Abstract

The enzyme laccase (EC.1.10.3.2: oxygen oxidoreductase) is an extracellular enzyme that has a benefit in various industrial fields. This enzyme catalyzes the oxidation of various inorganic and organic compounds such as phenolic compounds, diamine, aromatic amines and ascorbic acid (Mayer and Staples, 2002). Laccase enzyme was first isolated by Yoshida in 1883. He extracted from the Japanese lacquer tree (Rhus vernicifera) (Thurston, 1994; Risdianto, 2008 ; Shraddha dkk., 2011). Objectives of this study is to determine the commercial activity of the enzyme laccase derived from Rhus vernicifera using guaiacol as a substrate. The enzyme activity was measured using a UV-Vis spectrophotometer Thermo Scientific GENESYS 10S at a wavelength of 490 nm using guaiacol as substrate. The results of this study indicate that this enzyme has an activity of 0,0247 ± 0,0002 U/L (at pH 5,5 and a temperature of 40 oC).

Abstract

Fungus Sporothrix sp. LBKURCC43 is one of fungi which has been succeded to be isolated from Dahlia tuber (Dahlia variabilis). This reseach aimed to produce secondary metabolytes and antioxidant activity secondary metabolytes of fungus Sporothrix sp. LBKURCC43 in Huang medium. Measurement of antioxidant activity on secondary metabolytes were analyzed by DPPH (2,2-diphenyl-1-picryl-hydrazyl) showed the highest activity was found in 20th day secondary metabolytes in Huang medium with IC50 828,614 μg/mL.

Abstract

Helminthostacshyszeylanica is a plant of the family ophioglossaceae known as the tunjuk langit plant. The roots of this plant is used as a 100 days cough remedy, dysentery, nasal disease and lung disease. This plant contains saponins, andflavonoids. This study aims to isolate and characterizethe flavonoid compounds from the root of tunjuk langit. Isolation methods used polar fractionation method. Some 7.5 kg oftunjuk langit wasmacerated with methanol17 L for 3x24 hours later ultrasonicatide for 1 hour. Extract thick about 100 gr then partifioned with methanol mixture and n-hexan 2:1. Methanol fraction of partition result thickened then in chromatography liquid vacuum (VLC) in 12 fraction (n-hexane – etilacetate – methanol). Flavonoid compound obtained after VLC result on fraction 5 and 6 is combined then in flash chromatography. Flavonoid compound such yellow crictal on vial 19 is coded as HZ19. The purity of this compound was analized by TLC and HPLC as well as determination of the melting point range. Structure of pure compound was determined using spectroscopy method including UV-Vis, infrared (IR). Based on spectroscopic data HZ19 known that flavonoid is a Flavon kind in ugonin group.

Abstract

Identification of endophytic fungi isolated from orange flowering dahlia tubers of plants from outside Padang, West Sumatra by Lorenita et al., (2013) and pre-study by researchers showed different results, so that carried serially purification. Interest purification serially performed in this study is to obtain pure isolates. Purification is done by serial dilutions ranging from 10-1-10-30 fungi spores. Identification Microscopic morphology using a light microscope Olympus CX41. The results of the purification are serially obtained two isolates, namely LBKURCC40A and LBKURCC40B, and identification of macroscopic and microscopic morphology showed LBKURCC40A and LBKURCC40B is Aspergillus sp.

Abstract

A research of antifungal activity from the essential oil of Senna alata (L.) Roxb against fungus that causes opportunistic infections.This research was conducted by agar diffusion method to test fungus Aspergillus fumigatus, Candida albicans and Penicillium marneffei. The result of water vapor distillation of the leaves Senna alata obtained rendemen by 0,125%. The research had done in variation concentrastions respectively 25%, 20%, 15% ,10% and 5% obtained from observations of the average diameter of inhibitions on fungus Aspergillus fumigatus by 17.48 mm, 15.18 mm, 13.28 mm, 11.33 mm and 9.80 mm, Candida albicans by 12.86 mm and the Penicillium marneffei 19.46 mm, 16.06 mm, 14.48 mm, 13.16 mm, 12 .23 mm. Analyzed for essential oil constituents by GCMS. The quantitatively significant constituents of the leaf essential oil of Senna alata (L.) Roxb were 1,8 cineole (48.97%), α- pinene (24.96%), 1,4 cyclohexadine (9.53%), Trans- β- ocimene (7.48%) and P-cymene (4.53%).

