Abstract

This study determined the effect of cadmium (Cd) on the structure of the gills of catfish (Clarias batrachus). Animals were exposed to 0 (control), 1, 2, and 4 ppm of Cd during 14 days. The results showed that Cd affected the structure of the gills of catfish, such as edema, hyperplasia, and fusion gill lamela. The gills of control animals showed the lowest damage (13.9%), where as the level of gill damage of fish exposed to 1, 2, and 4 ppm were 45.7; 71.6; and 82.1% respectively.Keywords: cadmium, catfish, edema, hyperplasia, gills.

Abstract

The purpose of this research was to determine the effect of polysaccharide krestin (PSK) on the testicular protein profiles and testosterone levels of Mus musculus with variety of dosages. This research used a completely randomized design. It were devide into four treatment group i.e. control group, PSK treatment at a dose of  15, 30, 60 mg/kgBW. Each group had six replications. Testicular proteins were isolated by flushing technique and analized by SDS-PAGE. Testosterone levels were analized using ELISA technique at wavelength 450 nm. Protein bands analysis showed that there were no diversification between four treatments. Molecular weight of protein bands were 87, 63, 57, 35, and 30 kDa. The results of research showed that the testosterone levels at dosage 60 mg/kgBW had significanly different with control, PSK treatment of 15 and 30 mg/kgBW. PSK treatment of  60 mg/kgBW had lowest level at dosage, i.e. 25946.8 ρg/mL. It can be concluded that giving variety of dosages of polysaccharide krestin did not affect to testicular protein profiles but giving effect to testosterone levels of Mus musculus.

Abstract

  Staphylococcus aureus and Escherichia coli are among the most common species of gram-positive and gram-negative bacteria, which cause vaginitis, in infertile women. The Calamus rhizome (Acorus calamus L.) is an Indonesian plant that has antibacterial properties that can be used to treat vaginitis and increase fertility. The aim of this study was to determine the phytochemical and antibacterial activity of the calamus rhizoma in polar, semi-polar and non-polar solvents in the growth of S. aureus and E. coli. The antibacterial activity test was in the form of inhibitory test using the Kirby-Bauer, Minimum Inhibi-tion Concentration (MIC) and Minimum Bactericidal Concentration (MBC) by microdilution method with multilevel dilution (concentra-tions 50; 25; 12.5; 6.25; 3.13; 1.56; 0.78; and 0.39%). The screening results showed that ethanol and n-hexane extract contained alkaloids and triterpenoids, while chloroform extract was only triterpenoid. Chloroform extract produced the largest inhibition zone diameter of S. aureus and E. coli (7.26 and 3.28 mm), followed by ethanol extract (5.90 and 3.07 mm) and n-hexane extract (5.33 and 2.95 mm). The concentrations of 0.39 and 0.78% were the values of MIC and MBC for all three extracts, indicating that the extract of the calamus rhizome with several solvents in this study had the same antibacterial activity.

Abstract

The objective of the study was to determine the effect of cadmium on sperm quality and fertilization of C. carpio L. Sperm and eggs were collected by abdomen striping from the mature testis and ovary of C. carpio L. This study used one control and four treatment groups of variation on the cadmium concentration (0, 50, 100, 150, and 200 ppm) with eight replications. Sperm motility (mass motility, mass motility duration, and individual motility duration) and viability were measured after three to four seconds of incubation in the water. The percentage of fertility success was calculated by observing embryo development after the eggs were mixed with sperm and incubated in the water for 72 hours. The success of the fertilization process was indicated by a color change of the egg that darkens after successful fertilization, and white-milk if failed. The data were analyzed using analysis of variance (α = 0.05). The results of this study indicate that exposure of 50 ppm cadmium and control group shown success in term of sperm quality (motility and viability) and fertilization, but at 100 ppm or more decreased the sperm quality and fertilization rate. It can be concluded that cadmium exposure decreases sperm quality and fertility at 100 ppm or higher concentrations.

Abstract

This study aimed to determine the effect of arak bali on the quality of spermatozoa include morphology, motility, viability, membrane integrity of spermatozoa rat (Rattus norvegicus). The study was conducted in two phases: the first phase of the deployment of questionnaires conducted in five districts in Bali to determine the type and frequency of arak bali consumption and phase II made arak bali giving treatment in experimental animals. This study used 24 male rats (170-200 grams), divided into four groups: one control and three treatments (by arak bali containing 40% alcohol as much as 0.1 and 0.5 mL and 0.1 mL much alcohol synthesis, for 45 days. the results showed that of the five districts in Bali, most people consume arak bali commercial and most of the frequency of consumption of the week more than one bottle (350 mL). the provision of arak bali in experimental animals, degrade the quality (morphology, motility, viability, membrane integrity), the greater the volume given declining spermatozoa quality.

Abstract

This study aimed to determine the effects of arak bali on the fragmentation of spermatozoa and testosterone in rats (Rattus norvegicus). This study used 24 rats (170-200 g), divided into four groups: one control and three treatments (receiving arak bali containing 40% alcohol as much as 0.1 and 0.5 mL and synthetic alcohol (40%) as much as 0.1 mL, for 45 days). The observation of DNA fragmentation was done using acridine orange staining and the measurement of testosterone level used ELISA method. The results showed that the provision of arak bali in experimental animals increased the occurrence of spermatozoa DNA fragmentation. The higher the volume of arak bali given, the higher the fragmentation of spermatozoa DNA. The administration of arak bali also decreased testosterone level. The higher the volume of arak bali given, the lower the rats' testosterone level.

