Squalen publishes original and innovative research to provide readers with the latest research, knowledge, emerging technologies, postharvest, processing and preservation, food safety and environment, biotechnology and bio-discovery of marine and fisheries. The key focus of the research should be on marine and fishery and the manuscript should include a fundamental discussion of the research findings and their significance. Manuscripts that simply report data without providing a detailed interpretation of the results are unlikely to be accepted for publication in the journal.

Abstract

Despite the fact that fungi are important sources of both bioactive compounds and mycotoxins, and that they are very ubiquitous in our environment, their species identification is hampered by incomplete and often unclear literature. Fungi identification is primarily based on their phenotypic and physiological characteristics. Nowadays, many molecular methods to identify fungal species have been developed. One of the methods considered as a new concept to rapidly and accurately identify unknown fungal sample is DNA Barcoding. This literature review will outline the use of DNA barcoding approach to rapidly identify fungal species and the use of ITS region that recently has been designated as primary DNA barcode for fungal kingdom. “DNA barcode” is a short, highly variable and standardized DNA region with approximately 700 nucleotides in length, which is used as a unique pattern to identify living things. Internal Transcribed Spacer (ITS) region of nuclear DNA (rDNA) has become the most sequenced region to identify fungal taxonomy at species level, and even within species. ITS region is a highly polymorphic non-coding region with enough taxonomic units. Therefore, it is able to separate sequences into species level. Even though ribosomal ITS as a universal barcode marker for fungi is still hampered by few limitations, the ITS will remain as the key choice for fungal identification. The search for alternative regions as DNA marker to improve fungal identification, especially in specific heredities, has already started.

Abstract

With the aim of searching for new antibacterial compound from marine soft corals, the investigation had been conducted on antibacterial activity of an extract from soft corals Sarcophyton trocheliophorum in the n-hexane, ethyl acetate, n-butanol, and aqueous fractions. The antibacterial activity was tested against two Gram-positive bacteria, viz. Bacillus subtilis (ATCC 6633) and Staphylococcus aureus (ATCC 25923), and two Gram-negative bacteria, viz. Escherichia coli (ATCC 25922) and Vibrio cholerae (ATCC 14035) using the agar disc diffusion assay. Among them, the n-hexane fraction was the most active against three tested bacteria, viz. Bacillus subtilis, Staphylococcus aureus and Vibrio cholerae at the concentration 125 µg/ml, with inhibition zone 14.2, 18.2, 13.8 mm, respectively. Isolation and purification of the active component from the n-hexane fraction led to a known cembranoid-type diterpene, sarcophytoxide. The chemical structure of the isolated compound was determined by IR, MS and NMR, as well as compared to data from the literature. Sarcophytoxide showed moderate activity against B. subtilis, S. aureus and V. cholerae, with a minimum inhibitory concentration (MIC) of 125, 100, 125 mg/ml, respectively.

Abstract

Screening of alginate lyase producing bacteria associated with seaweed Sargassum crassifolium was carried out, and isolate S245, identified as Bacillus megaterium S245 was found to produce high alginate lyase activity. This research was conducted to evaluate activity of the alginate lyase enzyme at various pHs, temperatures and substrates. Polymannuronate and polyguluronate were used to evaluate substrate specificities. Alginate lyase activity was assayed by analysis of reducing sugar released using the 3,5 dinitrosalicylic acid (DNS) method. The research showed that the activity of alginate lyase was optimum at pH of 7.0 and  temperature of 45 0C. This enzyme was active for both polymannuronate and polyguluronate susbtrates. The Vmax and Km of this enzyme for polymannuronate and polyguluronate were 200 unit/ml/min and 79.8 mg/ml for polymannuronate substrate and 27.78 unit/ml/min and 13.17 mg/ml for polyguluronate substrate. This enzyme showed unique characteristic in working toward the two substrates.

Abstract

Unsaturated fatty acid in food has an effect to improve human health, while saturated fatty acid has the opposite impact. Ratio between both types of fatty acids is important to evaluate the biological material for nutraceutical product development. This paper aimed to evaluate the nutraceutical potency of sea cucumber from Kendari Bay Waters, South East Sulawesi, based on their fatty acids composition. The analysis was conducted by the 1H-NMR (Hidrogen-Nuclear Magnetic Resonance) method. Several species within 4 genera (Holothuria, Bohadschia, Actinopyga, and Stichopus) were selected as the subject of the study. Comparison of fatty acid composition has been done in genera level, the preliminary study found that variation between species among the same genera was insignificant (p >0.05). Results of the study detected that Holothuria contained with an even ratio of Poly Unsaturated Fatty Acids (PUFA) and Saturated Fatty Acids (SFA), but Actinopyga was detected with low PUFA/SFA ratio. It may suggest that Holothuria from this region is the most potential raw material for nutraceutical product development. Meanwhile, precaution is needed for genera that contained with low PUFA/SFA ratio. Furthermore, comparison with other studies detected that fatty acids composition is not morphological specific, but vary, according to food viability, environmental and geographic location. Therefore, preliminary screening is important in evaluation of sea cucumber material for nutraceutical product development.

Abstract

Marine algae contains bioactive secondary metabolites that  have potential to be developed as antioxidant. The aims of  this research were to investigate antioxidant activity and total phenolic compound of marine algae collected from Binuangeun Beach, Banten and to characterize antioxidant compounds from selected algae species. Antioxidant activity was determined by 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) assay and Ferric Ion Reducing Antioxidant Power (FRAP) Assay, while total phenolic content was estimated by Folin-Ciocalteau method. Isolation of bioactive antioxidant compound was performed using SiO2 column chromatography and preparative high performance liquid chromatography (HPLC). The antioxidan activity evaluation was conducted to methanol extracts  from 20 species (7 Chlorophyta, 9 Phaeophyta and 4 Rhodophyta). Results showed that Padina australis extract was found to be the most active. The major bioactive antioxidant compound was identified as fucoxanthin and a polar compound that was suspected as phenolic compound. The extracts of P. australis presented the highest phenolic content (58.59 mg GAE/g). A significant correlation between antioxidant capacity and total phenolic content was found, indicating that phenolic compounds are the major contributors to the antioxidant properties of P. australis.
