Abstract

Enzim L-asparaginase (E.C 3.5.1.1) adalah protein tetramer yang termasuk dalam kelompok keluarga amidohydrolases homolog. ). L-asparaginase secara selektif menghidrolisis L-aspargin (L-Asn) menjadi amonia (NH3) dan asam aspartat. Penelitian ini bertujuan untuk mendapatkan spesies bakteri penghasil enzim L-asparaginase yang diisolasi dari mangrove Avicennia marina yang berasal dari Pantai Bajul Mati, Malang, Jawa Timur, Indonesia. Penelitian ini dilakukan beberapa tahapan yaitu pengambilan dan preparasi sampel, skrining L-asparaginase, dan identifikasi menggunakan uji mycrobact system. Hasil penelitian menunjukkan bahwa isolat dengan aktivitas enzim L-asparaginase terbesar adalah isolat DAM6 yang berasal dari daun Avicennia marina. Isolat DAM6 adalah bakteri merupakan bakteri gram negatif, berbentuk basil, bentuk koloni oval, tepi koloni tidak rata dan elevansi cembung. Berdasarkan analisis microbact,  isolat DAM6 adalah bakteri spesies Pseudomonas aeruginosa.                                               AbstractThe L-asparaginase enzyme (E.C 3.5.1.1) is a tetramer protein belonging to the family group of amidohydrolases. L-asparaginase selectively hydrolyzes L-aspargin (L-Asn) to ammonia (NH3) and aspartic acid. This study aims to obtain species of L-asparaginase enzyme producing bacteria isolated from Avicennia marina mangroves from Bajul Mati Beach, Malang, East Java, Indonesia. This research carried out several stages such as took and prepared samples, screened L-asparaginase, and identified using the mycrobact system test. The results showed that the isolates with the largest L-asparaginase enzyme activity were DAM6 and it was originated from the leaves of Avicennia marina. DAM6 isolate is a gram-negative with an oval shape, uneven edges, and convex elevation. Based on microbact analysis, DAM6 isolates are Pseudomonas aeruginosa.

Abstract

AbstractThis study was carried out to investigate the effect of Asian seabass (Lates calcarifer) culture in floating net cages at the Brackishwater Aquaculture Development Center, Situbondo. Asian seabass were fed with commercial fish pellet (KPA) and fresh fish (trash fish) for 21 days. Seabass fed with trash fish and commercial fish pellet displayed the growth response to the average weight and length. The fish grew from an initial weight of 230g, 238g, and 244g in 21 days and the average length of seabass changed from 23 cm, 25 cm and 28 cm respectively. The fish were fed with 3-7% body weight of fish and the growth was monitored in 21 days. The results showed that trash fish can increase the significant growth response in Asian seabass. AbstrakStudi ini dilaksanakan untuk mengetahui pengaruh budidaya kakap putih Asia (Lates calcarifer) di keramba jarring apung di pusat pengembangan budidaya air payau, Situbondo. Kakap putih Asia diberi pelet ikan komersial (KPA) dengan ikan segar (limbah ikan) selama 21 hari. Kakap putih makan dengan limbah ikan dan pelet ikan komersial menunjukkan respon pertumbuhan rata-rata dari berat dan panjang. Pertumbuhan ikan dari berat awal 230g, 238g dan 244g pada hari ke- 21 dan terjadi perubahan panjang rata-rata kakap putih masing-masing menjadi 23 cm, 25 cm dan 28 cm. Ikan diberi makan dengan 3-7% dari berat badan ikan dan pertumbuhan diamati dalam 21 hari. Hasil penelitian menunjukkan bahwa limbah ikan dapat meningkatkan respon pertumbuhan yang signifikan di kakap putih Asia.

