Abstract

MC4R gene is known as an important candidate gene for the growth trait. The purpose of this research was to identify the MC4R gene in Kebumen Ongole grade cattle and examine its association with growth traits. Data of birth weight (BW), weaning weight (WW), birth body length (BBL), birth chest circumference (BCC), birth shoulder height (BSH), weaning body length (WBL), weaning chest circumference(WCC), weaning shoulder height (WSH) and average daily gain (ADG) were collected and used for analysis of MC4R gene. Sixty blood samples were collected for DNA isolation and PCR amplification. The single nucleotide polymorphisms (SNP) g.1133 C>G was used for genotyping by using PCR-RFLP methods. The frequenciy of G allele (0.59) was greater than C allele (0.41). The highest genotype frequencies have been detected in CG heterozygote animals (0.52) followed by GG (0.33) and CC (0.15) in homozygote animals. The results of Pearson ‘s Chi-square test indicated that the population was not deviate (P>0.05) from the Hardy-Weinberg equilibrium (HWE). The SNP g. 1133 C>G of MC4R gene indicated affecting high birth body length with GG genotype (P<0.05). In conclusion, the SNP g. 1133 C>G may can be a marker for birth body length of calf selection.

Abstract

Melanocortin 4 receptor gene is a potential marker gene for growth performance in ruminant and non-ruminant animals. The objective of this study was to perform allele and genotype frequencies, and association of melanocortin 4 receptor with growth traits in Bligon goat. Seventy seven blood samples were collected and DNA isolated successfully. One pair primers were used for PCR of MC4R gene (F: 5’-TCGGGCGTCTTGTTCATCAT-3’ and R: 5’-CAAGACTGGGCACTGCTTCA-3’). Two SNPs (g.998A/G and g.1079C/T) were identified in exon of Bligon MC4R by DNA sequencing and PCR-RFLP. The SNP g.998A/G identified amino acid change from isoleucine to methionine (I-M), while the SNP g.1079C/T was synonymous polymorphism. Based on relationship analysis between Genotype of MC4R gene (SNP g.998A/G) and growth traits indicated that GG genotype was lowest in weaning weight (WW; AA=13.0±1.7; AG=11.2±3.6; GG=9.2±3.9), weaning body length (WBL; AA=46.8±2.4; AG=46.3±6.0; GG=43.8±5.3) and weaning chest circumference (WCC; AA=50.9±4.0; AG=50.7±4.6; GG=47.1±7.3) compare to AA and AG genotype (p<0.05). Individual with CC and CT genotypes at SNP g.1079C/T had higher (P<0.05) in WW (CC=12.6±2.1; CT=12.5±2.5) and average daily gain (ADG; CC=81.2±21.8; CT=79.3±26.2) than TT genotype (WW=10.6±3.1; ADG=58.3±30.5). This results suggested that SNP g.998A/G and g.1079C/T of MC4R gene could be used as the genetic marker of growth traits for selection in Bligon goat.

Abstract

This study aimed to explore the socio-economic benefits of Jabres cattle as potential local livestock in two agro ecological zones, lowlands and middle zone in Bantarkawung Sub-District, Brebes, Central Java. The data were collected by Participatory Rural Appraisal (PRA) method, which is an approach to families or individuals to analyze the reality of their living conditions. This study involved 60 farmers which selected by purposive sampling based on three considerations. The analytical model used to test differences is the analysis of independent T-test. The result showed that the farmers in the middle zone are younger and most of their cattle keeping motivation are for livelihood. Meanwhile, the farmers in the low lands are older and most of their cattle keeping motivation is for saving only (2.90±0.31). This condition caused the socio-economic benefit difference between those locations where the value of net production (185±27 kg/y), added value (IDR 4.900.000±1.204.000), and total benefits (IDR 9.289.000±1.542.000) were higher in the medium land than in the lowlands. Based on these reasons, it can be concluded that middle zone farmers have more possibility to sustain their farm than those of lowlands farmers.

