Abstract

AbstractRandom Amplified Polymorphic DNA (RAPD) is one of molecular technique that can beused for DNA markers. RAPD gives better result, is more consistent, and is not influenced byenvironmental factors or growth phases than morphological characters. The research aimed foridentifiying specific DNA band and similarity level of tested rice lines and cultivars based onDNA band patterns of RAPD.DNA was extracted from single seed of rice by a modified CTAB DNA extractionmethod. The quantity and quality of extracted DNA was determined by agarose gelelectrophoresis and spectrophotometer. Result showed that the RAPD can differentiate alltested lines and cultivars. The RAPD revealed specific DNA band for Poso (OPA 9457), G12(OPA 91972), G13 (OPK 20227), G35 (OPA 9722), G39 (OPA 9341), Danau Tempe (OPK 10199 andOPA 244200) and Mentik Wangi (OPK 201487). Dendogram of genetic similarity showed that thelines closest to Mentik Wangi is G136, followed by G9, G10, G34, and G39. G12 hadsimilarity to Poso. G19 and G35 have similarity to Danau Tempe. Specific DNA band ofRAPD had a potential to be used as a tool for cultivar identification.Key words : Aromatic Upland Rice, RAPD, Cultivars Identification

Abstract

ABSTRAKPenelitian ini bertujuan untuk: (1) Mengetahui kelayakan pendirianindustri pengolahan salak skala kecil dalam aspek pasar dan pemasaran, teknisdan teknologi, manajemen operasional, hukum dan dampak lingkungan, danfinansial; (2) mengetahui industri pengolahan salak yang paling sesuaidikembangkan di Kabupaten Banjarnegara. Hasil penelitian menunjukkan,keempat industri pengolahan salak yang diteliti layak untuk dilaksanakan, namunindustri kripik salak lebih unggul dibandingkan industri pengolahan yang lain,terutama dalam aspek pasar dan pemasaran serta aspek finansial. Kripik salaksudah cukup dikenal dan disukai masyarakat sehingga dalam pemasaran lebihmudah. Dalam aspek finansial, industri kripik salak memiliki nilai NPV, IRR,ROI, dan PI yang paling tinggi, dengan PBP terpendek dan BEP terendah.Sehingga dapat disimpulkan, industri kripik salak merupakan industri pengolahanyang paling sesuai di Kabupaten Banjarnegara.Kata kunci: industri pengolahan salak, kripik salak, NPV, IRR, ROI, PI, PBP,BEP.

Abstract

SUMMARYNyamplung is a multy-function plant. Beside its wood is good forconstruction and furniture material, the almond is used for bio-diesel fuel´smaterial. The advantage in using Nyamplung as bio-diesel fuel´s material is theproductivity which may reach until 10 tons/hectare. Besides, Nyamplung has highendurance of environment, big amount of population, longer range of lifespan (1to 50 years), year-round productivity, especially during September to November.Nyamplung´s oil content may reach around 40% to 70%. Nyamplung´s oil is notused as food product (non edible). Thus, this plant will not disturb the foodstability program.This research was aimed to observe the extraction method, methanolconcentration, and the best Degumming method to produce Nyamplung´s biodiesel oil. So that Nyamplung may be considered as the alternative fuel to replacethe ordinary fuel which is continually decreased nowdays.This research was conducted at Agriculture Product ProcessingTechnology Laboratory and Agriculture Technic Laboratory of JenderalSoedirman University in Purwokerto, Quality control Laboratory of SMKN 1temanggung and PT Pertamina RU IV Cilacap, during May 2009 until October2009. The research was used Complete Randomized Block Design that consistedof three factors, namely: Extraction Method, Degumming Method and MethanolConcentration which was needed on Transeserification reaction. Observedvariables were Chemical composition of Nyamplung Biodiesel variable andbiodiesel´s performance to diesel engine as the Physical Variable.Based on the research, it was resulted the best treatment for Nyamplung´sseed extraction was Chemical method extraction. The greatest result ofdegumming method was reached by using 1% Sulfuric Acid (H3PO4) and 5%methanol concentration. In such a state, pH balance value was 6.58, viscocity oilas 5.645 cst, Flash Point was 157.295C, free fatty acid was 1.1%, acid numberwas 0.1, Iodine number was 82.775% and Alcyl Ester number was 93.545%.Based on visual observation, Nyamplung´s biodiesel oil has clear browncolor. The smell was similar like fresh oil. The stationere was 10 minute remindsstable. The soot produced was thin as it was resulted from perfect combustion.Key words: Nyamplung, bio diesel , extraction method, Degumming Method,Methanol Concentration, Transeserification reaction, biodiesel´s performance.

