Abstract

Tobacco plants are widely used as raw material for cigarettes, but the results of the study show that in tobacco leaves contain compounds that can be used in the pharmaceutical and health fields. Tobacco leaves contain essential oils that have a distinctive aroma. The study aims to examine the antibacterial activity of tobacco essential oil from six sources tobacco such as Temanggung, Yogyakarta, Purwodadi, Boyolali, Blitar and Probolinggo. Tobacco essential oil is obtained by the steam-water distillation method. Antibacterial activity evaluation was carried out by diffusion method and the minimum inhibitory concentration on two bacteria, Escherichia coli and Staphylococcus aureus. The yield of tobacco essential oil ranged from 0.54-3.39%. Tobacco essential oil had antibacterial activity against bacteria E. coli and S. aureus in the presence of inhibition zones in bacterial media. The biggest inhibition zone was in Yogyakarta tobacco essential oil of 26 mm for S. aureus bacteria, while for E. coli bacteria the biggest inhibition zone was Blitar tobacco essential oil which was equal to 21 mm. The inhibition zone was not much different from the inhibition zone in tetracycline antibiotic testing (positive control) which was 25 mm in S. aureus and 21.5 mm bacteria in E. coli bacteria. Tobacco essential oil from Probolinggo was able to inhibit the growth of S. aureus bacteria up to a concentration of 6.25% and E. coli bacteria to a concentration of 12.5%.

Abstract

Dental caries is a disease that many people experience, which is formed by the buildup of plaque on the teeth and the presence of bacteria. The bacteria known to be involved in the formation of dental caries is Streptococcus mutans. Dental caries used to be treated using antibiotics. However, it is known that many antibiotics are no longer effective against this bacteria hence it is necessary to look for another alternative. One alternative choice as an antibacterial is Noni fruit. This study aims to identify the bioactive compounds of noni fruit (Morinda citrifolia L) and to know the mechanism of action in inhibiting the growth of Streptococcus mutans. Noni fruit was macerated using 96% ethanol, then fractionated using n-hexane and ethyl acetate. Antibacterial activity test was done using agar difusion method, and identification of bioactive compounds was conducted using GCMS. The results showed that ethyl acetate fraction has the highest inhibitory activity on the growth of Streptococcus mutans with inhibition zone of ±17 mm and MIC (minimum inhibitor concentration) value of 10%. The inhibition mechanism is asumed to be done by making holes in cell membranes, which can be seen from the high concentration of proteins and nucleic acids coming out of cells after treatment that in turn means that cell leakage has occurred. The results of this study indicate that noni fruit (Morinda citrifolia L) contain bioactive quinolone compounds that can be a new source for treating dental caries

Abstract

Endophytes are potential as a source of active compound producer. Endophytes that is isolated from tropical medicinal plants has been getting attention due to its high biodiversity and active compound producing ability. L-asparaginase is the first enzyme used as chemotherapeutic agent for leukemia. The aim of this study is to collect the endophytic bacteria associated with tropical medicinal plants from Sumba Island and investigate the activity of L-asparaginase without L-glutaminase from endophytic bacteria isolates. The samples Gliricidia sepium, Pittosporum moluccanum, Clerodendrum buchanani, and Zingiberaceae are collected from Wanggameti, East Sumba, NTT. Samples are sterilized using surface-sterilization method and endophytic bacteria are isolated using plant piece method on R2A media. Selected endophytic bacteria are identified by 16S rDNA sequences. L-asparaginase screening is conducted using modified R2A with addition of L-asparagine and phenol red as colour indicator. A total of 34 isolates of endophytic bacteria were collected from 5 samples. A total of 14 genus consisted of 17 different bacterial species were obtained from 34 selected isolates. Endophytic bacteria of P. stutzeri strains of SMKL1 and R. radiobacter strains of SMKW2 from the Kahili plant were needed as L-glutaminase-free L-asparaginase and were potential to be candidate of leukemia cancer chemotherapy

Abstract

Drugs expenditure is the highest proportion in health services budget. financing for health services in National Universal Health Coverage era is carried out by BPJS payed to referral health facilities namely hospitals. The medicine cost at public service agency/District or Province public service agency is unknown certaintly. This descriptive study was advanced analyzes of the secondary data of 84 hospitals from health financing research in the year of 2016. The result showed that there were problems of drug availability in class B 94,6%, C 78,9% and A 77,8%. The highest proportion of generic drugs cost to total drug cost is class C hospital, followed by B class and A class hospitals i.e 40,57%, 37,83% dan 23,74% respectively. The cost of non generic drug compared to generic drugs at A, B and C class is 2,22 times, 1,15 times and 0,86 times respectively. The highest cost of generic drug is in outpatient specialist services in all class of hospitals. The cost of the drugs was increasing higher as well as the hospital class. Conclusion: The non generic drugs cost dominated in A and B class hospital while C class was generic drug cost. Generic drug use policy should be socialyzed by government and to be considered by management in order to be able to carried out cost efficiency.

Abstract

Inflammation is known as the basic mechanism underlying various chronic diseases. Macrophage activation by inflammatory stimulus induces the release of inflammatory mediators, thus the mediators becoming a promising target of anti-inflammatory drug development. Previous studies indicated that fucoidan has anti-inflammatory activity by inhibiting the release of proinflammatory mediators. The aim of this study is determining anti-inflammatory activity of fucoidan crude extract form Sargassum crassifolium Garut waters by observing its effect on proinflammatory cytokines TNF-α, IL-1β, dan IL-6. Fucoidan is a polysaccharide substance which has various characteristics, depending on the source and the extraction method which is influencing its bioactivity. Sargassum crassifolium collected from Garut-West Java is extracted using diluted HCl 0,1 M and precipitated with ethanol to obtain fucoidan crude extract. The crude extract is tested on LPS-induced RAW 264.7 cells to evaluate its effect on TNF-α, IL-1β, and IL-6 level using ELISA method. The result showed that fucoidan crude extract decreased the level of TNF-α by the dose of 25 and 50 μg/ml, and decreased the level of IL-1β and IL-6 by the dose of 25 μg/ml. The dose of 50 μg/ml failed to inhibit IL-1β and IL-6 production. This study showed that fucoidan crude extract derived from S. crassifolium has anti-inflammatory activity to RAW 264.7 cells by the dose of 25 μg/ml.

