Abstract

The aims of this study were to 1) determine the influence of curing time and extraction time, on the rendement and characteristics of essential oil of pandan wangi leaves 2) get curing time and extraction time procses to produce the highest yield and the best characteristics essential oil of pandan wangi leaves and 3) determine the type and composition of compounds in the essential oil of pandan wangi leaves. This research used randomized block design with factorial pattern. The first factor (curing time) consists of 3 level namely without curing, the 2 and 4 days process time, the second factor (extraction time) consists of 3 level using the 2, 3, and 4 hours process time. Each treatment grouped into 3 groups based on the time implementation. Objective variables measured were yield, and composition profiles of compounds in essential oil of pandan wangi leaves. Subjective variabel measured were the preference of aroma and aroma strength of the essential oil of pandan wangi leaves. The best treatment was determined with effectiveness index. The results showed that the curing time and extraction times had significant effect on the rendement and characteristics of essential oil of pandan wangi leaves. The extraction time of 3 hours and material conditions of curing 4 days was right treatment to produce essential oil of pandan wangi leaves with the highest rendement and the best characteristics, namely rendement 1.983 %, the aroma preference of 5.2 (between like and really like) and aroma strength 6.4. essential oil of pandan wangi contain 36 kinds of compounds with 16 compounds were not identified. Constituent compounds consists of classes alkane (46,66%), alkene (31,22%), aldehid (11,13%), and not identified (10,89%).

Abstract

This research aimed to isolate and identify potential yeast of bioethanol-producer from arak industri in Karangasem Bali. The isolated sample was taken from 3 different points from 2 villages in Karangasem Bali. Isolation was carried out using PYG media then purified to obtain pure isolates. The pure isolates were screened with several stages, namely a qualitative test of gas production, growth selection with the addition of antibiotics, and quantitative tests with an alcohol oxidation reaction. Determind by UV visible spectroscopy, 9 potential isolates was obtained to continue to the identification stage. At this stage fermentation was carried out in PYG media for 10 days using a starter from a potential isolate with a media glucose level of 20% of the media volume. Fementation results were then distilled. Of the obtained 9 potential isolates the best isolate. IS 258 isolates are determind the best isolates with 86.85 mL ethanol. The total ethanol produced by IS 258 is higher than with 60.73 mL alcohol control experiment (Alcotec). Isolate IS 258 was isolated from bayur skin samples taken from the sap fermentation process. IS 258 then identified macroscopically and microscopically. Based on the results of macroscopic and microscopic identification, IS 258 has many similarities with previous studies on yeast isolation, isolate IS 258 is presumably the yeast genus Saccharomyces sp. Further research is needed to optimize ethanol production of IS 258 and identify species from isolates IS 258. Abstract

This study aimed to isolate cellulolytic bacteria that have the potential to degrade cellulose taken compost samples from Temesi and Bondowoso, this study was also conducted to determine the ability of bacteria to degrade cellulose based on Congo red test and filter paper degradation test. All isolates were tested for cellulolytic activity using soluble Carboxymethyl Cellulose (CMC). Characterization was carried out by growing selected pure isolates on CMC media and then dripping 0.1% congo red to test the cellulolytic potential (cellulolytic potential was characterized by the emergence of clear zones around the colony). The results of isolation of bacteria obtained 38 isolates, namely 26 Bondowoso sample isolates and 12 Temesi isolates that were able to grow and utilize cellulose as a carbon source. But only fourteen isolates produced clear zones in the Congo red test with diameters ranging from 1.66 cm to 6.76 cm. Six isolates that have the largest diameters clear zone, were tested for degradation of filter paper (Whatman no. 1). Isolates bacteria of B2S8 obtained from Bondowoso compost samples has the highest ability to degrade cellulose on Whatman paper no. 1 as much as 51.30%. Abstract

The aims of  this study were  1) to find out the distribution strawberry that produce in Baturiti to Denpasar, 2) to find out post-harvest handling in each distribution line 3) to determine the marketing and profit margins of each distribution from Baturiti to Denpasar. This study was using survey method with 20 responden of strawberry farmer. The results of this study showed that there were 3 distribution line of strawberry from Baturiti to Denpasar and all lines had experienced in sorting and grading on the collectors and traders. The highest marketing margin was Rp 24,000 / kg margin was Rp 11,300 /kg which were found in line 2 (Farmer-Collectors-Retailer-Consumer). Meanwhile the lowest marketing margin was Rp 17,000 /kg and the lowest profits margin was Rp 3,814 /kg which were found in line 3 (Collectors-Farmer-Consumer Wholesalers Catering).