Abstract

KAJIAN KOMPARATIF AKTIVITAS ANTIOKSIDAN FORMULA PENGAWET ALAMI EKSTRAK KECOMBRANG (Nicolaia speciosa Horan) DAN POLA PEMISAHANKROMATOGRAFIS EKSTRAK BAGIAN-BAGIANTANAMAN KECOMBRANGOleh : PristiadiProgram Pascasarjana, Universitas Jenderal Soedirman, PurwokertoAbstractKecombrang (Nicolaia speciosa Horan) contains alkaloid compounds, saponin, tannin, phenolic, flavonoid,triterpenoid, steroid and glycoside which act as antimicrobial and antioxidant. Utilization kecombrang in the form of fresh or extract is easy to loss of volatile components, so that need to be formulated with the addition of fillers, stabilizers, NaCl and pH treatment. Formula that has antimicrobial activity investigated as total phenolic content and antioxidant activities. Tests performed on thin-layer chromatography of fruit extracts, rhizome extracts, stem extracts and flower extracts to get the chromatography pattern and Rf values. Experiment of eight formulas of kecombrang extracts have the highest measurement results obtaining on the formula F1 (15% fillers, 1% stabilizers,2.5% NaCl and pH 4) with total phenolic content of 323.25 mg/100 g and 58.49% antioxidant activity. The relationship between total phenolic content with antioxidant activity of formulas of kecombrang extracts obtained correlation value r = 0.92 this shows that 92% of the total phenolic effect on antioxidant activity. It shown the result of thin layer chromatography tests with GF254 stationary phase and ethyl acetate - formic acid - glacialacetic acid - water eluent (100:11:11:26) on stem and flower extracts (Rf 0.15; 0.54; 0.94), fruit extracts (Rf 0.09; 0.94) and rhizome extracts (Rf 0.15). The similarity of Rf values on kecombrang extracts shown similar compounds contained in it.Keywords : total phenolic, antioxidant activity

Abstract

ABSTRAKPeningkatan produksi tomat dapat dilakukan dengan berbagai cara, salahsatunya dengan teknologi pemupukan menggunakan EM4 dan inokulasi mikoriza.Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi EM4 terhadappertumbuhan dan hasil tomat, mengetahui pengaruh inokulasi mikoriza terhadappertumbuhan dan hasil tomat, mengetahui pertumbuhan dan hasil beberapa varietastomat dan mengetahui pengaruh interaksi konsentrasi EM4, varietas dan inokulasimikoriza terhadap pertumbuhan dan hasil tomat.Percobaan dilakukan dengan rancangan RAKL dengan 3 ulangan. Faktorpertama aplikasi mikoriza : tanpa mikoriza (M0), inokulasi mikoriza (M1); faktorkedua berupa varietas : Montera (V1), Lentana (V2), dan Permata (V3); faktor ketigaberupa konsentrasi EM4 : 0 ml EM4/L (E0), 2 ml EM4/L (E1), 5 ml EM4/L (E2) dan8 ml EM4/L (E3).Hasil penelitian menunjukkan bahwa varietas yang diuji mempunyaikeragaman tinggi tanaman, umur berbunga, umur panen, bobot buah per tanaman,serangan layu Fusarium dan penyakit virus Kuning. Varietas yang memberikan hasiltertinggi adalah Permata dengan produksi rata-rata 1,659 kg dan terendah adalahMontera yaitu 0,803 kg, sedangkan rata-rata produksi 1,338 kg/tanaman atau setara53,5 ton/ha. Konsentrasi EM4 tidak berpengaruh terhadap pertumbuhan dan hasiltomat. Inokulasi mikoriza tidak berpengaruh terhadap pertumbuhan dan hasil tomat.Interaksi antara konsentrasi EM4 dengan varietas dan inokulasi mikoriza, konsentrasiEM4 dan varietas hanya menunjukkan keragaman terhadap tinggi tanaman.
