Abstract

The purpose of the training is to socialize and provide comprehension to farmers about technologyinnovation of botanical pesticide from tobacco waste. Farmer- behavior change is expected to lead tofarmers’ desire to apply technological innovation of botanical pesticide. This research useddescriptive method. Determination of respondent is conducted by a census of all the farmers whoattended training on botanical pesticide technology (20 respondents). Indicators of farmer- behaviorchanges including cognitive, affective, and psychomotor about the benefits, how to make and apply thebotanical pesticide. The indicators are measured using a Likert scale. Data were analyzed usingdescriptive statistics. The results showed that the training of botanical pesticide technology has a goodinfluence on the behavior of farmers.

Abstract

Isoflavon merupakan senyawa fitokimia yang berperan dalam mekanisme pertahanan tanaman dan memiliki manfaat untuk kesehatan. Beberapa gen telah diketahui berperan dalam biosintesis isoflavon, diantaranya, yaitu: gen pal1, chs7 dan chi1b1. Namun demikian, pola ekspresi gen tersebut belum diketahui dengan baik pada varietas kedelai hitam (Glycine max (L.) Merr.). Studi pola ekspresi gen ini diharapkan dapat dijadikan sebagai studi pendahuluan metabolisme isoflavon. Penelitian ini bertujuan untuk menentukan pola ekspresi gen yang berperan penting pada produksi isoflavon di varietas UP 106, UP 115, UP 122, dan UP 157 pada kedelai hitam. Penelitian ini terdiri dari 5 tahap; pengambilan sampel daun pada tahap V3, R2, R5, R6, dan R7; isolasi RNA total; sintesis cDNA; konfirmasi dengan sekuensing; dan PCR kuantitatif. Pada penelitian ini, gen yang diisolasi adalah pal1, chs7, dan chi1b1. Hasil penelitian pada keempat varietas menunjukkan pada daun kedelai hitam, ekspresi gen pal1 bervariasi selama tahap perkembangan, sementara ekspresi gen chs7 menunjukan kecenderungan peningkatan ekspresi gen selama tahap pertumbuhan dan gen chi1b1menunjukkan peningkatan pada tahap awal reproduktif namun menurun di tahap akhir. Pola ekspresi masing-masing gen ini kemungkinan disebabkan oleh faktor tranksripsi yang khas berkaitan dengan masing-masing tahap pertumbuhan kedelai hitam.Kata kunci: isoflavon, kedelai hitam , pal1, chs7, chi1b1

Abstract

Cat’s whiskers plant (Orthosiphon aristatus B. Miq) is a medicinal plant containing various metabolites that act as antioxidants and anti-inflammatory, including rosmarinic acid. The domestic market demand for dry leaves was 20-40 tons and abroad it was 141-180 tons in the 1990-2005 period, while production from 2013 to 2017 decreased from 116 tons to 56 tons. Therefore, it is necessary to increase the cat's whiskers biomass production, one of which can be done through the engineering of growing media using organic fertilizers, including goat manure and microbes, e.g., Plant Growth Promoting Microorganisms (PGPM). This study aims to determine the optimal combination of PGPM – Goat Manure for biomass and the rosmarinic acid content in O. aristatus. The study was conducted using a 2-factor randomized block design (RBD) with 12 treatment combinations and 3 repetitions on cultivated land in the Tanjungsari area and analysis of the results was carried out in the SITH laboratory from October 2020 to March 2021. The rosmarinic acid content in the leaves ranged from 144.43 – 206.31 mg/g or 14.44 – 20.63%, with the highest content found in leaves treated with 3 ml/L PGPM and 86.2 g/plant goat manure fertilizer (P2O2).