Abstract

ABSTRACT Sweet potato (Ipomoea batatas (L.) lam) leaf has been reported contain flavonoids that can modulate GABAa. It a role in providing anticonvulsant effects. The standarization of extract and anticonvulsant effect of ethanol extract of sweet potato (Ipomoea batatas (L.) lam) leaves with striknin induced seizures in mice at dose levels 200, 400, 800 mg / kgbw an diazepam as comparison has been conducted. The test involved spesific and non spesific parameters according to standarisation method from legitimate literature. Result showed that on extract had a thick form, green-blackish, spesific smell, bitter taste, content of compounds solube in water 57,24%, solube in ethanol 40,11%, water content of 5,11%, ash content 7,84%, ash content solube in acid 5,48%, ash content unsolube in acid 0,72%, and thin layer chromatography profile of extract showed flavonoids, fenols, steroids, and alkaloids. Parameters of observation of anticonvulsion effect used were onset of seizure, recovery time, and death time. Result showed a diference significant, extract of sweet potato (Ipomoea batatas (L.) lam) significantly delayed the onset time of convulsions (314,8±16,27 seconds) and death time (323,8±8,10 seconds) in doses 200 mg/kgbw. The highest anticonvulsant effect was showe by doses 800 mg / kgbw, with onset time of convulsions (555.6 ± 11.63 seconds) and recovery time (83.4 ± 4.66 seconds). Result showed that increasing doses extract of sweet potato (Ipomoea batatas (L.) lam) leaves has a positive correlation to prolong onset time, accelerate the recovery time, and delay the death time. So that it can be concluded that increasing the dose can magnify the effects of anticonvulsant

Abstract

The stages of swamedication services include patient assessment, which is a patient assessment called information excavation. Information excavation is intended to assess patients who have carried out safety assessment, accuracy and rationality swamedikasi by patients. The purpose of this study is to determine the description of attitudes of pharmaceutical personnel in obtaining information about toothache at pharmacists Pekanbaru. This research is a descriptive observational study with a sampling method that consists of systematic random sampling of 100 people from pharmaceutical manpower. From the research that was done the number of staff found pharmacist assistants who perform the extracting information as much as 63%, the percentage score of extracting information obtained 19.25% with a gesture interpretation is lacking, while the number of pharmaceutical technical personnel that it Extracting information as much as 37%, the percentage of score to be obtained from information obtained 30.7% with less attitude and interpretation there are pharmacists who perform services swamedikasi extracting information about the dental pain. The total percentage of information extraction by the staff found pharmacist assistants and pharmaceutical technical personnel as 23.5% with less interpretation of the posture

Abstract

Alstonia spatulata Bl is a traditionally important medicinal plant. Roots, stems, leaves, and fruits of this genus have been used in traditionalmedicine to treat malaria, diarrhea, fever, fatigue, liver disease, dysentery, and diabetes. The aim of this research was to isolation and characterizationthe secondary metabolites of stem bark of A. spatulata . The result of fitochemistry test for the stembark of A. spatulata showed that it containssecondary metabolite of terpenoid and alkaloid groups. The result of isolation process yielded a white crystal of ASSB-C with n-heksan solution. Ithas melting point 196-198 0C. The analysis of 1H-NMR, 13C-NMR, HSQC, HMBC, COSY, NOESY and FT-IR spectra for ASSB-C showed that it isincluded into triterpenoid group which is called β-Amirin

Abstract

The study of anti-inflammatory activity and isolation of the methanolic extract, hexane and ethyl acetate fractions of the leaves of puwarkincung (Nicolaia speciosa Horan) have been done. Isolation was carried out by extraction, fractionation and multiple column chromatographicmethod, followed by recrystallization technique. Two compounds were isolated and labeled as F4 and S3. F4 was derived from n-hexanefraction as yellowish-white crystals (20 mg) with mp. 55–57oC and S3 from the ethyl acetate fraction (15 mg) as yellow crystals with mp.68–69oC. Anti-inflammatory activity of methanolic extract using Paw Edema method showed that the dose 250 and 1000mg/kgbw wereactive as anti inflammatory agents.

Abstract

Plectranthus amboinicus or bangun-bangun is a traditional medicinal plant used for postpartum recovery in North Sumatera. The aims ofthis study were to detemine antioxidant and antimicrobial activity of leaves Plectranthus amboinicus. The leaves of Plectranthus amboinicuswere extracted with using n-hexane, ethyl acetate and methanol, successively. The extracts were tested to antioxidant activity assay usingDPPH with two fold dillution tehnique and ascorbic acid was used as standard. The antimicrobial activity was conducted using agar diffusionmethod against Escherichia coli, Staphylococcus aureus and Candida albicans. The Amoxsan used as positive control for antibacterialassay, while ketokenazole was for antifungi test. Extracted solvents were used as negative control. The n-hexane and ethyl acetate extractsdid not show any antioxidant activity. While, the IC50 free radical DPPH of methanol extract was 90.96 mg/mL and ascorbic acid was 58.792mg/mL. The antimicrobial activity of n-hexane and Amoxsan againts Escherichia coli were 11.95 and 7.53 mm. The inhibition zone againtsStaphylococcus aureus were 11.30 and 11.57 mm. The antimicrobial activity of ethyl acetate and Amoxsan againts Escherichia coli were12.29 and 7.71 mm. The inhibition zone against Staphylococcus aureus were 13.72 and 11.57 mm. The n-hexane and ethyl acetate did notshow any antifungi activity towards Candida albicans. The methanol extract did not show any inhibition zone againts the selectedpathogens.