Abstract

Obesity is a co-morbidity of cardiovascular disease, diabetes, and other metabolic diseases, which the prevalence increases every year. The use of herbal medicine is an alternative choice to overcome obesity. Clinical observation of decoction of herbal medicine (Dutch teak, kemuning, rhei radix and tempuyung) has been conducted for obesity therapy which has been proven to be effective in reducing body weight and body mass index (BMI). Innovation in steeping form is considered more practical than stew. Randomized open label pararel design research was carried out to assess the equal efficacy of the herbs in stew and steeping form. The number of subjects in each group were 30 subjects, they were patients of of Rumah Riset Jamu (RRJ) Hortus Medicus during June-October 2016 who met the inclusion criteria. BW and BMI are measured on Day-0, 28 and 56, while body height is measured once at Day-0. Both stew and steeping groups drank herbs twice a day for 56 days. At the end of the study, it was found that the mean of declining BW of the decoction group was 67.35 kg to 66.37 kg, while the steeping group 67.41 kg to 66.45 kg. BMI of stew group was decreased 26.85 kg / m2 to 26.46 kg / m2, while in steeping group 26.67 kg / m2 to 26.29 kg / m2. Statistic test result to compare BW and BMI of both two groups before and after treatment showed no significant differences in the two groups (p> 0.05). The ability of herbal medicines in steeping or stew forms can reduce the BW and BMI equally.

Abstract

Pharmaceutical services is a direct service and responsible to patients relating to pharmaceutical products aimed to improve the quality of life of patients. Quality of pharmaceutical services can be assessed based on outpatient satisfaction. The aim of the study was to compare the satisfaction of outpatients with pharmaceutical services in hospitals and primary health care. This study used a cross-sectional comparative study design and was conducted in February-November 2017 in 11 provinces, each province consisting of 2 districts/cities selected purposively. Samples were outpatients who got medicines in hospitals or primary health care at least 31 patients in each pharmacy. Satisfaction was assessed by the dimension of responsiveness, reliability, collateral, friendliness, and physical evidence. Data was collected by the questionnaires and analyzed using chi-square test. The results of the study show that outpatient satisfaction with pharmaceutical services in hospital and primary health care was 90,9% and 96.6%, respectively. The largest percentage of outpatients in hospitals and primary health care are in the age group of 40-59 years, female, has further education, and not working/housewives. There were significant differences in the age group, gender, and education of outpatients between hospitals and primary health care. There was no significant relationship between the characteristics (age, sex, education, occupation) of outpatients and satisfaction with pharmaceutical services in hospitals and primary health care

Abstract

Celery (Apium graveolens L) is a very popular plant as a soup and many other vegetable menus. Celery has benefits as a medicinal plant and the efficacy has been known worlwide. Research on Medicinal Plants and Herbs (Ristoja) conducted in 2012, 2015, and 2017 has collected thousands of herbs and some of them contain celery. This study carried out further analysis on the empirical use of celery as an ingredient of traditional medicine by traditional healers (Hattra) through interviews and observations. Based on Ristoja's data, they found 90 herbs and among them there were 20 local names of herbs which are used by Hattra. The celery by Hattra was empirically identified for 10 types of diseases treatment. It was used mostly for hypertension or high blood pressure treatment and justificatied scientifically through other scientific literatures. It was recommended that celery could be use for self-medication a mild hypertension patient. Celery was safe although there are still contra indications. The celery treatment should be socialized through promotive activities in elderly posyandu activities remain under supervision of health workers

Abstract

Tuberculosis is a global health problem, and there is even an increase in cases of multidrug-resistant tuberculosis in the world. Therefore, research is needed that can find new anti-tuberculosis drugs (OAT) that are more effective for tuberculosis treatment. In this study, the effect of (-)-epigallocatechin-gallate (EGCG) of tea leaves (Camellia sinensis) combined with the first-line OAT will be observed, in order to find out whether EGCG has anti-tuberculosis activity and can increase the potential of first-line OAT in-vitro. The anti-tuberculosis activity of EGCG was determined by broth dilution method using Middlebrook 7H9 media at concentration of 50, 100, 150, dan 200 ppm, then the potential of first-line OAT before and after combined with the EGCG was observed. The results showed that the activity of EGCG at concentration 50 ppm and 100 ppm could inhibit the Mycobacterium tuberculosis growth by 80%, at concentration 150 ppm by 90%, and at concentration 200 ppm by 100%. First-line OAT activity before combined with EGCG was ≥ 90% at 5 ppm rifampicin, 0.5 ppm isoniazid, 50 ppm pyrazinamide, and 5 ppm ethambutol. Whereas after combined with EGCG, the potential of each drug increased, marked by anti-tuberculosis activity achieved ≥ 90% at lower concentrations, i.e. rifampicin 0.5 ppm, isoniazid 0.25 ppm, pyrazinamide 20 ppm, and ethambutol 2 ppm. These results indicated that the potential of each first-line OAT increases after being combined with EGCG, and EGCG has potentiation effect when combined with those drugs. In conclusion, EGCG can increase the first-line OAT activity
