Squalen publishes original and innovative research to provide readers with the latest research, knowledge, emerging technologies, postharvest, processing and preservation, food safety and environment, biotechnology and bio-discovery of marine and fisheries. The key focus of the research should be on marine and fishery and the manuscript should include a fundamental discussion of the research findings and their significance. Manuscripts that simply report data without providing a detailed interpretation of the results are unlikely to be accepted for publication in the journal.

Abstract

The presented study has been carried out in order to study the quality of Alkali Treated Cottonii (ATC) made from Eucheuma cottonii which being collected from different regions in Indonesia (Belitung, Nusa Tenggara Barat and Lampung). The quality variables analyzed were the characteristics of raw materials (Clean anhydrous weed and impurities) and the characteristics of ATC produced (moisture content, ash content, acid insoluble ash content, yield, gel strength, sulphate content, gelling -melting point). The analysis was done in 3 replicates and the data were statistically analyzed using SPSS 15 package software. Results indicated that the raw material from Lampung had a better quality than those from Nusa Tenggara Barat and Belitung. In addition, the characteristics of ATC produced from these three raw materials showed that seaweed from Lampung produced better quality ATC than those from Nusa Tenggara Barat and Belitung in terms of their gel strength, sulphate content and yield.

Abstract

Transposon is well-known as mobile element found abundant both in prokaryote and eukaryote genomes. In bacteria, transposon (famous name of a transposable DNA) could jump from chromosome to plasmid and its contrary. One type of transposons in bacteria known as insertion sequence (IS), it does not contain any additional genes except a gene encoding transposase, an enzyme that correlated to transponsition activities. The finding of transposon insertion unfortunately found during cloning of a fragment derived from deep-sea metagenomic DNA in this research. In the initial, this research was aimed to clone and characterize the á-amylase encoded gene derived from metagenomic DNA isolated from deep-sea water and sediment of Kawio Island, North Sulawesi. Metagenomic DNA has been isolated from deep-sea water and sediment and by using Whole Genome Amplification (WGA) technique, the DNA it could be increased in quantities to 146,31 ng for each 1 ng of metagenomic DNA. A fragment of ~1000 bp in length was obtained by using touchdown PCR method. The presence of a transposon in this DNA fragment is proposed as a hypothesis for losing ~700 bp leaving just 310 bp cloned sequence. Analysis of sequencing result showed a highest similarity between this 310 bp partial fragment with a replication protein (Rep) encoded gene from Pseudomonas putida (Query Coverage: 88%; Max. Identity: 80%, Positive: 86%) and this protein is known to be involved in plasmid replication where transposase encoding genes known usually presence together with this gene (Rep gene) in a bacterial plasmid.

Abstract

The needs of membrane biomaterials to prevent the migration process of epithelial cells into the bony area after tooth withdrawal process, for filler the holed tooth and to cure gingivitis have increased significantly. In this study, a biomembranes composed of chitosan (CTS) and hydroxyapatite (HA) were prepared. CTS was made of crab shell and HA was made of tuna fish bone. There were 4 groups of treatments, i.e.: K0: only 4 g CTS as a control; K1: combination of 4 g CTS and 2 g HA; K2: combination of 4 g CTS and 4 g HA; and K3: combination of 4 g CTS and 6 g HA. The dried and solid membrane was soaked in 5% Na2SO3 for 1 hour, and then dried in oven at 60 0C for 24 hours. The composite membrane properties, i.e.: water absorption, biodegradation, tensile strength, elongation at break, functional group (by FTIR), and microstructure (by SEM) were evaluated. Result showed that the increasing of HA content in membrane will decreasing the water absorption. The increasing of HA content in membrane and soaking time in Phosphate Buffered Saline (PBS), can increase in biodegradation rate. The biodegradation rate of membrane was about 4.44-6.01% during 27 days of soaking time. The increasing soaking time in PBS can decrease the tensile strength of membrane at about 18.33%-21.17% and decreased the elongation at break at about 26.64%-59.94%. FT-IR analysis indicated that there was a crosslinking between a CTS-HA composite membrane and Na2SO3. Among all the composites studied, CTS-HA (4/2 g/g) composite membrane is the best one for Guided Tissue Regeneration (GTR).

Abstract

Chemical investigation of the endophytic fungus Botryosphaeria australis isolated from Avicennia marina originally from Hainan Province, P.R. China, yielded a new compound botryosphaenin (1), from the class of napthoquinone, together with 5 known compounds, botryosterpene (2) and 5-hydroxy-2,7-dimethoxynaphthalene-1,4-dione (3) and its derivatives, 6-ethyl-5-hydroxy-2,7-dimethoxynaphthalene-1,4-dione (4), O-methylaspmenone (5), O-methylasparvenone (6) and 5-(carboxymethyl)-7-hydroxy-1,4a-dimethyl-6-methylene decahydron aphthalene-1-carboxylic acid (7). Their structures were determined on the basis of spectroscopic methods including 1D (1H, 13C, and DEPT) and 2D (COSY, HMQC, HMBC, and ROESY) NMR experiments and by mass spectroscopic measurements The new compounds, 1 showed activity against the bacterial pathogens Staphylococcus aureus, several Streptococcus species and Bacillus subtilis, but also against the eukaryotic cell lines THP-1 (human leukemia monocyte) and BALB/3T3 (mouse embryonic fibroblast).

Abstract

Alginate lyases are group of enzymes which catalyze depolymerization of alginate into oligosaccharides. Alginate lyase have been widely used in many applications such as in production of bioactive oligosaccharides, control of polysaccharide rheological properties, and polysaccharide structure analysis. The products of alginate lyase, polysaccharide structure analysis, alginate oligosaccharides (AOS) have many biological activities including act as prebiotics, immune modulator, anticoagulation, antioxidant, anticancer, growth promoting activities, promote production of antibiotics and ethanol. In relation to the importance of alginate lyases, their potential aplications and prospect in development of new bioactive products, we present review of the enzymes, sources, mechanism of activity and potential applications. This paper also discussed some new biological engineering in alginate lyase production.