Abstract

The objective of study was to estimate breeding value of sires of Kebumen Ongole grade cattle based on offsprings weaning weight. This research was carried out from August to December 2015 in Urut Sewu area, Kebumen. The calculation of genetic parameter estimation was heritability of weaning weight. The value of heritability was used to calculate the breeding value of sires based on offsprings weaning weight. Heritability was calculated using paternal half-sib correlation with the direct pattern variance analysis. Breeding Value (BV) of sire was calculated by absolute estimated breeding value. The materials used in this study included were data record during the last three years (2013-2015) consisted of 41 sires, 51 dams and 244 calves. Weaning weight data had been corrected based on age of dams, sex and weaning age of 120 days. The results showed that the heritability value of weaning weight was included in high category (0.36 ± 0.21). The highest BV was KJ.J608 (49.76) with a ratio of 231.45%. The results of this study were expected to be the basis recommendation for selecting sires in Kebumen.

Abstract

Melanocortin-4 receptor (MC4R) gene has an important role in the regulation of feed intake and energy balance control. The objective of this study was to identify the single nucleotide polymorphisms (SNPs) of MC4R gene and their association with growth traits in Madura cattle. A total of 198 calves were used in this study.Forward primer: 5’-GTCGGGCGTCTTGTTCATC-3’and reverse primer: 5’-GCTTGTGTTTAGCATCGCGT-3’ were used to amplify approximately 493 bp of MC4R gene. The results showed that two SNPs, g.1133C>G and g.1108C>T were identified by direct sequencing. The PCR-RFLP method was performed to genotype all individuals studied based on SNP g.1133C>G, and its SNP was significantly associated with shoulder height (SH) at yearling age (P<0.05). Animals with GG genotype had a higher SH (110.35±6.40cm) than those with CC (102.00±8.00 cm) and CG genotype (105.96±6.23 cm). The SNP g.1133 C>G changed amino acid from valine to leucine. In conclusion, the SNP g.1133C>G of the MC4R gene may be used as a marker-assisted selection for SH trait in Madura cattle.

Abstract

The aim of this study was to identify the polymorphism of leptin gene and its association with growth traits in Kebumen Ongole Grade cattle. One hundred blood samples were collected for molecular analysis. Polymorphism of the leptin gene was analyzed using Polymerase Chain Reaction - Restriction Fragment Length Polymorphism (PCR-RFLP) with HpyCH4V restriction enzyme and DNA sequencing. Association analysis of the leptin gene with growth traits was analyzed by T-test. The results showed that SNP g. 1180 C>T was found in the population.The SNP changed amino acid from arginine to cysteine. The SNP was significantly associated with a high chest circumference at weaning age in animal having CC genotype (P<0.05). There were two identified alleles, namely C and T, with frequencies were 0.885 and 0.115,respectively. The genotype frequencies of CC, CT and TT were 0.78, 0.21 and 0.01, respectively. Allelic and genotypic distribution in the studied population were in Hardy-Weinbergequilibrium. Animals with CC genotype had a higher circumference at weaning age (WCC) than those with CT genotype. In conclusion, SNP g. 1180 C> T in the leptin gene is potential as genetic marker for growth traits in Kebumen Ongole Grade cattle.

Abstract

This research is aimed to evaluate the nonlinear mathematical model in predicting the age and weight of Kebumen Ongole Grade (PO) cattle at puberty. The data used in this research was weight recordings of (768) PO cattle, ranging from calf (< 1 month) to adult (60 months), collected by Kebumen PO cattle Breeder Association (ASPOKEB) from 2013 to 2015. These cattle were reared by a group of farmers located in six sub-districts: Mirit, Lembu Purwo, Ambal, Puring, Petanahan, and Klirong in Kebumen district, Jawa Tengah Province. Brody, Bertalanfy, Logistic and Gompertz mathematical models were used to analyze the data. The analysis showed that the R2of the four models classify high in the category (R2 >0.70). Mature weight (A) of PO cattle predicted by Logistic (349 kg) and Gompertz (358 kg) were most appropriate. Out of the four models, the Logistic model was the most accurate in predicting the weight at puberty (174 kg) of PO cattle, but all observed cattle are still in the follicular phase.