Abstract

AbstrakEmbung adalah bekas galian tanah liat dari penambangan di PT. Semen Indonesia yang dimanfaatkan oleh warga sekitar sebagai sumber pengairan pada kegiatan dan tempat budidaya ikan dengan sistem Keramba Jaring Apung (KJA). Tujuan Penelitian ini untuk melakukan kajian teknis faktor abiotik embung bekas galian tanah liat PT. Semen Indonesia untuk budidaya perikanan dengan teknologi KJA. Metode penelitian menggunakan metode deskriptif dengan pengumpulan data survei yang dianalisa menggunakan WQI min dengan data diperoleh berupa kualitas air (suhu, pH, DO dan kecerahan). Hasil penelitian suhu pagi hari pada ke empat titik pengambilan sampel berkisar 32,3 – 32,7°C dan siang hari berkisar 32,7 - 33°C(kurang layak), pH pagi hari 8 – 8,1 siang hari 7,9 – 8 (layak), dissolved oxygen (DO) pagi hari 5,6 – 6,2 ppm, siang hari 6,1 – 6,7 ppm (layak) dan kecerahan  pagi hari 69,6 – 78.7 cm, siang hari 61,6 – 77 cm (layak) dengan mengacu pada standar kualitas air PP No. 82 Tahun 2001 dan BBPBAT (2010). Berdasarkan hasil analisa  faktor abiotik tersebut, maka direkomendasikan jenis ikan nila dapat dibudidayakan di embung bekas galian semen dengan teknologi KJA. AbstractEmbung is a former excavation of clay from mining at PT. Semen Indonesia is used by local residents as a source of irrigation in activities and fish farming sites using the floating net cage system (KJA). The purpose of this study was to conduct a technical study of abiotic factors used for clay excavation of PT. Semen Indonesia for aquaculture with KJA technology. The research method used the descriptive method with survey data collection analyzed using WQI min with data obtained in the form of water quality (temperature, pH, DO and brightness). The results of the morning temperature study at the four sampling points ranged from 32.3 - 32.7C and daytime ranged from 32.7 - 33 ° C (less feasible), morning pH 8 - 8.1 during the day 7.9 - 8 (feasible), dissolved oxygen (DO) in the morning 5.6 - 6.2 ppm, daytime 6.1 - 6.7 ppm (feasible) and morning brightness 69.6 - 78.7 cm, daytime 61, 6 - 77 cm (feasible) with reference to water quality standards PP No. 82 of 2001 and BBPBAT (2010). Based on the results of the analysis of the abiotic factor, it is recommended that the type of tilapia can be cultivated in used cement excavated ponds with KJA technology.

Abstract

AbstrakPelatihan industri telah dilakukan untuk mempelajari dan memahami teknik budidaya dari Kerapu Cantang (Epiphenelus sp.). Teknik budidaya kerapu Cantang merupakan salah satu faktor yang akan mempengaruhi tingkat pertumbuhannya. Pengambilan sampel ikan dilakukan selama 21 hari di Pusat Pengembangan Budidaya Perikanan Air Payau, Situbondo, Jawa Timur. Hasil penelitian ini menunjukkan bahwa pertumbuhan kerapu Cantang meningkat signifikan sebesar 2,29 gram/hari. Pengelolaan pakan dan kualitas air yang baik dianggap sebagai faktor utama yang berpengaruh pada pertumbuhan Kerapu Cantang selama proses budidaya. AbstractAn industrial training has been conducted to learn and understand the culture techniques of Cantang Grouper (Epiphenelus sp.). The culture technique of Cantang Grouper is one of the factors that affect its growth. The fish sampling was carried out for 21 days at Brackish Water Aquaculture Development Center, Situbondo, East Java. The result of this study demonstrated that growth of Cantang Grouper was increased significantly of 2.29 gram/day. Good management of feed and water quality are considered as the main factors that gave effect on the growth of Cantang Grouper during the cultivation process.

Abstract

AbstrakKerapu cantik merupakan kerapu hibrid hasil persilangan antara betina kerapu macan dan jantan kerapu batik. Salah satu permasalahan dalam pembenihan kerapu cantik secara umum, yaitu pertumbuhan stadia awal benih kerapu cantik yang lambat. Permasalahan tersebut dapat diatasi dengan menambahkan taurin pada pakan buatan untuk meningkatkan pertumbuhan benih ikan kerapu cantik.Penelitian ini bertujuan untuk mengetahui pengaruh penambahan taurin pada pakan buatan terhadap pertumbuhan dan sintasan benih ikan kerapu cantik. Penelitian ini menggunakan metode RAL dengan tiga perlakuan, yaitu pemberian pakan buatan dengan kadar taurin 0% (P1), 0,5% (P2) dan 1% (P3) yang diberikan pada benih ikan kerapu cantik dan masing-masing perlakuan diulang enam kali. Hasil pemeliharaan selama 45 hari menunjukkan bahwa penambahan taurin sebesar 1% pada pakan buatan memberikan pertumbuhan tertinggi dibandingkan perlakuan lainnya dan berbeda nyata (p<0,05). Penambahan taurin 1% pada pakan buatan tidak memberikan hasil sintasan yang berbeda dengan kontrol (taurin 0%). AbstractCantik grouper is hybrid grouper, it is the results of a crossover between females tiger grouper and males batik grouper. One of the problems in breeding cantik grouper in common is the growth of early stadia juvenile cantik grouper is slow. This problem could be solved by added taurine in the feed to increase the growth of juvenile cantik grouper. The purpose of this research is to find the influence of the addition of taurine in the feed on the growth and survival rate of juvenile cantik grouper. This research used completely randomized design (CRD) method with three treatments, which is giving taurine to feed with 0 % (P1), 0.5 % (P2) and 1 % (P3) concentration given to the juvenile cantik groupers and the treatment has been repeated six times. The results of breeding for 45 days showed that the addition of 1 % taurine in feed give the highest growth than other treatment and differs markedly ( p<0.05 ). The addition of 1% taurine gives no different result with the control variable (0% taurine).