Abstract

This study aimed to determine the effects of red fruit (Pandanus conoideus Lam) oil on MDA levels and spermatozoa quality in mice (Mus musculus) exposed to MSG. The quality includes motility, viability, concentration, and morphology of spermatozoa. This experimental study used randomized post-test only control group design. The subjects of this study were 25 mice (Mus musculus), divided into 5 groups (5 mice per group). K- group received distilled water for 35 days. K+ group received 4 mg/g BW MSG for 21 days. P1, P2, and P3 treatment groups received 4 mg/g BW MSG for 21 days and 0.02; 0.04; 0.08 ml/g BW red fruit oil, respectively, from day 22 to 35. The results showed that mean spermatozoa morphology in K-, K+, P1, P2, P3 groups were as follows: 0.86; 0.56; 0.67; 0.61; and 0.87 (%). The spermatozoa concentrations were sequentially as follows: 21; 10; 15; 32,8,19 (107 cells/ml). The spermatozoa's vitalities were as follows: 0,64; 0,14; 0,24; P2: 0.36; 0.68 (%). MDA levels were respectively: 0.29; 0.60; 0.35; 0.23; and 0.19 (nm). As a conclusion, testicular MDA levels in mice exposed to MSG and given with red fruit oil were lower than those in mice exposed to MSG without receiving red fruit oil. The quality of spermatozoa in mice exposed to MSG and receiving red fruit oil was higher than that of mice exposed to MSG without being given with red fruit oil.

Abstract

This study aimed to analyze the levels of metallothionein and damage to hepatocytes and gills of Barbonymus gonionotus in the Brantas River (upstream and downstream), and the correlation levels of metallothionein in the gills and liver of Barbonymus gonionotus with different sampling time variation (March, June, and September 2016). Fishes were caught using trawl in two stations (Karangkates Reservoir and the Kali Jagir river). Fish gills and livers were taken for histopathological analysis and levels of metallothionein were measured by ELISA method. Gills histopathological analysis showed that the highest damage in Karangkates reservoir dan Kali Jagir river is hyperplasia. Liver histopathological analysis in Karangkates reservoir showed normal hepatocytes meanwhile necrosis is highest damage found in Kali Jagir river. Metallothionein levels of fish in Karangkates reservoir for March, June, and September were 0.18 ng/mL; 0,18 ng/mL; and 0.2 ng/mL (gills fish); 0,18 ng/mL; 0,34 ng/mL; and 0,21 ng/mL (liver fish) respectively. Metallothionein levels of fish in Kali Jagir river for March, June, and September were 0,26 ng/mL; 0,18 ng/mL; and 0,18 ng/mL (gills fish); 0,2 ng/mL; 0,45 ng/mL; 0,19 ng/mL (liver fish) respectively. We conclude that the damage to the gills and liver of Barbonymus gonionotus was mostly found in Kali Jagir River and the highest metallothionein level of Barbonymus gonionotus also presented in Kali Jagir River.

Abstract

This research aimed to determine the water quality and fish diversity in Brantas river. Three station for sampling locations were in the upstream (one station on Karangkates reservoir) and downstream (two stations on the Surabaya and Jagir river). Water quality parameters were BOD, COD, DO, pH, temperature, and the levels of heavy metals (Pb, Cr, Cu, and Cd). Fish specimens was identified using fish identification book. The results of this research indicated that there was a difference in the water quality of upstream and downstream of the Brantas river. On the upstream, the source of pollution was mostly from fish catching, agriculture, and industrial activities. While on the downstream of the river it mostly dominated by industrial waste and domestic waste. The water quality in both stations had far exceeded the quality standards that was included in the polluted category. The concluded that Brantas river has been contaminated by waste (heavy metals), exceeded the water quality standard. The diversity of fish in the downstream (Surabaya river) has the highest diversity values, followed Jagir river and in the upstream (Karangkates reservoir) has the lowest value of diversity. Brantas river water was included in the category of moderate diversity.

Abstract

Research has done to investigate the effect of pumpkin seed extract on morphology and motility of the mices after given of 2-ME. Thirty male mices, 56–63 days with 23–27 g weight were used as experimental animals into 5 groups (2 groups is control and 3 groups is treatment), 6 mices each. The fi rst group is K0 which given 0.1 mL aquades by gavage for 40 days each day, and then K1 were given 0.05 mL 2-ME 200 mg/kg by intraperitoneally everyday within 5 days, and continued on the giving of aquadest. The next groups are treated groups which given 2-ME, in the fi rst 5 days, then continued to the givings of pumpkin seed extract by gavage. Cucurbita moschata seed extract were given in three doses: 0.5; 1.0; and 2.0 g/kg BB for 35 days. Sperm collected from cauda epididymis. Parameter of sperm quality are morphology and motility. The result shown that Cucurbita moschata seed extract doses 0.5–1.0 g/kg BB showed no significant effect and then doses 2.0 g/kg BB increased recovery process morphology and motility with signifi cant differences p < 0.05. The conclutions were Cucurbita moschata seed extract increased recovery process morphology and motility of the mices.