Abstract

The melanocortin-4 receptor (MC4R) gene plays an important role in sympathetic nerve activity, as well as adrenal and thyroid function and mediates the effects of leptin on energy homeostasis. This study aimed to investigate the effect of the 642 bpMC4R gene on feed intake, nutrient digestibility, average daily gain (ADG),and feed conversion ratio (FCR)at post-weaned in Bligon goat. Forty-six Bligon were used for the blood sample collection, and genomic DNA was extracted using the Geneaid isolation kit. Feed intake data were collected on 46 Bligon goats kept on 2 different animal houses, including the laboratory’s farm of Faculty of Animal Science Universitas Gadjah Mada (FAS UGM) with n=23 and Banyusoco farm (n=23). Variables observed were feed intake, nutrient digestibility,ADG,and FCR. All samples were genotyped using the PCR-RFLP method with the KpnIenzyme. One SNP was located in the exonic region (g.1079C/T). The t-test analysis revealed a significant association between SNP g.1079C/T and the digestibility of dry matter (DM), organic matter (OM),and total digestible nutrient (TDN) of Bligon goats at 7 months of age kept on FAS UGM, as well as the digestibility of DM, crude protein (CP), nitrogen-free extract (NFE),and TDN of Bligon goats at 7 months of age kept on Banyusoco. In conclusion, it is possible to use SNP g.1079C/T for animal genotyping and asaMAS for nutrient digestibility in Bligon goats

Abstract

The objective of this study was to assess the association of KiSS1 gene polymorphism with reproductive traits in Kaligesing goat. Genotypes of 48 ewes aged three years old were determined using PCR-RFLP and DNA sequencing. Sixteen out of 48 samples were monitored for the estrus cycle and determined their 17β-estradiol and progesterone levels using ELISA method. The results showed that KISS1 gene in the studied population was polymorphic with one single nucleotide polymorphism (SNP T125A). Then SNP was used to analyze genotype all individuals by PCR-RFLP method using MboII enzyme. Three genotypes (TT, TA and AA) were identified. Genotype frequencies of TA were 59.57%, AA were 40.43% while TT was not inculed in calculation for genetic diversity and assosication analysis since the number of genotype only one goat. The allele frequencies of T and A were 29.79% and 70.21%, respectively. The genotype distribution for the SNP was deviated from Hardy-Weinberg equilibrium (χ2=8.10; P<0.025). Hormonal analysis showed that the levels of 17β-estradiol in the follicular and luteal phase were 50.12±61.26 and 42.35±42.43 pg/ml, respectively, while the levels of progesterone hormone in the follicular and luteal phase were 7.87±10.59 and 12.39±15.93 ng/ml. No significant associations of the polymorphism were observed for any hormonal levels. However, it was first report about polymorfism in KiSS1 gene of local Indonesian goat especially Kaligesing goat.

Abstract

This study aimed to estimate natural increase (NI), net replacement rate (NRR), output, and population dynamics of Aceh cattle at Livestock Breeding and Forage Center in Indrapuri, Aceh province. Data of population structure, mortality, birth rate, the number of animals entering the herd, and the number of animals released from the herd during 2019were used for determining NIandNRR, and it isusedfor estimation of output. Population structure data (2015-2019) were used to estimate the population dynamics. The results showed that NI was 19.08% (medium category). The availability of replacement stock exceeded the need for replacement (140.08% for male and 73.33% for female); the NRR was 240.08% (male) and 173.33% (female). The output of Aceh cattle was 3.92% (culled male), 5.58% (culled female); the remaining replacement stocks were 5.49% (male) and 4.09% (female) of the total population. The population of Aceh cattle from 2020 to 2024 was expected to increase by 6.02% (65 heads) on average. In conclusion, that BPTU-HPT Indrapuri can be categorized as a beef cattle producing region, but still need efforts to increase natural increase by increasing birth rate minimum 32.77% and reducing mortality maximum 3.0%, in combination with better management systems.
