Abstract

This research was conducted from Maret until June 2008, in Big Hall of Development Marine Aquaculture Lampung.The research used experimental methodology, with 4 treatments and 3 replications. The aimed of research are toknow effects of vitamin C to increase a haemoglobin of blood and survival rate and measured effective C vitaminin prawn of humback grouper. The result showed that the highest haemoglobin rate of at PC (2 g/kg prawn), equalto 0,915 g/100 mL. The lowest haemoglobin found at treatment PA. equal to 0,066 g/100 mL. Result of variansianalysis indicate that addition of vitamin C at food give influence to increase the haemoglobin rate (P<0.05).Survival rate after experiment are 100 %. Parameter of water quality of were: temperature 29.3-30.2 oC, pH 7.81-8.27, salinitas 31-32 psu, DO 3,65-5 mg/L, and ammonia content (NH 3) 0.044-0.069 mg/1

Abstract

The fruits or seeds of Keben (Barringtonia asiatica L. Kurz) were used traditionally for fish poisons, to curvestomach and headeach. The aim of this research was to isolate and identify bioactive compound of n-butanolfraction of active compound against Artemia salina Leach larvae. Isolation and purification of n-butanol fractionwere carried out by column chomatography (SiO2, CHCl3-MeOH) and high pressure liquid chromatography (RP,MeOH). The result of purification was than tested using BSLT methode. The test showed that, BABU-2.4 had thehighest activity with LC 50 value was 30.19 ppm. BABU-2.4 was identified with FT-IR spectrophotometer and NMR(proton and carbon) spectrophotometer as derivate of glicoside.

Abstract

The contamination of toxic heavy metal Cadmium (Cd) in soils will be endanger the human health because it ismore available comparing to another toxic heavy metals. One method of Cd-contaminated soil bioremediation isusing exopolysachharide-producing bacteria Azotobacter. Exopolysachharides (EPS) can mobilize Cd through theformation of complex Cd-EPS which sequentially can increase the availability of Cd for plants uptake. A laboratoryexperiment has been done to study the EPS production and the viability of six Azotobacter isolates in the liquidculture containing 0.01, 0.1, and 1 mM CdCl2. The bacteria were cultured in liquid medium with and without CdCl2 for72 hours at room temperature. The EPS production was determined by gravimetric method after precipitationusing acetone and centrifugation at 7000 rpm. The result was that all of Azotobacter isolates produce EPS in thepresence of CdCl2. In the culture with 1 mM CdCl2, the density of Azotobacter sp. isolate BS3, LK5, LKM6 increasedsignificantly, and that of isolate LH16 decreased. No significant effect of CdCl2 on the density of isolate BS2 andLH15. This research suggested that some Azotobacter isolates were relatively resistence to the Cd and could bedeveloped as biological agents in Cd-contaminated soil bioremediation.

Abstract

Lactic acid bacteria is a group of bacteria with proteolytic activities enambling to grow on protein rich substratesuch as soymilk. This research was aim to isolate and identify lactic acid bacteria with have proteolytic activityfrom spontaneous fermented soy milk. Sixteen isolates out of 26 colonies isolated from fermented soymilk arepresumed as lactic acid bacteria. Among these 16 isolates, only 3 of them showed proteolytic activity. These threeisolates were further identify morphologically and only two isolates identified as Lactic Acid Bacteria, namelyR.1.3.2 and R.11.1.2. The ability of these isolates to produce acid and protease were observed. The results showedthat isolate R 1.2.3 higher ability in producing protease.

Abstract

Curcumin and β-carotene are two kinds of pigments serve as antitumor, anticancer and antioxidant agents. Theantioxidant activity of curcumin is stronger and more stable as compared to β-carotene, so it can protect β-carotene degradation from light and oxygen. Therefore curcumin has higher antioxidant activity as well. Theresearch was conducted by mixing curcumin and β-carotene in several molarity ratios followed by iradiation usingvolpi lamp for 0, 10, 30, 60, and 90 minutes. The result shows that curcumin has photoprotection effect because itcan protect photooxidation of β-carotene. Mixing of curcumin and β-carotene 1:1 was the most stable ratio forprotecting β-carotene.