Abstract

AbstrakPerairan Wonorejo merupakan salah satu daerah penghasil kepiting bakau di Surabaya. Namun perairan ini berpotensi mengalami pencemaran logam berat akibat aktivitas yang ada di daratan. Tembaga (Cu) merupakan salah satu jenis logam berat yang mencemari perairan Wonorejo dan jika dalam jumlah besar menyebabkan kerusakan hati organisme. Penelitian ini bertujuan untuk mengetahui konsentrasi logam berat tembaga pada kepiting bakau dan mengetahui keamanan pangan hasil tangkapan kepiting bakau di Sungai Wonorejo. Penelitian ini dilaksanakan pada bulan April 2018 dengan pengambilan sampel di tiga stasiun dengan waktu setiap minggu selama tiga kali. Analisis logam berat tembaga (Cu) pada kepiting bakau (Scylla sp.), air dan sedimen dilakukan di Laboratorium Gizi Fakultas Kesehatan Masyarakat Universitas Airlangga. Metode penelitian menggunakan metode observasi yang dilakukan di perairan dan tambak di Wonorejo Surabaya. Sampel daging kepiting bakau, air dan sedimen diuji menggunakan alat Atomic Absoption Spectrophotometry (AAS). Hasil kandungan kadar logam berat tembaga (Cu) pada kepiting bakau di Sungai Wonorejo pada stasiun 1 dan 2 yaitu 0,008 mg/kg dan pada stasiun 3 yaitu 0,002 mg/kg dimana nilai kadar ini masih berada di bawah ambang batas baku mutu logam berat Cu pada kepiting yaitu 20 mg/kg.AbstractWonorejo river as one area in Surabaya that has produce mud crab and potentially has heavy metal pollution caused by human activities. Copper (Cu) is on the heavy metals that pollute in the waters Wonorejo. Copper in large quantities can damage the liver. This research was conducted in April 2018 samples taken at three stations. Analysis of heavy metals copper (Cu) in samples of water, sediment, and meat of mud crab (Scylla sp.) is done in Laboratory Nutrition Department of Public Health, Universitas Airlangga. Other research methods of observation methods conducted in coastal areas and pond.Wonorejo Surabaya. Heavy metal testing of copper in samples of water, sediment, and meat of mud crab use Absorption Atomic Spectrophotometry (AAS). Results Cu content of heavy metal levels in the meat of mud crab in the Wonorejo river for site 1 and 2 was 0,008 mg/kg and for site 3 was 0,002 mg/kg. This shows the copper content in the meat of mud crab is under the allowable quality standards for copper of 20 mg/kg.

Abstract

Abstract Histology is science that learns about cell,organ, and body tissues in a microscopic condition. Whereas science that learns about morbidity or patology of a tissue that’s called as histopatology. Both of normal tissue’s structure and abnormal tissue’s stucturecan be learned by microscopic in a tissue preparation. This preparation made through processing of tissue until the preparation coloured. Then histology’s structure can be watched clearly so that make it easy to read. Field Work Practice purpose (PKL) this is to know the basic histoloy technique in fish. This Field Work Practice was held in Balai Uji Standar Karantina Ikan Pengendalian Mutu dan Keamanan Hasil Perikanan (BUSKIPM) Jl. Harapan I No. 01A Cilangkap, East Jakarta on 14th January 2013 until 14th February 2013. The method of this Field Work Practice is descriptive method by collecting data through primary data and secondary data. The histology technique in fish are tissue fixation, trimming (tissue selection), tissue dehydration, the tissue block making, tissue cutting, tissue colouring, and tissue observation. From the result got from the gourami’s tissue, there are edema and infestation of metacercaria parasites and myxosporidia in gill normal intestine tissue, normal liver tissue, and normal spleen tissue

Abstract

Abstract The increase in value-added of fishery products is one of which can be done by processing. In addition to maintaining the value of the protein in the fish raw material, it would be easier if the product is stored and increase the sale value of the fish itself. The ability to perform in the fish processing in Mandangin island is still very limited. Community service activities that have been carried out by a team of the Faculty of Fisheries and Marine Universitas Airlangga was introduce smoked fish product processing with liquid smoke,because this method easier and faster so that the products can be marketed. Liquid smoke was developed because the location is very minimal in the supply of raw materials such as firewood and coconut, so the use of liquid smoke is very appropriate. The resulting refined products are expected to be a source of protein Mandangin Island community itself and is able to become an alternative source of income for the community to increase the sale value of fishery products. Mentoring process being conducted by a team from the Faculty of Fisheries and Marine Universitas Airlangga so that the products can be marketed outside Mandangin Island or even to other areas in East Java.
