been getting popular in Indonesia. These surimi-based products can be made from various types of fish. These products generally exhibit good gel strength properties,  depending on the fish type and the processing method used. Transglutaminase plays an important role in generating such properties. Fish’s endogenous TGase reduces quickly after it is caught and is almost completely destroyed by freezing it, applying exogenous TGase may improve fish’s gel forming ability. Microbial transglutaminase (MTGase) can potentially be used to increase gel properties. In this research, a total of 228 Streptomyces strains from marine and terrestrial environments were screened and selected based on their ability to produce MTGase. Strain TTA 02 SDS 14 exhibited the highest activity; and therefore, it was selected for further study. The 16S-rRNA gene analysis showed that it shared 99% similarity to S. thioluteus. In order to optimize MTGase activity, enzyme production was carried out using six different formulas media, designated as media A, B, C, D, E, and F. The result shows that the highest MTGase activity was observed in medium B that contains pepton (1.5%), MgSO4.7H20 (0.1%), KH2PO4 (0.5%), Na2HPO4 (0.5%), soybean powder (2%), potato starch (2%), and glucose (1.5%). The MTGase activity reached the highest level (1.45 U/ml) after 4 days of incubation

Abstract

Microbial Transglutaminase (MTGase, EC 2.3.2.13) is an enzyme that catalyzes the transfer of acyl group. Many microbial strains that produce MTGase belong to Streptomyces members. This research was aimed at cloning of a MTGase gene. PCR–based screening of ten MTGase-producing streptomyces isolates from soil in West Nusa Tenggara led to detection of one potential isolate, designated as TTA 02 SDS 14. The partial  MTGase-encoding gene (470 bp)  was amplified by PCR and sequenced. The sequence result indicate its similarity of 93 % with that of Streptomyces cinnamoneus. The 16S rRNA gene analysis showed its identity as Streptomyces thioleteus. Fosmid-based construction of a genomic library from the isolate  and subsequent screening led to the isolation of  a ~40-Kb fosmid harboring a MTGase gene.

Abstract

Indonesian tropical soft corals are valuable resources that produce pharmacological cytotoxic cembranoids. However,  the manual structure determination in these compounds requires adequate knowledge of organic chemistry. This study presents the application of Logic Structure Determination (LSD) as a freeware Computer Assisted Structure Elucidation (CASE) for automatic molecular structure determination of cembranoid compounds from soft corals species. 12 Nuclear Magnetic Resonance (NMR) datasets of cytotoxic cembranoids were used to evaluate the accuracy of LSD in generating the possible structures. The results of this study shows that LSD generated numerous possible molecular structures as the data input files were only derived from 2D-NMR HMQC (Heteronuclear Multiple-Quantum Correlation), COSY (Homonuclear Correlation Spectroscopy), and HMBC (Heteronuclear Multiple Bond Correlation). The accuracy was significantly enhanced (only 2-4 possible cembranoid structures from each NMR dataset) with the addition of H2BC (Heteronuclear 2 Bond Correlation) experimental data. This may indicate that VLRC (Very Long-Range Correlations) significantly affects LSD capability. Furthermore, LSD with direct 2 bond NMR experimental data is a reliable CASE technique for cembranoid compounds identification. In general, this freeware-CASE has the potential to be applied  on other types of small molecule compounds and may serves as a solution for elucidation bottleneck step in studies on Indonesian natural products.

Abstract

The objectives of this research were to assess cytotoxic activity and examine characteristic of sea cucumber Actinopyga sp. methanolic extract. Cytotoxicity assay was conducted using MTT method against WiDr (colon cancer) and T47D (breast cancer) cell lines. Actinopyga sp. methanolic extract was characterized by using phytochemical screening, Transform-Infrared (FT-IR) spectroscopy, dan  Liquid Chromatography Ion Trap Time of Flight Mass Spectrophotometer (LC-IT-ToF-MS).  The result showed that Actinopyga sp. methanolic extract inhibited Widr and T47D cell lines viability with the IC50 value of 55.93 and 87.55 µg/mL, respectively. Functional groups analysis showed the presence of hydroxyl, amine, carboxylic acid, nitrate, amide, sulphur, ester, and ether. LC-IT-ToF-MS analysis of crude extract showed that it containing many steroids compound.