Abstract

Observations on pelagic fish catch activities using bagan Rambo in Makassar Strait had been conducted in 2005and 2006 to obtain physical, chemical, and biological data and their processes in the operation of bagan Rambo inthe observation sites. The objective of the study was to overcome the overfishing problems which have beenhappenning in Makassar Strait, using a model of the trophic level approaches. Nine observation stations were setin three transects perpendicular to the coast. The measurement of physical, chemical, and biological parameterswere conducted monthly. Biological parameters measured were chlorophyll-a, and the abundance of phytoplanktonand zooplankton. Phytoplankton primary productivity, grazing mortality of phytoplankton by zooplankton, numberof catch of fish, and fish feeding habit were also measured. Run of the built model based on the data obtainedshowed that with the present catch-pressure (assuming that this was the start of catching operation), there wouldbe an overfishing starting in the 26 th or 27th month onward. Meanwhile, if the catch were half of the present, thenthere would be an overfishing in the 31 st or 32nd month, but would back to the sustainable production months afterthat. The model is still being developed by finding and adding more accurate data and parameters.

Abstract

The purpose of the research was to find out the countent of nutrition of Ara (Ficus racemosa L) wich covers : thelevel of protein, fat, water, ash, fibrous part of the fruit, calcium, phosphor, vitamine C and E as well as amino acidwhich are very useful for enrichment of fish diets. The sample of Ara fruit were collected directly in the field. Therewere two types of processing Ara fruits used in this research, namely (1) cutting the fruits into thin layer and madeit into; (2) separating the fuit from the seed, dried, and made it into flour. The level of protein, fat, fibrous part of thefruit, water, and ash was analyzed by using proximate analysis, while the level of calcium was analyzed by usingAAS methods. The level of phosphor was measured by using spectrophotometer, the level of vitamine C and E wasanalyzed by using HPLC and amino acid by gas chromatography (CG). The study found that the flesh flour of Arafruit contained protein (10,63%), fat (8,66%), water (7,08 %), ash (11,57%), fibrous part of the fruit (16,15%), calcium514,88 mg/100 g), phosphor (418,29 mg/100 g), vitamine C (475,38 mg/100 g), and amino acid (3,013 %) higher thanAra flour with protein (8,98 %), fat (6,57%), water (10,26%), ash (18,23%), fibrous part of the fruit (20,31%), calcium(0,039 mg/100g), phosphor (0,002 mg/100g), vitamin C 25,48 mg/100 g ), and amino acid (1,24%). It can be concludedthat the Ara flour can be used as on of the ingredients for fish diets especially as the source of vitamine C, calciumand phosphor to incrase reproduction ability and growth.

Abstract

Nd-CeO2 nanoparticles have been synthesized with a simple heating method. The microstructure and morphologyof material were characterized by a Scanning Electron Microscopy (SEM), and the conductivity of the material wasmeasured by an AC Impedance Spectroscopy. At room temperature we obtain a sample prepared at 9% at/at. Ndhave the highest conductivity of 1.47 x 10-4. Scm-1. This conductivity surpass the value reported by Anefleous for Nd-CeO2 (NDC) material prepared by different method. Sample has been 49 nm crystals size obtained from FWHM .Thisresult proves that a simple heating method is the promising for synthesizing electrolyte materials for SOFCapplication.

Abstract

Screening to genomic library of soybean cultivar Slamet in ë phage to isolate the gene encoding for peroxidase(per) was performed by two steps. cDNA of per gene from Arabidopsis thaliana labeled by alkalin phosphatasewas used as probe. The first screening was done by using 105 recombinant pfu (plaque forming unit). The secondscreening was carried out on 100 recombinant pfu isolated from the first screening. The result of second screeningshowed that all screened clones were supposed containing per gene. The ë phage resulted from second screeningwas infected into E. coli BM 25.8. Due to the Cre recombinase in E. coli BM 25.8, two lox P sites flanking the insertDNA of soybean created recombination. The recombination in loxP sites resulted the excision and formedrecombinant plasmid containing per gene of soybean. To multiply, the recombinant plasmid was introduced into E.coli DH5á. Analysis southern hibridization of recombinant plasmid cut by Bam HI showed that Bam HI 3.7 kbfragmen of S52121R clone, 5,3 kb of S2412R clone, 2,4 and 5,3 kb fragments of S2532R clone were supposed tocontain per genes.

Abstract

Macroalgae research in Anambas Islands that is the reef flats Jemaja, Pemutus and Matak Waters was carried onJune 2005. Survey method on the research using quadrat transect. Macroalgae is a marine flora which containsChlorophyceae, Phaeophyceae and Rhodophyceae. Biodiversity in the research location was 18 species. Thehigher diversity index (H) was 0.24 and equitability index (e) was 0,105. The higher density of species were 207individuals. Dominant species was 22,83% by Sargassum polycystum.