Abstract

This study aims to determine the dibenzothiophene (DBT) degradation level of four potential sulfur aromatic degrading bacteria. Biodesulfurization activity test was done by using twophase system. First was water phase which used mineral salt sulfur free (MSSF) medium and the second phase was oil phase which use 200 ppm DBT in n-tetradecane. Isolates were incubated for 96 hours at 37oC and the DBT residu oil phase was analyzed using Gas Chromatography. The experiment was repeated twice. This isolate wa able to degrade DBT in level of 80,82% after 96 hours incubation. The growth of the bacteria in water phase reached OD6601,169. Abstract

The aim of this study were to determine the number of asparagus distribution lines and types of distribution, find out the impact of postharvest handling and to determine the impact of improvements postharvest on asparagus damage from farmers in Petang district to retailers in Denpasar. Postharvest improvements in this study by replacing the packaging using styrofoam box and placing the asparagus vertically. There are three lines in the distribution of asparagus. The first line (Farmers ? Cooperative ? Suppliers ? Retailer ? Consumer), line II (Farmer ? Middleman ? Suppliers ? Retailer ? Consumer) and line III (Farmer ? Middleman ? Suppliers ? Consumer). Postharvest handling includes harvesting, cleaning, cutting, sorting, grading, binding, storage, packaging, distribution and displaying. The impact of handling postharvest loss in the sorting stage at the farmers level is 2.5% (non significant), cooperative level is 1.8% (non significant), middleman level is 4% (insignificant), supplier level is 5.4% (significant) and retailers level is 6.69% (significant). Postharvest improvement by changing the packaging could reduce the level defect of asparagus to 7.81%. Keywords: asparagus, styrofoam packaging, CSAM, postharvest handling, improvements

Abstract

This study aims to know the types of agro-tourism can be an attraction in and formulate appropriate strategies for development. In this study there are several steps that are used starting agroindustry identification, collection and sampling, identification places agro-industry, External & Internal Environment Analysis, Strategy formulation using SWOT and final selection of alternative strategies using QSPM The results showed that the analysis of internal factors and external objects have been surveyed from the two sampled agro-tourism obtained IFE total weighted scores for 2,769 and 2,874 by placing second EFE agro-tourism that the cells in the matrix V IE. These positions describe both agro tourism can implement defense strategies. Priority selection strategy by using QSPM, recommending business development strategy based on six strategy alternatives derived from the SWOT matrix and matrix IE put alternative strategy Working with investors or with relevant government agencies to maximize the quality and quantity of agro products and agro-industry development opportunities in Denpasar to Bedugul tourist track on the top spot with a total score of weight 5,719 Keywords: agro-tourism, agro-industry, strategy, tourist atractions. Abstract

Coffee grounds residue are the primary waste obtained from brewed coffee. Furthermore, some reports showed that coffee grounds residues still containes of bioactive compounds like caffeine and total phenolic compounds. This study aims 1) to know the effect of ethanol solvent concentration and maceration temperature on extract characteristics of coffee grounds robusta extract, and 2) to determine the concentration of ethanol solvent and best maceration temperature to of coffee grounds robusta extract. This experiment uses Factorial Random Design with two factors. The first factor concentration of ethanol solvent consists of 3 levels: 70%, 80%, 90%. The second factor is the maceration temperature which consists of 3 levels: 45oC, 60oC, 75oC. From the above two factors obtained 9 treatment combinations, each grouped into 2 groups as 18 units of experiments. The result showed that etanol solvent concentration, maceration temperature and interaction between trearments had high significantly effect on yield extract and caffeine content. The ethanol solvent concentration treatment had a significant effect, while the treatment of the maceration temperature and the interaction between trearments had a significantly effect on the total phenol. The treatment of ethanol solvent concentration 90% and maceration temperature 60oC was the best characteristic with robusta coffee extract yield 7.87±0.05% yield, 0.70±0.01% caffeine content and total phenol 11,052.83±1,124.30 mg GAE/100g.
